Psoralidin, a major component of Psoraleae Fructus, induces inflammasome activation and idiosyncratic liver injury.

Idiosyncratic drug-induced liver injury (IDILI) is a rare but potentially fatal disease that is unpredictable and independent of the dose of the drug. Increasing evidence suggests that the majority of IDILI cases are immune-mediated, and the aberrant activation of inflammasome plays a vital role in progression. Psoraleae Fructus (PF), a tonic Chinese medicine, has been able to cause IDILI, but the precise mechanism of hepatotoxicity remains unclear. In this study, eight bioactive compounds involved in PF-induced inflammasome activation were investigated. The results demonstrated that psoralidi

Structural characterization and in vivo and in vitro anti-osteoporosis potential of two Psoralea corylifolia L. polysaccharides.

Psoralea corylifolia L. (PCL) has been widely used to improve osteoporosis for thousands of years, whereas the anti-osteoporosis activity of its polysaccharides has not been systematically reported. Here, we firstly proved that PCLP80 crude polysaccharides exhibited significant improvement effects (p < 0.05) about femur bone mineral density, bone trabecula, and muscle function performances including grip strengths, and forced swimming times in glucocorticoid-induced osteoporosis (GIOP) mice. Subsequently, a novel acidic (PCLP80-1) and a neutral (PCLP80-2) polysaccharides were purified from PCL

Analysis of molecular events triggered by furanocoumarins in Psoralea corylifolia L. via reactive metabolites and protein adducts.

ETHNOPHARMACOLOGICAL RELEVANCE: Psoralea Fructus (PF), the dried ripe fruit of Psoralea corylifolia L., is a widely utilized Chinese medicinal herb with a long-established history in clinical practice and is known for its diverse biological activities. Although furanocoumarin is potentially hepatotoxic, exposure levels at standard clinical doses are generally insufficient to induce liver injury. Moreover, traditional processing methods and herbal combinations in clinical settings can further alleviate this hepatotoxic risk. Nevertheless, owing to metabolic activation, it is essential to remain

Systematic identification of bacterial neuraminidase inhibitors from Psoralea corylifolia using ultrafiltration-UPLC-Q-Orbitrap-MS and molecular dynamics.

Bacterial neuraminidase (BNA) is a potential target for discovering anti-infective agents. Here, the BNA inhibitory activity of the ethanolic extract of Psoralea corylifolia (EEPC) was investigated using integrated experimental and computational approaches. Affinity ultrafiltration-UPLC-Q-Orbitrap-MS prioritised 11 representative constituents as putative BNA-interacting compounds. Five compounds (7-11) inhibited BNA with IC50 values of 4.67-34.09 μM. Bakuchiol (11) exhibited the strongest activity (IC50 = 4.67 ± 0.26 μM), and compounds 7, 9, and 10 were more active than the positive control, q
