Testing the priming effect in the deep ocean: are microorganisms too starved to consume recalcitrant organic carbon?

Deep ocean dissolved organic carbon (DOC) is one of the largest pools of reduced carbon on Earth. Many DOC compounds escape microbial degradation and persist for thousands of years in the ocean. Although many hypotheses have been proposed, the mechanisms responsible for this long-term stability remain unresolved. Heterotrophic microorganisms in the deep ocean are energetically starved and exhibit low metabolic activity. Here, we investigated whether the severe energy limitation in deep-sea environments acts as a barrier to microbial degradation of DOC. We hypothesized that alleviating this ene

Biogenic ROS mediated degradation mechanism of marine toxin domoic acid.

Domoic acid (DA) is a compound generated as a secondary metabolite during harmful algal blooms, has historically received attention as the potent neurotoxicity in marine environment. However, the aerobic degradation mechanism of DA and the DA-degrader remain largely unknown. Here, we revealed the mechanism of aerobic degradation of DA by a ubiquitous marine Pseudoalteromonas sp., and more importantly, we confirmed that the degradation of DA is mediated by biogenic reactive oxygen species (ROS), rather than direct enzyme-mediated as traditionally conceived. Results indicated that DA degradation

Biosynthetic Investigations of Ulbactins Unveil Two Novel Thiazolinyl Imine Reductases Crucial for the Generation of Siderophore Activity.

Siderophores are ferric ion chelating natural products secreted by microorganisms to survive in an iron-deficient environment. During screening for siderophores from marine bacteria, four interconvertible ulbactins C1-C4 and oxidized derivatives were identified in the bacterium Pseudoalteromonas flavipulchra S16. Iron-ion chelating activity assays revealed that the reduction of thiazoline rings is critical for the activity. The ulbactins biosynthetic gene cluster (ubt) was identified in the genome of P. flavipulchra S16 and validated via gene knockout and heterologous expression. A unique feat

Epibiotic Bacteria Isolated from the Non-Indigenous Species Codium fragile ssp. fragile: Identification, Characterization, and Biotechnological Potential.

Due to their richness in organic substances and nutrients, seaweed (macroalgae) harbor a large number of epiphytic bacteria on their surfaces. These bacteria interact with their host in multiple complex ways, in particular, by producing chemical compounds. The released metabolites may have biological properties beneficial for applications in both industry and medicine. In this study, we assess the diversity of culturable bacterial community of the invasive alga Codium fragile ssp. fragile with the aim to identify key groups within this epiphytic community. Seaweed samples were collected from t

Enzymatic preparation of chitooligosaccharides and their anti-obesity application.

Chitooligosaccharides (COS) are derived from chitosan, which can be used as nutraceuticals and functional foods. Because of their various biological activities, COS are widely used in the food, medicine, agriculture, and other fields. COS were prepared by chitosanase  from Pseudoalteromonas sp. SY39 and their anti-obesity activity was researched in mice in this study. The effects of hydrolysis time, temperature, the ratio of enzyme to chitosan, and pH on the productivity of COS were discussed. Preparation process of COS was established in a 5-L fermenter. COS were characterized and their anti-

Marine tubeworm metamorphosis induced by arrays of bacterial phage tail-like structures.

Many benthic marine animal populations are established and maintained by free-swimming larvae that recognize cues from surface-bound bacteria to settle and metamorphose. Larvae of the tubeworm Hydroides elegans, an important biofouling agent, require contact with surface-bound bacteria to undergo metamorphosis; however, the mechanisms that underpin this microbially mediated developmental transition have been enigmatic. Here, we show that a marine bacterium, Pseudoalteromonas luteoviolacea, produces arrays of phage tail-like structures that trigger metamorphosis of H. elegans. These arrays comp

Microdroplet-based high-throughput screening for antagonistic bacteria targeting penaeid shrimp pathogenic Vibrio harveyi.

Antagonistic bacteria that suppress the growth of specific bacteria have attracted attention as an antibiotics-independent strategy for infectious disease control in aquaculture. Microfluidics-based water-in-oil droplets (microdroplets) enable high-throughput screening of antagonistic bacteria in the field of medicine or agriculture. However, the use of this screening system in aquaculture has not yet been reported. In particular, penaeid shrimp aquaculture, one of the largest sectors of global aquaculture, has a strong demand for alternative disease control strategies because vaccination is i

Use of psychrophilic xylanases provides insight into the xylanase functionality in bread making.

The bread-improving potential of three psychrophilic xylanases from Pseudoalteromonas haloplanktis TAH3A (XPH), Flavobacterium sp. MSY-2 (rXFH), and unknown bacterial origin (rXyn8) was compared to that of the mesophilic xylanases from Bacillus subtilis (XBS) and Aspergillus aculeatus (XAA). XPH, rXFH, and rXyn8 increased specific bread volumes up to 28%, 18%, and 18%, respectively, while XBS and XAA gave increases of 23% and 12%, respectively. This could be related to their substrate hydrolysis behavior. Xylanases with a high capacity to solubilize water-unextractable arabinoxylan (WU-AX) dur

Heterotrophic inorganic carbon fixation varies with temperature and resource availability in marine and soil bacterial isolates.

