Second-Generation Polyhydroxybutyrate Production From Pretreated Corn Lignocellulosic Fraction as a Strategy to Repurpose by Products From the Corn-Processing Chain.

Lignocellulosic residues are non-cost, non-edible, renewable feedstocks that can be used in polyhydroxybutyrate (PHB) production. PHB is a bioplastic synthetized by microorganisms, formed intracellularly under stressful growth conditions; however, its production costs are high, representing the major drawback for fossil-based plastics replacement. The use of lignocellulosic waste in PHB production can help to reduce its costs. Therefore, this work aimed to optimize the second-generation (2G) PHB production process from the lignocellulosic corn fraction (LCF). The LCF is a lignocellulosic by-pr

A Coupled Adsorption-Biodegradation (CAB) Process Employing a Polyhydroxybutyrate (PHB)-Biochar Mini Pilot-Scale Reactor for Trichloroethylene-Contaminated Groundwater Remediation.

Actions for improving water quality are critical and include the remediation of polluted groundwater. The effectiveness of the remediation strategy to remove contamination by chlorinated solvents may be increased by combining physicochemical treatments (i.e., adsorption) and biological degradation (i.e., biological reductive dechlorination (BRD)). Recent studies have shown the potentialities of bio-based materials for bioremediation purposes, including polyhydroxybutyrate (PHB), a biodegradable microbial polyester tested as a fermentable source of slow-release electron donors. Further, a low-c

Polyhydroxybutyrate (PHB) production from sugar cane molasses and tap water without sterilization using novel strain, Priestia sp. YH4.

The production cost of biodegradable polymer like polyhydroxybutyrate (PHB) is still higher than that of petroleum-based plastics. A potential solution for reducing its production cost is using a cheap carbon source and avoiding a process of sterilization. In this study, a novel PHB-producing microbial strain, Priestia sp. YH4 was screened from the marine environment using sugarcane molasses as the carbon source without sterilization. Culture conditions, such as carbon, NaCl, temperature, pH, inoculum size, and cultivation time, were optimized for obtaining the highest PHB production by YH4 re

A closed-loop biorefinery approach for polyhydroxybutyrate (PHB) production using sugars from carob pods as the sole raw material and downstream processing using the co-product lignin.

A novel closed-loop biorefinery model using carob pods as the feed material was developed for PHB production. The carob pods were delignified, and as the second step, sugars present in the delignified carob pods were extracted using water. Ralstonia eutropha and Bacillus megaterium were cultivated on the carob pod extract and its performance was evaluated using Taguchi experimental design. R. eutropha outperformed the B. megaterium in terms of its capability to grow at a maximum initial sugar concentration of 40 g L-1 with a maximum PHB production of 12.2 g L-1. Finally, the concentrated ligni

Bio Polyhydroxybutyrate /Fiber castor Oil Hybrid Composite (CO-BPFHC) as adsorbent for simultaneous removal of Pb(II), Cd(II), and Cr(VI) from wastewater: mechanistic insights and thermodynamic evaluation.

Simultaneous contamination of industrial effluent discharges with Pb, Cd, and Cr(VI) poses a critical threat to the environment due to their toxicological profile, nonbiodegradability, and potential for causing cancer. Herein, a bio polymer/fiber hybrid composite (CO-BPFHC) derived from castor oil and prepared via isophoronediisocyanate (IPDI)-mediated urethane polymerization on a scaffold of alkali treated sisal/jute bi fibers is introduced for the simultaneous adsorptive removal of Pb(II), Cd(II), and Cr(VI) from contaminated industrial wastewater. Extensive characterization of CO-BPFHC was

Production of Bioplastic Compounds by Genetically Manipulated and Metabolic Engineered Cyanobacteria.

Direct conversion of carbon dioxide to valuable compounds is a desirable way to reduce the environmental burden and switch from fossil to renewable fuels. Cyanobacteria are photosynthetic bacteria that perform oxygenic photosynthesis and are able to produce valuable compounds from carbon dioxide in the air. Synechocystis and Synechococcus species, model unicellular cyanobacteria, can produce succinate and lactate, which are commodity chemicals used to generate bioplastics. Several cyanobacteria are also able to produce polyhydroxybutyrate, a biodegradable polyester that accumulates under nitro

Rewiring photosynthetic carbon flow: Engineered cyanobacterial factories for sustainable carbohydrate production and carbon-negative biomanufacturing.

Cyanobacteria are promising platforms for light-driven carbon fixation and carbohydrate biosynthesis. However, optimization strategies that focus solely on carbon allocation are insufficient to achieve substantial improvements in yield and sustainability. Here, Synechococcus elongatus PCC 7942 was engineered to enhance sucrose production by simultaneously increasing total carbon input and reinforcing the artificial sink. The engineered strain secreted 5.821 g L-1 sucrose, which was 27.4 times higher than the wild-type. Transcriptomic analysis revealed upregulation of abundant genes involved in

The Investigation of the Structure and Properties of Ozone-Sterilized Nonwoven Biopolymer Materials for Medical Applications.

