A comparative genomic analysis of two distant diptera, the fruit fly, Drosophila melanogaster, and the malaria mosquito, Anopheles gambiae.

Genome evolution entails changes in the DNA sequence of genes and intergenic regions, changes in gene numbers, and also changes in gene order along the chromosomes. Genes are reshuffled by chromosomal rearrangements such as deletions/insertions, inversions, translocations, and transpositions. Here we report a comparative study of genome organization in the main African malaria vector, Anopheles gambiae, relative to the recently determined sequence of the Drosophila melanogaster genome. The ancestral lines of these two dipteran insects are thought to have separated approximately 250 Myr, a long

Progress in mapping the yellow fever mosquito genome.

Mosquito-borne diseases cause significant problems for the human health. For this reason, the genomes of three most dangerous species of mosquitoes, including the yellow fever mosquito Aedes aegypti, were sequenced in last decade. The efficient vector of arboviruses. Ae. aegypti, is also a convenient model for laboratory research. The intensive genetic mapping of morphological and molecular markers conducted for this mosquito in the past was very successful. This mapping was also used as a tool to localize a number of quantitative trait loci related to the mosquito's ability to transmit variou

Chromosome rearrangement patterns of an SD chromosome (SDKona-2) in Drosophila melanogaster caused by hybrid dysgenesis.

The types and frequencies of spontaneous chromosome rearrangements caused by hybrid dysgenesis were studied in a second chromosome autosome of Drosophila melanogaster. This second chromosome, being an SD chromosome, had two important advantages over other autosomes for this study: (i) it had the two inversions characteristic of a standard SD-72 chromosome type, which distinguished it from its homolog in polytene chromosome spreads, and (ii) because of the meiotic drive associated with the segregation distorter system, it was preferentially transmitted to the next generation. The chromosome mut

Cloning, mapping and tissue-specific expression of Drosophila clathrin-associated protein AP50 gene.

The Drosophila homologue of AP50, the medium chain of clathrin-associated protein complex AP-2, was identified and characterized from the Drosophila Expressed Sequence Tag database. The Drosophila AP50 is 86% identical to that of mouse and human, and 80% identical to the Caenorhabditis elegans homologue. It is a single-copy gene with two mini-introns in the coding region and it maps to position 94B1-B2 on polytene chromosomes. Two P1 clones, DS01102 and DS0104, were identified that contain the AP50 gene. Alternative 5' UTR splicing is involved in the regulation of AP50 expression. AP50 express

Biased visibility in Hi-C datasets marks dynamically regulated condensed and decondensed chromatin states genome-wide.

BACKGROUND: Proximity ligation based techniques, like Hi-C, involve restriction digestion followed by ligation of formaldehyde cross-linked chromatin. Distinct chromatin states can impact the restriction digestion, and hence the visibility in the contact maps, of engaged loci. Yet, the extent and the potential impact of digestion bias remain obscure and under-appreciated in the literature. RESULTS: Through analysis of 45 Hi-C datasets, lamina-associated domains (LADs), inactive X-chromosome in mammals, and polytene bands in fly, we first established that the DNA in condensed chromatin had les

Partial-arm translocations in evolution of malaria mosquitoes revealed by high-coverage physical mapping of the Anopheles atroparvus genome.

BACKGROUND: Malaria mosquitoes have had a remarkable stability in the number of chromosomes in their karyotype (2n = 6) during 100 million years of evolution. Moreover, autosomal arms were assumed to maintain their integrity even if their associations with each other changed via whole-arm translocations. Here we use high-coverage comparative physical genome mapping of three Anopheles species to test the extent of evolutionary conservation of chromosomal arms in malaria mosquitoes. RESULTS: In this study, we developed a physical genome map for Anopheles atroparvus, one of the dominant malaria

A molecular mechanism for the stage specificity of the Drosophila prepupal genetic response to ecdysone.

