Systemic signaling during plant defense.

Systemic acquired resistance (SAR) is a type of pathogen-induced broad-spectrum resistance in plants. During SAR, primary infection-induced rapid generation and transportation of mobile signal(s) 'prepare' the rest of the plant for subsequent infections. Several, seemingly unrelated, mobile chemical inducers of SAR have been identified, at least two of which function in a feed-back regulatory loop with a lipid transfer-like protein. Signal(s) perception in the systemic tissues relies on the presence of an intact cuticle, the waxy layer covering all aerial parts of the plant. SAR results in chr

[Nonexpressor of pathogenesis-related genes 1 (NPR1): a key node of plant disease resistance signalling network].

The NPRI (nonexpressor of pathogenesis-related genes (1) gene, firstly cloned in Arabidopsis thaliana, is a key gene involved in regulation of plant disease resistance. It plays a pivotal role not only in systemic acquired resistance (SAR) and induced systemic resistance (ISR), but also in basic resistance and resistance (R) gene-dependent resistance. NPR1 monomerization induced by enhanced reducing condition after oxidative burst, and the accumulation of NPR1 monomers in the nuclei, are required and enough for expression of PR (pathogenesis-related) genes and SAR. NPR1 regulates PR gene expre

Multi-generational effects of salicylic acid on Lipaphis erysimi and induced defense in Brassica juncea.

Turnip aphid, Lipaphis erysimi Kalt. (Hemiptera: Aphididae), is an insecticide-resistant pest of Brassica spp. SA-salicylic acid, a hormonal regulator of systemic acquired resistance in plants, might prove to be a better option against resistant pests. Therefore, SA was applied exogenously in the four treatments with the concentrations of 0, 0.5, and 1 mM 24-hours before infesting the Brassica juncea (Chinese leaf mustard) plants with L. erysimi. Responses of aphid were noted for two generations (F1 and F2) on reproductive fitness parameters. Kaplan-Meier survivorship, developmental period, re

TGACG-BINDING FACTOR 1 (TGA1) and TGA4 regulate salicylic acid and pipecolic acid biosynthesis by modulating the expression of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g).

Salicylic acid (SA) and pipecolic acid (Pip) play important roles in plant immunity. Here we analyzed the roles of transcription factors TGACG-BINDING FACTOR 1 (TGA1) and TGA4 in regulating SA and Pip biosynthesis in Arabidopsis thaliana. We quantified the expression levels of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g), which encode two master transcription factors of plant immunity, and the accumulation of SA and Pip in tga1-1 tga4-1 mutant plants. We tested whether SARD1 and CBP60g are direct targets of TGA1 by chromatin immunoprecipitation-p

Systemic acquired resistance networks amplify airborne defense cues.

Salicylic acid (SA)-mediated innate immune responses are activated in plants perceiving volatile monoterpenes. Here, we show that monoterpene-associated responses are propagated in feed-forward loops involving the systemic acquired resistance (SAR) signaling components pipecolic acid, glycerol-3-phosphate, and LEGUME LECTIN-LIKE PROTEIN1 (LLP1). In this cascade, LLP1 forms a key regulatory unit in both within-plant and between-plant propagation of immunity. The data integrate molecular components of SAR into systemic signaling networks that are separate from conventional, SA-associated innate

Systemic acquired resistance-associated transport and metabolic regulation of salicylic acid and glycerol-3-phosphate.

Systemic acquired resistance (SAR), a type of long-distance immunity in plants, provides long-lasting resistance to a broad spectrum of pathogens. SAR is thought to involve the rapid generation and systemic transport of a mobile signal that prepares systemic parts of the plant to better resist future infections. Exploration of the molecular mechanisms underlying SAR have identified multiple mobile regulators of SAR in the last few decades. Examination of the relationship among several of these seemingly unrelated molecules depicts a forked pathway comprising at least two branches of equal impo

Salicylic acid: transport and long-distance immune signaling.

The small phenolic compound salicylic acid (SA) is a phytohormone that regulates many biological processes, although it is most well-known for its role in plant defense. SA is an important regulator of systemic acquired resistance (SAR), a type of systemic immunity that protects uninfected parts of the plant against secondary infections by a broad spectrum of pathogens. SAR involves the generation of mobile signal(s) at the primary infection site, which translocate to distal uninfected portions and activate systemic disease resistance. Although, SA was considered to not constitute the mobile S

Glycerol-3-phosphate metabolism in wheat contributes to systemic acquired resistance against Puccinia striiformis f. sp. tritici.

