Nuclear magnetic spin-lattice relaxation of water protons caused by metal cage compounds.

The nuclear magnetic spin-lattice relaxation rates of water protons are reported for solutions of manganese(II), copper(II), and chromium(III) cage complexes of the sarcophagine type. As simple aqueous solutions, the complexes are only modest magnetic relaxation agents, presumably because they lack protons on atoms in the first-coordination-sphere protons that are sufficiently labile to mix the large relaxation rate at the metal complex with that of the bulk solvent. The relaxation is approximately modeled using spectral density functions derived for translational diffusion of the interacting

NMR relaxation of water protons in normal and malignant hyperthermia-susceptible pig muscle.

(1)H-NMR has been used to study the evolution of water proton transversal relaxation times in ageing skeletal muscles of normal and halothane-positive Pietrain pigs. Malignant hyperthermia was confirmed by the caffeine contracture test. Lactic acid, creatine phosphate, and ATP levels in muscle biopsies were measured by biochemical analysis. The NMR dynamic results revealed malignant hyperthermia, but knowledge of animal age and muscle type improved significantly the detection ability. The NMR results revealed large differences between muscles. Halothane sensitivity detection seems to be less a

Correlation between membrane-localized protons and flash-driven ATP formation in chloroplast thylakoids.

Flash-driven ATP formation by spinach chloroplast thylakoids, using the luciferin luminescence assay to detect ATP formed in single turnover flashes, was studied under conditions where a membrane protein amine buffering pool was either protonated or deprotonated before the beginning of the flash trains. The flash number for the onset of ATP formation was delayed by about 10 flashes (from 15 to about 25) when the amine pool was deprotonated as compared to the protonated state. The delay was substantially reversed again by reprotonating the pool upon application of 20-30 single-turnover flashes

Chromosome aberrations induced by protons up to 31 MeV in cultured human cells.

Chromosome aberrations were induced in cultured human cells by proton beams of 31, 12, and 8 MeV. The frequencies of isochromatid breaks and dicentrics have been analysed as a function of proton energy and dose. Both effects are largely dependent on proton energy; isochromatid breaks increased linearly with the dose, whereas dicentrics show a definite parabolic behaviour. The experimental data were fitted to the analytic form Y = KDn and Y = alpha D + beta D2 and the best fitted values of the parameters are reported and discussed. The values of RBE for the isochromatid breaks are in the ratio

Proton perception and activation of a proton-sensing GPCR.

Maintaining pH at cellular, tissular, and systemic levels is essential for human health. Proton-sensing GPCRs regulate physiological and pathological processes by sensing the extracellular acidity. However, the molecular mechanism of proton sensing and activation of these receptors remains elusive. Here, we present cryoelectron microscopy (cryo-EM) structures of human GPR4, a prototypical proton-sensing GPCR, in its inactive and active states. Our studies reveal that three extracellular histidine residues are crucial for proton sensing of human GPR4. The binding of protons induces substantial

Free-base porphyrins as CEST MRI contrast agents with highly upfield shifted labile protons.

PURPOSE: CEST has become a preeminent technology for the rapid detection and grading of tumors, securing its widespread use in both laboratory and clinical research. However, many existing CEST MRI agents exhibit a sensitivity limitation due to small chemical shifts between their exchangeable protons and water. We propose a new group of CEST MRI agents, free-base porphyrins and chlorin, with large exchangeable proton chemical shifts from water for enhanced detection. METHODS: To test these newly identified CEST agents, we acquired a series of Z-spectra at multiple pH values and saturation fie

Critical role for the protons in FRTL-5 thyroid cells: nuclear sphingomyelinase induced-damage.

Proliferating thyroid cells are more sensitive to UV-C radiations than quiescent cells. The effect is mediated by nuclear phosphatidylcholine and sphingomyelin metabolism. It was demonstrated that proton beams arrest cell growth and stimulate apoptosis but until now there have been no indications in the literature about their possible mechanism of action. Here we studied the effect of protons on FRTL-5 cells in culture. We showed that proton beams stimulate slightly nuclear neutral sphingomyelinase activity and inhibit nuclear sphingomyelin-synthase activity in quiescent cells whereas stimulat

Two protons, two positrons, and four electrons: Covalent bond with van der Waals characteristics.

Classifying interactions is key in the physical sciences, and bonding mechanisms in matter-antimatter systems remain particularly enigmatic. Here, we focus on a paradigmatic example of positronium hydride (PsH) dimer composed of two protons, two positrons, and four electrons, whose bonding nature has been previously described as either ionic, covalent, or van der Waals like. Accurate quantum Monte Carlo calculations show that the two positrons occupy a delocalized molecular orbital that envelopes the two hydrogen anions and responds as a collective dipole to an applied electric field. This pos

Protons Versus Photons Postmastectomy Radiation Therapy Effects on Breast Reconstruction Outcomes and Dosimetry Analysis.

PURPOSE: To compare the impact of proton versus photon postmastectomy radiation therapy (PMRT) on implant-related complications. METHODS AND MATERIALS: The records of patients with breast cancer treated with mastectomy and expander and/or implant reconstruction followed by PMRT at our institution between 2011 and 2022 were reviewed. Patients were divided into 2 groups by treatment modality: proton and photon groups. All identified patients in the proton group were treated using conventional fractionation, and radiobiological effectiveness (RBE) was scaled to 1.1. Recorded complications includ

Radiation Therapy for Primary Adenoid Cystic Carcinoma of the Trachea: Photons, Protons, or Carbon.

