Reproductive stress response in male black molly Poecilia sphenops: Evidence for receptor specific regulation by GABA.

Gamma-aminobutyric acid (GABA) is a primary inhibitory neurotransmitter involved in several physiological functions, including reproduction in vertebrates. However, its role in stress-induced reproductive modulation in viviparous teleosts remains unclear. This study examined the receptor-specific GABAergic mechanisms influencing gonadal activity in response to stress in the black molly Poecilia sphenops. Four experiments were conducted. In the first experiment, fish were subjected to chronic intermittent stress, whereas in the second experiment, the effect of gabazine (a GABAA receptor antagon

Cholinergic alterations by exposure to pesticides used in control vector: Guppies fish (Poecilia reticulta) as biological model.

Spinosad and temephos are two of the most used pesticides in Mexico for the control of vector causing disease such as dengue, chikungunya and Zika. The aim of this study was to compare the neurotoxic effects of these two pesticides using guppy fish (Poecilia reticulata) as a model organism. Guppies were exposed for 7 and 21 days to technical grade temephos and spinosad at 1.0 and 0.07 g/L, respectively, (10 and 0.5 mg/L of active substance; concentrations recommended by the Ministery of Health of the State (Secretaría de Salud de Nayarit (SSN) Mexico)). Subsequently, acetylcholinesterase activ

Unique evolutionary trajectories in repeated adaptation to hydrogen sulphide-toxic habitats of a neotropical fish (Poecilia mexicana).

Replicated ecological gradients are prime systems to study processes of molecular evolution underlying ecological divergence. Here, we investigated the repeated adaptation of the neotropical fish Poecilia mexicana to habitats containing toxic hydrogen sulphide (H2 S) and compared two population pairs of sulphide-adapted and ancestral fish by sequencing population pools of >200 individuals (Pool-Seq). We inferred the evolutionary processes shaping divergence and tested the hypothesis of increase of parallelism from SNPs to molecular pathways. Coalescence analyses showed that the divergence occu

GABAA and GABAB Receptor Agonists Promote the Hypothalamic-Pituitary-Ovarian Axis in the Postfertilization Model Poecilia sphenops.

Although gamma-aminobutyric acid (GABA) is known to play a role in the regulation of several physiological functions in vertebrates, the role of GABA in regulating the reproductive axis in viviparous teleosts remains opaque. This study examined the effects of GABAA and GABAB receptor agonists, muscimol (MUSC) and baclofen (BAC), on the hypothalamic-pituitary-ovarian (HPO) axis in the black molly Poecilia sphenops. A dose-dependent upregulation of gonadotropin-releasing hormone-I (GnRH-I) mRNA expression in the preoptic hypothalamus was observed in 0.5 and 5 µg MUSC-treated fish compared to exp

Phylogenetic characteristics, virulence properties and antibiogram profile of motile Aeromonas spp. isolated from ornamental guppy (Poecilia reticulata).

Aeromonas spp. are opportunistic pathogenic bacteria related to an assembly of infectious diseases in ornamental fish. In the present study, virulence properties and antibiotic susceptibility of 52 guppy-borne Aeromonas spp. were investigated. The isolates were identified as A. veronii (n = 34), A. dhakensis (n = 10), A. hydrophila (n = 3), A. caviae (n = 3) and A. enteropelogenes (n = 2) by gyrB gene sequencing. The gyrB sequence deviation within and among the species ranged from 0 to 2.6% and 2.7-9.2%. Each species formed a distinct group in the unrooted neighbor-joining phylogenetic tree. T

Detection of changes in mitochondrial hydrogen sulfide i n vivo in the fish model Poecilia mexicana (Poeciliidae).

In this paper, we outline the use of a mitochondria-targeted ratiometric mass spectrometry probe, MitoA, to detect in vivo changes in mitochondrial hydrogen sulfide (H2S) in Poecilia mexicana (family Poeciliidae). MitoA is introduced via intraperitoneal injection into the animal and is taken up by mitochondria, where it reacts with H2S to form the product MitoN. The MitoN/MitoA ratio can be used to assess relative changes in the amounts of mitochondrial H2S produced over time. We describe the use of MitoA in the fish species P. mexicana to illustrate the steps for adopting the use of MitoA in

Genome-scale data reveal that endemic Poecilia populations from small sulphidic springs display no evidence of inbreeding.

Populations with limited ranges can be highly vulnerable to changes in their environment and are, thus, of high conservation concern. Populations that experience human-induced range reductions are often highly inbred and lack genetic diversity, but it is unknown whether this is also the case for populations with naturally small ranges. The fishes Poecilia sulphuraria (listed as critically endangered) and Poecilia thermalis, which are endemic to small hydrogen sulphide-rich springs in southern Mexico, are examples of such populations with inherently small habitats. We used geometric morphometri

Ependymal cells of the filum terminale in fish (Poecilia sphenops) adapted to freshwater and saltwater: electron microscopic study.

The ependymal lining of the central canal of the filum terminale and spinal cord in the vicinity of the caudal neurosecretory system in P. sphenops was examined in this study. Two general cell types based on shape and location were observed in the ependymal lining: cuboidal ependyma located in dorsal aspects of the filum terminal and columnar to pseudostratified ependymal cells found in ventrolateral and ventral aspects of the filum terminale. Comparison of the ependymal lining was made in animals adapted to saltwater and freshwater. In animals adapted to saltwater there was an increase in the

Female Choice Undermines the Emergence of Strong Sexual Isolation between Locally Adapted Populations of Atlantic Mollies (Poecilia mexicana).

