Genotypic analyses of IncHI2 plasmids from enteric bacteria.

Incompatibility (Inc) HI2 plasmids are large (typically > 200 kb), transmissible plasmids that encode antimicrobial resistance (AMR), heavy metal resistance (HMR) and disinfectants/biocide resistance (DBR). To better understand the distribution and diversity of resistance-encoding genes among IncHI2 plasmids, computational approaches were used to evaluate resistance and transfer-associated genes among the plasmids. Complete IncHI2 plasmid (N = 667) sequences were extracted from GenBank and analyzed using AMRFinderPlus, IntegronFinder and Plasmid Transfer Factor database. The most common IncHI2

SHIP: identifying antimicrobial resistance gene transfer between plasmids.

MOTIVATION: Plasmids are carriers for antimicrobial resistance (AMR) genes and can exchange genetic material with other structures, contributing to the spread of AMR. There is no reliable approach to identify the transfer of AMR genes across plasmids. This is mainly due to the absence of a method to assess the phylogenetic distance of plasmids, as they show large DNA sequence variability. Identifying and quantifying such transfer can provide novel insight into the role of small mobile elements and resistant plasmid regions in the spread of AMR. RESULTS: We developed SHIP, a novel method to qu

Evolution and exchange of plasmids in pathogenic Neisseria.

Horizontal gene transfer (HGT) is a major influence in driving the spread of antimicrobial resistance (AMR) in many bacteria. A conjugative plasmid which is widespread in Neisseria gonorrhoeae, pConj, prevented the use of tetracycline/doxycycline for treating gonococcal infection. Here, we show that pConj evolved in the related pathogen, Neisseria meningitidis, and has been acquired by the gonococcus from the meningococcus on multiple occasions. Following its initial acquisition, pConj spread to different gonococcal lineages; changes in the plasmid's conjugation machinery associated with anoth

Plasmids of Distinct IncK Lineages Show Compatible Phenotypes.

IncK plasmids are some of the main carriers of blaCTX-M-14 and blaCMY-2 genes and show high similarity to other plasmids belonging to the I complex, including IncB/O plasmids. Here, we studied the phylogenetic relationship of 37 newly sequenced IncK and IncB/O plasmids. We show that IncK plasmids can be divided into two compatible lineages named IncK1 and IncK2.

Distribution of Plasmids in Distinct Leptospira Pathogenic Species.

Leptospirosis, caused by pathogenic Leptospira, is a worldwide zoonotic infection. The genus Leptospira includes at least 21 species clustered into three groups--pathogens, non-pathogens, and intermediates--based on 16S rRNA phylogeny. Research on Leptospira is difficult due to slow growth and poor transformability of the pathogens. Recent identification of extrachromosomal elements besides the two chromosomes in L. interrogans has provided new insight into genome complexity of the genus Leptospira. The large size, low copy number, and high similarity of the sequence of these extrachromosomal

[Characteristics of penicillinase plasmids from Staphylococcus aureus strains].

Penicillinase plasmids are present in most MRSA strains. They are very varying in their genotype and phenotype they confer. Penicillinase plasmids were transduced from 80 hospital MRSA strains to NCTC 8325 and the phenotype as well as the incompatibility group of plasmid were determined. Resistance to cadmium (high and low level), resistance to organic and nonorganic mercury compounds, arsenate/arsenite/antimonium resistance, resistance to bismuth and hypersensitivity to bismuth, resistance to macrolides as well as beta-lactamase production and its inductibility were checked. Among the examine

[Function of large plasmids of the plague pathogen].

Function of the largest plasmid of Yersinia pestis was studied. Assay of four strains of highly virulent Y. pestis isolated from different natural sources demonstrated that the characters of endotoxin production, sugar and alcohol fermentation, nitrate reduction as well as requirement in nutrients are mediated by no Y. pestis plasmids. On the contrary, FI and FII antigens are determined by the largest plasmid in some strains and are chromosome-mediated in others. It may well be that the genes for these antigens are comprised within the transposon-like DNA fragment.

Plasmids as epidemiological markers in nosocomial Legionnaires' disease.

