Translocator protein 18 kDA (TSPO) imaging using positron emission tomography (PET) is widely used to assess neuroinflammation in Alzheimer's disease (AD). However, the significance of the increase in brain TSPO levels in AD pathophysiology is not known. Here, we show that in the 5XFAD transgenic mouse model, brain TSPO levels increase in an age-, brain region-, and sex-dependent fashion. TSPO levels were first increased in the subiculum at 1.5 months of age in male and female 5XFAD mice compared to wildtype mice. The TSPO increase in the subiculum of 1.5-month 5XFAD mice coincided with the ap

Alzheimer's disease (AD) is an irreversible, progressive neurodegenerative disorder of the central nervous system that causes severe cognitive impairment. One of the most significant pathological features of AD is the accumulation of β‑amyloid (Aβ) peptide in the brain. Resveratrol (Res) is a polyphenol derived from peanuts, red grapes and other plants, which has received increasing attention due to its neuroprotective features. Tg6799 mice are transgenic mice with five familial AD (FAD) mutations that are also known as 5XFAD mice. The present study aimed to investigate the effects of Res on T

Alzheimer's disease (AD) is a complex disorder for which various in vivo models exist. The TgCRND8 mouse, transgenic for the human amyloid precursor protein, is an aggressive early onset model of brain amyloid deposition. Preliminary studies revealed that when the transgene is expressed on an A/J genetic background, these mice not only survive longer but also deposit less parenchymal amyloid-beta (Abeta) peptides as compared to those on a C57BL/6 background. We performed a genome-wide study of an F2 intercross between TgCRND8 on an A/J background and C57BL/6 mice, to identify genetic modulator

In this paper a review of the most recent investigations of the pathology of amyloid and of amyloid plaque is presented. The purpose of this report is to describe various clinical conditions (including their etiopathology and clinical manifestations) in which amyloid deposits (especially amyloid plaques) are significant features of the disease. Similarly, to define the neuropathological picture of amyloid deposits, molecular composition of amyloid fibrils and the genes which encode them.

In a hyper-aging society, taking measures against dementia, especially against dementia due to Alzheimer's disease, which accounts for about two thirds of dementia cases, is an important task socially and economically. Forty years have passed since amyloid β was isolated from a patient, 30 years have passed since the amyloid cascade hypothesis was proposed and antibody drugs based on this hypothesis have emerged, which has led to the new era of Alzheimer's disease treatment. This article reviews the significance of amyloid plaque removal with amyloid β-targeting antibody treatment for Alzheime

Substantial evidence suggests a role for immunotherapy in treating Alzheimer's disease (AD). While the precise pathophysiology of AD is incompletely understood, clinical trials of antibodies targeting aggregated forms of β amyloid (Aβ) have shown that reducing amyloid plaques can mitigate cognitive decline in patients with early-stage AD. Here, we describe what we believe to be a novel approach to target and degrade amyloid plaques by genetically engineering macrophages to express an Aβ-targeting chimeric antigen receptor (CAR-Ms). When injected intrahippocampally, first-generation CAR-Ms have

Formation of senile plaques composed of amyloid beta peptide, a pathological hallmark of Alzheimer disease, in human brains precedes disease onset by many years. Noninvasive detection of such plaques could be critical in presymptomatic diagnosis and could contribute to early preventive treatment strategies. Using amyloid precursor protein (APP) transgenic mice as a model of amyloid beta amyloidosis, we demonstrate here that an intravenously administered (19)F-containing amyloidophilic compound labels brain plaques and allows them to be visualized in living mice by magnetic resonance imaging (M

Moderate numbers of amyloid plaques with associated argyrophilic dystrophic neurites and cerebral amyloid angiopathy (CAA) but no neurofibrillary tangles (NFTs) were found in the brains of 3 middle-aged common marmosets (Callithrix jacchus) inoculated intracerebrally (i.c.) 6-7 years earlier with brain tissue from a patient with early onset Alzheimer's disease. The plaques and vascular amyloid stained positively with antibodies to beta (A4)-protein. The brains of 3 age-matched control marmosets from the same colony did not show these neuropathological features. beta-amyloid plaques and CAA (bu

Amyloid protein was isolated from the cerebral meninges of 4 aged dogs with cerebral amyloid angiopathy. By immunoblot analysis, antiserum against synthetic oligo-peptide consisting of 1-28 amino acid of amyloid beta protein recognized prominent wide band ranging from 14 to 18 kilodalton (kd). When amyloid samples were solubilized by formic acid, the antiserum recognized lower molecular weight band ranging from 3 to 4 kd. Immunohistochemical studies on cerebral amyloid angiopathy and senile plaques were performed in 17 aged dogs. Anti-amyloid beta protein serum labeled amyloid deposits in cere

