Plesiomonas shigelloides as an Emerging Pathogen in Catfish Aquaculture: A Case from a South Texas Commercial Farm.

During the summer of 2023, a spontaneous disease outbreak occurred in intensively stocked hybrid catfish (♀ channel catfish, Ictalurus punctatus × ♂ blue catfish, I. furcatus) in earthen ponds on a commercial aquaculture farm in South Texas. The farmer reported 50 to 80 dead fish per pond daily for a month. The fish were market size (1.0 ± 0.3 kg on average), resulting in substantial economic losses. Fifteen moribund fish were submitted for laboratory examination. Grossly, the fish showed distended abdomens, erythematous fins, and inflamed vents. Autopsy demonstrated visceral congestion, diste

Lipopolysaccharide of Plesiomonas shigelloides O68:H2 (CNCTC 138/92). Expanding the array of P. shigelloides strains with unusual hydrophobic properties of the O-antigens.

The complete structure of lipopolysaccharide from P. shigelloides O68:H2 (strain CNCTC 138/92) has been investigated by NMR spectroscopy, mass spectrometry and chemical methods. It was confirmed that the P. shigelloides O68 O-specific polysaccharide (O-PS) is composed of a trisaccharide repeating-unit (RU) with a new structure: →4)-α-d-Glcp6OAc-(1 → 4)-β-d-GlcpNAcyl3NAc-(1 → 3)-β-FucpNAc4N-(1→ in which a 3-acetamido-2,3-dideoxy-glucosamine was acylated with d-3-hydroxybutyrate. The O-PS is attached to a non-phosphorylated undecasaccharide through →3)-β-d-GlcpNAc-(1 → residue in the outer core

New UDP-GlcNAc C4 epimerase involved in the biosynthesis of 2-acetamino-2-deoxy-L-altruronic acid in the O-antigen repeating units of Plesiomonas shigelloides O17.

Plesiomonas shigelloides is a ubiquitous waterborne pathogen responsible for diseases such as diarrhea and bacillary dysentery, commonly afflicting infants and children. This bacterium is endowed with an O-antigen gene cluster consisting of 10 consecutive reading frames. One of these, designated wbgU (orf3), has been overexpressed and biochemically characterized to show that it encodes a uridine diphosphate-N-acetylglucosamine (UDP-GlcNAc) C4 epimerase, only the second microbial enzyme characterized to have this activity. Epimerization is an equilibrium reaction resulting in a 70:30 ratio of U

Characterisation of Plesiomonas shigelloides strains that share type-specific antigen with Shigella flexneri 6 and common group 1 antigen with Shigella flexneri spp. and Shigella dysenteriae 1.

Three strains of Plesiomonas shigelloides isolated from patients with diarrhoea were agglutinated with Shigella flexneri 6 antiserum in slide and tube tests. All the strains were also agglutinated with a monoclonal antibody to the common group 1 antigen shared between S. flexneri serotypes and S. dysenteriae type 1. Further studies with one strain also showed sharing of antigenicity in an enzyme-linked immunosorbent assay. The results suggest that the strains share type-specific antigen with S. flexneri 6 and the common group 1 antigen with S. flexneri serotypes and S. dysenteriae 1. The shari

Complete lipopolysaccharide of Plesiomonas shigelloides O74:H5 (strain CNCTC 144/92). 1. Structural analysis of the highly hydrophobic lipopolysaccharide, including the O-antigen, its biological repeating unit, the core oligosaccharide, and the linkage between them.

The lipopolysaccharide of Plesiomonas shigelloides serotype O74:H5 (strain CNCTC 144/92) was obtained with the hot phenol/water method, but unlike most of the S-type enterobacterial lipopolysaccharides, the O-antigens were preferentially extracted into the phenol phase. The poly- and oligosaccharides released by mild acidic hydrolysis of the lipopolysaccharide from both phenol and water phases were separated and investigated by (1)H and (13)C NMR spectroscopy, MALDI-TOF mass spectrometry, and sugar and methylation analysis. Prevalence of bacterial pathogens and their anti-microbial resistance in Tilapia and their pond water in Trinidad.

In Trinidad, Tilapia (Oreonchromis spp.) is one of the most important fresh water food fish and the number of farms has been increasing annually. A study was conducted in the local tilapia industry to determine the microbial quality of pond water, prevalence of bacterial pathogens and their anti-microbial resistance using the disk diffusion method. Seventy-five apparently healthy fish and 15 pond water samples from three of the four commercial tilapia fish farms in the country were processed. The 202 bacterial isolates recovered from fish slurry and 88 from water, belonged to 13 and 16 genera
