The known, unknown, and unknown unknowns of cell-cell communication in planarian regeneration.

Planarians represent the most primitive bilateral triploblastic animals. Most planarian species exhibit mechanisms for whole-body regeneration, exemplified by the regeneration of their cephalic ganglion after complete excision. Given their robust whole-body regeneration capacity, planarians have been model organisms in regenerative research for more than 240 years. Advancements in research tools and techniques have progressively elucidated the mechanisms underlying planarian regeneration. Accurate cell-cell communication is recognized as a fundamental requirement for regeneration. In recent de

Differential accumulation of PAHs within planarian cephalic and posterior body parts.

It has been observed that freshwater planarians can accumulate cadmium in their head portions, with neurotoxicity and head loss accompanying this phenomenon. Since planarians exhibit head loss and symptoms of neurotoxicity in response to PAHs, we investigated the differential accumulation of pyrene and B[a]P in the body and head portions of Girardia tigrina, a freshwater planarian. It is evidenced that planarian head fragments present higher amounts of pyrene- and B[a]P-equivalents than body fragments, indicating a differential distribution of these compounds within planarian tissues.

Production of asexual and sexual offspring in the triploid sexual planarian Dugesia ryukyuensis.

Certain freshwater planarians reproduce asexually as well as sexually, and their chromosomal ploidies include polyploidy, aneuploidy and mixoploidy. Previously, we successfully performed an experiment in which a clonal population produced by asexual reproduction of the Dugesia ryukyuensis (OH strain) switched to the sexual mode of reproduction. Worms of this strain are triploid with a pericentric inversion on Chromosome 4. The worms were switched to sexual reproduction after being fed with sexually mature Bdellocephala brunnea, which is a sexually reproducing species. The resulting sexualized

Production of diploid and triploid offspring by inbreeding of the triploid planarian Dugesia ryukyuensis.

Triploidy has generally been considered to be an evolutionary dead end due to problems of chromosomal pairing and segregation during meiosis. Thus, the formation of tetraploids and diploids from triploid types is a rare phenomenon. In the present study, we demonstrated that inbreeding of the triploid planarian Dugesia ryukyuensis resulted in both diploid and triploid offspring in nature. In the triploids of D. ryukyuensis, chiasmata between homologous chromosomes were observed in both female and male germ lines. This result suggests that both diploid and triploid offspring of this species are

Psilocybin induces stereotyped movements and reduces defensive responding in planarians through 5-hydroxytryptamine mechanisms.

Psilocybin is a serotonergic 5-HT 2A R agonist that causes psychedelic and anxiolytic effects in human users. To delineate conservation of psilocybin pharmacology, we investigated behavioral effects of psilocybin in planarians ( Dugesia dorotocephala ), the simplest living animal with cephalization that also has a well defined serotonin (5-hydroxytryptamine [5-HT]) system. We quantified stereotyped movements (e.g. head bops, twists, scrunches, and C-shapes) and defensive responding (negative phototaxis) and probed a 5-HT 2A R mechanism for psilocybin using a selective 5-HT 2A R antagonist (vol

Xylazine, veterinary anesthetic and recreational drug adulterant, reduces cephalic regeneration and alters defensive responding in planarians.

Xylazine is a veterinary anesthetic and α2-adrenoceptor agonist found in illicit drug supplies. Injection of opioids adulterated with xylazine is correlated with necrotic tissue injury and a withdrawal syndrome including anxiety. Planarians are flatworms with a simple nervous system and no circulatory system that exhibit several characteristics, including regenerative capacity and negative phototaxis (light avoidance). Because regeneration reflects tissue repair and negative phototaxis constitutes defensive behavior, planarians provide a simple in vivo system to explore possible effects of xyl

Behavioral and redox effects of lithium and curcumin in a planarian model of sublethal stress.

Freshwater ecosystems are increasingly contaminated by neuroactive compounds, including lithium (Li) from battery waste and curcumin (CUR) from consumer products, but their sublethal effects on aquatic invertebrates are poorly characterized. Linking behavioral alterations to biochemical responses under sublethal exposure remains a challenge. We used Girardia tigrina to assess behavioral and redox responses to mechanical, photic, and predator odor stress, followed by Li or CUR treatment. Mechanical and predator-odor stress increased time in the illuminated compartment by 40-60% (p < 0.0001), wh

Unraveling the role of Djstat5 in planarian regeneration by modulating cell proliferation and apoptosis.

Signal transducer and activator of transcription (STAT) proteins function as key mediators of cytokine signaling and are critically involved in stem cell regulation, tissue regeneration, and immune responses in vertebrates. However, their roles in invertebrate regeneration, particularly in adult pluripotent stem cell systems like those of planarians, remain largely unexplored. In this study, we investigated the function of Djstat5, a STAT5 homolog in the planarian Dugesia japonica, and demonstrated that it is dynamically upregulated during the early regenerative phases. RNA interference (RNAi)

A niche-derived non-ribosomal peptide triggers planarian sexual development.

Germ cells are regulated by local microenvironments (niches), which secrete instructive cues. Conserved developmental signaling molecules act as niche-derived regulatory factors, yet other types of niche signals remain to be identified. Single-cell RNA-sequencing of sexual planarians revealed niche cells expressing a non-ribosomal peptide synthetase (nrps). Inhibiting nrps led to loss of female reproductive organs and testis hyperplasia. Mass spectrometry detected the dipeptide β-alanyl-tryptamine (BATT), which is associated with reproductive system development and requires nrps and a monoamin

Inhibition of ATM kinase rescues planarian regeneration after lethal radiation.