Two successive pulses of ecdysone signal the ends of larval and prepupal development in Drosophila, inducing early and late puffs in the salivary gland polytene chromosomes. Early puff induction in prepupae is dependent on a preceding period of protein synthesis and low ecdysone concentration. We demonstrate here that the competence acquired during this interval can be provided by beta FTZ-F1, a nuclear hormone receptor superfamily member derived from the 75CD mid-prepupal puff. We show that beta FTZ-F1 represses its own transcription and is repressed by ecdysone, explaining its brief expressi

Biodiversity of Simulium metallicum sensu lato (Diptera: Simuliidae), a complex of Neotropical vectors associated with human onchocerciasis.

The polytene chromosomes of 130 larvae of the Neotropical Simulium metallicum complex from Brazil, Costa Rica, and Ecuador revealed five cytoforms, including three ('M', 'N', and 'O') that are new and two ('B' and 'J') that represent range extensions of up to 850km. The discovery of three new cytoforms brings the total number in the complex to 17. Cytoforms 'B', 'J', and 'N' are reproductively isolated from one another, and their species status is corroborated by morphological evidence. None of the three new cytoforms is known from current or historical onchocerciasis foci, although 'M' inhabi

The Tryptophan oxygenase gene of Anopheles gambiae.

The Anopheles gambiae gene encoding tryptophan oxygenase, a homolog of the Drosophila melanogaster vermilion gene, has been molecularly cloned and characterized. Unlike Drosophila, where it is X-linked, the A. gambiae gene maps to chromosome 2R, subdivision 12E, by in situ hybridization to the polytene chromosomes. Of the six introns present, four are positioned identically to those of the Drosophila homolog, one is similarly positioned, and one is novel. A 1 955 nt cDNA potentially encodes a 392 amino acid protein of an estimated 45 kDa. Amino acid comparisons between the deduced protein and

Molecular identification of onchocerciasis vector sibling species in black flies (Diptera: Simuliidae).

A number of sibling species in the Simulium damnosum species complex transmit River Blindness (onchocerciasis) in tropical Africa. Black fly sibling species can be reliably recognized only through analysis of the larval polytene chromosomes. The routine, unambiguous assignment of the potentially disease-transmitting adult females to a particular species is virtually impossible. Two molecular assays to identify the sibling species of biting female flies are presented. The first relies on PCR-mediated amplification of internal transcribed spacers of the nuclear rDNA loci while the second involve

Molecular genetic mapping of three X-linked avirulence genes, vH6, vH9 and vH13, in the Hessian fly.

Three X-linked avirulence genes, vH6, vH9, and vH13 in the Hessian fly, Mayetiola destructor, confer avirulence to Hessian fly resistance genes H6, H9, and H13 in wheat. We used a combination of two- and three-point crosses to determine the order of these genes with respect to each other, the white eye mutation and three X-linked molecular markers, G15-1, 020, and 021, developed from genomic lambda clones, lambda G15-1, lambda 020, and lambda 021. The gene order was determined to be vH9-vH6-G15-1-w-vH13-020-021. In situ hybridization of lambda G15-1, lambda 020, and lambda 021, on the polytene

Chromosome-Level Genome of Simulium vittatum Links Black Fly Cytogenetics to Genome Organization and Evolution.

Black flies (Diptera: Simuliidae) are important vectors of pathogens affecting human and animal health, yet the absence of a chromosome-level nuclear reference genome has constrained molecular and evolutionary studies of the family. Here, we present the first chromosome-level nuclear genome and a developmental transcriptomic resource for the long-established IS-7 laboratory lineage of Simulium vittatum . Combining Oxford Nanopore long-read sequencing with Hi-C scaffolding, we assembled a 340.4-Mb genome, with 99.1% of the assembly resolved into three chromosome-length scaffolds (N50=104.8 Mb,

Distribution of B52 within a chromosomal locus depends on the level of transcription.

