Medial prefrontal cortex as an action-outcome predictor.

The medial prefrontal cortex (mPFC) and especially anterior cingulate cortex is central to higher cognitive function and many clinical disorders, yet its basic function remains in dispute. Various competing theories of mPFC have treated effects of errors, conflict, error likelihood, volatility and reward, using findings from neuroimaging and neurophysiology in humans and monkeys. No single theory has been able to reconcile and account for the variety of findings. Here we show that a simple model based on standard learning rules can simulate and unify an unprecedented range of known effects in

The role of the ventromedial prefrontal cortex in memory consolidation.

"System-level memory consolidation theory" posits that the hippocampus an initially links the neocortical representations, followed by a shift to a hippocampus-independent neocortical network. With consolidation, an increase in activity in the human subgenual ventromedial prefrontal cortex (vmPFC) has repeatedly been shown. Previously we and others have proposed that this area might link the neocortical representational areas in remote memory, similarly as has been proposed for the rodent anterior cingulate cortex (ACC). Here, we review literature involving the human vmPFC to investigate if th

Understanding the role of the prefrontal cortex in phonological processing.

Lesion studies have demonstrated impairments of specific types of phonological processes. However, results from neuropsychological studies of speech sound processing have been inconclusive as to the role of specific brain regions because of a lack of a one-to-one correspondence between behavioural patterns and lesion location. Functional neuroimaging studies have contributed more detailed information about the involvement of specific brain regions in a wide range of phonological tasks. A framework developed by Hickok and Poeppel to account for these neuropsychological and neuroimaging results

Methylphenidate sensitization is prevented by prefrontal cortex lesion.

Methylphenidate (MPD), also known as Ritalin, is a widely used treatment for Attention Deficit Hyperactivity Disorder. Repeated administration of MPD causes dose-dependent sensitization. MPD binds to dopamine (DA) transporters, and DA, therefore, remain in the synaptic cleft for longer time, resulting in an indirect DA agonist effect. MPD affects neurotransmission in brain regions including the prefrontal cortex (PFC). The mechanisms of sensitization to MPD are not clear. The aim of this study was to investigate the role of prefrontal cortex in effects of acute and chronic MPD administration,

Prefrontal cortex dysfunction in patients with suicidal behavior.

BACKGROUND: Abnormal serotonergic neurotransmission has long been demonstrated in suicidal behavior. The dorsal and median raphe nuclei housing the main serotonergic cell bodies and the prefrontal cortex (PFC), particularly the ventral part innervated by the serotonergic system, have therefore been studied extensively in suicidal behavior research. However, only a few studies have described neuropsychological function impairment in suicidal patients. We investigated PFC-related neuropsychological function in patients with suicidal behavior, separating dorsolateral PFC (DLPFC)- and orbitofronta

Schizophrenia, the prefrontal cortex, and a mechanism of genetic susceptibility.

Information-processing in the prefrontal cortex is abnormal in patients with schizophrenia. From functional neuroimaging and other studies, it appears that neurons of the dorsolateral prefrontal cortex are effectors of core clinical and biological phenomena associated with the illness. Cognitive deficits qualitatively similar to those in patients with schizophrenia are also found in their healthy siblings and other relatives. The evidence for abnormal prefrontal function in healthy siblings suggests that the deficit in information-processing is part of the biology of genetic susceptibility. Th

Sensorimotor remapping drives task specialization in prefrontal cortex.

The prefrontal cortex (PFC) plays a key role in flexible, context-dependent decision-making. Yet, the population-level mechanisms that support this flexibility remain unclear: is there a generalist neural population involved in representing useful information across tasks, or are representations distributed over multiple modules of task-specialized subpopulations? Here, we investigate whether the population structure modularity in medial prefrontal cortex (mPFC) increases with sensorimotor remapping between task rules in three different paradigms. In a paradigm where all stimuli were remapped,

Distributed cell assemblies spanning prefrontal cortex and striatum.

Highly synchronous neuronal assembly activity is deemed essential for cognitive brain function. In theory, such synchrony could coordinate multiple brain areas performing complementary processes. However, cell assemblies have been observed only in single structures, typically cortical areas, and little is known about their synchrony with downstream subcortical structures, such as the striatum. Here, we demonstrate distributed cell assemblies activated at high synchrony (∼10 ms) spanning prefrontal cortex and striatum. In addition to including neurons at different brain hierarchical levels, sur

Prefrontal cortex function in remitted major depressive disorder.

BACKGROUND: Recent models of major depressive disorder (MDD) have proposed the rostral anterior cingulate (rACC) and dorsomedial prefrontal cortex (dmPFC) as nexus sites in the dysfunctional regulation of cognitive-affective state. Limited evidence from remitted-state MDD supports these theories by suggesting that aberrant neural activity proximal to the rACC and the dmPFC may play a role in vulnerability to recurrence/relapse within this disorder. Here we present a targeted analysis assessing functional activity within these two regions of interest (ROIs) for groups with identified vulnerabil

Efficiency and robustness in three cortical areas: frontal pole cortex, dorsolateral prefrontal cortex and orbitofrontal cortex.

The prefrontal cortex in primates has a diverse evolutionary history, characterized by distinct phases of development. Granular regions of the orbital prefrontal cortex (PFo) emerged early in primate evolution, while other granular areas, such as the dorsolateral (PFdl) and polar (PFp) regions, evolved later during anthropoid development. This study explored the functional differences among PFp, PFdl, and PFo in macaque monkeys, focusing on their coding mechanisms, specifically regarding robustness and efficiency. Efficiency was estimated using the 'contrast entropy', defined as the entropy of

Dorsolateral prefrontal cortex to subgenual cingulate anticorrelation predicts subgenual cingulate response from prefrontal stimulation.

