Poly(D,L)lactide and polylactide-co-glycolide drug-loaded microspheres were prepared with lipopholic (bupivacaine and etidocaine) and hydrophilic (mepivacine and lidocaine) local anaesthetics. Formulations of drug-loaded microspheres were characterized by the drug content, the in-vitro release kinetics and by the physical state of the drug within the microspheres. Release rates of the local anaesthetics from the microspheres were different and could not be accounted for by the intrinsic dissolution rates of the drugs. The encapsulation efficiency was highly dependent on the lipophilicity of th

Polymeric nanoparticles (NPs) are a versatile delivery platform for non-viral genetic therapies. However, a key shortcoming of polymeric NPs (and other non-viral delivery vehicles) is the often-high accumulation of NPs within the liver and the spleen after systemic intravenous (IV) administration in vivo. This phenomenon is largely the result of the mononuclear phagocytic system (MPS), a class of phagocytic cells responsible for native immune response and toxin clearance within the body. One strategy to overcome NP clearance by the MPS is the use of phagocytic modulating pre-treatments to inte

Poly(lactic-co-glycolic acid) (PLGA) microspheres are a clinically established platform for sustained and controlled drug delivery, offering tunable degradation and reduced dosing frequency. This review critically appraises PLGA microsphere technology, linking materials science with clinical translation. We examine how molecular parameters-molecular weight, lactide-to-glycolide ratio, and terminal group chemistry-affect degradation and release kinetics. Key fabrication methods (emulsion-solvent evaporation, spray drying, membrane emulsification, and microfluidics) are compared for their contro

Poly(lactic-co-glycolic acid) (PLGA) is an FDA-approved, biodegradable, and biocompatible polymer, which makes it a promising starting material for the development of nanoparticles. However, in vivo studies have revealed a short biological half-life due to recognition and consequently internalization of these nanoparticles by cells of the mononuclear phagocyte system, resulting in their accumulation in the liver and spleen. In this study, we analyzed the adsorption pattern of proteins on PLGA nanoparticles after incubation with human plasma and human serum. For this analysis, different nanopar

The oral route is the most patient-friendly option for drug administration, yet biological barriers often limit its effectiveness. Chief among these is the mucus layer along the gastrointestinal (GI) tract, which restricts the transport of drugs and carriers. Strategies such as mucolytics, mucus-inert materials, and anisotropic nanosystems have been employed to enhance penetration. We developed urease-powered poly(lactic-co-glycolic acid) (PLGA) nanomotors for drug delivery, featuring either random (isotropic) or spatially localized (anisotropic, Janus-like) urease surface functionalization. Autologous adipose tissue is commonly used for tissue engraftment for the purposes of soft tissue reconstruction due to its relative abundance in the human body and ease of acquisition using liposuction methods. This has led to the adoption of autologous adipose engraftment procedures that allow for the injection of adipose tissues to be used as a "filler" for correcting cosmetic defects and deformities in soft tissues. However, the clinical use of such methods has several limitations, including high resorption rates and poor cell survivability, which lead to low graft volume retention and inc

AIMS: The poly(lactic-co-glycolic) acid (PLGA) nanoparticles of tubercular drugs have been demonstrated to have a sustained release profile over 7 days. There is no information on the location or mode of release of these nanoparticles in living systems. Therefore, we have planned the study to explore the pharmacokinetics and biodistribution of PLGA rifampicin nanoparticles in healthy human volunteers. We aim to study the distribution pattern of PLGA-loaded nano-formulation of radiolabelled rifampicin in humans.
METHODS: Rifampicin was labelled with 99m Tc by indirect method and nanoparticles

Poly-(lactic-co-glycolic) acid (PLGA) is a biodegradable, biosafe, and biocompatible copolymer. The Aspergillus section Nigri causes otomycosis localized in the external auditory canal. In this research, Aspergillus brasiliensis, a species belonging to the Nigri section, was tested. Coumarin 6 and pterostilbene loaded in poly-(lactic-co-glycolic) acid nanoparticles (PLGA-coumarin6-NPs and PLGA-PTB-NPs) were tested for fungal cell uptake and antifungal ability against A. brasiliensis biofilm, respectively. Moreover, the activity of PLGA-PTB-NPs in inhibiting the A. brasiliensis infection was te

Due to the excellent safety profile of poly (D,L-lactide-co-glycolide) (PLGA) particles in human, and their biodegradability, many studies have focused on the application of PLGA particles as a controlled-release vaccine delivery system. Antigenic proteins/peptides can be encapsulated into or adsorbed to the surface of PLGA particles. The gradual release of loaded antigens from PLGA particles is necessary for the induction of efficient immunity. Various factors can influence protein release rates from PLGA particles, which can be defined intrinsic features of the polymer, particle characterist

