The last 3 decades have been a revolution in the area of sol-gel-derived materials. They can be used to encapsulate biomolecules such as enzymes, antibodies, hormones, and proteins retaining their functional state. Proteins can be immobilized in many ways but it is crucial that they retain their native conformational structure and, therefore, bioactivity. Porous silica gel matrixes with modified surfaces offer unlimited possibilities to control the protein-solid interaction behavior. The bioimmobilization process on sol-gel biomaterials with chemically engineered surface has driven application

De novo proteins represent a challenging frontier at the intersection of evolutionary biology and synthetic biology. Broadly, the term encompasses two distinct categories: naturally evolved de novo proteins, which arise from previously non-coding DNA, and synthetically designed de novo proteins, which are created from scratch through computational and experimental methods. This review provides an overview of the definitions, history, functional significance, and investigation methods for both categories. By examining potential challenges in current de novo protein research, this review highlig

beta-Glucuronidase (GUSB) is a lysosomal enzyme that could be developed as a brain therapy for Type VII Mucopolysaccharidosis. However, GUSB does not cross the blood-brain barrier (BBB). To enable BBB transport of the enzyme, human GUSB was re-engineered as a fusion protein with the chimeric monoclonal antibody (MAb) to the human insulin receptor (HIR). The HIRMAb crosses the BBB on the endogenous insulin receptor, and acts as a molecular Trojan horse to ferry into brain the GUSB. The 611 amino acid GUSB was fused to either the carboxyl or amino terminus of the heavy chain of the HIRMAb. This

Protein recognition mimicry is of great interest in the field of molecular bioengineering and rational design, with mutagenesis frequently employed to analyze the effects of altering amino acids involved in molecular recognition. The conformational and energetic effects of such alterations can be investigated in detail with the help of molecular dynamics (MD) methodologies. While existing MD-based computational tools can be used to explore a particular set of mutations at a time, suitable for small-scale studies, high-throughput (HTP) exploration of protein recognition for engineering purposes

Applying unconstrained generative protein models to fixed structural scaffolds can produce systematic design artifacts, including glycine enrichment, grammar violations, and discordant complex predictions. We developed DARPinMPNN, a scaffold-constrained computational framework that restricts production sequence generation to permitted DARPin Greek Grammar residue pools. A state-aware chimeric multiple sequence alignment strategy supported AlphaFold2-Multimer (AF2) screening, followed by complementary AlphaFold 3 (AF3) evaluation. Of 15,000 generated variants targeting mesothelin, VEGF-A, and H

The glomerular filtration barrier poses a significant challenge for circulating proteins, with molecules below ~60-70 kDa facing rapid renal clearance. Endogenous proteins have evolved sophisticated evasion mechanisms including oligomerization, carrier binding, electrostatic repulsion, and FcRn-mediated recycling. Understanding these natural strategies provides blueprints for engineering therapeutic proteins with improved pharmacokinetics. This review examines how endogenous proteins resist filtration, evaluates their application in protein engineering, and discusses clinical translation inclu

Targeted protein degradation (TPD) has revolutionized the therapeutic landscape of cancer therapy. TPD aids in avoiding cellular malfunction by recruiting the protein of interest (POI) into intracellular degradation machinery. The proteolysis-targeting chimeras (PROTACs) degradation hijacks the protein's disposal system and co-opts ubiquitin E3 ligase to degrade disease-associated proteins. Numerous PROTACs have come to the forefront in cancer therapy, and several are in clinical trials. PROTACs are on the verge of U.S. Food and Drug Administration (FDA) approval. However, the low bioavailabil

Human immunodeficiency virus (HIV) is an enveloped virus with a remarkable capacity for genetic diversification, enabling rapid escape from host immune responses and therapeutic interventions. Despite extensive global efforts, the development of an effective vaccine has remained elusive owing to the virus's high genetic variability and antigenic diversity. Consequently, considerable effort has been directed toward the development of therapeutic agents targeting viral entry, reverse transcriptase, integrase, protease, and more recently, capsid. Although antiretroviral therapy (ART) remains the

Glycosylation is an effective means to alter the structure and properties of plant compounds, influencing the pharmacological activity of natural products (NPs) to obtain highly active NPs. In nature, glucosides are the most widely distributed, while other glycosides such as xylosides are less common and present in lower quantities. This is due to the scarcity of xylosyltransferases with substrate promiscuity in nature, and the modification of their catalytic function is also quite challenging. In this study, we first performed a phylogenetic analysis of reported UDP-glycosyltransferases (UGTs

