The SMA Modifier Plastin 3 Targets Cell Membrane-Associated Proteins in Motoneurons.

Loss of the Survival Motor Neuron (SMN) gene inevitably leads to spinal muscular atrophy (SMA), one of the most common fatal neuromuscular diseases in children with FDA and EMA approved therapies. However, the cellular mechanisms leading to neuromuscular junction (NMJ) dysfunction due to impaired Ca2+ homeostasis in the presynaptic compartment remain largely unexplained. In the last decade, the so-called SMA modifiers have gained attention. The F-actin bundler Plastin 3 (PLS3) is one of them and counteracts neurotransmission defects, including altered vesicle endocytosis, in Smn-deficient NMJs

Epigenetic regulation of plastin 3 expression by the macrosatellite DXZ4 and the transcriptional regulator CHD4.

Dysregulated Plastin 3 (PLS3) levels associate with a wide range of skeletal and neuromuscular disorders and the most common types of solid and hematopoietic cancer. Most importantly, PLS3 overexpression protects against spinal muscular atrophy. Despite its crucial role in F-actin dynamics in healthy cells and its involvement in many diseases, the mechanisms that regulate PLS3 expression are unknown. Interestingly, PLS3 is an X-linked gene and all asymptomatic SMN1-deleted individuals in SMA-discordant families who exhibit PLS3 upregulation are female, suggesting that PLS3 may escape X chromos

Meningeal Architecture of the Jugular Foramen: An Anatomic Study Using Plastinated Histologic Sections.

OBJECTIVE: This 3-dimensional histologic study aimed to provide a precise description of the meningeal structures in the jugular foramen. METHODS: 22 posterior skull base tissue blocks containing the jugular foramen region were obtained from 11 human cadaveric heads. These blocks were plastinated and cut into serial sections. After staining, these sections were examined under an optical microscope and used to reconstruct a 3-dimensional visualization model. RESULTS: At the intracranial orifice of the jugular foramen, the meningeal dura formed 2 separate dural perforations: the glossopharynge

L-plastin regulates the stability of the immune synapse of naive and effector T-cells.

T-cells need to be tightly regulated during their activation and effector phase to assure an appropriate defence against cancer or pathogens and - vice versa - to avoid autoimmune reactions. Regulatory signals are provided via the immune synapse between T-cells and antigen-presenting cells (APCs) or target cells. The stability and kinetics of immune synapse formation is critical for proper T-cell functions. It requires dynamic rearrangements of the actin cytoskeleton necessary for organized spatio-temporal redistribution of receptors and adhesion molecules. We identified glucocorticoid-sensiti

Creation of plastinated placentas as a novel teaching resource for medical education in obstetrics and gynaecology.

INTRODUCTION: Knowledge of the gross anatomy of the placenta is fundamental in order to help identify potential complications during pregnancy. The placenta is difficult to study without a three-dimensional appreciation of its structure. The aim of this study was to develop a collection of plastinated placenta specimens and accompanying clinical educational materials to provide learning resources for placental abnormalities and their associated pregnancy outcomes. These plastinates and educational modules were used as teaching resources for both undergraduate and post-graduate medical trainees

L-plastin is essential for alveolar macrophage production and control of pulmonary pneumococcal infection.

We report that mice deficient for the hematopoietic-specific, actin-bundling protein L-plastin (LPL) succumb rapidly to intratracheal pneumococcal infection. The increased susceptibility of LPL(-/-) mice to pulmonary pneumococcal challenge correlated with reduced numbers of alveolar macrophages, consistent with a critical role for this cell type in the immediate response to pneumococcal infection. LPL(-/-) mice demonstrated a very early clearance defect, with an almost 10-fold-higher bacterial burden in the bronchoalveolar lavage fluid 3 h following infection. Clearance of pneumococci from the

T-plastin expression downstream to the calcineurin/NFAT pathway is involved in keratinocyte migration.

Cutaneous wound healing requires keratinocyte proliferation, migration and differentiation to restore the barrier function of the skin. The calcineurin/nuclear factor of activated-T-cell (NFAT) signaling pathway has been recently shown to be involved in keratinocyte growth, differentiation and migration. It is induced by an increased intracellular calcium rate and its inhibition results in decreased capacities of keratinocytes to migrate. Nevertheless, the link between calcineurin activation and keratinocyte migration remains unknown. Recently, Orai1, a pore subunit of a store-operated calcium

Inducible expression and pathophysiologic functions of T-plastin in cutaneous T-cell lymphoma.

A molecular feature of Sézary syndrome (SS) is the abnormal expression of T-plastin by malignant T cells. Herein, we investigated the molecular mechanisms involved in T-plastin synthesis and the functions of this actin-binding protein, with a special interest in chemoresistance and migration. We confirm the specific expression of T-plastin in peripheral blood lymphocytes (PBLs) from SS patients and its total absence in PBLs from patients with mycosis fungoides, inflammatory cutaneous or hematologic diseases, and from healthy volunteers. Only 3 of 4 SS patients did constitutively express T-plas

Regulation of synthesis of the transformation-induced protein, leukocyte plastin, by ovarian steroid hormones.

Previous studies indicated that, among normal cells, only those of hemopoietic lineages expressed the abundant leukocyte phosphoprotein, L-plastin, and that activation of the L-plastin gene frequently occurred in malignant cells of solid tumors. We discovered that the gene encoding L-plastin contains potential estrogen and progesterone response elements upstream from its promoter, suggesting that L-plastin expression is subject to ovarian steroid regulation. To determine if L-plastin synthesis is regulated by ovarian steroids (estrogens and progestins), we examined cultured uterine endometrial

Plastination of biological materials using a vinyl-based polymer as an alternative method for preserving anatomical specimens.

