QnrD is a plasmid-mediated quinolone resistance (PMQR) determinant first reported in clinical Salmonella enterica isolates from China, located on nonconjugative plasmids of 4270 bp. Since then, the qnrD gene has been mostly found on plasmids around 2683 bp in Proteus and Morganella genera. However, Providencia spp. strains carrying qnrD-harboring plasmids have only been reported among clinical samples, in France and China. In this paper we describe two plasmids carrying qnrD in Providencia spp. isolated from Brazilian food and coastal waters. These plasmids present high coverage and identity w

BACKGROUND/OBJECTIVES: A rise in infections associated with carbapenem-resistant Providencia species (CRPS) has been observed worldwide. This study presents a genomic analysis of CRPS isolates from four hospitals in Croatia and the outpatient setting, in order to determine the extent of the spread of CRPS in Croatia. In the present study, we applied a combination of phenotypic characterization and molecular analysis of resistance traits to determine the mechanisms and the routes of spread of CRPS.
MATERIAL AND METHODS: The antibiotic susceptibility testing was performed using disk-diffusion a

Providencia rettgeri (P. rettgeri) is a ubiquitous organism that is infrequently associated with human disease. Here we report the isolation of this organism from a polymicrobial wound infection resulting from ruptured tophi on a 54-year-old male patient from Guam. We describe the identification and confirmation of this organism, and propose metabolic synergy as a possible mechanism of pathogenesis. To our knowledge, this is the first published report of a wound infection colonized by P. rettgeri from Guam, and the first report to speculate upon the role of bacterial synergy in P. rettgeri pat

A laboratory model system was developed to investigate the progressive colonization of urinary catheters by Escherichia coli Providencia stuartii, prominent organisms in the polymicrobial bacteriuria of the long-term catheterized urinary tract. We hypothesized that colonization of the catheter and artificial urine by E. coli is influenced by the presence of P. stuartii. E. coli or P. stuartii in pure culture both rapidly colonized the artificial urine and catheters, and both persisted throughout all experiments. In systems containing both organisms, P. stuartii occurred in significantly higher

We obtained a clinical isolate of Providencia stuartii showing a high level of resistance to norfloxacin and to other 4-quinolones, whose target is the enzyme DNA-gyrase. This strain showed resistance also to beta-lactam and aminoglycoside antibiotics. In order to detect modification of DNA-gyrase, we performed supercoiling assays in vitro in presence of norfloxacin and ciprofloxacin. Furthermore, outer membrane proteins, which are involved in permeability mechanisms, were analyzed on SDS-polyacrylamide gels. Results showed that both modifications in DNA-gyrase and changes in outer membrane pr

Antigenic material obtained by phenol-water extraction from Providencia rettgeri strains, Escherichia coli O:14 strains, and mutants of the E. coli O:14 strain were examined by the passive (indirect) hemagglutination technique, by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and by immune blotting (lipopolysaccharide (LPS) blotting). Providencia rettgeri 965, like E. coli O:14, was demonstrated to have an enterobacterial common antigen (ECA) in the immunogenic form but, unlike E. coli O:14, it possessed characteristics of a smooth strain. Two populations of molecules were observe

Providencia stuartii is an emerging opportunistic pathogen in humans and belongs to a genus that has shown great genomic diversity. P. stuartii has shown relevant traits in antimicrobial resistance because of its intrinsic resistance to multiple antibiotics and its ability to acquire resistance genes that limit therapeutic options. The present study describes the molecular epidemiology of P. stuartii based on the multilocus sequence typing (MLST) scheme developed for this purpose. This non-PCR-based scheme uses seven complete housekeeping genes. In total, 518 genomes obtained from public datab

INTRODUCTION AND IMPORTANCE: Providencia rettgeri, a member of the Morganellaceae family within the Enterobacterales order, is predominantly associated with urinary tract infections in hospitalized individuals, particularly those with indwelling urinary catheters. However, wound infections caused by P. rettgeri are exceedingly rare, with an estimated incidence of around 0.1 %. Here, we present a case of wound infection in a healthy child caused by P. rettgeri, highlighting the rarity of the organism and emphasizing the importance of prompte identification and appropriate antibiotic therapy.

