First Report of Streptomyces stelliscabiei Causing Potato Common Scab in Michigan.

Potato (Solanum tuberosum L.) common scab can be caused by multiple Streptomyces spp., with S. scabies as a predominant species (2,3). However, according to our survey in August 2007, many symptomatic potato tubers did not have S. scabies in Michigan. To identify the pathogen, potato tubers with scab symptoms were collected from two fields in Michigan, and Streptomyces spp. were isolated using Streptomyces selective medium (STR) (2). Pure cultures of the isolates were obtained by transferring single colonies and incubation at 28°C on STR. Three isolates, designated HER21, HER24, and HER26, wer

A simple and effective method to enhance the level of gamma-aminobutyric acid in Chinese yam tubers while preserving its original appearance.

Hot-air drying is an effective method to enhance the levels of gamma-aminobutyric acid (GABA) in edible tubers/tuberous roots. However, consumers prefer fresh food to processed food. Therefore, this study aims to develop an effective method to increase the GABA levels in the tubers of Chinese yam (CY tubers) and the tubers/tuberous roots of other plants while preserving its original appearance. Among nitrogen treatment (treatment under a nitrogen atmosphere), carbon dioxide (CO2) treatment (treatment under a CO2 atmosphere), vacuum treatment, and water immersion, CO2 treatment was the most eff

Antiproliferative Effects of Fractionated Crude Lectins From Boiled Japanese Taro Tubers (Colocasia esculenta (L.) Schott).

Lectins are carbohydrate-binding proteins with diverse biological functions, including antitumor and immunomodulatory activities. Taro (Colocasia esculenta) tubers, commonly consumed in cooked form, are a potential bioactive lectin source; however, the effects of thermal processing on their functionality remain insufficiently understood. This study investigated the biological profiles of lectins fractionated from both raw and boiled Japanese taro tubers (JTT). Lectins were fractionated using carboxymethyl-Sepharose and diethylaminoethyl-Sepharose chromatography, depending on their solubility f

Effect of the level of "Candidatus Liberibacter solanacearum" infection on the development of zebra chip disease in different potato genotypes at harvest and post storage.

Potato psyllid (Bactericera cockerelli Sulc)-transmitted "Candidatus Liberibacter solanacearum" (Lso) has been negatively impacting the potato industry in the United States as well as other potato-producing countries. Lso has been linked to a condition known as zebra chip (ZC) that affects yield and quality of potato tubers. Efforts to find sources of resistance to ZC have primarily focused on greenhouse evaluations based on a single inoculation time prior to harvest. Plant response to infection, however, could be influenced by the developmental stage of the host plant, and ZC may continue to

Expression and assembly of cholera toxin B subunit and domain III of dengue virus 2 envelope fusion protein in transgenic potatoes.

The rates of mosquito-transmitted dengue virus infection in humans have increased in tropical and sub-tropical areas. Domain III of dengue envelope protein (EDIII) is involved in cellular receptor binding and induces serotype-specific neutralizing antibodies. EDIII fused to the B subunit of Vibrio cholera (CTB-EDIII) was expressed in potatoes to develop a plant-based vaccine against dengue virus type 2. CTB-EDIII fused to an endoplasmic reticulum (ER) retention signal, SEKDEL, was introduced into potatoes by A. tumefaciens-mediated gene transformation. The integration of the CTB-EDIII fusion g

Occurrence of Turnip Scab Caused by Phytotoxin-Producing Streptomyces spp.

Turnips (Brassica rapa L var. rapifera) purchased from a New Brunswick supermarket in the winter of 2003 contained discernable corky scab-like lesions on their surfaces. The turnips were surface-disinfested with a warm-water rinse, and microorganisms from excised lesions were isolated with methodologies developed for the specific isolation of Streptomyces spp. (1). Nine isolates morphologically characteristic of Streptomyces scabies strains (2) were subsequently examined for pathogenicity on potato mini-tubers and production of the scab phytotoxin, thaxtomin A (1). Four of the nine isolates pr

Synthesis and assembly of anthrax lethal factor-cholera toxin B-subunit fusion protein in transgenic potato.

A DNA encoding the 27-kDa domain I of anthrax lethal factor protein (LF), was linked to the carboxyl terminus of the cholera toxin B-subunit (CTB-LF). The CTB-LF fusion gene was transferred into Solanum tuberosum cells by Agrobacterium tumefaciens-mediated in vivo transformation methods and antibiotic-resistant plants were regenerated. The CTB-LF fusion gene was detected in transformed potato leaf genomic DNA by polymerase chain reaction (PCR)-mediated DNA amplification. Immunoblot analysis with anti-CTB and anti-LF primary antibodies verified the synthesis and assembly of biologically active

Remodeling of the tuberization-controlling FLOWERING LOCUS T genes from non-tuberizing ancestor to potato.

Potato (Solanum section Petota) originated from a natural hybridization between tomato and Etuberosum ancestors, through which the trait of tuber formation evolved. Genetically, tuberization in potato is controlled by FLOWERING LOCUS T (FT) homologs. However, from non-tuberous ancestors to potato, what remodeling of these tuber formation determinants emerged remains unclear. Using homology-based discovery, expression profiling, CRISPR knockouts, and genetic complementation, we here show that tuberization-controlling FT-like genes underwent Petota-specific expansion, restructuring, and divergen

[Isolation of protoplasts from vegetable tissues using extracellular lytic enzymes from fusarium oxysporum f.sp. melonis].

Fusarium oxysporum f.sp. melonis, a pathogen of melon (Cucumis melo L.), was grown in shaken cultures at 26 degrees C in a mineral salts medium containing glucose, xylan and apple pectin as carbon sources. The extracellular enzymic complex obtained from these cultures showed lytic activity on plant tissues, causing maceration of melon fruits, potato tubers and carrot roots. Protoplasts were isolated from melon fruits when the maceration was carried out under appropriate osmotic conditions. This fact suggest a possible relationship between the enzymes produced by Fusarium oxysporum f.sp. meloni

Identification of a New Haplotype of 'Candidatus Liberibacter solanacearum' in Solanum tuberosum.

In 2017, potato tubers suspected of being infected with the bacterium 'Candidatus Liberibacter solanacearum' were received from the Animal and Plant Health Inspection Service in the United States. A total of 368 chipping tubers were observed for internal symptoms of zebra chip disease, which is associated with 'Ca. L. solanacearum' infection in the United States, Mexico, Central America, and New Zealand. A single tuber sliced at the stem end showed classic zebra chip symptoms of darkened medullary rays, with streaking and necrotic flecking. The symptomatic tuber was confirmed positive for the
