Arbuscular Mycorrhizal Fungi Promote Nodulation and N2 Fixation in Soybean by Specific Root Exudates.

Legume plants commonly associate with both arbuscular mycorrhizal (AM) fungi and rhizobia and thus enhance the acquisition of phosphorus (P) and nitrogen (N) nutrition. Inoculation with AM fungi can promote nodulation and N2 fixation of legume plants; however, the underlying mechanisms remain poorly understood. Here, root exudates collected from AM-colonised soybean plants showed greater accumulation of the specific flavonoids (daidzein and genistein) and phenolic acids (benzoic acid and p-Hydroxybenzoic acid), and significantly promoted nodulation. Furthermore, the exudates from AM-colonised

Leguminous plants nodulated by selected strains of Cupriavidus necator grow in heavy metal contaminated soils amended with calcium silicate.

Increasing concern regarding mining area environmental contamination with heavy metals has resulted in an emphasis of current research on phytoremediation. The aim of the present study was to assess the efficiency of symbiotic Cupriavidus necator strains on different leguminous plants in soil contaminated with heavy metals following the application of inorganic materials. The application of limestone and calcium silicate induced a significant increase in soil pH, with reductions in zinc and cadmium availability of 99 and 94 %, respectively. In addition, improved nodulation of Mimosa caesalpini

Silicon-mediated alleviation of mercury toxicity requires coordinated regulation of antioxidant defense, metal homeostasis, and nodule function in mung bean.

Mercury (Hg) contamination and accumulation in agricultural soil represent a major hazardous environmental concern, posing serious threats to living organisms, including plants. Silicon (Si) has been widely recognized to mitigate heavy metal (loid) toxicity; however, the underlying mechanism of Si-mediated mitigation of Hg-stress in mung bean remains unclear. In this study, we addressed this research gap by thoroughly examining the potential effects of Si supplementation on Hg-stressed mung bean plants, with particular emphasis on investigating the possible effects of Si on plant biomass, nodu

SCARN a Novel Class of SCAR Protein That Is Required for Root-Hair Infection during Legume Nodulation.

Rhizobial infection of legume root hairs requires a rearrangement of the actin cytoskeleton to enable the establishment of plant-made infection structures called infection threads. In the SCAR/WAVE (Suppressor of cAMP receptor defect/WASP family verpolin homologous protein) actin regulatory complex, the conserved N-terminal domains of SCAR proteins interact with other components of the SCAR/WAVE complex. The conserved C-terminal domains of SCAR proteins bind to and activate the actin-related protein 2/3 (ARP2/3) complex, which can bind to actin filaments catalyzing new actin filament formation

Spontaneous symbiotic reprogramming of plant roots triggered by receptor-like kinases.

Symbiosis Receptor-like Kinase (SYMRK) is indispensable for the development of phosphate-acquiring arbuscular mycorrhiza (AM) as well as nitrogen-fixing root nodule symbiosis, but the mechanisms that discriminate between the two distinct symbiotic developmental fates have been enigmatic. In this study, we show that upon ectopic expression, the receptor-like kinase genes Nod Factor Receptor 1 (NFR1), NFR5, and SYMRK initiate spontaneous nodule organogenesis and nodulation-related gene expression in the absence of rhizobia. Furthermore, overexpressed NFR1 or NFR5 associated with endogenous SYMRK

The geographic scale of population-level variation in growth and nodulation differs for two species of prairie clover.

PREMISE: Prairies are among the most threatened biomes due to changing patterns of climate and land use, yet information on genetic variation in key species that would inform conservation is often limited. We assessed evidence for the geographic scale of population-level variation in growth of two species of prairie clover and of their symbiotic associations with nitrogen-fixing bacteria. METHODS: Seed representing two species, Dalea candida and D. purpurea, from the same five source populations were planted into an experimental site in Minnesota. We assessed variation within and among source

Inoculation of new rhizobial isolates improve nutrient uptake and growth of bean (Phaseolus vulgaris) and arugula (Eruca sativa).

BACKGROUND: In the current agricultural model, the massive use of chemical fertilizer causes environmental and economic losses. Inoculation of plant-growth-promoting (PGP) nitrogen-fixing bacteria is an alternative to the use of nitrogen, phosphorus and potassium fertilizers. In this study, rhizobia strains isolated from common bean (Phaseolus vulgaris) root nodules were evaluated in an effort to identify an efficient nitrogen-fixing rhizobia strain able to improve bean germination and growth. RESULTS: Common bean plants were collected from seven sites in southern Brazil, and 210 native rhizo

Drought-tolerant rhizobacterial consortia with diverse plant growth promoting traits enhance wheat and faba bean growth under water and low-P availability promising multi-traits.

BACKGROUND: Staple crops like wheat and faba bean are increasingly subjected to multiple and simultaneous stresses, resulting in substantial yield reduction. Although vast available knowledge about the role of root-rhizosphere microbes in enhancing crop tolerance to single stress, few is known about the potential of bacterial consortia rationally assembled from strains with defined and complementary ecological functions to improve crop tolerance to combined drought and phosphorus (P) deficiency. This study evaluated wheat (Triticum durum) or faba bean (Vicia faba) morpho-physiological response

Post translational modifications of Trifolitoxin: a blue fluorescent peptide antibiotic.