Nowadays, the development and research of nonwoven medical fibrous materials based on biopolymers is an area of a great practical interest. One of the most promising methods for producing nonwoven materials with a highly developed surface is electrospinning (ES). In this article, the possibility of efficient sterilization of ultrathin fibers based on polyhydroxybutyrate (PHB) by ozone treatment was considered. The purpose of this work was to select the most optimal morphology of nonwoven materials for medical purposes and to establish the correlation between the supramolecular structure and th

Biosynthesis and accumulation of poly(3-hydroxybutyrate-co-3-hydroxyvalerate)-polyethylene glycol, a hybrid co-polymer by endophytic Bacillus cereus RCL 02.

Co-polymerization of microbial polyesters, polyhydroxyalkanoates (PHAs), with synthetic polymers has become an established and promising tool in the recent past for improving the material and biological properties of the biopolyesters. Bacillus cereus RCL 02, a leaf endophytic bacterium of the oleaginous plant Ricinus communis L., has been reported to produce a significant amount of poly(3-hydroxybutyrate) [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] under batch cultivation. The present study demonstrates the synthesis and accumulation of poly(3-hydroxybutyrate-co-

Bioprocess performance and COD partitioning for PHA/EPS production from activated sludge fed with lactate, ethanol and citrate.

Lactate-, ethanol- and citrate-rich side streams are increasingly available from waste fermentation and organic-acid industries, yet their effects on carbon allocation in mixed-culture activated sludge remain insufficiently compared under identical conditions. Here, we operated three parallel sequencing batch reactors fed with lactate, ethanol, or citrate and quantified treatment performance together with chemical oxygen demand (COD) partitioning to intracellular polyhydroxyalkanoates (PHA) and extracellular polymeric substances (EPS). All reactors maintained stable removals of COD, total nitr

Protective T Cell and Antibody Immune Responses against Hepatitis C Virus Achieved Using a Biopolyester-Bead-Based Vaccine Delivery System.

Hepatitis C virus (HCV) infection is a major worldwide problem. Chronic hepatitis C is recognized as one of the major causes of cirrhosis, hepatocellular carcinoma, and liver failure. Although new, directly acting antiviral therapies are suggested to overcome the low efficacy and adverse effects observed for the current standard of treatment, an effective vaccine would be the only way to certainly eradicate HCV infection. Recently, polyhydroxybutyrate beads produced by engineered Escherichia coli showed efficacy as a vaccine delivery system. Here, an endotoxin-free E. coli strain (ClearColi) w

Quantitative determination of release kinetics from fibrous poly(3-hydroxybutyrate) scaffolds.

A fibrous scaffold was prepared from poly(3-hudroxybutirate) (PHB) by wet spinning from chloroform solution. The fibers were precipitated into excess ethanol and they had a regular, cylindrical shape. The diameter of the fibers was regulated by the rate of spinning. A model drug, quercetin, was dissolved in the fiber in various amounts. The rate of drug release could be controlled by the adjustment of fiber diameter. A novel approach is proposed in the paper for the quantitative analysis of release kinetics. The model is based on Fick's laws, and does not use any simplification or introduce em

Value-added utilisation of industrial by-products from bioenergy processes for growth of the PHB synthesising bacterium Cupriavidus necator.

The utilisation of low-value by-products derived from the production of bioenergy as nutrient media for microbial growth was investigated using the polyhydroxybutyrate (PHB) synthesising bacterium Cupriavidus necator as a case study. The potentially suitable by-products crude glycerol (derived from biodiesel production as carbon source) and digestate (resulting from biogas production as source for nitrogen and other nutrients) contain significant amounts of nutrients but also dissolved and solid impurities. In order to minimise negative effects such as clogging in bioreactor infrastructure and

Enhanced aromatics production via catalytic pyrolysis of poly(3-hydroxybutyrate) using ZSM-5 catalysts.

The valorization of biodegradable plastics into high-value chemicals offers a sustainable pathway for plastic waste management. In this study, catalytic pyrolysis of poly(3-hydroxybutyrate) (PHB), a representative bioplastic, was systematically investigated over ZSM-5 zeolites with varying degrees of mesoporosity, aiming to maximize the BTEX yield. Catalyst characterization confirmed that mesopore introduction increased external surface area and accessibility of acid sites, while relatively reducing strong Brønsted acidity. Catalytic performance was evaluated, as well as the effects of differe

Harnessing the potential of Cupriavidus necator for CO2 capture from alcoholic fermentation and its bioconversion into poly(3-hydroxybutyrate).

The fermentation process in alcoholic beverage production converts sugars into ethanol and CO2, releasing significant amounts of greenhouse gases. Here, Cupriavidus necator DSM 545 was grown autotrophically using gas derived from alcoholic fermentation, using a fed-batch bottle system. Nutrient starvation was applied to induce intracellular accumulation of poly(3-hydroxybutyrate) (PHB), a bioplastic polymer, for bioconversion of CO2-rich waste gas into PHB. Grape marc, another by-product of wine production, was evaluated as a low-cost carbon source for the heterotrophic growth of C. necator, w
