Long noncoding RNAs (lncRNAs) are a class of transcripts longer than 200 nucleotides with little or no protein-coding capacity. They represent a substantial fraction of eukaryotic genomes and function as molecular architects that coordinate diverse biological processes and physiological plasticity, including multilayered plant defence systems, through versatile regulatory interactions at multiple levels of gene regulation. Recent advances in high-throughput RNA sequencing technologies have greatly expanded the identification of plant lncRNAs, revealing their diverse roles as scaffolds, decoys,

Global food production faces persistent threats from environmental challenges and pathogenic attacks, leading to significant yield losses. Conventional strategies to combat pathogens, such as fungicides and disease-resistant breeding, are limited by environmental contamination and emergence of pathogen resistance. Herein, we engineered sunlight-sensitive and biodegradable carbon dots (CDs) capable of generating reactive oxygen species (ROS), offering a novel and sustainable approach for plant protection. Our study demonstrates that CDs function as dual-purpose materials: priming plant immune r

Climate change has profound impacts on plant growth, productivity, and immunity. Photosynthesis, a key biological process that sequesters atmospheric CO2, plays a crucial role in mitigating the effects of climate change. However, photorespiration, a process that consumes O2 instead of CO2 and is metabolically linked with photosynthesis, is often viewed as a wasteful process that reduces photosynthetic efficiency by nearly 48 % in C3 plants. This reduction has a significant impact on crop yield, given the direct contribution of photosynthesis to biomass accumulation. Over the years, numerous ef

Plant chemical defenses are known to influence herbivore performance, yet their effects on herbivore immunity are poorly understood. In this study, we manipulated both the mean and variance of the concentration of a plant toxin in an artificial diet to assess the effects on herbivore immune function and survival. Our results revealed a biphasic immune response across increasing mean toxin concentrations, with exposure to low toxin concentrations enhancing immune function, creating a 'window of enhanced immunity', and exposure to higher concentrations suppressing it relative to toxin-free diets

When organisms encounter pathogens, they rapidly activate complex defense programs to ensure survival. While these immune responses are vital, they often also incur trade-offs, such as reduced growth and development and must therefore be tightly controlled. In this study, we reveal that the steroid hormones brassinosteroids (BRs) contribute to this control in Arabidopsis thaliana by repressing immunity-related genes. We provide evidence that the BR-regulated basic helix-loop-helix (bHLH) transcription factor CESTA (CES), along with its homologs BR ENHANCED EXPRESSION (BEE)1-3, mediate DNA meth

DREB1A, a pivotal transcription factor, has long been known to regulate plant abiotic stress tolerance. However, its role in plant biotic stress tolerance and the underlying mechanisms have remained a mystery. Our research reveals that the maize ZmDREB1A gene is up-regulated in maize seedlings when the plants are infected by Rhizoctonia solani (R. solani). The maize ZmDREB1A knock-out mutant exhibits increased disease resistance against the pathogen R. solani. Further investigation showed that ZmDREB1A regulates salicylic acid (SA) metabolism by inhibiting ZmSARD1 gene and activating ZmSAGT ge

Plants have evolved sophisticated mechanisms to regulate gene expression to activate immune responses against pathogen infections. However, how the translation system contributes to plant immunity is largely unknown. The evolutionarily conserved thiolation modification of transfer RNA (tRNA) ensures efficient decoding during translation. Here, we show that tRNA thiolation is required for plant immunity in Arabidopsis. We identify a cgb mutant that is hyper-susceptible to the pathogen Pseudomonas syringae. CGB encodes ROL5, a homolog of yeast NCS6 required for tRNA thiolation. ROL5 physically i

In plants, the reactive oxygen species (ROS) formed during normal conditions are essential in regulating several processes, like stomatal physiology, pathogen immunity and developmental signaling. However, biotic and abiotic stresses can cause ROS over-accumulation leading to oxidative stress. Therefore, a suitable equilibrium is vital for redox homeostasis in plants, and there have been major advances in this research arena. Salicylic acid (SA) is known as a chief regulator of ROS; however, the underlying mechanisms remain largely unexplored. SA plays an important role in establishing the hyp

Arabidopsis SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g) are two master transcription factors that regulate many defense-related genes in plant immunity. They are required for immunity downstream of the receptor-like protein SUPPRESSOR OF NPR1-1, CONSTITUTIVE 2 (SNC2). Constitutive defense responses in the gain-of-function autoimmune snc2-1D mutant are modestly affected in either sard1 or cbp60g single mutants but completely suppressed in the sard1 cbp60g double mutant. Here we report that CBP60b, another member of the CBP60 family, also function

Pathogen-associated molecular patterns (PAMPs) activate innate immunity in both animals and plants. Although calcium has long been recognized as an essential signal for PAMP-triggered immunity in plants, the mechanism of PAMP-induced calcium signalling remains unknown1,2. Here we report that calcium nutrient status is critical for calcium-dependent PAMP-triggered immunity in plants. When calcium supply is sufficient, two genes that encode cyclic nucleotide-gated channel (CNGC) proteins, CNGC2 and CNGC4, are essential for PAMP-induced calcium signalling in Arabidopsis3-7. The circadian clock in plants synchronizes biological processes that display cyclic 24-h oscillation based on metabolic and physiological reactions. This clock is a precise timekeeping system, that helps anticipate diurnal changes; e.g., expression levels of clock-related genes move in synchrony with changes in pathogen infection and help prepare appropriate defense responses in advance. Salicylic acid (SA) is a plant hormone and immune signal involved in systemic acquired resistance (SAR)-mediated defense responses. SA signaling induces cellular redox changes, and degradation and rhythmic nuc