Primary adenoid cystic carcinoma of the trachea (ACC-T) is an extremely rare cancer of the central bronchial system. It is usually associated with an excellent prognosis. Surgery is the standard treatment for resectable tumors, while radiation therapy is used for unresectable tumors or medically inoperable patients. Radiation therapy can be delivered with photons, protons, or carbon ion therapy. In this report, we review a case of unresectable ACC-T in a middle-aged female patient who was treated with radiation therapy and review the potential benefits of the different types of radiation thera

Calcium and protons affect the interaction of neurotransmitters and anesthetics with anionic lipid membranes.

We study how zwitterionic and anionic biomembrane models interact with neurotransmitters (NTs) and anesthetics (ATs) in the presence of Ca(2+) and different pH conditions. As NTs we used acetylcholine (ACh), γ-aminobutyric acid (GABA), and l-glutamic acid (LGlu). As ATs, tetracaine (TC), and pentobarbital (PB) were employed. By using differential scanning calorimetry (DSC), we analyzed the changes such molecules produce in the thermal properties of the membranes. We found that calcium and pH play important roles in the interactions of NTs and ATs with the anionic lipid membranes. Changes in pH

Signaling Pathways in Proton and Non-proton ASIC1a Activation.

Acid-sensing ion channels (ASICs) regulate synaptic activities and play important roles in neurodegenerative diseases as well as pain conditions. Classically, ASICs are described as transiently activated by a reduced pH, followed by desensitization; the activation allows sodium influx, and in the case of ASIC1a-composed channels, also calcium to some degree. Several factors are emerging and extensively analyzed as modulators, activating, inhibiting, and potentiating specific channel subunits. However, the signaling pathways triggered by channel activation are only starting to be revealed. The c

A simulation study of in-beam visualization system for proton therapy by monitoring scattered protons.

Recently, in-beam positron emission tomography (PET) has been actively researched for reducing biological washout effects and dose monitoring during irradiation. However, the positron distribution does not precisely reflect the dose distribution since positron production and ionization are completely different physical processes. Thus, a novel in-beam system was proposed to determine proton dose range by measuring scattered protons with dozens of scintillation detectors surrounding the body surface. While previous studies conducted a preliminary experiment with a simple phantom, we simulated m

The production, buffering and efflux of protons in human skeletal muscle during exercise and recovery.

We show how quantitative information about proton handling in human skeletal muscle in exercise and recovery can be obtained by 31P MRS and illustrate this with data from metabolic disorders. Proton production, proton efflux and passive buffering can be distinguished by comparing changes in [phosphocreatine] and pH at the end of exercise and by calculating ATP turnover during ischaemic exercise and in the 'natural experiment' of myophosphorylase deficiency (McArdle's disease). We calculate the effective buffer capacity to be 20-30 mmol/L/pH unit (slykes), somewhat lower than published measurem

Construction of Successive Proton Conduction Channels to Accelerate the Proton Conduction Process in Flexible Proton Exchange Membranes.

Successive proton conduction channels are constructed with the spin coating method in flexible proton exchange membranes (PEMs). In this research, phosphoric acid (PA) molecules are immobilized in the multilayered microstructure of Kevlar nanofibers and polystyrene-block-poly(ethylene-ran-butylene)-block-polystyrene (SEBS) polymer molecular chains. As a result, successive proton conduction channels can accelerate the proton conduction process in the prepared membrane with the multilayered microstructure. Additionally, the microstructure fractures of the composite membranes from the external fo

Long TE PRESS and STEAM for measuring the triglyceride glycerol CH2 protons at 3 T.

The glycerol methylene proton resonances (4-4.5 parts per million, ppm), which arise from the triglyceride backbone, are relevant to fat composition assessment and can be measured with proton MRS. The purpose of the presented work is to determine long TE (echo time) point resolved spectroscopy (PRESS) and stimulated echo acquisition mode (STEAM) values at 3 T to resolve the glycerol resonances from that of overlapping water. The response of the glycerol methylene protons of nine edible oils as a function of PRESS and STEAM TE (mixing time, TM = 20 ms) was investigated. In addition, high resolu

The role of buffer anions and protons in secretion by the rabbit mandibular salivary gland.

1. The role of extracellular HCO3- and H+ in the formation of primary saliva and its subsequent modification by the glandular ducts has been investigated in the isolated perfused mandibular salivary gland of the rabbit. 2. Variation of extracellular HCO3- concentration between 12.5 and 50.0 mmol/l was without effect on salivary flow rate or on Na+ and K+ excretion, even though salivary HCO3- (and Cl-) content altered with changes in the extracellular concentration of the two anions. 3. Complete replacement of perfusate HCO3- by Cl- reduced fluid secretion by 34% and almost abolished ductal Na+

Size-dependent femtosecond proton transfer in protonated methanol clusters.

Methanol clusters (CH3OH)n are prototypical hydrogen-bonded systems that bridge isolated molecules and bulk liquid methanol. Strong-field ionization of these clusters can trigger ultrafast charge migration, proton transfer, and isomerization; however, the onset timescales of these pathways remain largely unexplored despite extensive steady-state mass-spectrometric evidence for protonation and chemical rearrangement. Here we use femtosecond time-resolved strong-field ionization and disruptive probing to directly track the early-time dynamics of methanol clusters. Transient mass spectra yield si
