Evolutionary tuning of the molecular charge state of UBP24 shapes responses to high temperature.

Protein evolution is shaped by sequence variation that modulates protein properties-for example, through the gain or loss of post-translational modifications. Among these, reversible phosphorylation alters a protein's overall electrical charge and enables organisms to dynamically respond to environmental fluctuations. In plants, the hydro-active opening of stomata, microscopic pores that regulate gas exchange and leaf temperature, is governed by phosphorylation-dependent signalling. Here we identify a mechanism involving the deubiquitylase UBIQUITIN-SPECIFIC PROTEASE 24 (UBP24) that promotes s

ATP-dependent binding cassette transporter G family member 16 increases plant tolerance to abscisic acid and assists in basal resistance against Pseudomonas syringae DC3000.

Plants have been shown previously to perceive bacteria on the leaf surface and respond by closing their stomata. The virulent bacterial pathogen Pseudomonas syringae pv tomato DC3000 (PstDC3000) responds by secreting a virulence factor, coronatine, which blocks the functioning of guard cells and forces stomata to reopen. After it is inside the leaf, PstDC3000 has been shown to up-regulate abscisic acid (ABA) signaling and thereby suppress salicylic acid-dependent resistance. Some wild plants exhibit resistance to PstDC3000, but the mechanisms by which they achieve this resistance remain unknow

Thermal study of plants in a dynamic environment using thermovision.

We examined the effect of light-induced heating of leaves in wild-type (Col-0) and tmm-1 Arabidopsis plants. The results showed that the tmm-1 mutant exhibits accelerated cooling of rosettes in response to changes in light intensity, as demonstrated by thermal imaging measurements. This is evident in the time constants determined for temperature kinetics, which were reduced during the transition from light to dark. Our findings indicate that these dynamics can be reliably assessed under various light conditions. Measurements of gas exchange showed that stomatal conductance, transpiration rate,

Methanol chemoreceptor MtpA- and flagellin protein FliC-dependent methylotaxis contributes to the spatial colonization of PPFM in the phyllosphere.

Pink-pigmented facultative methylotrophs (PPFMs) capable of growth on methanol are dominant and versatile phyllosphere bacteria that provide positive effects on plant growth through symbiosis. However, the spatial behavior of PPFMs on plant surfaces and its molecular basis are unknown. Here, we show that Methylobacterium sp. strain OR01 inoculated onto red perilla seeds colonized across the entire plant surface in the phyllosphere concomitant with the plant growth. During its transmission, strain OR01 was found to be present on the entire leaf surface with a preference to sites around the peri

Wide vessels sustain marginal transpiration flux and do not optimize inefficient gas exchange activity under impaired hydraulic control and salinity.

Plants optimize water use and carbon assimilation via transient regulation of stomata resistance and by limiting hydraulic conductivity in a long-term response of xylem anatomy. We postulated that without effective hydraulic regulation plants would permanently restrain water loss and photosynthetic productivity under salt stress conditions. We compared wild-type tomatoes to a transgenic type (TT) with impaired stomatal control. Gas exchange activity, biomass, starch content, leaf area and root traits, mineral composition and main stems xylem anatomy and hydraulic conductivity were analyzed in

Recent insights into air space formation in plant shoots.

An adequate supply of oxygen and carbon dioxide is essential for plant survival. Although plant cell walls are somewhat porous, their hydrated nature hampers gas diffusion. Furthermore, the cuticular wax coating the epidermal layer of aerial tissues strongly inhibits gas exchange. Because plants lack specialized systems that bind and transport gases, gases must be directly delivered to the target cells. This necessitates the establishment of effective gas transport pathways connecting stomata to the target cells. However, our understanding of this process remains fragmented. Recent studies hav

A stomate by any other name? The open question of hornwort gametophytic pores, their homology, and implications for the evolution of stomates.

Advances in bryophyte genomics and the phylogenetic recovery of hornworts, mosses, and liverworts as a clade have spurred considerable recent interest in character evolution among early embryophytes. Discussion of stomatal evolution, however, has been incomplete; the result of the neglect of certain potential stomate homologues, namely the two-celled epidermal gametophytic pores of hornworts (typically referred to as 'mucilage clefts'). Confusion over the potential homology of these structures is the consequence of a relatively recent consensus that hornwort gametophytic pores ('HGPs' - our te

Automated calibration of stomatal conductance models from thermal imagery by leveraging synthetic images generated from Helios 3D biophysical model simulations.

Stomatal conductance (gs) is indicative of plant carbon dioxide uptake via photosynthesis and water loss via transpiration, making it a crucial plant biophysical trait. Direct measurement of gs is labor-intensive and usually not scalable to large fields. Using manual measurements to estimate parameters of gs models is even more labor-intensive and prone to sampling errors. This study aimed to develop an automated pipeline for gs measurement and model calibration using thermal imagery data, which not only disentangles the impacts of genotype-specific stomatal traits and environmental conditions

Climate-resilient crops: Lessons from xerophytes.

Developing climate-resilient crops is critical for future food security and sustainable agriculture under current climate scenarios. Of specific importance are drought and soil salinity. Tolerance traits to these stresses are highly complex, and the progress in improving crop tolerance is too slow to cope with the growing demand in food production unless a major paradigm shift in crop breeding occurs. In this work, we combined bioinformatics and physiological approaches to compare some of the key traits that may differentiate between xerophytes (naturally drought-tolerant plants) and mesophyte

Cell wall water shields stomata against falling leaf airspace humidity.

