OBJECTIVE: Increased protein phosphatase magnesium-dependent 1A (PPM1A) levels in patients with ankylosing spondylitis regulate osteoblast differentiation in bony ankylosis; however, the potential mechanisms that regulate osteoclast differentiation in relation to abnormal bone formation remain unclear. This study was undertaken to investigate the relationship of PPM1A to osteoclast differentiation by generating conditional gene-knockout (PPM1Afl/fl ;LysM-Cre) mice and evaluating their bone phenotype.
METHODS: The bone phenotypes of LysM-Cre mice (n = 6) and PPM1Afl/fl ;LysM-Cre mice (n = 6) w

PPM1A and PTEN emerged as novel suppressors of chronic kidney disease (CKD). Since loss of PPM1A and PTEN in the tubulointerstitium promotes fibrogenesis, defining molecular events underlying PPM1A/PTEN deregulation is necessary to develop expression rescue as novel therapeutic strategies. Here we identify TGF-β1 as a principle repressor of PPM1A, as conditional renal tubular-specific induction of TGF-β1 in mice dramatically downregulates kidney PPM1A expression. TGF-β1 similarly attenuates PPM1A and PTEN expression in human renal epithelial cells and fibroblasts, via a protein degradation mec

Bud endodormancy is an important, complex process subject to both genetic and epigenetic control, the mechanism of which is still unclear. The endogenous hormone abscisic acid (ABA) and its signaling pathway play important roles in the endodormancy process, in which the type 2C protein phosphatases (PP2Cs) is key to the ABA signal pathway. Due to its excellent effect on endodormancy release, hydrogen cyanamide (HC) treatment is considered an effective measure to study the mechanism of endodormancy release. In this study, RNA-Seq analysis was conducted on endodormant floral buds of pear (Pyrus

Wip1 belongs to the protein phosphatase C (PP2C) family, of which expression is up-regulated by a number of external stresses, and serves as a stress modulator in normal physiological conditions. When overexpressed, premature dephosphorylation of stress-mediators by Wip1 results in abrogation of tumor surveillance, thus Wip1 acts as an oncogene. Previously, the functional regulation of Wip1 in cell-cycle progression by counteracting cellular G1 and G2/M checkpoint activity in response to DNA damage was reported. However, other than in stress conditions, the function and regulatory mechanism of

Radiotherapy resistance remains a major obstacle in nasopharyngeal carcinoma (NPC). This study delineates the role of protein phosphatase 2 regulatory subunit B gamma (PPP2R2C) in NPC radioresistance and its underlying mechanism to identify therapeutic targets. Through integrated bioinformatic analysis, PPP2R2C was identified as a candidate radioresistance driver. In vitro and in vivo functional assays demonstrated that PPP2R2C depletion significantly impaired NPC cell proliferation, migration, and radioresistance, while its overexpression enhanced these phenotypes. Mechanistic investigations

BACKGROUND: Infections by viruses including severe acute respiratory syndrome coronavirus 2 could cause organ inflammations such as myocarditis, pneumonia and encephalitis. Innate immunity to viral nucleic acids mediates antiviral immunity as well as inflammatory organ injury. However, the innate immune mechanisms that control viral induced organ inflammations are unclear.
METHODS: To understand the role of the E3 ligase TRIM18 in controlling viral myocarditis and organ inflammation, wild-type and Trim18 knockout mice were infected with coxsackievirus B3 for inducing viral myocarditis, influe

PPM1K is a significant metal-dependent phosphatase predominantly located in the mitochondrial matrix, where it plays a crucial role in the metabolism of branched-chain amino acids (BCAAs). It is implicated in cellular function and development across various tissues and is associated with diseases like Alzheimer's, cardiomyopathy, and maple syrup urine disease (MSUD). This article reviews PPM1K's impact on mitochondrial function and cellular metabolism, as well as its role in disease progression. The regulation of PPM1K expression and activity by various factors is complex and highlights its th

Truncating mutations in the last and penultimate exons of the PPM1D gene were recently described as a cause for mild to severe intellectual disability in fourteen patients. Feeding difficulties, periods of fever and vomiting as well as a high pain threshold were described as additional characteristic features and the disorder was subsequently termed "intellectual developmental disorder with gastrointestinal difficulties and high pain threshold (IDDGIP)" in the OMIM database (MIM # 617450). Here we report on an additional patient carrying a novel de novo truncating mutation NM_003620.3: c.1535d

BACKGROUND: Sustained cardiac pressure overload-induced hypertrophy and pathological remodeling frequently leads to heart failure. Casein kinase-2 interacting protein-1 (CKIP-1) has been identified to be an important regulator of cell proliferation, differentiation, and apoptosis. However, the physiological role of CKIP-1 in the heart is unknown.
METHODS AND RESULTS: The results of echocardiography and histology demonstrate that CKIP-1-deficient mice exhibit spontaneous cardiac hypertrophy with aging and hypersensitivity to pressure overload-induced pathological cardiac hypertrophy, as well.

