A New Approach to Harness Probiotics Against Common Bacterial Skin Pathogens: Towards Living Antimicrobials.

In this study, the potential of certain lactic acid bacteria-classified as probiotics and known to be antimicrobially active against pathogens or food-poisoning microorganisms-was evaluated with respect to their activity against bacterial skin pathogens. The aim of the study was to develop a plaster/bandage for the application of inhibitory substances produced by these probiotics when applied to diseased skin. For this purpose, two Streptococcus salivarius strains and one Lactobacillus plantarum were tested for production of antimicrobials (bacteriocin-like substances) active against Gram-posi

The impact of maternal gestational diabetes mellitus on the microbiome of offspring: A systematic review.

Gestational diabetes mellitus (GDM) adversely affects maternal and offspring health, and the early-life microbiome is implicated in developmental programming. This systematic review quantitatively evaluated the association between maternal GDM and alterations in the gut and oral microbiomes of offspring. We systematically searched PubMed, Web of Science, and Embase (up to August 2025) for eligible studies adopting high-throughput sequencing techniques. Random-effects meta-analysis was performed for alpha diversity metrics, while findings on beta diversity and microbial taxonomy were summarized

ORAL MICROBIOME OF BENNETT'S (NOTAMACROPUS RUFOGRISEUS) AND YELLOW-FOOTED (PETROGALE XANTHOPUS) ROCK WALLABIES AND THE IMPACT OF INTRAORAL DISEASE.

Intraoral disease, including macropod progressive periodontal disease, is one of the leading causes of morbidity and mortality for wallabies under human care. Clinical signs associated with intraoral disease vary, and diagnostic findings can be difficult to interpret without intraoral radiographs or advanced imaging; therefore, this disease process can be challenging to detect in its early stages. Previous studies have investigated the effects of intraoral disease on the normal oral microbiome of various domestic species. Results from these studies demonstrate specific changes to the oral micr

Changes in the composition of the upper respiratory tract microbial community in granulomatosis with polyangiitis.

Dysbiosis¸ i.e. changes in microbial composition at a mucosal interface, is implicated in the pathogenesis of many chronic inflammatory and autoimmune diseases. To assess the composition of the microbial upper respiratory tract (URT) community in patients with granulomatosis with polyangiitis (GPA), we used culture-independent high-throughput methods. In this prospective clinical study, nasal swabs were collected from patients with GPA, patients with rheumatoid arthritis (RA, disease control), and healthy controls. Nasal bacterial taxa were assessed using V3-V4 region 16S rRNA amplicon sequenc

A comprehensive study of intratumor bacteria in clear-cell renal cell carcinoma identifies Cutibacterium acnes as a protector via suppression of macrophage M2 polarization.

UNLABELLED: Intratumor bacteria (ITB) have emerged as key modulators in various malignancies; however, their role in clear-cell renal cell carcinoma (ccRCC) remains unclear and elusive. In this study, we analyzed seven independent cohorts (comprising one 16S rRNA sequencing and six bulk RNA sequencing data sets) encompassing 517 ccRCC and 65 paired adjacent normal tissues. Following a rigorous decontamination pipeline, we found that neither microbial diversity nor compositional profiles differed significantly between cancerous and adjacent normal tissues. Thirteen specific genera-including Str

Azure A as a promising candidate for antimicrobial photodynamic therapy of acne-related bacteria.

Antimicrobial photodynamic inactivation (aPDT) is an effective technique for treating acne vulgaris, due to its efficiency in inactivating multiresistant bacteria and the absence of systemic side effects. This study investigated the potential of the dye Azure A (AZA), belonging to the phenothiazine class, as a photosensitizer (PS) in aPDT to combat the gram-positive bacteria Staphylococcus epidermidis and Cutibacterium acnes, both associated with inflammatory acne vulgaris. The efficiency of AZA was compared to Orthotoluidine Blue (OTB), evaluating the absorbance spectra, singlet oxygen genera

Identification of Cutibacterium modestum in Spondylitis by Metagenomics Analysis.

BACKGROUND/AIM: Identifying pathogens with culture-negative pyogenic spondylitis is difficult. Shotgun metagenomic sequencing is an unbiased and culture-free approach in the diagnosis of infectious diseases. There are, however, a variety of contaminating factors that can confound the precision of metagenomic sequencing. CASE REPORT: In a 65-year-old man suffering from culture-negative L3-5 spondylitis, metagenomics was applied to facilitate the diagnosis. The patient underwent percutaneous endoscopic lumbar discectomy. We applied metagenomic sequencing with a robust contamination-free protoco

Do Melanocytes Have a Role in Controlling Epidermal Bacterial Colonisation and the Skin Microbiome?

