Ectomycorrhizas with Paxillus involutus enhance cadmium uptake and tolerance in Populus × canescens.

Ectomycorrhizas (EMs), which are symbiotic organs formed between tree roots and certain fungi, can mediate cadmium (Cd) tolerance of host plants, but the underlying physiological and molecular mechanisms are not fully understood. To investigate EMs mediated Cd tolerance in woody plants, Populus × canescens was inoculated with Paxillus involutus (strain MAJ) to establish mycorrhizal roots. Mycorrhizal poplars and non-mycorrhizal controls were exposed to 0 or 50 μM CdSO4 . EMs displayed higher net Cd(2+) influx than non-mycorrhizal roots. Net Cd(2+) influx was coupled with net H(+) efflux and in

High temperature exposure did not affect induced 2n pollen viability in Populus.

High temperature exposure is widely used as a physical mutagenic agent to induce 2n gametes in Populus. However, whether high temperature exposure affects induced 2n pollen viability remains unknown. To clarify whether high temperature exposure affected the induced 2n pollen viability, 2n pollen induced by 38 and 41 °C temperatures, pollen morphology, 2n pollen germination in vitro, and crossing induced 2n pollen with normal gametes to produce a triploid was, based on observations of meiosis, conducted in Populus canescens. We found that the dominant meiotic stages (F = 56.6, p < .001) and the

Phytotoxicity of chlorinated aliphatics to hybrid poplar (Populus deltoides x nigra DN34).

Effects of a series of chlorinated ethenes and ethanes on hybrid poplar (Populus deltoides x nigra DN34) were assessed in laboratory experiments. Poplar cuttings were grown in sealed reactors with hydroponic solutions and were exposed to a chlorinated solvent for a period of two weeks. Exposure concentrations ranged from 0 to 0.4 mM for perchloroethylene to 0 to 8.4 mM for 1,1-dichloroethane. Effects were assessed by gravimetrically monitoring transpiration and measuring change in cutting mass. The zero-growth concentrations of the chemicals tested were 0.3 mM perchloroethylene, 0.9 mM trichlo

PtoHsfB1 regulates growth and salt response by affecting ABA biosynthesis in Populus tomentosa.

Poplar is an important commercial timber species and a model organism for forest molecular biology. Here, we identified PtoHsfB1, a subgroup B heat shock transcription factor from Populus tomentosa that is homologous to AtHsfB1 and PtrHsfB1, and contains the conserved repression motif '-LFGV-'. To elucidate its function, transgenic poplars overexpressing PtoHsfB1 were generated. Overexpression significantly enhanced growth rate and biomass accumulation. Histological analyses revealed increased cambial cell layers and enlarged phloem caps, indicating a positive role in cambial activity and phlo

Isolation, Characterization, and Pathogenicity of Two Pseudomonas syringae Pathovars from Populus trichocarpa Seeds.

Pseudomonas syringae is a ubiquitous plant pathogen, infecting both woody and herbaceous plants and resulting in devastating agricultural crop losses. Characterized by a remarkable specificity for plant hosts, P. syringae pathovars utilize a number of virulence factors including the type III secretion system and effector proteins to elicit disease in a particular host species. Here, two Pseudomonas syringae strains were isolated from diseased Populustrichocarpa seeds. The pathovars were capable of inhibiting poplar seed germination and were selective for the Populus genus. Sequencing of the ne

Specificity of Herbivore Defense Responses in a Woody Plant, Black Poplar (Populus nigra).

The specificity of woody plant defense responses to different attacking herbivores is poorly known. We investigated the responses of black poplar (Populus nigra) to leaf feeding by three lepidopteran species (Lymantria dispar, Laothoe populi and Amata mogadorensis) and two leaf beetle species (Phratora vulgatissima and Chrysomela populi). Of the direct defenses monitored, increases in trypsin protease inhibitor activity and the salicinoid salicin were triggered by herbivore damage, but this was not herbivore-specific. Moreover, the majority of leaf salicinoid content was present constitutively

UHPLC-ESI/TOFMS determination of salicylate-like phenolic gycosides in Populus tremula leaves.

Associations of salicylate-like phenolic glycosides (PGs) with biological activity have been reported in Salix and Populus trees, but only for a few compounds, and in relation to a limited number of herbivores. By considering the full diversity of PGs, we may improve our ability to recognize genotypes or chemotype groups and enhance our understanding of their ecological function. Here, we present a fast and efficient general method for salicylate determination in leaves of Eurasian aspen that uses ultra-high performance liquid chromatography-electrospray ionization/time-of-flight mass spectrom

Genome-wide analysis of the structural genes regulating defense phenylpropanoid metabolism in Populus.

Salicin-based phenolic glycosides, hydroxycinnamate derivatives and flavonoid-derived condensed tannins comprise up to one-third of Populus leaf dry mass. Genes regulating the abundance and chemical diversity of these substances have not been comprehensively analysed in tree species exhibiting this metabolically demanding level of phenolic metabolism. Here, shikimate-phenylpropanoid pathway genes thought to give rise to these phenolic products were annotated from the Populus genome, their expression assessed by semiquantitative or quantitative reverse transcription polymerase chain reaction (P

Overexpression of AspAT alleviates the inhibitory effects of ammonium on root development in Populus tomentosa.

