Determination of Flavonoid and Proanthocyanidin Profile of Hungarian Sour Cherry.

Hungarian sour cherries (SC) are excellent source of anthocyanin (concentrations (100⁻300 mg in 100 g fresh fruit) and melatonin (0.15 mg in 100 g fresh fruit), but other flavonoid derivatives also can be isolated by aqueous alcoholic extraction. We have developed a new process for extracting non-extractable procyanidines bound to the membrane, proteins, and fibers. These compounds were seperated with UHPLC-MS methods, and the structure of individual components were identified on the basis of their mass fragmentation spectra. The antioxidant capacity of soluble and non-soluble antioxidants wer

Co-opted genes of algal origin protect C. elegans against cyanogenic toxins.

Amygdalin is a cyanogenic glycoside enriched in the tissues of many edible plants, including seeds of stone fruits such as cherry (Prunus avium), peach (Prunus persica), and apple (Malus domestica). These plants biosynthesize amygdalin in defense against herbivore animals, as amygdalin generates poisonous cyanide upon plant tissue destruction.1,2,3,4 Poisonous to many animals, amygdalin-derived cyanide is detoxified by potent enzymes commonly found in bacteria and plants but not most animals.5 Here we show that the nematode C. elegans can detoxify amygdalin by a genetic pathway comprising cysl

Anti-auxin treatment and co-expression networks reveal auxin involvement in the transition to ripening in sweet cherry fruits.

The transition from development to ripening is governed by several hormones that interplay with abscisic acid (ABA) in non-climacterics, including auxin. In sweet cherry (Prunus avium), auxin has been shown to promote fruit coloration and ABA accumulation, in contrast to other non-climacteric models. Thus, this work aims to explore the role of auxin during the transition to ripening in sweet cherry by assessing the global gene expression response to an auxin homeostasis disturbance. For this, we used 5d, a fluorescently labeled compound with previously reported anti-auxin activity, which was a

L‒asparaginase activity in some endophytic fungi: Glutaminase‒free and low urease co‒activities.

In this study, L‒asparaginase production in several endophytic fungi was evaluated along with their L‒glutaminase and urease co-activities. The effect of L‒asparagine and different culture media on L-asparaginase production were also evaluated. Among the 62 investigated isolates, 49 isolates exhibited L‒asparaginase activity, and the maximum zone index (6.58) was observed in Cladosporium perangustum EL1. Evaluation of L‒glutaminase and urease co-activities in L‒asparaginase-positive isolates screened 19 isolates with no L‒glutaminase activity and four isolates with minimum urease production. L

Plastome architecture and maternal relationships of cultivated almond (Prunus dulcis) and its wild relatives.

Plastome structure and gene content are invariant across almond and its wild relatives; the spacers rpl33-rps18, rps2-rpoC2 and psbB-psbT are the most variable regions and strongest candidate markers for Amygdalus. Almond (Prunus dulcis) is a major tree nut crop whose domestication narrowed its genetic base, whilst its wild relatives retain adaptive diversity. Complete plastomes remain scarce for almond. We sequenced three new plastomes (P. arabica, P. argentea and P. dulcis) from Türkiye and compared them with ten further Prunus plastomes from public reads, giving 13 uniformly annotated genom

Temperature-Mediated Kill and Oviposition of Western Cherry Fruit Fly (Diptera: Tephritidae) in the Presence of Spinosad.

Western cherry fruit fly, Rhagoletis indifferens Curran (Diptera: Tephritidae), is a quarantine pest of sweet cherry (Prunus avium (L.) L.) that is managed using insecticides, including spinosad, an organic compound that can be applied in low spray volumes. Identifying factors that can increase the efficacy of spinosad can be useful for improving fly control. Here, the major objective was to determine if temperature mediates kill and oviposition of R. indifferens in the presence of low spinosad coverage in the laboratory. Experiments were conducted by placing flies in cages with cherries and w

Prevalence of Listeria spp. on food contact surfaces in commercial cherry packinghouses in Washington and California.

Recent recalls for tree fruit due to possible contamination with Listeria monocytogenes have highlighted the importance of understanding the high-risk food contact surfaces (FCS) in commercial packinghouses in order to manage the risk of cross-contamination within the packing environment. The objectives of this study were to determine the prevalence of Listeria spp. on FCS in Washington State and California cherry packinghouses and to identify locations with the greatest likelihood to harbor Listeria spp. Four commercial cherry packinghouses were visited three times each during two consecutive

Ethephon as a bloom-delaying agent for frost-sensitive tree crop flowers at risk from a changing climate.

BACKGROUND: Climate change increases the risk of late spring frosts, which damage flowers and developing fruitlets in tree crops. Ethephon, a plant growth regulator, may delay flowering and potentially reduce frost damage. METHODS: We investigated the effects of ethephon on flowering time and fruit set in two sweet cherry (Bellise and Kordia) and two apple cultivars (Elstar and Kanzi). Treatments included two ethephon concentrations (500 and 750 ppm) applied either in early or late autumn. RESULTS: Ethephon significantly delayed flowering in cherry cultivar Kordia, but not cultivar Bellise,

Regulation of light-induced anthocyanin biosynthesis in sweet cherry by the WRKY transcription factor PavWRKY18.

