Adjuvant activity of PCP-II, a polysaccharide from Poria cocos, on a whole killed rabies vaccine.

Adjuvants are important components of vaccination strategies because they boost and accelerate the immune response. The aim of this study was to investigate the adjuvant activity of PCP-II, a polysaccharide isolated from Poria cocos, together with an inactivated rabies vaccine. The polysaccharide PCP-II was compared with the common veterinary rabies vaccine adjuvant Alhydrogel by co-administration of either adjuvant with the inactivated rabies virus rCVS-11-G to mice via the intramuscular route. Blood samples were collected to determine the virus-neutralizing antibody (VNA) titer and assess ac

A novel DNA topoisomerase inhibitor: dehydroebriconic acid, one of the lanostane-type triterpene acids from Poria cocos.

Traditional Chinese medicinal plants are a treasure house for screening novel inhibitors of DNA polymerases and DNA topoisomerases from mammals; in the present study, nine lanostane-type triterpene acids were found in sclerotium of Poria cocos. Among the nine compounds, only dehydroebriconic acid could potently inhibit DNA topoisomerase II (topo II) activity (IC(50) = 4.6 microM), while the compound moderately inhibited the activities of DNA polymerases alpha, beta, gamma, delta, epsilon, eta, iota, kappa and lambda only from mammals, to similar extents. Another compound, dehydrotrametenonic a

Inflammatory Response and Oxidative Stress as Mechanism of Reducing Hyperuricemia of Gardenia jasminoides-Poria cocos with Network Pharmacology.

Hyperuricemia (HUA) is a metabolic disease, closely related to oxidative stress and inflammatory responses, caused by reduced excretion or increased production of uric acid. However, the existing therapeutic drugs have many side effects. It is imperative to find a drug or an alternative medicine to effectively control HUA. It was reported that Gardenia jasminoides and Poria cocos could reduce the level of uric acid in hyperuricemic rats through the inhibition of xanthine oxidase (XOD) activity. But there were few studies on its mechanism. Therefore, the effective ingredients in G. jasminoides

Feeding Effect and Antioxidant Capacity of Fermentation Broths Based on Strain-Specific Utilization of Poria cocos Polysaccharide Derivatives.

This study investigated the prebiotic properties of the supernatant from gluconic acid-hydrolyzed alkali-soluble Poria cocos polysaccharide (s-GA-PCP). s-GA-PCP directly promoted the proliferation of Bifidobacterium and Lactobacillus strains. Among the s-GA-PCP-utilizing strains, strain L10 (Bifidobacterium pseudolongum) exhibited the broadest feeding capability, as its fermentation broth supported the growth of all six non-utilizing strains. Analyses revealed that L10 produced a more diverse oligosaccharide profile and accumulated higher levels of amino acid and peptide metabolites. Furthermo