Drosophila B52 protein is a homologue of human ASF/SF2 that functions in vitro as an essential pre-mRNA splicing factor. Immunofluorescence analysis of polytene chromosomes has shown that B52 generally colocalizes with RNA polymerase II; however, in contrast to other splicing factors, B52 brackets RNA polymerase II at highly active heat-shock puffs. Also, UV cross-linking in nonpolytene cells has shown that B52 cross-links in vivo to DNA flanking the highly active transcription units. Here, we find that the distribution of cross-linked B52 at heat-shock loci depends on transcription levels. He

A note on the maternal effect mutants daughterless and abnormal oocyte in Drosophila melanogaster.

Two deficiencies for, and a dominant enhancer of, the second chromosome maternal effect mutant, "daughterless" (da), were induced with X-irradiation. Their properties were studied with respect to both da and the linked maternal effect mutant, "abnormal oocyte" (abo), with the following conclusions. (1) The most probable map positions of da and abo are: J-(1/2)-da-2(1/2)-abo, where J is a dominant marker located at 41 on the standard map. (2) The da locus is in bands 31CD-F on the polytene chromosome map; abo is to the right of 32A. (3) Because homozygous da individuals survive while individual

A systematic review: is Anopheles vagus a species complex?

BACKGROUND: Anopheles vagus (subgenus Cellia) has been identified as a vector for malaria, filariasis, and Japanese encephalitis in Asia. Sporozoites of Plasmodium falciparum and Plasmodium vivax have been found in this zoophilic mosquito in Asia and Indonesia. This study systematically reviews publications regarding An. vagus species, variation, bio-ecology, and malaria transmission in various localities in Asia, especially Indonesia, to determine whether the current data support An. vagus as a species complex. METHODS: The databases Pubmed, Scopus, Europe PMC, and Proquest were searched to

Cytological and isoenzyme analysis of the Bucay and Quevedo cytotypes of the Onchocerciasis vector Simulium exiguum (Diptera: Simuliidae) in Ecuador.

Four cytotypes of Simulium exiguum occur in Ecuador, where this morphospecies is the primary vector of onchocerciasis. In this paper, we give the first full description of the banding pattern of the larval polytene chromosomes of the Quevedo cytotype and assess its degree of genetic separation from the Bucay cytotype. Both cytotypes differ from the chromosomal standard sequence (of the Cayapa cytotype) by the fixed inversions IIL-5 and IIL-6. The Quevedo cytotype additionally differs from the standard and Bucay cytotypes by possessing a differentiated X chromosome, which is indicated by the in

A three-locus system of interspecific incompatibility underlies male inviability in hybrids between Drosophila buzzatii and D. koepferae.

In hybrids between the sibling species D. buzzatii and D. koepferae, both sexes are more or less equally viable in the F1. However, backcross males to D. buzzatii are frequently inviable, apparently because of interspecific genetic incompatibilities that are cryptic in the F1. We have performed a genetic dissection of the effects of the X chromosome from D. koepferae. We found only two cytological regions, termed hmi-1 and hmi-2, altogether representing 9% of the whole chromosome, which when introgressed into D. buzzatii cause inviability of hybrid males. Observation of the pattern of asynapsi

Chromosomal identification of eight species of the subgenus Edwardsellum near and including Simulium (Edwardsellum) damnosum Theobald (Deptera: Simuliidae).

Examinations of the banding pattern of polytene chromsomes from larval salivary glands of the population in west Africa once thought to be a single species, Simulium (Edwardsellum) damnosum, revealed eight species all within the Nile subgroup of the complex. Mapping a quantitative trait locus (QTL) conferring pyrethroid resistance in the African malaria vector Anopheles funestus.

BACKGROUND: Pyrethroid resistance in Anopheles funestus populations has led to an increase in malaria transmission in southern Africa. Resistance has been attributed to elevated activities of cytochrome P450s but the molecular basis underlying this metabolic resistance is unknown. Microsatellite and SNP markers were used to construct a linkage map and to detect a quantitative trait locus (QTL) associated with pyrethroid resistance in the FUMOZ-R strain of An. funestus from Mozambique. RESULTS: By genotyping 349 F2 individuals from 11 independent families, a single major QTL, rp1, at the telom