Glycerol-3-phosphate (G3P) is a proposed regulator of plant defense signaling in basal resistance and systemic acquired resistance (SAR). The GLY1-encoded glycerol-3-phosphate dehydrogenase (G3PDH) and GLI1-encoded glycerol kinase (GK) are two key enzymes involved in the G3P biosynthesis in plants. However, their physiological importance in wheat defense against pathogens remains unclear. In this study, quantification analysis revealed that G3P levels were significantly induced in wheat leaves challenged by the avirulent Puccinia striiformis f. sp. tritici (Pst) race CYR23. The transcriptional

Light acclimation, retrograde signalling, cell death and immune defences in plants.

This review confronts the classical view of plant immune defence and light acclimation with recently published data. Earlier findings have linked plant immune defences to nucleotide-binding site leucine-rich repeat (NBS-LRR)-dependent recognition of pathogen effectors and to the role of plasma membrane-localized NADPH-dependent oxidoreductase (AtRbohD), reactive oxygen species (ROS) and salicylic acid (SA). However, recent results suggest that plant immune defence also depends on the absorption of excessive light energy and photorespiration. Rapid changes in light intensity and quality often c

New molecules in plant defence against pathogens.

Plants host a multipart immune signalling network to ward off pathogens. Pathogen attack upon plant tissues can often lead to an amplified state of (induced) defence against subsequent infections in distal tissues; this is known as systemic acquired resistance (SAR). The interaction of plants with beneficial microbes of the rhizosphere microbiome can also lead to an induced resistance in above-ground plant tissues, known as induced systemic resistance. Second messengers such as calcium (Ca2+), reactive oxygen species (ROS), and nitric oxide (NO) are necessary for cell-to-cell signal propagatio

Grasping Molecular Biology Mechanisms to Optimize Plant Resistance and Advance Microbiome Role Against Phytonematodes.

Plant-parasitic nematodes (PPNs) cause big crop losses globally. Safe/reliable methods for their durable management strategies can harness various beneficial relationships among the plant immune system and related microbiomes. Molecular mechanisms basic to these relations reveal wide arrays of significant roles for plant-healthy growth. This review focuses on such relations of microbiomes to prime and immunize plants against PPNs. It also highlights molecular issues facing PPN-resistant varieties with possible solutions such as genetic breeding/engineering, grafting, PPN-antagonistic root exud

Dual regulation role of GH3.5 in salicylic acid and auxin signaling during Arabidopsis-Pseudomonas syringae interaction.

Salicylic acid (SA) plays a central role in plant disease resistance, and emerging evidence indicates that auxin, an essential plant hormone in regulating plant growth and development, is involved in plant disease susceptibility. GH3.5, a member of the GH3 family of early auxin-responsive genes in Arabidopsis (Arabidopsis thaliana), encodes a protein possessing in vitro adenylation activity on both indole-3-acetic acid (IAA) and SA. Here, we show that GH3.5 acts as a bifunctional modulator in both SA and auxin signaling during pathogen infection. Overexpression of the GH3.5 gene in an activati

Floro-pyrazolo[3,4-d]pyrimidine derivative as a novel plant activator induces two-pathway immune system.

Plants are known to develop a multi-faceted innate immune system for pathogenic defense. Systemic acquired resistance (SAR) and induced systemic resistance (ISR) are the two main pathways. Many chemical inducers known as plant activators that activate innate immunity to defend against pathogens have been discovered. Currently, the exploitation of new plant activators is mainly done to develop analogs of salicylic acid as SAR-signaling molecules; however, the ISR pathway is hardly investigated for new plant activators. Based on recent studies on 1-methyl pyrazolo [3,4-d]pyrimidine bioactivity a

Characterization of plant immunity-activating mechanism by a pyrazole derivative.

