Diverse Evolutionary Histories Characterise Polyploidy in the Deep-Sea Brittle Star Genus Ophiosabine.

Polyploidy is rare in animals, where it is often associated with unusual modes of reproduction. Here we use phased exon capture data to identify ploidy and investigate the subgenome composition of four polyploid taxa in the deep-sea brittle star genus Ophiosabine (Echinodermata: Ophiuroidea). Of the four, O. anomala and O. vivipara are hybrid allopolyploids, the latter particularly complex comprising at least three progenitor lineages. The other two undescribed taxa are better regarded as autopolyploids. While at first the four appeared to be phylogenetically distinct, closer analysis reveals

Harnessing polyploidy for climate-resilient crops: Lessons from the evolutionary model, allotetraploid cotton.

Escalating pressures of global climate change necessitate developing agricultural systems and crop varieties with enhanced resilience. Polyploidy, the state of possessing multiple complete sets of chromosomes arising from whole genome duplication (WGD), is a major evolutionary force in plants, often conferring novel genetic and regulatory capacities that facilitate adaptation. Allotetraploid cotton (Gossypium spp.), which formed through the merger of distinct A and D subgenomes approximately 1 to 1.6 Mya, is an exemplary model for elucidating polyploid genome evolution and molecular mechanisms

Aneuploidy, polyploidy, and loss of heterozygosity distinguish serial bloodstream isolates of Candida albicans.

BACKGROUND: The opportunistic pathogen Candida albicans is the leading species causing invasive Candida infections worldwide. Genomic variation is widespread in clinical isolates and complicates identification of genetic variants underlying antifungal drug resistance and tolerance. Our understanding of genomic and phenotypic diversity during invasive infections is limited and studies of serial isolates from individual patients are uncommon. We performed comparative analyses of 101 C. albicans bloodstream isolates from 55 patients in the Minnesota Minneapolis-Saint Paul region, including serial

Regional specialization, polyploidy, and seminal fluid transcripts in the Drosophila female reproductive tract.

Sexual reproduction requires the choreographed interaction of female cells and molecules with sperm and seminal fluid. In internally fertilizing animals, these interactions are managed by specialized tissues within the female reproductive tract (FRT), such as a uterus, glands, and sperm storage organs. However, female somatic reproductive tissues remain understudied, hindering insight into the molecular interactions that support fertility. Here, we report the identification, molecular characterization, and analysis of cell types throughout the somatic FRT in the premier Drosophila melanogaster

Polyploidy Spectrum Correlates with Immunophenotype and Shapes Hepatocellular Carcinoma Recurrence Following Liver Transplantation.

PURPOSE: Patients receiving liver transplantation (LT) for hepatocellular carcinoma (HCC) are at high risk of tumor recurrence. Polyploidy is a fascinating characteristic of the liver and correlates with HCC development and progression. This study aims to investigate the association between hepatocyte polyploidy spectrum and HCC recurrence after LT. PATIENTS AND METHODS: Thirty-two paired HCC, peritumoral, cirrhotic, and normal liver specimens were employed to examine the hepatocyte polyploidization pattern during liver tumorigenesis. Clinicopathological implications of polyploidy spectrum fo

Disaggregating polyploidy, parental genome dosage and hybridity contributions to heterosis in Arabidopsis thaliana.

Heterosis is the phenomenon whereby hybrid offspring of genetically divergent parents display superior characteristics compared with their parents. Although hybridity and polyploidy can influence heterosis in hybrid plants, the differential contributions of hybridity vs polyploidy to heterosis effects remain unknown. To address this question, we investigated heterosis effects on rosette size and growth rate of 88 distinct F1 lines of Arabidopsis thaliana consisting of diploids, reciprocal triploids and tetraploids in isogenic and hybrid genetic contexts. 'Heterosis without hybridity' effects o

The Dynamic Fungal Genome: Polyploidy, Aneuploidy and Copy Number Variation in Response to Stress.

Fungal species have dynamic genomes and often exhibit genomic plasticity in response to stress. This genome plasticity often comes with phenotypic consequences that affect fitness and resistance to stress. Fungal pathogens exhibit genome plasticity in both clinical and agricultural settings and often during adaptation to antifungal drugs, posing significant challenges to human health. Therefore, it is important to understand the rates, mechanisms, and impact of large genomic changes. This review addresses the prevalence of polyploidy, aneuploidy, and copy number variation across diverse fungal

A pangenome-based approach reveals genes associated with polyploidy and apomixis in Eragrostis curvula.

Eragrostis curvula is a forage grass species in which multiple cytotypes coexist and shifts between sexual and apomictic reproduction are common. This study investigates genetic variation among these cytotypes in relation to ploidy levels and reproductive modes. To this end, a panel of accessions was assembled to capture the species' intraspecific diversity. The panel was used to construct a whole-genome sequencing-based linear pangenome by iteratively mapping Illumina short reads and de novo assembling unmapped reads, thereby expanding the available reference genome and improving the represen

Histological diagnosis of polyploidy discriminates an aggressive subset of hepatocellular carcinomas with poor prognosis.

BACKGROUND: Although genome duplication, or polyploidization, is believed to drive cancer evolution and affect tumor features, its significance in hepatocellular carcinoma (HCC) is unclear. We aimed to determine the characteristics of polyploid HCCs by evaluating chromosome duplication and to discover surrogate markers to discriminate polyploid HCCs. METHODS: The ploidy in human HCC was assessed by fluorescence in situ hybridization for multiple chromosomes. Clinicopathological and expression features were compared between polyploid and near-diploid HCCs. Markers indicating polyploid HCC were

Low Temperature and Polyploidy Result in Larger Cell and Body Size in an Ectothermic Vertebrate.