Inorganic carbon use in cellular metabolism is ubiquitous in all living organisms. The assimilation of inorganic carbon into biomass via heterotrophic inorganic carbon fixation (HICF) is estimated to contribute significantly to global carbon fixation, yet the controls on this process in strictly heterotrophic microorganisms remain poorly understood. To identify potential drivers of HICF, we cultivated well-characterized heterotrophic Gammaproteobacteria (Pseudomonas putida and Pseudoalteromonas carrageenovora) at two temperatures (optimal vs colder) and resource treatments (high vs low organic

Low-molecular-mass purine nucleoside phosphorylase: characterization and application in enzymatic synthesis of nucleoside antiviral drugs.

Purine nucleoside phosphorylase (PNP) that catalyzes the reversible phosphorolysis of various purine nucleosides is widely distributed in prokaryotes and eukaryotes. Four pnp genes from Bacillus subtilis 168, Escherichia coli K-12 and Pseudoalteromonas sp. XM2107 were cloned by PCR and expressed in E. coli XL1-Blue. Recombinant PNPs (rPNPs) were purified by Ni(2+)-NTA chromatography. Compared with other rPNPs, PNP(816) was a low-molecular-mass homotrimer, which exhibited 11-, 4- and 1.5-fold higher values in k (cat)/K (m) using inosine as the substrate at 37°C. Simultaneous removal of nitrogen, Cu2+, and bisphenol A in a hydrogel-biochar-AQDS immobilized bioreactor with added bicarbonate: Performance and metagenomic insights.

As the complexity of industrial wastewater pollution continues to increase, the simultaneous removal of nitrogen, metal contaminants, and persistent organic pollutants under low carbon conditions has become a key challenge for biological treatment systems. To address the operational instability and dependence on carbon sources observed in immobilized systems when exposed to copper (Cu2+) and bisphenol A (BPA), the Pseudoalteromonas japonicus strain LY0623 was integrated into a hydrogel-biochar-AQDS composite carrier to construct a multifunctional immobilized biofilm system. The Challenge of the Sponge Suberites domuncula (Olivi, 1792) in the Presence of a Symbiotic Bacterium and a Pathogen Bacterium.

Sponges, which are in close contact with numerous bacteria in prey/predator, symbiotic and pathogenic relationships, must provide an appropriate response in such situations. This starts with a discriminating recognition of the partner either by a physical contact or through secreted molecules or both. We investigated the expression of the Toll-like receptor, Caspase 3/7, Tumor Necrosis Factor receptor-associated factor 6, Bcl-2 homology protein-2 and macrophage expressed genes of axenic sponge cells in the presence of a symbiotic bacterium (Endozoicomonas sp. Hex311), a pathogen bacterium (Pse

Polarity-Driven Marine Antifouling Coating with Dynamic Surface: Elucidating Mechanism for Disrupting Mussel Adhesion.

Mussels can adhere to the surfaces of marine structures, which leads to the formation of fouling communities that disrupt the performance of these facilities. However, their robust adhesion mechanism presents a substantial challenge to conventional antifouling coatings that depend on the flow rate of seawater, which can even cause the coatings to fail in static marine environments. Herein, a dynamic interface engineering strategy is employed to develop an antifouling coating that intervenes with mussel adhesion by disrupting biological functions along multiple pathways. A dynamic surface is ac

The dual-function enzyme PpLipO protects polar marine bacteria from phospholipid peroxidation.

Membrane phospholipid peroxidation is a deleterious process in which reactive oxygen species (ROS) attack unsaturated fatty acids embedded in cell membranes, generating phospholipid hydroperoxides and triggering structural damage that can ultimately lead to cell death. While mammalian strategies to mitigate peroxidation, primarily through the combined activities of phospholipase A2 (PLA2) and subsequent reduction of resultant fatty acid hydroperoxides with glutathione peroxidases/peroxiredoxins have been well characterized for more than two decades, mechanisms by which prokaryotes contend with

Enhancing flavor quality in low-salt dried large yellow croaker (Pseudosciaena crocea) through secondary fermentation: insights into microbial drivers.

Large yellow croaker is highly perishable and traditionally preserved by salting. This study evaluated secondary fermentation as an improved processing method compared to traditional techniques. A low-salt (5%) curing process, combined with secondary fermentation under reduced‑oxygen conditions and baijiu spraying, was applied to produce dried fish with lower sodium and enhanced flavor. This approach significantly suppressed lipid oxidation and protein degradation. A total of 65 volatile flavor compounds were identified, with substantial increases in aldehydes, esters, and aromatic compounds d

Transposon end recognition and excision mechanisms of type I-F CRISPR-associated transposases.

CRISPR-associated transposons (CASTs) are Tn7-like elements that have co-opted RNA-guided CRISPR effectors for targeted DNA insertion. CASTs have been adapted as genome editing tools for programmable, site-specific integration. Among them, the type I-F system from Pseudoalteromonas (PseCAST) shows exceptionally robust activity in human cells, yet its mechanistic basis remains poorly understood. Here, we present structural and biochemical analysis of the PseCAST transposase TnsAB. Biochemical reconstitution of transposon DNA excision defines key characteristics of the transposition mechanism. C

Probiotic and postbiotic treatment improves coral health and promotes specific metabolic and microbiome changes in situ during a heatwave.

Microbial therapies are emerging as promising tools for coral protection against heat stress, yet such application was not tested in situ. We tested two coral-derived probiotic consortia and their heat-killed counterparts (postbiotics) on bleaching Acropora cf. valida in the Red Sea during a marine heatwave. Over 15 days, both live and one of the heat-killed treatments maintained photosynthetic efficiency (Fv/Fm), whereas placebo-treated corals exhibited significant thermal stress-induced decline. 16S rRNA gene sequencing profiling showed enrichment of putatively beneficial genera (e.g., Teras