In addition to producing melanin to protect epidermal keratinocytes against DNA damage, melanocytes may have important roles in strengthening innate immunity against pathogens. We have developed a functional, pigmented, human full-thickness 3D skin equivalent to determine whether the presence of melanocytes impacts epidermal bacterial growth and regulates the expression of genes involved in the immune response. We introduced primary epidermal melanocytes to construct a 3-cell full-thickness skin equivalent with primary dermal fibroblasts and epidermal keratinocytes. Immunohistochemistry verifi

Intradermal injection of Cutibacterium acnes and staphylococcus: A pustular acne-like murine model.

BACKGROUND: Skin 16S microbiome diversity analysis indicates that the Staphylococcus genus, especially Staphylococcus aureus (S. aureus), plays a crucial role in the inflammatory lesions of acne. However, current animal models for acne do not fully replicate human diseases, especially pustular acne, which limits the development of anti-acne medications. AIMS: The aim is to develop a mouse model for acne, establishing an animal model that more closely mimics the clinical presentation of pustular acne. This will provide a new research platform for screening anti-acne drugs and evaluating the ef

Pathogenic genetic variations of C. acnes are associated with clinically relevant orthopedic shoulder infections.

Many surgeons continue to face the clinical dilemma of interpreting a positive aspiration or unexpected positive Cutibacterium acnes (C. acnes) culture. There are factors that complicate the interpretation of positive cultures including variations in both frequency of false positive cultures and virulence properties. As indices of virulence, hemolytic strains, from previously confirmed clinically infected shoulders, were compared with non-hemolytic isolates determined to be contaminants, by RNA-sequencing (RNA-Seq). Six C. acnes isolates from patients who underwent revision total shoulder arth

Preparation and evaluation the effects of retinoic acid loaded proliposomal nanofibers on microbial biofilm inhibition.

The electrospinning method involves the production of different drug delivery systems using various polymers. The production of proliposomes with electrospinning provides the hybridization of two novel drug delivery systems. Retinoic acid, also known as all-trans retinoic acid (ATRA), is a common and effective drug for acne therapy. This study aimed to prepare ATRA-loaded proliposomal nanofibers and evaluate their effectiveness on microbial biofilm inhibition. Blank and ATRA-loaded proliposomal nanofiber formulations were fabricated in various polyvinylpyrrolidone, phosphatidylcholine and chol

Biofilm of Cutibacterium acnes: a target of different active substances.

BACKGROUND: Acne vulgaris is a chronic inflammatory dermatosis. Cutibacterium acnes plays a crucial role in the acne pathophysiology. Recent works present evidence of C. acnes growing as a biofilm in cutaneous follicles. This development is currently considered one of the leading causes of C. acnes in vivo persistence and resistance to antimicrobials used to treat acne. OBJECTIVE: Our objective was to evaluate the effects of various active compounds (clindamycin, erythromycin, doxycycline, and myrtle extract) on eight distinct, well-characterized strains of C. acnes following their growth in

Spatial Distribution and Functional Impact of Human Scalp Hair Follicle Microbiota.

Human hair follicles (HFs) constitute a unique microbiota habitat that differs substantially from the skin surface. Traditional HF sampling methods fail to eliminate skin microbiota contaminants or assess the HF microbiota incompletely, and microbiota functions in human HF physiology remain ill explored. Therefore, we used laser-capture microdissection, metagenomic shotgun sequencing, and FISH to characterize the human scalp HF microbiota in defined anatomical compartments. This revealed significant compartment-, tissue lineage-, and donor age-dependent variations in microbiota composition. Gr

Skin microbiome modulation induced by probiotic solutions.

BACKGROUND: The skin is colonized by a large number of microorganisms, most of which are beneficial or harmless. However, disease states of skin have specific microbiome compositions that are different from those of healthy skin. Gut microbiome modulation through fecal transplant has been proven as a valid therapeutic strategy in diseases such as Clostridium difficile infections. Therefore, techniques to modulate the skin microbiome composition may become an interesting therapeutic option in diseases affecting the skin such as psoriasis or acne vulgaris. METHODS: Here, we have used mixtures o