Previous studies reported that low temperatures result in increases in both cell size and body size in ectotherms that may explain patterns of geographic variation of their body size across latitudinal ranges. Also, polyploidy showed the same effect on body size in invertebrates. In vertebrates, despite their having larger cells, no clear effect of polyploidy on body size has been found. This article presents the relationship between temperature, cell size, growth rate, and body size in diploid and polyploid hybridogenetic frog Pelophylax esculentus reared as tadpoles at 19° and 24°C. The size

Polyploidy in myelofibrosis: analysis by cytogenetic and SNP array indicates association with advancing disease.

BACKGROUND: Myelofibrosis occurs as primary myelofibrosis or as a late occurrence in the evolution of essential thrombocythaemia and polycythaemia vera. It is the rarest of the three classic myeloproliferative neoplasms (MPN). Polyploidy has only rarely been reported in MPN despite the prominent involvement of abnormal megakaryocytes. The use of peripheral blood samples containing increased numbers of haematopoietic progenitors has improved the output from cytogenetic studies in myelofibrosis and together with the use of single nucleotide polymorphism arrays (SNPa) has contributed to an improv

Polyploidy influences sexual system and mating patterns in the moss Atrichum undulatum sensu lato.

BACKGROUND AND AIMS: Evolutionary transitions between separate and combined sexes have frequently occurred across various plant lineages. In mosses, which are haploid-dominant, evolutionary transitions from separate to combined sexes are often associated with genome doubling. Polyploidy and hermaphroditism have strong effects on the inbreeding depression of a population, and are subsequently predicted to affect the mating system. METHODS: We tested the association between ploidy (haploid, diploid or triploid gametophytes) and mating system in 21 populations of Atrichum undulatum sensu lato, w

Domain-autonomous MKLP1 modules divergently regulate mitotic fidelity and polyploidy in a holocentric system.

While the kinesin MKLP1 is a well-established regulator of cytokinesis, whether its individual domains possess autonomous functions has remained unexplored. Here, using the holocentric Bombyx mori BmN4 system, we show that isolated head, stalk, and tail domains of MKLP1 exhibit distinct subcellular localization patterns and exert divergent control over mitosis. These domains differentially regulate cell-cycle progression, with the head and stalk inducing G2/M arrest, whereas the tail, and to a lesser extent the head, promotes polyploidization. Despite these divergent outcomes, all three domain

Polyploidy in germ cell of brown bear Ursus arctos (Ursidae, Mammalia): A case report.

In this work, we present the first detailed study of the normal meiotic karyotype of the brown bear based on the analysis of synaptonemal complexes. Microphotographs of the key stages of meiotic prophase I were obtained. The level of meiotic recombination, features of XY sex bivalent assembly during meiotic prophase I were analyzed. A refined SC karyotype ideogram was constructed, consisting of 37 pachytene bivalents: 36 autosomal bivalents and and XY sex bivalent. We report a rare finding in mammalian germinal cells - a polyploid spermatocyte after genome duplication in the brown bear Ursus a

Effects of polyploidization and their evolutionary implications are revealed by heritable polyploidy in the haplodiploid wasp Nasonia vitripennis.

Recurrent polyploidization occurred in the evolutionary history of most Eukaryota. However, how neopolyploid detriment (sterility, gigantism, gene dosage imbalances) has been overcome and even been bridged to evolutionary advantage (gene network diversification, mass radiation, range expansion) is largely unknown, particularly for animals. We used the parasitoid wasp Nasonia vitripennis, a rare insect system with heritable polyploidy, to begin addressing this knowledge gap. In Hymenoptera the sexes have different ploidies (haploid males, diploid females) and neopolyploids (diploid males, tripl

Fructose Causes Liver Damage, Polyploidy, and Dysplasia in the Setting of Short Telomeres and p53 Loss.

Studies in humans and model systems have established an important role of short telomeres in predisposing to liver fibrosis through pathways that are incompletely understood. Recent studies have shown that telomere dysfunction impairs cellular metabolism, but whether and how these metabolic alterations contribute to liver fibrosis is not well understood. Here, we investigated whether short telomeres change the hepatic response to metabolic stress induced by fructose, a sugar that is highly implicated in non-alcoholic fatty liver disease. We find that telomere shortening in telomerase knockout

Flow cytometric studies of the survival of cytochalasin-induced polyploidy in Chinese hamster ovary cells.

A method has been developed to estimate the post-irradiation survival of cytochalasin B-induced polyploidization of adherent Chinese hamster ovary cell using the flow cytometer. After exposure to radiation, surviving cells are allowed to become polyploid in the presence of cytochalasin, and are detached using trypsin, fixed by the addition of glutaraldehyde and stained using mithramycin. DNA content distributions are polymodal, and the absolute number of cells per culture in any given ploidy class is estimated by reference to a non-fluorescent bead internal standard, detected using forward sca

[Proliferation and polyploidy of hepatocytes as indicators of liver regeneration after exposure to environmental chemical factors].

The study revealed that a comprehensive assessment of proliferation and hepatocyte polyploidy serves as an integral indicator of compensator liver regeneration and its resistance. The development of reparative regeneration is correlated with the accumulation of the significant amount of destructive cells and is regarded as an adequate reaction to the damaging effect of toxic agents. Thus it is possible to consider reparative regeneration as an indicator of hepatotoxic effect under hygienic regulation and to justify its examination under the effect of environmental hygienic factors.