Plasmid analysis was used in the investigation of an outbreak of nosocomial Legionnaires' disease in four patients. Serogroup 1 strains were isolated from two patients, the air-conditioning cooling tower, and two hot-water tanks. All serogroup 1 strains contained two plasmids with approximate molecular masses of 21 and 48 megadaltons (Mdal). The serogroup 1 strain found in the cooling-tower isolate also contained an additional 1.9 Mdal-plasmid. Restriction-endonuclease analysis of the 21-Mdal plasmid that was present in patient and hot water-tank isolates revealed identical EcoRI and HaeIII fr

[Characteristics of the resistance of Pseudomonas aeruginosa plasmids].

A total of 503 clinical strains of Ps. aeruginosa isolated in hospitals of 4 towns in 1976 - 1978 were studied. It was shown that a significant number of the isolates were resistant to high concentrations of streptomycin (83.3 per cent), kanamycin (71 per cent), sulfanilamide (70.2 per cent) and mercuric chloride (61.6 per cent). Strains resistant to gentamicin (29.8 per cent) and carbenicillin (20.2 per cent) occurred comparatively rare. Resistance to polymyxin and rifampicin was recorded in 2 and 4 per cent f the isolates respectively. It was found that 77 per cent of the isolates could tran

beta-lactamases and R-plasmids of Haemophilus influenzae.

The emergence of resistance to ampicillin and other antibiotics in Haemophilus influenzae has been a relatively recent event. In contrast, drug resistance has been rampant in the Enterobacteriaceae for many years. Ampicillin-resistance in H. influenzae is almost invariably attributable to possession of the TEM (Type III a)beta-lactamase. As is common in other bacteria the gene specifying this enzyme is plasmid-borne in Haemophilus. Some ampicillin-resistant strains of H. influenzae can transfer the TEM beta-lactamase gene to other strains of Haemophilus, to Escherichia coli and to Pseudomonas

Variability of plasmid fitness effects contributes to plasmid persistence in bacterial communities.

Plasmid persistence in bacterial populations is strongly influenced by the fitness effects associated with plasmid carriage. However, plasmid fitness effects in wild-type bacterial hosts remain largely unexplored. In this study, we determined the fitness effects of the major antibiotic resistance plasmid pOXA-48_K8 in wild-type, ecologically compatible enterobacterial isolates from the human gut microbiota. Our results show that although pOXA-48_K8 produced an overall reduction in bacterial fitness, it produced small effects in most bacterial hosts, and even beneficial effects in several isola

Curing of plasmid pXO1 from Bacillus anthracis using plasmid incompatibility.

The large plasmid pXO1 encoding the anthrax toxin is important for the virulence of Bacillus anthracis. It is essential to cure pXO1 from B. anthracis to evaluate its role in the pathogenesis of anthrax infection. Because conventional methods for curing plasmids (e.g., curing agents or growth at elevated temperatures) can induce mutations in the host chromosomal DNA, we developed a specific and reliable method to eliminate pXO1 from B. anthracis using plasmid incompatibility. Three putative replication origins of pXO1 were inserted into a temperature-sensitive plasmid to generate three incompa

Control of replication of FII plasmids: comparison of the basic replicons and of the copB systems of plasmids R100 and R1.

The copy numbers of the FII plasmids R1 and R100 were determined in four different ways and found to be identical. Deletion of one of the copy number control genes, copB, together with its promoter gives rise to plasmid copy mutants with an increased copy number. The increase was found to be 8- and 3.5-fold for plasmids R1 and R100, respectively. These deletion derivatives were found to be extremely sensitive to the presence of CopB activity from their own parent plasmid but not to that of the other plasmid. Hence, the CopB protein and its target are plasmid-specific and not FII-group-specific

Plasmid DNA vaccination.

Plasmid DNA vaccination against tuberculosis is a very powerful and easy method for the induction of strong humoral responses, CD4+ mediated secretion of Th1 cytokines and CD8+ mediated CTL activity in mice. Tuberculosis DNA vaccines have not been assessed so far in humans, and clinical trials with DNA in general have been somewhat disappointing. However, numerous studies have reported on the potent priming capacity of plasmid DNA for Th1 and CD8+ mediated immune responses, which can be boosted subsequently by recombinant protein or recombinant pox-viruses. With respect to tuberculosis, prime/

Plasmid diversity in Chlamydia.