Divergent selection between ecologically dissimilar habitats promotes local adaptation, which can lead to reproductive isolation (RI). Populations in the Poecilia mexicana species complex have independently adapted to toxic hydrogen sulfide and show varying degrees of RI. Here, we examined the variation in the mate choice component of prezygotic RI. Mate choice tests across drainages (with stimulus males from another drainage) suggest that specific features of the males coupled with a general female preference for yellow color patterns explain the observed variation. Analyses of male body colo

Associations between trace metals in sediment, water, and guppy, Poecilia reticulata (Peters), from urban streams of Semarang, Indonesia.

The present study aims to answer the question whether the amount of metal in aquatic biota reflects the concentrations in the sediment and water, and whether the physico-chemical properties of the water and sediment have any influence on the suspected relationship. A study was made of 101 small streams in the city of Semarang, Central Java, Indonesia. Data on fish occurrence in 63 streams of the greater Semarang indicated that the guppies did not avoid the highly polluted sites. No significant difference in body weight between sites was found. Significant differences were found in metal body c

A genome-wide association study for the red-eyed trait (ocular albinism) in an F2 population of guppy (Poecilia reticulata).

Guppy (Poecilia reticulata) is a popular tropical ornamental fish with high phenotypic diversity. This study investigates the genetic basis of the red-eyed trait (ocular albinism) in guppy through hybridization between the sky-blue strain (red-eyed) and the red-fun strain (black-eyed). Our results demonstrated black eye to be the dominant trait relative to red eye, and that the ratio of the number of black-eyed guppies to that of red-eyed ones in the F2 generation is consistent with 3:1, which indicates that the red-eyed trait in the sky-blue strain guppies conforms to the model of Mendelian m

Gyrodactylus in the spotlight: how exposure to light impacts disease and the feeding behavior of the freshwater tropical guppy (Poecilia reticulata).

Artificial light at night (ALAN) negatively impacts organisms in many ways, from their feeding behaviors to their response and ability to deal with disease. Our knowledge of ALAN is focused on hosts, but we must also consider their parasites, which constitute half of all described animal species. Here, we assessed the impact of light exposure on a model host-parasite system (Poecilia reticulata and the ectoparasitic monogenean Gyrodactylus turnbulli). First, parasite-free fish were exposed to 12:12 h light:dark (control) or 24:0 h light:dark (ALAN) for 21 days followed by experimental infectio

Molecular characterization and confocal laser scanning microscopic study of Pygidiopsis macrostomum (Trematoda: Heterophyidae) parasites of guppies Poecilia vivipara.

Pygidiopsis macrostomum and Ascocotyle (Phagicola) pindoramensis (Digenea: Heterophyidae) parasitize guppies as intermediate hosts and, respectively, fish-eating mammals or birds as definitive hosts. Heterophyids have zoonotic potential, and molecular studies associated with morphological and ecological aspects have helped to clarify their taxonomy and phylogeny. Poecilia vivipara naturally parasitized by metacercariae of both species (100% prevalence) exhibit no external signs of parasitism. In this work, four new sequences of P. macrostomum (18S rDNA, 28S rDNA and ITS2 rDNA) and one new sequ

Differences in Salinity Performance Curves between Two Closely Related Poecilia Species: Implications for Species Turnover along a Salinity Gradient.

AbstractMany locomotor traits exhibit predictable variation along environmental gradients, which in turn influence the lower-level cellular and physiological processes that determine whole-organism performance (i.e., performance curves). While the impact of salinity on locomotor performance has been previously studied, few have investigated the ecological consequences of salinity-induced changes in performance. On the island of Trinidad, Poecilia reticulata are confined to freshwater, while the closely related Poecilia picta primarily inhabit lowland brackish water and saltwater. However, both

Histology of Liver Vascular-Biliary Tracts and Associated Exocrine Pancreas in Guppies (Poecilia reticulata) at Three Post-Hatch Ages.

To describe the structural development of the liver in young guppies (Poecilia reticulata) and complement the adult characterisation, we conducted a histological study at 7, 17, and 28 days post-birth. We used serial sections to trace the organisation of blood vessels, bile ducts, and pancreatic tissue within and around the liver. At all ages, the liver was highly cellular and composed mainly of hepatocytes. Pigmented melanomacrophage centres or aggregates were not observed, whereas rodlet cells were rare and restricted to extrahepatic connective tissue. At 7 days, we identified four distinct

Interaction between female mating preferences and predation may explain the maintenance of rare males in the pentamorphic fish Poecilia parae.

Variation in mating preferences coupled with selective predation may allow for the maintenance of alternative mating strategies. Males of the South American live-bearing fish Poecilia parae fall in one of five discrete morphs: red, yellow, blue, stripe-coloured tail (parae) and female mimic (immaculata). Field surveys indicate that the red and yellow morphs are the rarest and that their rarity is consistent across years. We explored the role of variable female mating preference and selective predation by visual predators in explaining the rarity of red and yellow males, and more generally, the

Co-exposure of iron oxide nanoparticles and glyphosate-based herbicide induces DNA damage and mutagenic effects in the guppy (Poecilia reticulata).

Iron oxide nanoparticles (IONPs) have been tested to remediate aquatic environments polluted by chemicals, such as pesticides. However, their interactive effects on aquatic organisms remain unknown. This study aimed to investigate the genotoxicity and mutagenicity of co-exposure of IONPs (γ-Fe2O3 NPs) and glyphosate-based herbicide (GBH) in the fish Poecilia reticulata. Thus, fish were exposed to citrate-functionalized γ-Fe2O3 NPs (0.3 mg L-1; 5.44 nm) alone or co-exposed to γ-Fe2O3 NPs (0.3 mg L-1) and GBH (65 and 130 μg of glyphosate L-1) during 14 and 21 days.