The preservation of anatomical specimens is essential for teaching and research, enabling detailed analysis of tissue morphology and structure. Traditional methods such as formaldehyde fixation have been increasingly questioned due to their toxicity and associated health risks. Plastination offers a safer, more durable alternative, producing dry, odorless, permanently preserved specimens. This study evaluated polyvinyl acetate (PVAc) as an innovative, low-cost, and non-toxic polymer for the plastination of biological tissues. Acetic acid and glycerol were tested as additives, using bovine hear

Investigation of meningomyovertebral structures within the upper cervical epidural space: a sheet plastination study with clinical implications.

BACKGROUND CONTEXT: Over the past two decades, soft-tissue structures communicating with the dura mater within the epidural space have become the focus of many anatomical and histopathologic studies. The relationship between these bridging structures has yet to be evaluated in situ. PURPOSE: This is the first study that used E12 sheet plastination to investigate the epidural space of the upper cervical spine in situ and its associated bridging structures. Given the complexity of this space, this study may prove useful to clinical anatomists and surgeons who operate within this region. STUDY

[Plastination-histological investigations on the inserting extensor pollicis brevis tendon on the proximal phalanx of the thumb].

By help of thick transparent cross-sections of 10 thumbs, manufactured by plastination histology, it has been shown that the most fibres of the extensor pollicis brevis tendon insert on a special tubercle of the basis of the proximal phalanx. Our studies prove that the shape of this tubercle is configurated in the phalanx proximalis longitudinally between the first and second fifth part and in the wideness of the base, seen from radial to ulnar between the forth and fifth tenth part.

A study on the sectional anatomy of the oculomotor nerve and its related blood vessels with plastination and MRI.

To obtain normal images and sectional anatomical data of the oculomotor nerve and its related arteries, the optimal angles and the length of intracisternal segment of the oculomotor nerve were measured on MPR images. Meanwhile, the relationships between the nerve and the basilar, posterior cerebral, superior cerebellar and posterior communicating arteries were observed from plastination slices, original images, MPR and MIP images. MRI revealed similar results to the plastination sections. The intracisternal segment of oculomotor nerve formed an angle with the posterior plane of the brainstem.

Distribution of muscle spindles in the extensor digitorum and hallucis brevis muscles of the macaque as determined by plastination.

The structure-function relationships of the macaque extensor digitorum and hallucis brevis muscle (EDB and EHB), the single intrinsic and four-tendoned muscle of the dorsum of the foot, were investigated in the macaque after preparation of samples by two plastination methods, using gelatin and epoxy resin. These preparative methods clearly revealed the muscle fibers, spindles and myelinated axons in the EDB and EHB. In the more medially located two bellies (the medial two bellies of the EDB), the mean relative proportion of red fibers was larger than in the more laterally located two bellies (

Integrating plastinate-derived three-dimensional scans into computer-based anatomy practical examinations.

Anatomy instruction increasingly incorporates three-dimensional (3D) digital resources; however, the assessment of practical anatomy knowledge often relies on two-dimensional (2D) images. This misalignment may limit the authenticity of computer-based anatomy practical examinations (APEs). We implemented 3D scans of plastinated human specimens in APEs and examined student usage patterns and perceptions. High-resolution 3D scans of plastinated specimens were embedded into APE items. Fifteen anatomy practical questions were analyzed across three consecutive cohorts of first-year medical students.

Plastin 3 rescues cell surface translocation and activation of TrkB in spinal muscular atrophy.

Plastin 3 (PLS3) is an F-actin-bundling protein that has gained attention as a modifier of spinal muscular atrophy (SMA) pathology. SMA is a lethal pediatric neuromuscular disease caused by loss of or mutations in the Survival Motor Neuron 1 (SMN1) gene. Pathophysiological hallmarks are cellular maturation defects of motoneurons prior to degeneration. Despite the observed beneficial modifying effect of PLS3, the mechanism of how it supports F-actin-mediated cellular processes in motoneurons is not yet well understood. Our data reveal disturbed F-actin-dependent translocation of the Tropomyosin

Plastin 1 drives metastasis of colorectal cancer through the IQGAP1/Rac1/ERK pathway.

Tumor metastasis is the dominant cause of death in colorectal cancer (CRC) patients, and it often involves dysregulation of various cytoskeletal proteins. Plastin 1 (PLS1) is an actin-bundling protein that has been implicated in the structure of intestinal epithelial microvilli; however, its role in CRC metastasis has not yet been determined. In this study, we demonstrated that PLS1 is highly expressed in 33.3% (45/135) of CRC patients and is correlated with lymph node metastasis and poor survival. In in vitro and in vivo experiments, PLS1 induced the migration and invasion of CRC cells and th

Plastin 3 is upregulated in iPSC-derived motoneurons from asymptomatic SMN1-deleted individuals.

Spinal muscular atrophy (SMA) is a devastating motoneuron (MN) disorder caused by homozygous loss of SMN1. Rarely, SMN1-deleted individuals are fully asymptomatic despite carrying identical SMN2 copies as their SMA III-affected siblings suggesting protection by genetic modifiers other than SMN2. High plastin 3 (PLS3) expression has previously been found in lymphoblastoid cells but not in fibroblasts of asymptomatic compared to symptomatic siblings. To find out whether PLS3 is also upregulated in MNs of asymptomatic individuals and thus a convincing SMA protective modifier, we generated induced
