Maize Inositol Polyphosphate 5-phosphatase Gene Family Analysis Reveals Zm5PTase25 as a Negative Regulator of Low Phosphate-Induced Leaf Senescence.

Phosphate (Pi) deficiency promotes premature leaf senescence in plants through complex molecular networks; however, the underlying mechanisms remain poorly understood. Inositol polyphosphate 5-phosphatases (5PTases) are key regulators of inositol phosphate metabolism, but the maize (Zea mays L.) 5PTase gene family and its roles in low Pi (LP)-induced leaf senescence have not been systematically investigated. Here, we identified 31 members of the Zm5PTase family in the maize genome. Phylogenetic and structural analyses revealed marked divergence among clades. Among these genes, Zm5PTase25 was p

The C2H2-type zinc finger protein ZOS202 regulates leaf senescence in rice by targeting senescence-associated transcription factors and ROS homoeostasis.

Leaf senescence is a tightly regulated developmental process orchestrated by multiple transcription factor (TF) families. Although C2H2-type zinc finger proteins are known to participate in various aspects of plant growth and abiotic stress responses, their specific role in regulating leaf senescence remains poorly understood. Here, we show that knockout mutation of the C2H2-type zinc finger TF gene ZOS202 (LOC_Os02g02424) resulted in delayed leaf senescence under both dark-induced and natural conditions, while overexpression lines exhibit accelerated leaf senescence, as indicated by reactive

Molecular characterization of a novel senescence-associated gene SPA15 induced during leaf senescence in sweet potato.

The structure and expression of a novel senescence-associated gene (SPA15) of sweet potato were characterized. The protein coding region of the gene consists of 13 exons encoding 420 amino acids. Apparent homologues of this sweet potato gene are found in a variety of dicot and monocot plants, but not in animals or microorganisms. Examination of the expression patterns of the SPA15 gene in sweet potato reveals that the transcripts of SPA15 are specifically induced in the senescing leaves, and the temporal profile of SPA15 protein accumulation is correlated with that of SPA15 transcripts. Role of reactive oxygen species and antioxidants in plant disease resistance.

Membrane damage caused by the non-specific fungal toxin fusaric acid was less on pretreated than on control leaves when tobacco leaves were pretreated with anti-senescence plant hormones, such as kinetin, benzyladenine or the anti-ozonant N-[2-(2-oxo-1-imidazolidinyl)]ethyl-N'-phenylurea. Similarly, the necrosis caused by mercuric chloride was reduced by the above anti-senescence agents. In addition, in in vitro tests, leaves from selected paraquat-tolerant tobacco plants were less sensitive to Alternaria alternata (Fr) Keissler infection than those of the control paraquat-sensitive tobacco le

Preservation of integrity and activity of Haberlea rhodopensis photosynthetic apparatus during prolonged light deprivation.

The effect of prolonged light deprivation on ultrastructure, pigment composition and functions of photosynthetic apparatus of the resurrection plant Haberlea rhodopensis Friv. (Gesneriaceae) was studied. For this purpose, intact plants were kept in darkness for up to 6 months. Haberlea rhodopensis demonstrated extraordinary ability to preserve its photosynthetic machinery intact despite complete absence of light. During the first 4 weeks of light deprivation, we observed preservation of pigment content, chloroplast ultrastructure and a decrease in the rate of CO(2) assimilation. The signs of d

Lyso-phosphatidylethanolamine primes the plant immune system and promotes basal resistance against hemibiotrophic pathogens.

BACKGROUND: Lyso-phosphatidylethanolamine (LPE) is a natural phospholipid that functions in the early stages of plant senescence. Plant innate immunity and early leaf senescence share molecular components. To reveal conserved mechanisms that link-up both processes, we tried to unravel to what extent LPE coordinates defense response and by what mode of action. RESULT: We found that LPE-treatment induces signaling and biosynthesis gene expression of the defensive hormone salicylic acid (SA). However, jasmonic acid and ethylene triggered gene induction levels are indistinguishable from the contr

Transcription factor NtWRKY75 accelerates dark-induced leaf senescence through orchestrating hydrogen sulfide homeostasis in Nicotiana tabacum.