A newly identified chemical, 4-{3-[(3,5-dichloro-2-hydroxybenzylidene)amino]propyl}-4,5-dihydro-1H-pyrazol-5-one (BAPP) was characterized as a plant immunity activator. BAPP enhanced disease resistance in rice against rice blast disease and expression of a defense-related gene without growth inhibition. Moreover, BAPP was able to enhance disease resistance in dicotyledonous tomato and Arabidopsis plants against bacterial pathogen without growth inhibition, suggesting that BAPP could be a candidate as an effective plant activator. Analysis using Arabidopsis sid2-1 and npr1-2 mutants suggested t

N-hydroxy-pipecolic acid-mediated signal transduction and resistance requires DAWDLE.

Plant systemic acquired resistance (SAR) is a pathogen-induced, long-distance immune response that primes uninfected tissue against future pathogen attacks. N-hydroxy-pipecolic acid (NHP) is a mobile signal required to initiate and amplify SAR signaling in different plant species. However, the key regulators involved in activating NHP-mediated signaling and systemic resistance remain unclear. In this study, we identified an Arabidopsis (Arabidopsis thaliana) mutant, dawdle (ddl), that exhibits a compromised systemic resistance phenotype upon NHP treatment and pathogen infection. Biosynthesis and Regulation of Salicylic Acid and N-Hydroxypipecolic Acid in Plant Immunity.

Salicylic acid (SA) has long been known to be essential for basal defense and systemic acquired resistance (SAR). N-Hydroxypipecolic acid (NHP), a recently discovered plant metabolite, also plays a key role in SAR and to a lesser extent in basal resistance. Following pathogen infection, levels of both compounds are dramatically increased. Analysis of SA- or SAR-deficient mutants has uncovered how SA and NHP are biosynthesized. The completion of the SA and NHP biosynthetic pathways in Arabidopsis allowed better understanding of how they are regulated. In this review, we discuss recent progress

A vitamin B3-driven root bacterial metabolite primes systemic immunity in Arabidopsis.

The microbiota is being increasingly recognized for its ability to regulate host physiology through the production of small bioactive molecules. However, how host-derived nutrients are metabolically transformed by root-associated microbes to influence plant immunity remains poorly understood. Here, we show that vitamin B3 (VB3; niacin) secreted by plant roots shapes the assembly of a functionally specialized root microbiota, which, in turn, metabolizes VB3 into an immune-active signal that enhances plant disease resistance. VB3 secretion selectively increases the abundance of root-associated b

Simultaneous Quantification of Salicylic Acid, Pipecolic Acid, and N-Hydroxypipecolic Acid in Plants Using High-Performance Liquid Chromatography Coupled with Tandem Mass Spectrometry.

Plant immune activation in response to disease-causing pathogens entails proper spatiotemporal regulation of plant hormones and defense-activating metabolites. In Arabidopsis thaliana and numerous plant species, disease resistance to biotrophic and hemibiotrophic pathogens is underpinned by accumulation of these important immune signals, which include salicylic acid (SA), pipecolic acid (Pip), and N-hydroxypipecolic acid (NHP). SA, Pip, and NHP are essential for activating plant defense against various pathogens in local infected tissues, as well as priming distal, uninfected plant tissues in

The Janus face of reactive oxygen species in resistance and susceptibility of plants to necrotrophic and biotrophic pathogens.

Plant pathogens can be divided into biotrophs and necrotrophs according to their different life styles; biotrophs prefer living, while necrotrophs prefer dead cells for nutritional purposes. Therefore tissue necrosis caused by reactive oxygen species (ROS) during pathogen infection increases host susceptibility to necrotrophic, but resistance to biotrophic pathogen. Consequently, elevation of antioxidant capacity of plants enhances their tolerance to development of necroses caused by necrotrophic pathogens. Plant hormones can strongly influence induction of ROS and antioxidants, thereby influe

The role of methyl salicylate in plant growth under stress conditions.

Methyl salicylate is a volatile compound, the synthesis of which takes place via the salicylic acid pathway in plants. Both compounds can be involved in the development of systemic acquired resistance and they play their role partly independently. Salicylic acid transport has an important role in long-distance signalling, but methyl salicylate has also been suggested as a phloem-based mobile signal, which can be demethylated to form salicylic acid, inducing the de-novo synthesis of salicylic acid in distal tissue. Despite the fact that salicylic acid has a protective role in abiotic stress res
