What the pox? Review of poxviruses affecting humans.

The re-emergence of human mpox with the multi-country outbreak and a recent report of borealpox (previously Alaskapox) resulting in one death has heightened awareness of the significance of the Poxviridae family and their zoonotic potential. This review examines various poxviruses affecting humans, with discussion of less commonly encountered Poxviridae members, including pathogenesis, epidemiology, and diagnostic methods. Poxvirus treatment is beyond the intended scope of this review and will not be discussed.

Cross-species transmission and host range genes in poxviruses.

The persistent epidemic of human mpox, caused by mpox virus (MPXV), raises concerns about the future spread of MPXV and other poxviruses. MPXV is a typical zoonotic virus which can infect human and cause smallpox-like symptoms. MPXV belongs to the Poxviridae family, which has a relatively broad host range from arthropods to vertebrates. Cross-species transmission of poxviruses among different hosts has been frequently reported and resulted in numerous epidemics. Poxviruses have a complex linear double-strand DNA genome that encodes hundreds of proteins. Genes related to the host range of poxvi

Adventures with poxviruses of vertebrates.

Because they were the largest of all viruses and could be visualised with a light microscope, the poxviruses were the first viruses to be intensively studied in the laboratory. It was clear from an early date that they caused important diseases of humans and their domestic animals, such as smallpox, cowpox, camelpox, sheeppox, fowlpox and goatpox. This essay recounts some of the early history of their recognition and classification and then expands on aspects of research on poxviruses in which the author has been involved. Studies on the best-known genus, Orthopoxvirus, relate to the use of in

Historical vignette: a life with poxviruses and publishers.

Although I started science investigating the physical anthropology of Australian aboriginals and then spent six years in the Australian Army during the 1939-45 war, largely working on malaria control, the poxviruses have been the focal point of my research--at the bench, in the field, on committees, and in front of my word processor. I have had a relatively short period as a scientist at the bench, just over twenty years out of the sixty years since I graduated. For the last thirty years the pipette has been replaced by the pen and the word processor, and contacts with publishers have become a

Host Range of Carp Edema Virus (CEV) during a Natural Mortality Event in a Minnesota Lake and Update of CEV Associated Mortality Events in the USA.

Mass mortality events of common carp (Cyprinus carpio, carp) associated with carp edema virus (CEV) alone or in coinfections with koi herpesvirus (KHV), is an emerging issue. Despite recent outbreaks of CEV in wild carp populations, the host range of North American species has not been well studied. To that end, we intensively sampled carp (n = 106) and co-habiting native fish species (n = 5 species; n = 156 total fish) from a CEV-suspect mass-mortality event of carp in a small Minnesota lake (Lake Swartout). Additionally, fecal and regurgitant samples (n = 73 each) from double-crested cormora

Divergent poxvirus identified in a non-native black rat from Madagascar.

Non-native rodents serve as bridges between anthropogenic and natural landscapes. They have expanded across the planet alongside humans while bringing competition, predation, and pathogens, such as poxviruses, to naïve ecosystems. Although rodents serve as reservoirs for multiple zoonotic poxviruses, limited research has focused on rodents for identification of unknown poxviruses. Here, we characterized a divergent metagenome-assembled poxvirus, madamurpox virus, from the oral swab of a black rat in southeastern Madagascar. While madamurpox virus shares a phylogenetic relationship with human-i

Continued poxvirus research: From foe to friend.

The eradication of smallpox is one of the greatest medical successes in history. Vaccinia virus was made famous by being the virus used in the live vaccine that enabled this feat. Nearly 40 years on from that success, this prototypical poxvirus continues to empower the exploration of fundamental biology and the potential to develop therapeutics against some of the major causes of death and disease in the modern world.

Modulation of chemokines by poxvirus infections.

Viruses that successfully replicate within the host have devised strategies to subvert or evade the challenges posed by the innate and adaptive immune responses. Many investigators are now beginning to dissect the diverse and complex interactions involving chemokines, chemokine receptors and viral infection. In recent years, much attention has been focused on the role of chemokines in antiviral defense.

Pox virus infection in captive juvenile caimans (Caiman crocodilus fuscus) in South Africa.

Light grey macules developed on the skin and in the mouths of juvenile caimans, (Caiman crocodilus fuscus) (n = 8), kept in the quarantine section of the reptile park at the National Zoological Gardens, Pretoria, Republic of South Africa. The gross, histopathological and ultrastructural features of the lesions were commensurate with pox virus infection. This outbreak closely resembled the disease described elsewhere in 3 juvenile captive caimans.

Monkeypox: another pandemic in the making?

Monkeypox is a zoonotic disease caused by the monkeypox virus, which is a member of the Poxviridae family of viruses. It is transmitted through direct or indirect contact with fluid secretions. Initial symptoms include fever, chills, headache, and malaise, followed by a maculopapular rash that starts on the face and progresses centrifugally. Polymerase chain reaction is the preferred laboratory test for the diagnosis, and management is mostly supportive. The clinical presentation of monkeypox is quite similar to that of another member of the Poxviridae family: smallpox, which wreaked havoc in

Molluscum Contagiosum of the Gluteal Cleft: Observations and Implications for Management in Five Children.