As the last stage of leaf development, senescence is orchestrated by an intricate network of endogenous factors and external signals to ensure an efficient recycling of nutrients. Hydrogen sulfide (H2S) serves as an important gaseous signaling molecule in plants, mediating a myriad of physiological processes like leaf senescence. However, the molecular mechanisms underlying H2S accumulation and its regulation during leaf senescence in tobacco are still not fully elucidated. In this work, we demonstrate that NtWRKY75, a WRKY transcription factor in tobacco (Nicotiana tabacum), serves as a negat

Exogenous nitric oxide extends longevity in cut Lilium tigrinum flowers by orchestrating biochemical and molecular aspects.

Senescence represents a developmentally orchestrated and precisely regulated cascade of events, culminating in the abscission of plant organs and ultimately leading to the demise of the plant or its constituent parts. In this study, we observed that senescence in cut Lilium tigrinum flowers is induced by elevated ABA levels and the hyperactivation of lipoxygenase (LOX) activity. This cascade increased ROS concentrations, heightened oxidative damage, and disrupted cellular redox equilibrium. This was evidenced by elevated lipid peroxidation, attenuated antioxidant machinery, and reduced membran

Cellular senescence in skin-related research: Targeted signaling pathways and naturally occurring therapeutic agents.

Despite the growing interest by researchers into cellular senescence, a hallmark of cellular aging, its role in human skin remains equivocal. The skin is the largest and most accessible human organ, reacting to the external and internal environment. Hence, it is an organ of choice to investigate cellular senescence and to target root-cause aging processes using senolytic and senomorphic agents, including naturally occurring plant-based derivatives. This review presents different aspects of skin cellular senescence, from physiology to pathology and signaling pathways. Cellular senescence can ha

Transcriptome Analysis Reveals Root Endophyte Serendipita indica-Mediated Growth Promotion in Wheat Is Associated with Enhanced Photosynthesis.

Serendipita indica (S. indica), a root endophytic fungus of the Sebacinaceae family, promotes growth and increases biomass accumulation in a wide range of plant species. However, the mechanism underlying S. indica-mediated growth promotion in wheat (Triticum aestivum L.), particularly its effects on chlorophyll accumulation, remains poorly understood. In this study, colonization by S. indica significantly enhanced shoot growth, biomass accumulation, plant height, fresh weight, dry weight and chlorophyll content in wheat seedlings. Transcriptome analysis revealed extensive transcriptional repro

Phenological stage-dependent hierarchical responses mediate extreme drought impacts on carbon fluxes of a semiarid grassland.

Ecosystem carbon balance is influenced by plant phenological stage-dependent hierarchical responses. However, the molecular and physiological mechanisms that underlie extreme drought-induced alterations in these responses, as well as their cascading effects on carbon fluxes in semiarid grasslands, remain unresolved. Extreme drought was imposed during the green-up, peak growing season, and senescence phase in a semiarid grassland. We measured plant genes, photosynthetic physiology, phenological phase, and CO2 fluxes at molecular, individual, community, and ecosystem levels, respectively. The hi

PROTEIN PHOSPHATASE 2A-B'γ Controls Botrytis cinerea Resistance and Developmental Leaf Senescence.

Plants optimize their growth and survival through highly integrated regulatory networks that coordinate defensive measures and developmental transitions in response to environmental cues. Protein phosphatase 2A (PP2A) is a key signaling component that controls stress reactions and growth at different stages of plant development, and the PP2A regulatory subunit PP2A-B'γ is required for negative regulation of pathogenesis responses and for maintenance of cell homeostasis in short-day conditions. Here, we report molecular mechanisms by which PP2A-B'γ regulates Botrytis cinerea resistance and leaf

Probing the role of cell wall feruloylation during maize development by differential expression of an apoplast targeted fungal ferulic acid esterase.

While many aspects of the growth of maize are well understood, the role of cell wall feruloylation particularly during internode elongation has not been firmly established, but results so far indicate that it has significant implications for both biofuel feedstock conversion and for crop yield. The growth of the cell wall is achieved by synthesis, integration and cross-linking between wall polymers. As ferulate oxidative coupling of arabinoxylan side chains constitutes a significant type of cross-link in grass cell walls, it is expected to have a crucial role in plant growth. Making use of pla

Interactions between ScNAC23 and ScGAI regulate GA-mediated flowering and senescence in sugarcane.