Trifolitoxin (TFX, C41H63N15O15S) is a selective, ribosomally-synthesized, post-translationally modified, peptide antibiotic, produced by Rhizobium leguminosarum bv. trifolii T24. TFX specifically inhibits α-proteobacteria, including the plant symbiont Rhizobium spp., the plant pathogen Agrobacterium spp. and the animal pathogen Brucella abortus. TFX-producing strains prevent legume root nodulation by TFX-sensitive rhizobia. TFX has been isolated as a pair of geometric isomers, TFX1 and TFX2, which are derived from the biologically inactive primary amino acid sequence: Asp-Ile-Gly-Gly-Ser-Arg-

Reconnoitering the capabilities of nodule endophytic Pantoea dispersa for improved nodulation and grain yield of chickpea (Cicer arietinum L.).

Microorganisms belonging to root and soil provide a wide range of services and benefits to the plant by promoting plant growth and controlling phytopathogens. This study aimed to isolate endophytic bacteria from the root nodules of chickpea (Cicer arietinum L.) and determine their potential in improving plant growth. A total of nineteen different bacterial morphotypes were isolated from root nodules of chickpea and characterized in vitro for plant growth promotion abilities. All bacterial isolates were able to produce indole acetic acid at varying levels, out of which MCA19 was screened as the

Diverse plant growth-promoting bacteria as microsymbionts in nodules of Leucaena leucocephala.

This study characterized 18 endophytic bacterial isolates in association with the root nodules of Leucaena leucocephala through phenotypic and genotypic analyses. The endophytes were associated with the plants and exhibited diverse plant growth-promoting (PGP) traits. Phosphate solubilization was observed in 39% of isolates at high levels and 33.3% at moderate levels. Siderophore production was prevalent, with 38.9% displaying high and 33.3% moderate production, aiding iron uptake. Indole-3-acetic acid (IAA) production varied (32.15 to 86.28 µg/ml) among the isolates. Notably, 94.4% of isolate

Development of a controlled-environment assay to induce iron deficiency chlorosis in soybean by adjusting calcium carbonates, pH, and nodulation.

BACKGROUND: Soybean iron deficiency chlorosis (IDC) is an important nutrient stress frequently found in high pH and/or soils high in calcium carbonates. To advance the understanding of IDC resistance in soybean, a rapid (21-day) controlled-environment assay was developed to investigate the effects of nodulation, pH, and calcium carbonate levels on soybean iron deficiency traits. This system was tested on four genotypes known to exhibit differences in iron efficiency, including two standard IDC check cultivars and a pair of near-isogenic lines exhibiting variation at an IDC resistance quantitat

Persulfidation of host NADPH oxidase RbohB by rhizobial 3-mercaptopyruvate sulfurtransferase maintains redox homeostasis and promotes symbiotic nodulation in soybean.

Reactive oxygen species play a crucial role in various stages of the legume-rhizobia symbiosis, from initial nodulation signaling to nodule senescence. However, how rhizobial redox-related proteins regulate symbiotic nodulation in legumes remains largely unknown. By combining transcriptomics, proteomics, and biochemical and molecular genetics, we investigated the role of the Sinorhizobium fredii Q8 enzyme 3-mercaptopyruvate sulfurtransferase (3MST). Although 3MST was not the primary enzyme responsible for hydrogen sulfide (H2S) production under our conditions, its absence significantly impaire

Plant-microbe-microbe interactions influence the faba bean nodule colonization by diverse endophytic bacteria.

Legume root nodules harbor rhizobia and other non-nodulating endophytes known as nodule-associated bacteria (NAB) whose role in the legume symbiosis is still unknown. We analysed the genetic diversity of 34 NAB isolates obtained from the root nodules of faba bean grown under various soil conditions in Egypt using 16S rRNA and concatenated sequences of three housekeeping genes. All isolates were identified as members of the family Enterobacteriaceae belonging to the genera Klebsiella, Enterobacter and Raoultella. We identified nine enterobacterial genospecies, most of which have not been previo

3D MicroCT Imaging of Medicago sativa Root Nodules.

The symbiotic relationship between the legume Medicago sativa and the soil bacteria Sinorhizobium meliloti results in the formation of nitrogen-fixing root nodules. Traditional destructive methods, including paraffin sectioning, vibratome sectioning, and cryosectioning, have been applied to visualize how bacteria occupy the nodule, making it extremely difficult to obtain reliable three-dimensional information. These approaches are often combined with fluorescent labeling or staining, which can introduce additional stress affecting plant growth and nodule formation. MicroCT has emerged as a rel

Bioformulations of Entrophospora lutea enriched with biostimulants for growth promotion and control of Rhizoctonia root rot in lupine.

Arbuscular mycorrhizal fungus (AMF) Entrophospora lutea was previously selected as a highly compatible isolate for lupine, and the main objective of the present study was to further improve its performance and practical applicability by developing two E. lutea formulations using peat and vermiculite carriers enriched with various biostimulants. Incorporating biostimulants into AMF formulations can enhance fungal performance and root symbiosis. The first formulation contained proline, ascorbic acid, and hemicellulose, whereas the second contained proline, humic acid, and mannitol. Both formulat

Reactive Sulfur Species Interact with Other Signal Molecules in Root Nodule Symbiosis in Lotus japonicus.

Reactive sulfur species (RSS) function as strong antioxidants and are involved in various biological responses in animals and bacteria. Few studies; however, have examined RSS in plants. In the present study, we clarified that RSS are involved in root nodule symbiosis in the model legume Lotus japonicus. Polysulfides, a type of RSS, were detected in the roots by using a sulfane sulfur-specific fluorescent probe, SSP4. Supplying the sulfane sulfur donor Na2S3 to the roots increased the amounts of both polysulfides and hydrogen sulfide (H2S) in the roots and simultaneously decreased the amounts