Molluscum contagiosum (MC) is a common, self-limited skin infection caused by a double-stranded DNA virus of the family Poxviridae. Although the morphology of the disease is well described, MC presenting at unusual sites can exhibit atypical morphology, resulting in misdiagnosis. This observational, retrospective case series discusses the novel morphology of MC presenting in the intergluteal cleft of five children seen in the outpatient dermatology clinic of an academic medical center. We present this case series to highlight this novel presentation of MC in children and to emphasize a conserv

Human monkeypox - After 40 years, an unintended consequence of smallpox eradication.

Monkeypox is one of the many zoonotic viruses that belong to the Orthopoxvirus genus of the Poxviridae family with a similar clinical appearance to smallpox. The symptoms of monkeypox include fever, headache, muscle aches, and lymphadenopathy. The transmission of monkeypox occurs from infected animals to humans or through direct contact (sexual or skin-to-skin), respiratory droplets, and clothing such as towels. The incidence of monkeypox is rising drastically over the world. This short communication discusses the causes of the rising monkeypox cases and emphasizes strategies to prevent the sp

The evolutionary biology of poxviruses.

The poxviruses (family Poxviridae) are a family of double-stranded viruses including several species that infect humans and their domestic animals, most notably Variola virus (VARV), the causative agent of smallpox. The evolutionary biology of these viruses poses numerous questions, for which we have only partial answers at present. Here we review evidence regarding the origin of poxviruses, the frequency of host transfer in poxvirus history, horizontal transfer of host genes to poxviruses, and the population processes accounting for patterns of nucleotide sequence polymorphism.

Review of poxvirus: emergence of monkeypox.

This article reviews the different types of poxvirus infections. Smallpox, although eradicated, must continue to be monitored because of the potential risk of accidental or voluntary (by bioterrorism) reintroduction. Monkeypox and cowpox viruses are considered to be emergent today ; their high risk of dissemination is due to the increase in international transport as well as trends for new animals as pets and the loss of vaccinal protection against smallpox. Molluscum contagiosum (molluscipoxvirus) causes mild infections, is particularly frequent in children ; in adults it is a marker of the r

Poxvirus structural biology for application to vaccine design.

The upsurge of mpox (formerly known as monkeypox) in Africa and its global spread highlight the need for improved vaccines. The development of new recombinant vaccines, including mRNA and protein nanoparticles, depends on understanding the biology of poxviruses and selecting the most protective immunogens. Animal studies demonstrate that vaccines need to target the antigens of both infectious forms - the mature virion and the enveloped virion - which display surface proteins responsible for cell entry and cell-to-cell spread, respectively. Although some of these proteins have been shown to ind

Tecovirimat: A journey from discovery to mechanistic insights in poxvirus inhibition.

Tecovirimat (ST-246 or TPOXX) is an antiviral agent developed as part of a U.S. biodefense initiative aimed at addressing Orthopoxvirus infections, including smallpox and mpox. Although smallpox was declared eradicated in 1980, the potential for its reemergence as a biothreat persists due to illegal stockpiling and the possibility of laboratory synthesis. The F13 protein, which plays a critical role in the formation of extracellular viral particles, serves as the primary target for tecovirimat, inhibiting the transition from intracellular mature viruses (IMVs) to intracellular enveloped viruse

Poxvirus Epidemiology.

Illnesses caused by poxviruses have plagued humanity for millennia. The most notable are caused by viruses comprising the orthopoxvirus genus. Smallpox (caused by Variola virus) became the first eradicated disease in 1980. Devotion to smallpox eradication required significant orthopoxvirus research and discovery including vaccine and medical countermeasure development. Here we describe historical and current poxvirus epidemiology, with particular emphasis on orthopoxviruses including Monkeypox virus and Vaccinia virus.

Poxvirus in farmed Nile crocodiles.

In an outbreak of wart-like skin lesions in nine-month-old farmed Nile crocodiles (Crocodylus niloticus) the lesions were especially common on the head and neck but occurred on all parts of the body except the tail. Eosinophilic intracytoplasmic inclusion bodies were present in epithelial cells and further examination of the lesions by transmission electron microscopy revealed the presence of a poxvirus. An autogenous vaccine was produced from material from the lesions and in six vaccinated crocodiles the lesions healed more quickly than in unvaccinated controls.

Natural infections of Vaccinia virus (VACV)-the prototype species of the Orthopoxvirus genus, from the family Poxviridae and subfamily Chordopoxvirinae-cause an occupational emergent zoonotic disease that is primarily associated with the handling of infected dairy cattle. In humans, VACV infection is characterized by skin lesions, primarily on the hands, and accompanied by systemic symptoms such as fever, myalgia, headache, and lymphadenopathy. The diagnosis of VACV is usually performed according to the methods described for other orthopoxviruses. This unit describes the methods utilized to ob

Screening of potential inhibitors against structural proteins from Monkeypox and related viruses of Poxviridae family via docking and molecular dynamics simulation.

Monkeypox virus (MPXV) is an orthopoxvirus which causes zoonotic infection in humans. Even though sporadic cases of this infection are limited to the African continent, but if the infection continues to increase unabated, it can be a cause of serious concern for the human populace. Smallpox vaccination has been in use against monkeypox infection but it only provides mild protection. In the current study, we have screened novel small molecules (estrone fused heterocycles (EH1-EH7)) exhibiting good binding with monkeypox virus protein and related proteins from Poxviridae family of viruses via co