Bydureon® is a once-weekly injection of poly(lactide-co-glycolide) (PLGA) microspheres containing exenatide acetate, a synthetic analog of the GLP-1 receptor agonist exendin-4. These microspheres are formulated by coacervation (i.e., phase separation), using a single-emulsion method. There remains a knowledge gap between how formulation variables affect product attributes and performance. We aimed to bridge this gap by evaluating the effect of formulation variables on encapsulating exenatide in PLGA microspheres at similar compositions to Bydureon®. We first screened process variables without

Peripheral nerve injury is one of the most debilitating pathologies that severely impair patients' life. Although many efforts have been made to advance in the treatment of such a complex disorder, successful strategies to ensure full recovery are still scarce. The aim of the present work was to develop flexible and mechanically resistant platforms intended to act as a support and guide for neural cells during the regeneration process of peripheral nerve injury. For this purpose, poly(lactic-co-glycolic acid) (PLGA)/poly(d,l-lactic acid) (PDLLA)/poly(ethylene glycol) 400 (PEG)-multichannel-bas

The development of biodegradable antifouling coatings represents a promising route to environmentally compatible marine protection. However, most polyester-based systems exhibit limited interfacial evolution and a poor long-term biofouling resistance. Here, we report poly(lactic-co-glycolic acid) (PLGA)-based polyurethane (PU) coatings that bear covalently tethered eugenol (EU) side groups (PLGA-PU-EUx), designed to couple hydrolytic degradation with interfacial bioactivity. Compared with PLA-based analogues, the PLGA-PU-EUx coatings show greater mass loss, lower water contact angles during se

Intrinsic hemostasis is an innate body response to prevent bleeding based on the sol-gel transition of blood. However, it is often inadequate for exceptional situations, such as acute injury and coagulation disorders, which typically require immediate medical intervention. Herein, we report the preparation of an efficient hemostatic powder, composed of tannic acid (TA), poly(ethylene glycol) (PEG), and poly(d,l-lactide-co-glycolide)-b-poly(ethylene glycol)-b-poly(d,l-lactide-co-glycolide) triblock copolymer (TB), for biomimetic hemostasis at the bleeding sites. TA has a high affinity for biomo

BACKGROUND: Myelomeningocele (MMC) is a devastating congenital neurologic disorder that can lead to lifelong morbidity and has limited treatment options. This study investigates the use of poly(lactic-co-glycolic acid) (PLGA) microparticles (MPs) loaded with fibroblast growth factor (FGF) as a platform for in utero treatment of MMC.
STUDY DESIGN: Intra-amniotic injections of PLGA MPs were performed on gestational day 17 (E17) in all-trans retinoic acid-induced MMC rat dams. MPs loaded with fluorescent dye (DiO) were evaluated 3 hours after injection to determine incidence of binding to the MM

The degradation time is a crucial factor in evaluating the performance of poly (lactic-co-glycolic acid) (PLGA) stents. Bulk degradation mode was commonly used to analyze the stent degradation behavior by finite element approach. However, the PLGA stents may present surface degradation more than bulk degradation under certain conditions, which will greatly affect the degradation time after implantation. In this study, the degradation processes of the poly (lactic-co-glycolic acid) stent were reproduced utilizing finite element analysis. Both bulk degradation and surface degradation modes were

Ionizing radiation is routinely used for sterilization of medical and biomedical products. The most commonly used radiation type for this purpose, gamma rays, features several drawbacks related to the high energy of the radiation, such as being restricted to dedicated external facilities. Low energy electron irradiation (LEEI) is an alternative type of irradiation, suitable for sterilization with significantly lower material penetration, allowing in-process implementation with smaller shielding, but also requiring more involved sample preparation. In this study, we investigated the effect of c

Osteoarthritis (OA) is an inflammatory disease accompanied by synovial joint inflammation, and IL-36 plays an important role in this process. Local application of IL-36 receptor antagonist (IL-36Ra) can effectively control the inflammatory response, thereby protecting cartilage and slowing down the development of OA. However, its application is limited by the fact that it is rapidly metabolized locally. We designed and prepared a temperature-sensitive poly(lactic-co-glycolic acid)-poly(ethylene glycol)-poly(lactic-co-glycolic acid) (PLGA-PEG-PLGA) hydrogel (IL-36Ra@Gel) system carrying IL-36Ra

For the first time, the interaction of the Poly lactic-co-glycolic acid (PLGA) and Chitosan (CH) with Zirconium dioxide (ZrO2) nanotube was studied using density functional theory (DFT). The binding energies of the most stable configurations of PLGA and CH monomers absorbed on ZrO2 were calculated using density functional theory (DFT) methods. The obtained results indicate that both CH and PLGA monomers were chemisorbed on the surface of ZrO2. The interaction between PLGA and ZrO2 is stronger than that of CH due to its shorter equilibrium interval and higher binding energy. In addition, the el