As stem cells divide, they acquire mutations that can be passed on to daughter cells. To mitigate potentially deleterious outcomes, cells activate the DNA damage response (DDR) network, which governs several cellular outcomes following DNA damage, including repairing DNA or undergoing apoptosis. At the helm of the DDR are three PI3-like kinases including Ataxia-Telangiectasia Mutated (ATM). We report here that knockdown of ATM in planarian flatworms enables stem cells to withstand lethal doses of radiation which would otherwise induce cell death. In this context, stem cells circumvent apoptosi

Morphology of parenchymally implanted foreign bodies indicates copulatory wounding in a planarian.

Traumatic mating, that is, copulation that involves wounding the partner's body, is a widespread phenomenon that is particularly prevalent in hermaphroditic animals. Traumatic mating is generally a collateral side effect of diverse strategies (from physical anchorage to injection of substances to manipulate the partner), but the trauma could also be adaptive by itself if it delays remating by the injured partner. In the Tricladida (the clade of planarian flatworms), reciprocal sperm transfer is often assumed to occur by means of a 'regular' nontraumatic copulation, that is, insertion of the pe

Characterization of tyramine beta-hydroxylase in planarian Dugesia japonica: cloning and expression.

The planarian Dugesia japonica has a relatively well-organized central nervous system (CNS) consisting of a brain and ventral nerve cords (VNCs), and can completely regenerate it CNS utilizing pluripotent stem cells present in the mesenchymal space. This remarkable capacity has begun to be exploited for research on neural regeneration. Recently, several kinds of molecular markers for labeling of neural subtypes have been reported in planarians. These molecular markers are useful for visualizing the distinct neural populations in planarians. In this study, we isolated a cDNA encoding tyramine b

Combinatorial mechanisms specify cellular location and neurotransmitter identity during planarian neurogenesis.

Regenerative neurogenesis can drive replacement of neurons in the right types and locations to faithfully restore form and function after injury. The genetic mechanisms underlying successful regenerative neurogenesis, including mechanisms that produce neuronal diversity and spatial organization, remain poorly understood. Planarians are flatworms with extraordinary capacity for brain regeneration made possible by pluripotent stem cells throughout the body that undergo neurogenesis to form a complex nervous system anew after injury. Here, we focus on the dopaminergic neuron identity and report t

DjFNDC3A is associated with DjSmad4 to regulate cranial nerve regeneration in planarian (Dugesia japonica).

FNDC3A protein, widely expressed in various tissues, is involved in a variety of biological processes, such as, tail bud development and caudal fin regeneration in zebrafish. However, functional studies of FNDC3A in planarians have not been reported. In this study, we examined the spatiotemporal expression pattern of DjFNDC3A and investigated its functions in planarians regeneration. Knockdown of DjFNDC3A resulted in severe defects, including poorly defined brain lobes, complete loss of brain lateral branches and reduction of multiple neuronal populations. Silencing DjFNDC3A did not affect the

Melastatin subfamily Transient Receptor Potential channels support spermatogenesis in planarian flatworms.

The Transient Receptor Potential superfamily of proteins (TRPs) form cation channels that are abundant in animal sensory systems. Amongst TRPs, the Melastatin-related subfamily (TRPMs) is composed of members that respond to temperature, pH, sex hormones, and various other stimuli. Some TRPMs exhibit enriched expression in gonads of vertebrate and invertebrate species, but their contributions to germline development remain to be determined. We identified twenty-one potential TRPMs in the planarian flatworm Schmidtea mediterranea and analyzed their anatomical distribution of expression by whole-

Morphogenic Effect of Exogenous Glucocorticoid Hormones in the Girardia tigrina Planarian (Turbellaria, Tricladida).

We have studied the effect of two glucocorticoid hormones: hydrocortisone and its synthetic analogue methylprednisolone on the regeneration activity of head and tail blastema of the Girardia tigrina planarian. The regeneration activity was studied in head and tail blastema formed after resection by means of lifetime computer morphometry and immunohistochemical labeling of neoblasts. The search for orthologous proteins-glucocorticoid receptors (hydrocortisone) was performed using the SmedGD database of the Schmidtea mediterranea planarian. The results indicate that both hormones influence the r

Toxic effects and mechanistic insights of cadmium telluride quantum dots on the homeostasis and regeneration in planarians.

The widespread application of quantum dots (QDs) in recent years has raised concerns about potential environmental and human health risks. Although the toxicity of cadmium telluride quantum dots (CdTe QDs) has been partially studied, their effects on stem cells, tissue regeneration, neurodevelopment, and neurobehavioral toxicity remain unclear. This study aimed to investigate the combined toxic effects and mechanisms of CdTe QDs on planarians at the individual, tissue, cellular, and molecular levels. The results showed that exposure to CdTe QDs led to tissue damage, abnormal motor behavior, de

Measuring functional brain recovery in regenerating planarians by assessing the behavioral response to the cholinergic compound cytisine.

Planarians are traditional model invertebrates in regeneration and developmental biology research that also display a variety of quantifiable behaviors useful to screen for pharmacologically active compounds. One such behavior is the expression of seizure-like movements (pSLMs) induced by a variety of substances. Previous work from our laboratory showed that cocaine, but not nicotine, induced pSLMs in intact but not decapitated planarians. Interestingly, as decapitated planarians regenerated their heads, they gradually recovered their sensitivity to cocaine. These results suggested a method to

[Regeneration of photoreceptor organs in freshwater planarians at different levels of accumulation of natural methylmercury compounds].

The effects of natural methylmercury compounds on regeneration of photoreceptor organs were studied in three freshwater planarians: Polycelis tenuis, Dugesia lugubris, and D. tigrina. Accumulation of methyl mercury in the planarian body suppressed regeneration of P. tenuis with numerous photoreceptor organs to a greater extent than in two other planarians that have only two eyes. High methyl mercury concentrations inhibited the restoration of photoreceptor organs in asexual and sexual D. tigrina races.