OBJECTIVE: Transcranial magnetic stimulation (TMS) of the dorsolateral prefrontal cortex (DLPFC) may achieve its antidepressant effect via transsynaptic activation of the subgenual cingulate (SGC). SGC activation by DLPFC TMS can be quantified by concurrent EEG (TMS-EEG) via source localization, while the connection between the DLPFC and SGC can be quantified by resting state functional connectivity (RSFC). Here, the relationship between SGC source activation and DLPFC-SGC anticorrelation is characterized in depression patients. METHODS: TMS-EEG and fMRI were collected from 42 depression pati

Decreased GABA levels in anterior cingulate cortex/medial prefrontal cortex in panic disorder.

Changes of various brain metabolites including γ-aminobutyric acid (GABA), measured by 1H-magnetic resonance spectroscopy (MRS), have been reported in panic disorder (PD). Deficits in GABA have been implicated in the pathophysiology of PD. Furthermore, it has been suggested that cortical metabolite changes in PD are familial. Eleven PD patients, including five with and six without a PD family history, and eight age- and gender-matched healthy controls without a family history of psychopathology were recruited. Each subject underwent MRS exams and behavioral assessments (resting visual analog a

Dissociating the Role of Dorsolateral Prefrontal Cortex and Ventrolateral Prefrontal Cortex in Cognitive Control in Depression: A Combined HD-tDCS and fNIRS Study.

Major depressive disorder (MDD) is marked by cognitive and affective dysfunctions associated with altered prefrontal cortical activity. While high-definition transcranial direct current stimulation (HD-tDCS) shows promise in modulating these deficits, little is known about the differential effects of targeting specific prefrontal subregions. This study investigated whether HD-tDCS over the dorsolateral (DLPFC) or ventrolateral (VLPFC) prefrontal cortex produces distinct behavioural and neurophysiological effects in patients with MDD, focusing on cognitive control, mood, and functional brain co

Spike prediction on primary motor cortex from medial prefrontal cortex during task learning.

Objectives. Brain-machine interfaces (BMIs) aim to help people with motor disabilities by interpreting brain signals into motor intentions using advanced signal processing methods. Currently, BMI users require intensive training to perform a pre-defined task, not to mention learning a new task. Thus, it is essential to understand neural information pathways among the cortical areas in task learning to provide principles for designing BMIs with learning abilities. We propose to investigate the relationship between the medial prefrontal cortex (mPFC) and primary motor cortex (M1), which are acti

Prefrontal cortex, temporal cortex, and hippocampus volume are affected in suicidal psychiatric patients.

Suicide is a leading cause of death in America, with over 40,000 reported suicides per year. Mental illness is a major risk factor for suicidality. This study attempts to validate findings of volumetric differences from studies on suicidality. Psychiatric inpatients classified as having mildly severe or severe depression were separated into two groups: suicide attempted in the past two months (SA; n=20), non-suicidal control group (DA; n=20); these patients were all depressed and not significantly different for age, gender, race, marital status, education level, anxiety level, and substance ab

Inferior temporal cortex leads prefrontal cortex in response to a violation of a learned sequence.

Primates learn statistical regularities that are embedded in visual sequences, a form of statistical learning. Single-unit recordings in macaques showed that inferior temporal (IT) neurons are sensitive to statistical regularities in visual sequences. Here, we asked whether ventrolateral prefrontal cortex (VLPFC), which is connected to IT, is also sensitive to the transition probabilities in visual sequences and whether the statistical learning signal in IT originates in VLPFC. We recorded simultaneously multiunit activity (MUA) and local field potentials (LFPs) in IT and VLPFC after monkeys w

The role of posterior parietal cortex and medial prefrontal cortex in distraction and mind-wandering.

Distraction reflects a drift of attention away from the task at hand towards task-irrelevant external or internal information (mind-wandering). The right posterior parietal cortex (PPC) and the medial prefrontal cortex (mPFC) are known to mediate attention to external information and mind-wandering, respectively, but it is not clear whether they support each process selectively or rather they play similar roles in supporting both. In this study, participants performed a visual search task including salient color singleton distractors before and after receiving cathodal (inhibitory) transcrania

GABAergic interneurons in local circuits of primate prefrontal cortex.

Cortical GABAergic interneurons exhibit remarkable diversity of morphological, electrophysiological and molecular features, which underlies their specialized roles in circuit functions. Molecular characteristics have proven to be a reasonable starting point in the past for classification, grouping them into parvalbumin (PV), somatostatin (SST), vasoactive intestinal polypeptide (VIP) and "others" (e.g. LAMP5). While Patch-seq studies provide valuable insight into human cortical interneurons, similar results from non-human primates are lagging behind. This is problematic because the studying of

Cell-specific protein expression in Alzheimer's disease prefrontal cortex.

INTRODUCTION: Analyzing the proteomes of different brain cell types is fundamental for understanding the pathophysiology of Alzheimer's disease (AD). However, spatial analysis of these diverse and limited cell populations poses significant challenges. METHODS: The GeoMx Digital Spatial Profiler (DSP) platform was used to analyze protein level in the prefrontal cortex of AD and non-AD brains. The platform interrogated 76 proteins and used immunofluorescence to distinguish between three cell types. RESULTS: Neprilysin (NEP), which promotes amyloid beta degradation, was significantly higher in
