Animal and Human Tissue Models of Vertical Listeria monocytogenes Transmission and Implications for Other Pregnancy-Associated Infections.

Intrauterine infections lead to serious complications for mother and fetus, including preterm birth, maternal and fetal death, and neurological sequelae in the surviving offspring. Improving maternal and child heath is a global priority. Yet, the development of strategies to prevent and treat pregnancy-related diseases has lagged behind progress made in other medical fields. One of the challenges is finding tractable model systems that replicate the human maternal-fetal interface. Animal models offer the ability to study pathogenesis and host defenses in vivo However, the anatomy of the matern

Probiotic-rich fermented milk from Lactiplantibacillus plantarum IIA-1A5: Effects on pregnancy health in the animal model.

Previous studies of Lactiplantibacillus plantarum IIA-1A5 have shown its potential as a probiotic in modulating gut microbiota and providing health benefits; however, its effects during pregnancy remain underexplored. The aim of this study was to assess the safety of fermented milk enriched with L. plantarum IIA-IA5 in pregnant mice. An experimental study was conducted at Universitas Andalas, Padang, Indonesia. Two groups of pregnant mice (Mus musculus L.) were used, each with six mice. The control group received sterilized milk (10 mL/kg body weight (BW)), while the intervention group was giv

Metabolic and Microbial Changes Associated With Diet and Obesity During Pregnancy: What Can We Learn From Animal Studies?

The intestinal microbiota changes throughout pregnancy and influences maternal metabolic adaptations to support fetal growth. Obesity induces alterations to the microbiota that include decreased microbial diversity and shifts in microbial composition, though specific species changes are inconsistent between published studies. In animal models, probiotics and exercise moderate maternal weight gain and partially correct the maternal microbiota. Supplemental Escherichia coli, however, exacerbate maternal obesity during the perinatal period, lending weight to the theory that inflammation-induced g

Higher Dietary Intake of Animal Protein Foods in Pregnancy Is Associated with Lower Risk of Adverse Birth Outcomes.

BACKGROUND: The prevalence of adverse birth outcomes is highest in resource-limited settings such as sub-Saharan Africa. Maternal consumption of diets with adequate nutrients during pregnancy may protect against these adverse outcomes. OBJECTIVES: The objective was to determine the association between maternal dietary consumption of animal source foods (ASFs) and the risk of adverse birth outcomes among HIV-negative pregnant women in Tanzania. METHODS: Using dietary intake data from 7564 HIV-negative pregnant women, we used Poisson regression with the empirical variance (generalized estimati

[Study of animal model of hypertensive disorder complicating pregnancy in pregnant rats stimulated by homocysteine and monosodium glutamate].

OBJECTIVE: To determine whether homocysteine (Hcy) and monosodium glutamate (MSG) could lead to animal model of hypertensive disorder complicating pregnancy and its mechanism. METHODS: Female adult Wistar rats were randomly divided into 4 groups: pregnant control group (PN), pregnant Hcy group (PH), pregnant glutamic acid group (PG) and pregnant Hcy and glutamic acid group (PHG). The rats of each group were injected with Hcy 200 mg/kg or physiological saline every day intraperitoneally and with MSG or 0.9% saline every other day via Hcy injection from the 10th day to the 20th day of pregnancy

Vaping during pregnancy and maternal, fetal, and infant health outcomes: A scoping review of human and animal studies.

BACKGROUND: The prevalence of vaping, defined as the use of e-cigarettes, during pregnancy is increasing, but its effects on maternal, fetal, and infant health remain incompletely understood. Human studies are heterogeneous and largely observational with a primary focus on neonatal and infant outcomes and limited assessment of maternal physiology. In contrast, pre-clinical animal studies suggest potential biological effects across maternal vascular, respiratory, metabolic, and neurodevelopmental systems. A comprehensive mapping of the existing evidence is therefore needed to characterise the s

Air pollutants and adverse pregnancy outcomes: A multi-method integration of Mendelian randomization, meta-analysis, and animal models.

BACKGROUND: Exposure to air pollutants is associated with adverse pregnancy outcomes, but establishing a causal relationship remains challenging. METHODS: We integrated Mendelian randomization (MR) and animal experiments to investigate causality. MR analyses leveraged data from FinnGen consortium and UK Biobank data to assess effects of air pollutants (particulate matter ≤2.5 μm in diameter [PM2.5], particulate matter 2.5-10 μm in diameter [PM2.5-10], and nitrogen oxide [NOx]) on pregnancy complications (preeclampsia, intrauterine fetal growth restriction, gestational hypertension, placental

SAMe, Choline, and Valproic Acid as Possible Epigenetic Drugs: Their Effects in Pregnancy with a Special Emphasis on Animal Studies.