The emerging advance of targeted protein degradation (TPD) technology offers a novel option for protein modulation, ensuring a more durable and precise therapeutic impact for cancer management. While promising, complete degradation of the proteins of interest (POIs) in both pathological and normal tissues may cause severe side effects. Furthermore, insufficient accumulation of the protein degraders at the target tissues also limits the clinical translation of TPD. It remains an unmet need to achieve spatiotemporally tunable degradation of the POI at the tumor lesion. In recent years, our group

Surgical sealants have been used for sealing or reconnecting ruptured tissues but often have low adhesion, inappropriate mechanical strength, cytotoxicity concerns, and poor performance in biological environments. To address these challenges, we engineered a biocompatible and highly elastic hydrogel sealant with tunable adhesion properties by photocrosslinking the recombinant human protein tropoelastin. The subcutaneous implantation of the methacryloyl-substituted tropoelastin (MeTro) sealant in rodents demonstrated low toxicity and controlled degradation. All animals survived surgical procedu

The cardiac sarcomere is the functional unit for myocyte contraction. Ordered arrays of sarcomeric proteins, held in stoichiometric balance with each other, respond to calcium to coordinate contraction and relaxation of the heart. Altered sarcomeric structure-function underlies the primary basis of disease in multiple acquired and inherited heart disease states. Hypertrophic and restrictive cardiomyopathies are caused by inherited mutations in sarcomeric genes and result in altered contractility. Ischemia-mediated acidosis directly alters sarcomere function resulting in decreased contractility

Conventional intake of drugs and active substances is most often based on oral intake of an appropriate dose to achieve the desired effect in the affected area or source of pain. In this case, controlling their distribution in the body is difficult, as the substance also reaches other tissues. This phenomenon results in the occurrence of side effects and the need to increase the concentration of the therapeutic substance to ensure it has the desired effect. The scientific field of tissue engineering proposes a solution to this problem, which creates the possibility of designing intelligent sys

Cytokines play a crucial role in cell communication and immunity, making them interesting potential therapeutics for immune-mediated conditions. However, cytokine therapeutics' clinical translation is hampered by their short blood half-lives and unfavorable biodistribution, resulting in toxicity and poor pharmacokinetics. In this study, we present a strategy to improve cytokines' pharmacokinetic profile by engineering fusions of apolipoproteins and cytokines, which are formulated into apolipoprotein-based nanoparticles (cytokine-aNPs). After establishing chemical and recombinant fusion approac

To engineer biotherapeutics that can specifically target glioma, a highly lethal and recurrent type of brain tumor, we created a class of recombinant chimeric proteins containing multiple functional modules, such as a blood-brain barrier (BBB)-penetrating domain and an immune checkpoint blockade (ICB) agent, PD-L1 nanobody, through fusion protein engineering. The recombinant proteins that could prolong circulation time, cross the BBB, target brain tumors, penetrate cell membranes, and release PD-L1 nanobody were demonstrated. Further, by covalently linking doxorubicin (DOX), an inducer of immu

The human body may suffer multiple injuries and losses due to various external factors, such as tumors, diseases, traffic accidents, and war conflicts. Under such circumstances, engineered protein-based materials, as an innovative adjunctive material, can not only effectively promote the natural repair process of tissues, but also greatly circumvent the negative effects and complications that may be associated with conventional surgery. In this review, we first trace the definition and development of engineered protein-based materials and explain in detail their mechanism of action in promotin

BACKGROUND: Effective cancer vaccines require delivery platforms that can simultaneously enhance immune responses, maintain safety, and simplify the formulation process. This study aimed to develop a novel vaccine delivery system based on the Staphylococcus aureus-derived formyl peptide receptor-like 1 inhibitory protein (FLIPr), which naturally targets Fcγ receptors on antigen-presenting cells.
METHODS: A recombinant variant, rF9R, was engineered by adding nine arginine residues to the C-terminus of FLIPr to facilitate electrostatic binding with anionic components. To optimize this interacti

Formaldehyde is an attractive one-carbon building block for sustainable biomanufacturing, but efficient conversion of formaldehyde into higher-value chemicals remains challenging. Formolases, a class of engineered thiamine pyrophosphate-dependent enzymes, catalyze the condensation of formaldehyde to produce glycolaldehyde or 1,3-dihydroxyacetone (DHA), yet their catalytic performance is often insufficient for practical application. This chapter presents a workflow for engineering a benzoylformate decarboxylase (BFD)-derived formolase through a channel-modulating helix strategy. The protocol co

Isolation of various groups of protein preparations is a crucial task in many areas of biochemical research. Creating an effective, simple and economically feasible system for recombinant protein production is highly valuable for both basic research and applied biotechnology. Most laboratory studies focus on bacterial systems for heterologous gene expression. However, such systems can encounter major difficulties with target proteins that have specific properties. Some proteins can aggregate, form insoluble inclusion bodies or undergo proteolytic cleavage during overproduction. Moreover, they