Medulloblastoma (MB) is the most common malignant pediatric brain tumor. Current therapies are associated with substantial morbidity, and prognosis remains poor in high-risk subgroups, particularly those with TP53 mutations or relapsed disease. Cellular senescence is a tumor-suppressive program implicated in MB, but its role in antitumor immunity remains incompletely understood. We found that protein phosphatase 2A (PP2A) regulated immunogenic senescence in MB. Genetic ablation of the PP2A catalytic subunit PP2Ac or depletion of the regulatory subunit PP2A-B56α induced senescence in MB models.

Nucleotide excision repair (NER) is the only mechanism in humans to repair UV-induced DNA lesions such as pyrimidine (6-4) pyrimidone photoproducts and cyclobutane pyrimidine dimers (CPDs). In response to UV damage, the ataxia telangiectasia mutated and Rad3-related (ATR) kinase phosphorylates and activates several downstream effector proteins, such as p53 and XPA, to arrest cell cycle progression, stimulate DNA repair, or initiate apoptosis. However, following the completion of DNA repair, there must be active mechanisms that restore the cell to a prestressed homeostatic state. An important p

As a powerful clinical strategy for cancer treatment, immune checkpoint blockage targeting the PD-1/PD-L1 axis is frequently hampered by diverse resistance mechanisms. An in-depth elucidation of the molecular mechanisms governing PD-L1 homeostasis is crucial for improving the immunotherapy response rate. Here, we demonstrate that the PPM1L-E3s-SERPINB1 cascade plays a pivotal role in regulating PD-L1 stability. Specifically, PPM1L-modulated phosphorylation status of PD-L1 dictates its recognition by distinct E3 ubiquitin ligases, including ARIH1, RING1, and RNF2, thereby promoting its proteaso

Intervertebral disc degeneration (IDD) is a prevalent disease with an increasing incidence, and aging is a key risk factor for its progression. Therefore, this study aimed to explore the role of aging-related genes in IDD through bioinformatics analysis. Differentially expressed aging-related genes were obtained from the CellAge, GSE150408 and GSE124272 datasets, followed by biomarker screening via machine learning. Finally, the identified biomarkers were validated using reverse transcription-quantitative polymerase chain reaction (RT-qPCR), Western Blot (WB), and immunohistochemistry (IHC) as

Glc7 is the only catalytic subunit of the protein phosphatase type 1 in the yeast S. cerevisiae and, together with its regulatory subunits, is involved in many essential processes. Analysis of the non-essential mutants in the regulatory subunits of Glc7 revealed that the lack of Reg1, and no other subunit, causes hypersensitivity to unfolded protein response (UPR)-inducers, which was concomitant with an augmented UPR element-dependent transcriptional response. The Glc7-Reg1 complex takes part in the regulation of the yeast AMP-activated serine/threonine protein kinase Snf1 in response to gluco

Mutations in the LRRK2 gene are a major genetic cause of Parkinson's disease (PD) and induce hyperphosphorylation of RAB GTPases through increased LRRK2 kinase activity. The phosphatase PPM1H counteracts this activity by selectively dephosphorylating LRRK2-phosphorylated RAB proteins. Here, we used primary PPM1H knockout neurons to investigate how loss of PPM1H and the resulting hyperphosphorylation of RAB proteins affect the distribution of alpha-synuclein (aSyn), a key protein in PD pathology, at presynaptic terminals. We found that PPM1H deficiency significantly increased the distance betwe

Water deficit limits maize establishment, but coordinated hormonal and metabolic responses remain incompletely understood. Physiological phenotyping, RNA sequencing, widely targeted metabolomics, and phytohormone profiling were integrated to characterize B73 seedlings during PEG-induced osmotic stress and early recovery. Stress reduced growth and leaf water status and increased the level of oxidative injury. The 266 differentially expressed genes (DEGs) and 47 differentially accumulated metabolites (DAMs) were associated with abscisic acid (ABA) and jasmonate (JA) signaling, amino acid (AA) me