Plants lose water by transpiration through stomatal pores. However, it remains a matter of debate whether relative humidity (RH) in the substomatal cavity may fall below saturation and guard cells experience strong differences water potential driven by RH in the cavity. We developed a gas exchange chamber to control RH and CO2 at the inner epidermal surface. Vicia faba L. stomata remained open with high stomatal conductance (gs), even when RH inside was reduced substantially below saturation. Concurrent measurements showed no resolvable decline in bulk cell wall water potential, even with 50 %

Foliar water and solute absorption: an update.

The absorption of water and solutes by plant leaves has been recognised since more than two centuries. Given the polar nature of water and solutes, the mechanisms of foliar uptake have been proposed to be similar for water and electrolytes, including nutrient solutions. Research efforts since the 19th century focussed on characterising the properties of cuticles and applying foliar sprays to crop plants as a tool for improving crop nutrition. This was accompanied by the development of hundreds of studies aimed at characterising the chemical and structural nature of plant cuticles from differen

Effects of major vein blockage and aquaporin inhibition on leaf hydraulics and stomatal conductance.

The density and architecture of leaf veins determine the network and efficiency of water transport within laminae and resultant leaf gas exchange and vary widely among plant species. Leaf hydraulic conductance ( Kleaf) can be regulated by vein architecture in conjunction with the water channel protein aquaporin. However, our understanding of how leaf veins and aquaporins affect leaf hydraulics and stomatal conductance ( gs) remains poor. By inducing blockage of the major veins and inhibition of aquaporin activity using HgCl2, we examined the effects of major veins and aquaporins on Kleaf and g

Stomatal Decoupling From Photosynthesis Under High Temperatures Is Consistent With Stomatal Optimisation.

Stomatal pores on plant leaves regulate the gain of carbon through photosynthesis and the loss of water through transpiration. Through their responses to environmental conditions, stomata can constrain plant productivity and transpiration fluxes, exerting a strong control on climate feedbacks over land. Although mechanistic modelling of stomata remains a challenge, semi-empirical and optimisation models have been successfully applied to improve the simulation of land-atmosphere fluxes of water and carbon. Optimisation approaches assume that some aspect of plant function, such as photosynthesis

OsCYCD7;1 rigidly determines guard mother cell division in rice.

Functional stomata depend on paired guard cells that arise from a single symmetric division of the guard mother cell (GMC). However, the mechanism governing this division in grasses with unique dumbbell-shaped guard cells remains poorly understood. Here, we identify OsCYCD7;1 as an essential component for GMC division in rice. OsCYCD7;1 was strictly confined to pre-divisional GMCs, and its knockout prevented GMC division. OsSMR9, a plant CDK (cyclin-dependent kinase) inhibitor, physically interacted with OsCYCD7;1 and suppressed GMC division. Yeast one-hybrid (Y1H) and EMSA assays revealed OsM

Aba-induced active stomatal closure in bulb scales of Lanzhou lily.

Abscisic acid (ABA) mediated stomatal closure is a highly effective mode of active stomatal regulation under drought stress. Previous studies on stomatal regulation have primarily focused on the leaves of vascular plants, while research on the stomatal behavior of bulbous plants remains unknown. In addition, ABA-induced stomatal regulation in bulbs has yet to be explored. Therefore, we aim to investigate the ABA-induced active regulation in the bulb of the Lanzhou lily (Lilium davidii var. unicolor). The morphological characteristics of epidermal strips were analyzed along with a stomatal aper

How does the VPD response of isohydric and anisohydric plants depend on leaf surface particles?

Atmospheric vapour pressure deficit (VPD) is the driving force for plant transpiration. Plants have different strategies to respond to this 'atmospheric drought'. Deposited aerosols on leaf surfaces can interact with plant water relations and may influence VPD response. We studied transpiration and water use efficiency of pine, beech and sunflower by measuring sap flow, gas exchange and carbon isotopes, thereby addressing different time scales of plant/atmosphere interaction. Plants were grown (i) outdoors under rainfall exclusion (OD) and in ventilated greenhouses with (ii) ambient air (AA) o

More shade or sun? How fluctuating light affects photosynthesis in cotton.

Natural light conditions are inherently dynamic, yet their effects on plant light acclimation remain ambiguous. We established two light regimes with the same daily quantum input [constant light (CL) and fluctuating light (FL)] with cotton (Gossypium hirsutum L.) cultivar Xinkang4 as the object, and measured leaf anatomy, chlorophyll fluorescence, photosynthetic gas exchange, and light induction kinetics. (1) Compared with CL, FL induced more shade-acclimated leaf morphology, with total leaf thickness reduced by 8.2%, palisade layer thickness by 11.3%, and total stomatal density by 16.7%. (2)

Nocturnal pollutant uptake contributes significantly to the total stomatal uptake of Mangifera indica.

DO3SE (Deposition of Ozone for Stomatal Exchange), is a dry deposition model, designed to assess tropospheric ozone risk to vegetation, and is based on two alternative algorithms to estimate stomatal conductance: multiplicative and photosynthetic. The multiplicative model has been argued to perform better for leaf-level and regional-level application. In this study, we demonstrate that the photosynthetic model is superior to the multiplicative model even for leaf-level studies using measurements performed on Mangifera indica. We find that the multiplicative model overestimates the daytime stom

Enhancing Rubisco gene expression and metabolites accumulation for better plant growth in Ficus deltoidea under drought stress using hydrogen peroxide.

Growth improvement of the medicinal plant, Ficus deltoidea (Mas Cotek) under drought conditions is a vital issue in Malaysia since it is a slow-growing plant and disposed to leaf damage under the stresses of drought. Therefore, investigation was done to examine the outcomes of hydrogen peroxide (H2O2) application on Rubisco gene expression and metabolites accumulation of stressed F. deltoidea plants, and thereby to record the changes in leaf histology, photosynthesis, biochemical properties, and the growth of the plant. H2O2 at the rates of 0, 5, 10, 15, and 20 mM were foliar sprayed biweekly