Recent trends in neurodegeneration research have been aimed at developing new amyloid ligands for the neuroimaging of dementia. Among the positron emission tomography (PET) radiotracers, fluorodeoxyglucose F 18 ((18)F-FDG) is the compound most widely used in the diagnosis of neurodegenerative dementias. However, this compound shows a level of specificity and sensitivity for early Alzheimer's disease detection that is lower than that provided by high-affinity ligands for β-amyloid (Aβ). Among the new widely available fluorine 18 ((18)F)-labeled Aβ ligands, florbetapir F 18 ((18))F-AV-45; Amyvid

Blood vessels with amyloid angiopathy and senile plaques in the cortices of the brains with Alzheimer's disease and senile dementia were observed by means of a scanning electron microscope. the results obtained were as follows: The blood vessels with amyloid angiopathy were surrounded by solid substances. The senile plaques consisted of rough solid substances, contained degenerated cell processes, and almost all plaques existed around the degenerated capillaries with amyloid angiopathy. From the above described findings, we suggest that the senile plaque has an extremely cose relationship to t

Regional differences in senile plaques immunostained by antibodies against beta-amyloid A4 (beta-A4), amyloid precursor protein 695 (APP) and apolipoprotein E (apo E) were studied in the hippocampus and the entorhinal, temporal and occipital cortices both quantitatively and semiquantitatively with respect to the laminar cortical distribution of the plaques. These patterns were related to the staging of Alzheimer's disease in regard to the distribution of neurofibrillary tangles (Braak and Braak (1991) Acta Neuropathol 82: 239-259). In the hippocampus and especially in sector CA 1, no significa

BACKGROUND: Research in recent years firmly established that microglial cells play an important role in the pathogenesis of Alzheimer's disease (AD). In parallel, a series of studies showed that, under both homeostatic and pathological conditions, microglia are a heterogeneous cell population. In AD, amyloid-β (Aβ) plaque-associated microglia (PAM) display a clearly distinct phenotype compared to plaque-distant microglia (PCM), suggesting that these two microglia subtypes likely differently contribute to disease progression. So far, molecular characterization of PAM was performed indirectly us

BackgroundAmyloid-β peptides (Aβ) applied to Alzheimer's disease (AD) brain bind to amyloid plaques (APs) and this binding is saturable. Meanwhile, anti-immunoglobulin G (IgG) antibodies label APs. Considering the saturable binding of Aβ to APs and the localization of IgGs in APs, specific binding proteins (BPs) for the applied Aβ could be anti-Aβ IgG antibodies (anti-AβAbs) in APs.ObjectiveOur objective is to demonstrate whether anti-AβAbs are the BPs for administered Aβ in APs.MethodsTo identify the binding specificity, we established a tissue competition assay utilizing the competitiveness

INTRODUCTION: Amyloid plaque deposition is a defining feature of Alzheimer's disease (AD), yet how plaque-associated cellular alterations are reflected in noninvasive imaging remains unclear.
METHODS: We integrated high-resolution diffusion magnetic resonance imaging (dMRI) with spatial transcriptomics (ST) in an AD mouse model. Spatial correlations were assessed across the whole brain and in disease-control and plaque-non-plaque comparisons. Neural network models were trained to predict AD-associated cell types from dMRI metrics.
RESULTS: dMRI metrics showed moderate spatial correlations (r

Apolipoprotein E4 (ApoE4) is the most important and prevalent risk factor for late-onset Alzheimer's disease (AD). The isoelectric point of ApoE4 matches the pH of the early endosome (EE), causing its delayed dissociation from ApoE receptors and hence impaired endolysosomal trafficking, disruption of synaptic homeostasis, and reduced amyloid clearance. We have shown that enhancing endosomal acidification by inhibiting the EE-specific sodium-hydrogen exchanger 6 (NHE6) restores vesicular trafficking and normalizes synaptic homeostasis. Remarkably and unexpectedly, loss of NHE6 (encoded by the g

INTRODUCTION: Genome-wide association studies have identified MS4A4A, a microglia-enriched gene, as a modulator of Alzheimer's disease (AD) risk. Common variants in MS4A4A affect AD susceptibility, gene expression, triggering receptor expressed on myeloid cells 2 (TREM2) signaling, and microglial transcriptional states, but the gene's functional role remains unclear.
METHODS: Using a novel model, we investigated the impact of Ms4a4a loss in the 5xFAD mouse model of amyloid beta (Aβ) accumulation.
RESULTS: Ms4a4a deficiency reduced steady-state Aβ levels and shortened its half-life in brain i

Alzheimer's disease (AD) is the most common neurodegenerative brain disease and represents the most frequent type of dementia characterized by cognitive impairment and amnesia. AD neuropathology is connected to the development of synaptic dysfunction and loss of synaptic homeostasis due to an imbalance in the production and clearance of β-amyloid (Aβ) and intracellular neurofibrillary tangles (NFTs). However, AD neuropathology is complex and may relate to the deposition of other misfolded proteins, such as alpha-synuclein (α-Syn). Of note, α-Syn, which is involved in the pathogenesis of Parkin