Chlamydiae exhibit low interspecies DNA homology and plasmids from different chlamydial species can be readily distinguished by Southern blot analysis and restriction enzyme profiling. In contrast, available plasmid sequence data from within the species Chlamydia trachomatis indicate that plasmids from human isolates are highly conserved. To evaluate the nature and extent of plasmid variation, the complete nucleotide sequences were determined for novel plasmids from three diverse non-human chlamydial isolates: pCpA1 from avian Chlamydia psittaci (N352); pCpnE1 from equine Chlamydia pneumoniae

Transfer of plasmids from Escherichia coli to Pseudomonas aeruginosa: characterization of a Pseudomonas aeruginosa mutant with enhanced recipient ability for enterobacterial plasmids.

Pseudomonas aeruginosa strain GT424, derived by the transfer of plasmid R45 to strain GT1, was shown to have an Era+ phenotype (enhanced recipient ability) for enterobacterial plasmids from incompatibility (Inc) groups C, FI, FII, J, N, W, and X. Increase in transfer frequency ranged from 10 to 10(6)-fold. Plasmids of IncFII, IncJ, and IncX were found to be transmissible only to the Era+ strain. Plasmids of Inc groups A-C, M, and T transferred at low frequency (less than 10(-7)) to both GT1 and GT424 and did not respond to the Era+ character of the latter. Also not responsive to the Era charac

Conjugative plasmid pAW63 brings new insights into the genesis of the Bacillus anthracis virulence plasmid pXO2 and of the Bacillus thuringiensis plasmid pBT9727.

BACKGROUND: Bacillus cereus, Bacillus anthracis and Bacillus thuringiensis belong to the genetically close-knit Bacillus cereus sensu lato group, a family of rod-shaped Gram-positive bacteria. pAW63 is the first conjugative plasmid from the B. cereus group to be completely sequenced. RESULTS: The 71,777 bp nucleotide sequence of pAW63 reveals a modular structure, including a 42 kb tra region encoding homologs of the Type IV secretion systems components VirB11, VirB4 and VirD4, as well as homologs of Gram-positive conjugation genes from Enterococcus, Lactococcus, Listeria, Streptococcus and St

Plasmid diversity in neisseriae.

Horizontal gene transfer constitutes an important force in prokaryotic genome evolution, and it is well-known that plasmids are vehicles for DNA transfer. Chromosomal DNA is frequently exchanged between pathogenic and commensal neisseriae, but relatively little is known about plasmid diversity and prevalence among these nasopharyngeal inhabitants. We investigated the plasmid contents of 18 Neisseria lactamica isolates and 20 nasopharyngeal Neisseria meningitidis isolates. Of 18 N. lactamica strains, 9 harbored one or more plasmids, whereas only one N. meningitidis isolate contained a plasmid.

Plasmidomics: studying plasmids as ecological entities beyond their hosts.

Plasmids are mobile genetic elements with autonomous replication, whose ecology extends beyond individual bacterial hosts. As molecular symbionts, they have the potential to traverse bacterial taxa and environments, disseminating adaptive genes and shaping microbial community structure through dynamics that are likely decoupled from host taxonomy. Plasmidomics - the omics discipline dedicated to the study of plasmids - has revealed that plasmid diversity and distribution respond to environmental gradients independently of their hosts, underscoring their roles as ecological entities. However, t

Comparative genomic analysis of the blaOXA-23-carrying plasmids.

UNLABELLED: The global dissemination of the carbapenem resistance gene blaOXA-23 is partly facilitated by plasmids, yet a comprehensive characterization of these vectors is lacking. This study analyzed the blaOXA-23-carrying plasmids selected from the National Center for Biotechnology Information RefSeq database to elucidate their general characteristics and genomic diversity. The blaOXA-23-carrying plasmids were predominantly hosted by Acinetobacter baumannii (94.06%), and their sizes varied widely, from 8.22 to 336.05 kb. Replicon typing revealed that the majority belonged to the RP Rep fami