BACKGROUND Anorexia nervosa, predominantly affecting young females, significantly compromises the immune system, rendering patients vulnerable to infections and contributing to a notably high mortality rate. The complexities involved in managing infections in immunocompromised patients with anorexia nervosa are often underappreciated. This case report aims to shed light on these challenges and the potential complications arising from treatment interventions. CASE REPORT We present the case of a 30-year-old woman with a long-standing history of anorexia nervosa, who was admitted with severe sys

In the last years, an increasing number of untreatable infections caused by drug-resistant microbes have impacted the health care system. Worldwide, infections caused by carbapenem-resistant (CR) Gram-negative bacilli have dramatically increased. Among the CR-Gram-negative bacilli, those producing carbapenemases, such as NDM-1, are the main concern. Different Enterobacterales harboring NDM-1 have been reported lately. Providencia stuartii, a member of the Morganellaceae family, is ubiquitous in the environment, but is also known to cause nosocomial infections. Here we describe the genomic anal

CntA oxygenase is a Rieske 2S-2Fe cluster-containing protein that has been previously described as able to produce trimethylamine (TMA) from carnitine, gamma-butyrobetaine, glycine betaine, and in one case, choline. TMA found in humans is exclusively of bacterial origin, and its metabolite, trimethylamine oxide (TMAO), has been associated with atherosclerosis and heart and renal failure. We isolated four different Rieske oxygenases and determined that there are no significant differences in their substrate panels. All three had high activity toward carnitine/gamma-butyrobetaine, medium activit

The natural antibiotic susceptibility of 38 Providencia rettgeri, 35 P. stuartii, 23 P. alcalifaciens and 20 P. rustigianii strains was examined. MIC values were determined by a microdilution procedure and evaluated by a table calculation programme. P. stuartii was the least susceptible Providencia sp. and was naturally resistant to tetracyclines, some penicillins, older cephalosporins, sulphamethoxazole and fosfomycin and to antibiotics to which other species of Enterobacteriaceae are also resistant. It was naturally sensitive to modern penicillins and cephalosporins, carbapenems and aztreona

Providencia rettgeri is a Gram-negative bacterium, rarely reported as a cause of diabetic wound infections. Its frequent resistance to β-lactams and carbapenems limits treatment options, especially in immunocompromised patients. We present the case of a 59-year-old unhoused male living with diabetes, peripheral artery disease, and prior bilateral transmetatarsal amputations who presented with a University of Texas 3B diabetic foot ulcer. Magnetic resonance imaging showed osteomyelitis of metatarsals 2-4, and bone biopsy grew Providencia rettgeri and Proteus mirabilis. The patient declined furt

We describe the emergence of Providencia stuartii infection and subsequent bacteremia in a bedridden patient with poorly controlled diabetes mellitus. Despite empirical treatment, the disease progressed, leading to septic shock and below-knee amputation. We examine the timing and clinical progression of the infection, highlighting that P. stuartii wound infections in diabetic patients can lead to severe complications. Early detection and appropriate intervention are crucial to prevent such outcomes.

The occurrence of carbapenemases encoding genes in Providencia rettgeri is a critical public health concern since this species has intrinsic resistance to several antimicrobials, including polymyxins. The identification of this multidrug-resistant (MDR) pathogen outside the hospital setting has become increasingly frequent, and raises an alert for the global health agencies, as they indicate a possible spread of such pathogens. Herein, we described three MDR P. rettgeri isolates carrying a diversity of antimicrobial resistance genes (ARGs) isolated from stool samples of swine and bovine in Bra

We report here the draft genome sequence of the first NDM-1-producing Providencia stuartii strain isolated in Portugal. Sequence analyses revealed the presence of an incompatibility group A/C2 (IncA/C2) plasmid and of diverse acquired genes conferring resistance to β-lactams, aminoglycosides, tetracycline, macrolides, chloramphenicol, and sulfonamides. This sequence contributes to the evaluation of the spread of NDM-1 producers.