The impacts of rising atmospheric CO2 concentrations on plant disease have received increasing attention, but with little consensus emerging on the direct mechanisms by which CO2 shapes plant immunity. Furthermore, the impact of sub-ambient CO2 concentrations, which plants have experienced repeatedly over the past 800 000 yr, has been largely overlooked. A combination of gene expression analysis, phenotypic characterisation of mutants and mass spectrometry-based metabolic profiling was used to determine development-independent effects of sub-ambient CO2 (saCO2 ) and elevated CO2 (eCO2 ) on Ara

The ubiquitin-proteasome system (UPS) has been shown to play vital roles in diverse plant developmental and stress responses. The UPS post-translationally modifies cellular proteins with the small molecule ubiquitin, resulting in their regulated degradation by the proteasome. Of particular importance is the role of the UPS in regulating hormone-responsive gene expression profiles, including those triggered by the immune hormone salicylic acid (SA). SA utilizes components of the UPS pathway to reprogram the transcriptome for establishment of local and systemic immunity. Emerging evidence has sh

Ferredoxins, the major distributors for electrons to various acceptor systems in plastids, contribute to redox regulation and antioxidant defence in plants. However, their function in plant immunity is not fully understood. In this study, we show that the expression of the major leaf ferredoxin gene Fd2 is suppressed by Pseudomonas syringae pv. tomato (Pst) DC3000 infection, and that knockout of Fd2 (Fd2-KO) in Arabidopsis increases the plant's susceptibility to both Pst DC3000 and Golovinomyces cichoracearum. On Pst DC3000 infection, the Fd2-KO mutant accumulates increased levels of jasmonic

Ethylene is a key signal in the regulation of plant defense responses. It is required for the expression and function of GDSL LIPASE1 (GLIP1) in Arabidopsis (Arabidopsis thaliana), which plays an important role in plant immunity. Here, we explore molecular mechanisms underlying the relationship between GLIP1 and ethylene signaling by an epistatic analysis of ethylene response mutants and GLIP1-overexpressing (35S:GLIP1) plants. We show that GLIP1 expression is regulated by ethylene signaling components and, further, that GLIP1 expression or application of petiole exudates from 35S:GLIP1 plants

The nuclear pore complex (NPC) was initially viewed as a passive gateway for molecular transport, but it is now recognized as an active and highly selective signaling hub that orchestrates nucleocytoplasmic communication. In plants, dissection of the NPC's intricate architecture has revealed its critical role in innate immunity: specific nucleoporins selectively gate the nuclear entry of key immune regulators, while plant‑specific components dynamically control pore permeability or form phase‑separated condensates that assemble specialized transcription factories. Strikingly, the NPC recruits

Systemic acquired resistance (SAR) is a highly desirable form of resistance that protects against a broad-spectrum of related or unrelated pathogens. SAR involves the generation of multiple signals at the site of primary infection, which arms distal portions against subsequent secondary infections. The last decade has witnessed considerable progress, and a number of chemical signals contributing to SAR have been isolated and characterized. The diverse chemical nature of these chemicals had led to the growing belief that SAR might involve interplay of multiple diverse and independent signals. H

Selective autophagy has emerged as a critical component of plant immunity, yet its role as a dedicated antimicrobial mechanism, xenophagy, remains conceptually underdeveloped. In plants, xenophagy extends beyond bulk degradation to function as a highly regulated, cargo-specific pathway that targets invading pathogens and their effector molecules for autophagic clearance. Recent evidence demonstrates that selective autophagy receptors, particularly NBR1, integrate ubiquitin-mediated recognition with ATG8-dependent sequestration, enabling the elimination of bacterial, fungal, and viral component

The 1988 papers by Mauch, Mauch-Mani, Hadwiger and Boller on pea antifungal hydrolases captured a simple but powerful idea: plants can attack pathogens by directly targeting the structural polymers that hold their cell walls together. At that time, chitinases and β-1,3-glucanases were known to accumulate after infection, wounding and ethylene treatment, but whether these proteins actually restricted fungal growth remained unclear. By resolving distinct hydrolase isoforms and showing that inducible chitinase and β-1,3-glucanase act synergistically to inhibit fungal growth and lyse hyphal tips,

Plants possess a multilayered immune system that detects and responds to pathogen invasion through pattern-triggered immunity (PTI) and effector-triggered immunity (ETI). We recently developed a high-throughput screening platform using transgenic Arabidopsis thaliana expressing the β-glucuronidase (GUS) reporter gene under the control of the promoter of the PTI marker gene FRK1 (pFRK1::GUS) to identify chemical inducers of plant immunity. Among the ∼10,000 screened compounds, we identified 10 candidate compounds with the potential to activate FRK1 expression. In this study, we focused on one o