Ammonium toxicity, resulting from prolonged use of ammonium as the sole nitrogen source, can lead to physiological and morphological disorders, ultimately stunting plant growth. Enhancing ammonium assimilation efficiency has been extensively explored as a strategy to mitigate ammonium toxicity. However, the role of AspAT, a key enzyme in nitrogen assimilation, remains underexplored. This study investigates the function of AspAT in alleviating ammonium toxicity and uncovers the underlying physiological mechanisms. The results show that two Populus AspAT genes, AspAT13 and AspAT15, exhibit the h

Morphological, Transcriptome, and Hormone Analysis of Dwarfism in Tetraploids of Populus alba × P. glandulosa.

Breeding for dwarfism is an important approach to improve lodging resistance. Here, we performed comparative analysis of the phenotype, transcriptome, and hormone contents between diploids and tetraploids of poplar 84K (Populus alba × P. glandulosa). Compared with diploids, the indole-3-acetic acid (IAA) and gibberellin (GA3) contents were increased, whereas the jasmonic acid (JA) and abscisic acid (ABA) contents were decreased in tetraploids. RNA-sequencing revealed that differentially expressed genes (DEGs) in leaves of tetraploids were mainly involved in plant hormone pathways. Most DEGs as

Intraspecific variation of physiological and molecular response to cadmium stress in Populus nigra L.

Little is known about the variability of response to heavy metal stress within tree species, although it could be a key for a better understanding of tolerance mechanisms and for breeding. The aim of the present study was to characterize the natural variation of response to cadmium (Cd) in Populus nigra L. in order to understand the mechanisms of Cd tolerance. For that, two P. nigra genotypes, originating from contrasting environments in northern (genotype 58-861) and southern (genotype Poli) Italy, were exposed to Cd stress in hydroponics for 3 weeks. The effect of stress was estimated by mea

CENL1 expression in the rib meristem affects stem elongation and the transition to dormancy in Populus.

We investigated the short day (SD)-induced transition to dormancy in wild-type hybrid poplar (Populus tremula x P. tremuloides) and its absence in transgenic poplar overexpressing heterologous PHYTOCHROME A (PHYA). CENTRORADIALIS-LIKE1 (CENL1), a poplar ortholog of Arabidopsis thaliana TERMINAL FLOWER1 (TFL1), was markedly downregulated in the wild-type apex coincident with SD-induced growth cessation. By contrast, poplar overexpressing a heterologous Avena sativa PHYA construct (P35S:AsPHYA), with PHYA accumulating in the rib meristem (RM) and adjacent tissues but not in the shoot apical meri

Biotechnological adaptation of high-yielding, gibberellin-overproducing Populus trees to seasonal growth patterns.

Tree growth is central to both terrestrial ecology and the forestry industry. Biotechnology for inducing the overproduction of phytohormones such as gibberellins (GAs) is a powerful tool for improving tree yield and wood properties. Augumentation of GAs in trees in temperate and boreal regions, however, prevents the induction of vegetative dormancy, which results in reduced yield and tree loss over time. Hybrid Populus trees (Populus tremula × P. tremuloides; Ptt) use an internal circadian clock to synchronise their daily metabolism and seasonal growth with predictable light and temperature ch

Genome-wide identification and characterization of novel lncRNAs in Populus under nitrogen deficiency.

Long non-coding RNAs (lncRNAs) have been identified as important regulatory factors of gene expression in eukaryotic species, such as Homo sapiens, Arabidopsis thaliana, and Oryza sativa. However, the systematic identification of potential lncRNAs in trees is comparatively rare. In particular, the characteristics, expression, and regulatory roles of lncRNAs in trees under nutrient stress remain largely unknown. A genome-wide strategy was used in this investigation to identify and characterize novel and low-nitrogen (N)-responsive lncRNAs in Populus tomentosa; 388 unique lncRNA candidates belon

Timing of photoperiodic competency causes phenological mismatch in balsam poplar (Populus balsamifera L.).

Plant phenology is expected to be sensitive to climate warming. In boreal trees, spring flush is primarily temperature driven, whereas height growth cessation and autumn leaf senescence are predominantly controlled by photoperiod. Cuttings of 525 genotypes from the full range of balsam poplar were planted into two common gardens (Vancouver and Indian Head, Canada) at similar latitudes, but with differing winter temperatures and growing seasons. There was clinal variation in spring and, particularly, summer and fall phenology. Bud flush and, despite milder climate, bud set and leaf drop were ea

PalKCH1 and PalKCH2 are required for tension wood formation and gelatinous layer deposition in Populus.

Tension wood (TW) is a graviresponsive xylem tissue exclusive to angiosperm trees that counteracts gravitational stress. Its hallmark is a gelatinous layer (G-layer) in fiber secondary walls, rich in cellulose and low in lignin. Although kinesins mediate microtubule-microfilament crosstalk, their role in G-layer biogenesis during gravistimulation is unclear in woody angiosperms. We studied two kinesin paralogs, PalKCH1 and PalKCH2, in Populus alba var. pyramidalis during TW development. Both belong to the Group I KCH clade, possessing a kinesin motor domain and a calponin-homology (CH) domain.