Anthocyanin production and storage are crucial for determining the color and quality of sweet cherry (Prunus avium) fruits. Although WRKY transcription factors are recognized as critical regulators of secondary metabolism, their specific roles and underlying mechanisms regarding coloration in sweet cherry remain elusive. Through two comparative transcriptomic analysis of sweet cherry in different ripening stages and lighting treatments, we identified the candidate gene PavWRKY18. The expression patterns of PavWRKY18 exhibits specifically high expression during late fruit ripening and showed a

Integrated Packaging and Ethylene Scavenging Strategies to Extend the Shelf Life and Quality of Sweet Cherry cv. 'Regina' Under Ambient and Refrigerated Storage.

Sweet cherry cv. 'Regina' is highly susceptible to rapid postharvest deterioration due to high respiration rate, moisture loss, and ethylene sensitivity, resulting in reduced firmness, stem browning, and decay. This study evaluated the effectiveness of two commercial modified atmosphere packaging films (Fresh Mama and Keep It Fresh) in combination with ethylene scavengers such as potassium permanganate, activated carbon, and their combination in extending the shelf life and preserving fruit quality under ambient (25°C) and refrigerated (4°C) storage. Fruits were assessed for physiological loss

Postharvest CO2 treatment and cold storage for Drosophila suzukii (Diptera: Drosophilidae) fruit infestation control.

The invasive pest, spotted wing drosophila (Drosophila suzukii (Matsumura, 1931) or SWD), damages various soft-skinned fruits, severely impacting orchards and vineyards economically. Current sorting practices in commercial production may overlook early-stage SWD infestations, as visible signs take a few days to appear. Our study focused on managing SWD infesting fruits (blueberry, cherry, and raspberry) without visible signs using an artificial atmosphere with elevated CO2 and low temperature. We hypothesized that these factors affect SWD survival and possibly interact, with potential variatio

The effects of fruit consumption in patients with hyperuricaemia or gout.

The consumption of fructose has gained increased attention as a potential cause of hyperuricaemia since fructose metabolism produces urate as a byproduct. In addition to sucrose and high fructose corn syrup, fresh fruits also contain fructose, suggesting that patients with hyperuricaemia or gout might also avoid fresh fruit. However, the effect of fruits is complex. Some studies reported that fruit intake was associated with gout flares while other studies showed that fruits rather lowered the risk for gout. Thus, fruits should not be simply viewed as a source of fructose. The complexity of fr

Effect of Low-Oxygen Conditions Created by Modified Atmosphere Packaging on Radiation Tolerance in Drosophila suzukii (Diptera: Drosophilidae) in Sweet Cherries.

Modified atmosphere packaging (MAP) creates a low-oxygen (O2) environment that can increase the shelf life of fresh produce by decreasing respiration and the growth of pathogens. Low oxygen may also increase insect tolerance to irradiation (IR), and the use of MAP with products treated by IR to control quarantine pests before export may inadvertently compromise treatment efficacy. Spotted wing drosophila, Drosophila suzukii Matsumura (Diptera: Drosophilidae) is a quarantine pest of stone and small fruits and a potential target for postharvest IR treatment. The effect of low oxygen generated by

Montmorency tart cherry protects against age-related bone loss in female C57BL/6 mice and demonstrates some anabolic effects.

PURPOSE: Age-related bone loss is a consequence of endocrine and immune changes that disrupt bone remodeling. Functional foods (e.g., tart cherries) with antioxidant, anti-inflammatory and prebiotic activity can potentially counter this age-related phenomenon. The aim of this study was to determine if Montmorency tart cherry protects against early age-related bone loss and the culpable alterations in bone metabolism. METHODS: Female, 5-month-old, C57BL/6 mice were assigned to baseline or treatment groups: AIN-93M diet supplemented with 0, 1, 5, or 10% tart cherry for 90 days. Bone mineral den

In vitro and in vivo cysticidal activity of extracts and isolated flavanone from the bark of Prunus serotina: A bio-guided study.

Currently, neurocysticercosis treatment involves two drugs: albendazole and praziquantel; however, their efficacy is suboptimal and new cysticidal drugs are needed. The present paper reports the cysticidal activity of extracts of the bark from Prunus serotina against Taenia crassiceps cysts and the isolation and identification of the main components of the most active extract. Results showed that all extracts displayed in vitro cysticidal activity (EC50=17.9-88.5μg/mL), being the methanolic the most active and selective. Also, methanolic extract exhibited in vivo efficacy at 300mg/kg which was

Influence of environmental conditions on the production of nutraceuticals in Italian edible plant landraces.

Autochthonous plant varieties, also referred to as landraces, represent an important genetic resource, being well-adapted to the environment in which they have been selected. Landraces usually show profiles rich in nutraceuticals, making them an effective and valuable alternative to commercial agri-products, as well as potential candidates for crop improvement programs. Basilicata region is recognized as an Italian hotspot for agrobiodiversity, due to its complex orography. Thus, this work aimed to characterize and monitor, for two successive years, the content of secondary metabolites and rel