Control of gene transcription is crucial to regulate plant growth and development events, such as flowering, leaf senescence, and seed germination. Here we identified a NAC transcription factor (ScNAC23) isolated from sugarcane (cv. ROC22). Analysis by qRT-PCR indicated that ScNAC23 expression was strongly induced in mature leaves and flowering varieties and was also responsive to exogenous treatment with the hormone gibberellin (GA). Ectopic expression of ScNAC23 in Arabidopsis accelerated bolting, flowering, and leaf senescence compared to wild type plants. Furthermore, Arabidopsis overexpre

Genome-wide identification of the UGT gene family in foxtail millet and preliminary functional analysis of SiUGT26 during leaf senescence.

Plant uridine diphosphate glycosyltransferases (UGTs) catalyze glycosylation, a pivotal biochemical modification essential for diverse metabolic and physiological processes. Foxtail millet (Setaria italica), a drought-tolerant C4 cereal crop, serves as an emerging model for studying abiotic stress adaptation. Here, we identified 173 UGT genes (SiUGTs) in the foxtail millet genome, unevenly distributed across nine chromosomes and classified into 17 phylogenetically distinct subfamilies. Collinearity analysis revealed nine segmental duplicated duplication pairs under purifying selection, highlig

Copper deficiency alters shoot architecture and reduces fertility of both gynoecium and androecium in Arabidopsis thaliana.

Copper deficiency reduces plant growth, male fertility, and seed set. The contribution of copper to female fertility and the underlying molecular aspects of copper deficiency-caused phenotypes are not well known. We show that among copper deficiency-caused defects in Arabidopsis thaliana were also the increased shoot branching, delayed flowering and senescence, and entirely abolished gynoecium fertility. The increased shoot branching of copper-deficient plants was rescued by the exogenous application of auxin or copper. The delayed flowering was associated with the decreased expression of the

Hydrogen Sulfide: A Gaseous Molecule in Postharvest Freshness.

Hydrogen sulfide (H2S), as a signaling molecule, is involved in the regulation of growth and development in plants. Recent studies have indicated that H2S also plays important roles in regulating postharvest senescence of horticultural products. The focus of this review is to summarize the synthesis of H2S in plants and its potential roles in alleviating the senescence of cut flowers, fruits, and vegetables during postharvest storage. During postharvest of horticultural products, H2S could scavenge reactive oxygen species via promoting the activity of antioxidant enzymes, thereby, sustaining t

WRKY55 transcription factor positively regulates leaf senescence and the defense response by modulating the transcription of genes implicated in the biosynthesis of reactive oxygen species and salicylic acid in Arabidopsis.

Reactive oxygen species (ROS) and salicylic acid (SA) are two factors regulating leaf senescence and defense against pathogens. However, how a single gene integrates both ROS and SA pathways remains poorly understood. Here, we show that Arabidopsis WRKY55 transcription factor positively regulates ROS and SA accumulation, and thus leaf senescence and resistance against the bacterial pathogen Pseudomonas syringaeWRKY55 is predominantly expressed in senescent leaves and encodes a transcriptional activator localized to nuclei. Both inducible and constitutive overexpression of WRKY55 accelerates le

A sucrose-jasmonic acid feedback loop drives stem cell senescence in Arabidopsis.

The lifespan of annual plants is tightly linked to stem cell senescence and the cessation of proliferative activity in the inflorescence meristem (IM). A "senescence factor" originating in developing fruits has long been proposed to trigger proliferative arrest in the IM. Here, we show that jasmonic acid (JA) accumulates in the aging IM, where the JA-responsive transcription factor MYC2 directly represses WUSCHEL (WUS) expression and cytokinin accumulation. We demonstrate that sucrose originating in the siliques triggers local JA biosynthesis in the IM and induces stem cell senescence and meri

The chloroplast-localised metalloprotease EGY1 and Mg-dechelatase SGR1 control chloroplast development and senescence by modulating photosynthetic functions.

The chloroplast plays an essential role in the production of biochemical energy and metabolism, as well as in plant development, including senescence. We describe a mutant with unusual progression of senescence and identify it as a new allele of ethylene-dependent gravitropism-deficient and yellow-green 1 (egy1-5). EGY1 encodes a chloroplast-localized metalloprotease that regulates chlorophyll metabolism. The single egy1-5 mutant exhibits various defects in chloroplast function, including abnormal accumulation of photosynthetic complexes and defects in photosynthetic parameters, as well as alt