In this review, we discuss the functions and main effects on pregnancy outcomes of three agents that have the ability to induce epigenetic modifications: valproic acid (VPA), a well-known teratogen that is a histone deacetylase inhibitor; S-adenosylmethionine (SAMe), the most effective methyl donor; and choline, an important micronutrient involved in the one methyl group cycle and in the synthesis of SAMe. Our aim was to describe the possible effects of these compounds when administered during pregnancy on the developing embryo and fetus or, if administered postnatally, their effects on the de

Association Between Proximity to Animal Agriculture During Pregnancy and Spontaneous Preterm Birth Among Nulliparous Women in a California Cohort.

BACKGROUND: Environmental exposures during pregnancy may influence risk of spontaneous preterm birth, a serious adverse outcome. Concentrated Animal Feeding Operations (CAFOs) near maternal residences may emit pollutants associated with preterm birth. METHODS: We analyzed n = 811,953 births to nulliparous women in California (2007-2011), including 7487 spontaneous very preterm (20-31 weeks) and 37,026 spontaneous moderate preterm (32-36 weeks) births. Locations of n = 2121 CAFOs were obtained from a near-census of facilities. Using time-to-event and Bayesian methods, we estimated hazard and o

Biological, Behavioral and Physiological Consequences of Drug-Induced Pregnancy Termination at First-Trimester Human Equivalent in an Animal Model.

Given the significant physiological changes that take place during and resulting from pregnancy, as well as the relative absence of such information in relation to pregnancy termination, this study investigated the potential for developing a valid animal model to objectively assess the biological, physiological and behavioral consequences of drug-induced pregnancy termination. Female Long-Evans rats were divided into four groups (n = 19-21/group), controlling for drug [mifepristone (50 mg/kg/3 ml, i.g.)/misoprostol (0.3 mg/kg/ml, i.g.) or vehicle (1% Carboxymethylcellulose Sodium/0.2% Tween® 8

Evaluating the Risks of Systemic Maternal Ivermectin Exposure During Pregnancy in Human and Vertebrate Animals: A Scoping Review.

BACKGROUND: Ivermectin is widely used for the treatment of neglected tropical diseases associated with adverse maternal and fetal outcomes when the infection complicates the pregnancy. However, the United States FDA classification and current distribution protocols limit ivermectin treatment in pregnant women because antepartum safety has not been well-established. OBJECTIVE: We conducted a scoping review to address the question of what is known from both human and vertebrate animal studies about the safety of systemic ivermectin exposure during pregnancy. METHODS: We searched PubMed for adv

Relationship between maternal global nutrient restriction during pregnancy and offspring kidney structure and function: a systematic review of animal studies.

Maternal undernutrition during pregnancy is prevalent across the globe, and the origins of many chronic diseases can be traced back to in utero conditions. This systematic review considers the current evidence in animal models regarding the relationship between maternal global nutrient restriction during pregnancy and offspring kidney structure and function. CINAHL, Cochrane, EMBASE, MEDLINE, and Scopus were searched to November 2017. Preferred Reporting Items for Systematic Reviews and Meta-analysis guidelines were followed, and articles were screened by two independent reviewers. Twenty-eigh

Brain-derived neurotrophic factor/tyrosine kinase B signaling regulates human trophoblast growth in an in vivo animal model of ectopic pregnancy.

Although medical treatment of unruptured ectopic pregnancy using methotrexate has been established, development of more potent and safer medical treatment is needed due to limited indications and side effects of methotrexate. Brain-derived neurotrophic factor (BDNF) signals through its receptor tyrosine kinase B (TrkB) to regulate the growth of malignant trophoblastic, choriocarcinoma cell. We investigated possible involvement of this signaling system in nonmalignant human trophoblast growth in both ectopic and intrauterine pregnancy. Here, we demonstrated the expression of BDNF in syncytiotro

[Linear salpingotomy in the treatment of unruptured tubal pregnancy. Animal experiment study, comparison of 3 laser systems (CO2, Nd:YAG contact, argon)].

The efficacy of three different laser systems for linear salpingotomy was tested in an animal model. For that purpose, the uterus horns of 22 white New Zealand rabbits were incised with the CO2-Laser, the Nd: YAG contact laser and the argon laser using the "bare fiber" technique. The length of uterotomy was between twice and three times of the diameter of the uterine horn. Secondary healing of the incision site was investigated macroscopically (patency, fistulas, formation of adhesions) and microscopically (quantity and quality of the scars) six weeks after the operation. Despite the different

Safety aspects of Chinese herbal medicine in pregnancy-re-evaluation of experimental data of two animal studies and the clinical experience.

INTRODUCTION: Chinese herbal medicine is an increasingly popular worldwide medical therapy which also has an impact in pregnancy. However, the question of its drug safety during pregnancy remains unresolved. Potential problems include teratogenicity, abortion, perinatal toxicity, pre- and postnatal developmental abnormalities, and eventually an increased risk for carcinomas in the offspring. Standard Materia Medica textbooks contain unreliable information when it comes to risks during pregnancy. Wang and co-workers conducted an experimental study (WS) on mice in which they investigated the eff
