To determine the usefulness of the assays of maternal serum alpha-fetoprotein, human placental lactogen, and pregnancy-specific beta 1-glycoprotein in prenatal diagnosis of fetal alcohol syndrome, these proteins were followed in 35 problem drinkers (greater than 100 gm of ethanol weekly) and in 14 abstinent women throughout gestation. BACKGROUND: Mature spermatozoa contain thousands of mRNA transcripts. It has been recently shown that human sperm can deliver RNA into the oocyte, suggesting that mRNAs might have a role before or after fertilization. Human embryos express PSG1 (pregnancy-specific beta-1-glycoprotein 1) and HLA-E (human leukocyte antigen-E), molecules playing a role in implantation and early development. The relationship between the increase in maternal serum concentration of pregnancy-specific beta 1-glycoprotein (SP1) and levels of components of hemostasis system was investigated. The concentrations of SP1 were measured in the blood sera of 234 pregnant women with recurrent spontaneous abortions, in 115 somatically healthy pregnant women and 75 donors. A direct correlation between serum SP1 and levels of components of hemostasis system was shown. Results suggest that disturbances in hemostasis system influence synthesis of some placental proteins, including SP1.

Human placental lactogen (HPL) and beta 1-SP-1, a placental glycoprotein specific for pregnancy, were determined in 177 pregnant women with pre-eclampsia of varying severity. The serum levels of both hormones decrease during pregnancy in concordance to the severity of pre-eclampsia. In women with severe pre-eclampsia more than 50% showed extreme decreases of the serum concentrations of both hormones. Because of the simple methodology of the test, rapid availability of the results and a lower cost, the determination of the beta-1-SP-1 is perferable to the determination of HPL.

Fetal trophoblast populations in ectopic fallopian tube pregnancy have been characterised using immunohistochemical techniques. The distribution of trophoblast membrane antigens, low molecular weight epithelial cytokeratins, epithelial membrane antigen, human chorionic gonadotrophin, human placental lactogen, and pregnancy-specific beta 1-glycoprotein in ectopic tubal pregnancy appears similar to that reported in normal early intrauterine pregnancy. Serum human chorionic gonadotrophin (hCG), pregnancy-specific beta 1-glycoprotein (SP 1) and placental proteins 5 (PP5) levels have been measured by radioimmunoassay in 20 patients (116 samples) with hydatidiform mole, one patient (nine samples) with invasive mole and 10 patients (103 samples) with choriocarcinoma. Measurement of both serum SP1 and hCG are useful in the monitoring of these diseases. The presence of PP5 in hydatidiform mole and its absence in choriocarcinoma support the hypothesis that PP5 is closely associated with the invasive activity of malignant trophoblast.
