Infection-induced viscerosensory signals from the gut enhance anxiety: implications for psychoneuroimmunology.

Infection and inflammation lead to changes in mood and cognition. Although the "classic" sickness behavior syndrome, involving fatigue, social withdrawal, and loss of appetites are most familiar, other emotional responses accompany immune activation, including anxiety. Recent studies have shown that gastrointestinal bacterial infections lead to enhanced anxiety-like behavior in mice. The bacteria-induced signal is most likely carried by vagal sensory neurons, and occurs early on (within 6h) during the infection. These signals induce evidence of activation in brain regions that integrate viscer

The neutrophil-to-lymphocyte ratio: a psychoneuroimmunological nexus linking depression and cardiovascular comorbidity.

BACKGROUND: A significant worldwide health concern linked to ongoing low-grade inflammatory processes is depression concomitant with cardiovascular disease (CVD). There aren't many pertinent clinical biomarkers, though. This study aims to clarify the underlying mechanisms of the neutrophil-to-lymphocyte ratio (NLR) and ascertain if it is associated with depression-CVD comorbidity. METHODS: Multivariable logistic regression and restricted cubic spline (RCS) analysis were used to examine the nonlinear connection between NLR and depression-cardiovascular comorbidity based on data from the Nation

At the extreme end of the psychoneuroimmunological spectrum: delirium as a maladaptive sickness behaviour response.

Delirium is a common and severe neuropsychiatric syndrome characterised by acute deterioration and fluctuations in mental status. It is precipitated mainly by acute illness, trauma, surgery, or drugs. Delirium affects around one in eight hospital inpatients and is associated with multiple adverse consequences, including new institutionalisation, worsening of existing dementia, and death. Patients with delirium show attentional and other cognitive deficits, altered alertness (mostly reduced, but some patients develop agitation and hyperactivity), altered sleep-wake cycle and psychoses. The path

Stress and cancer from an integrative psychoneuroimmunology perspective: Neurobiological mechanisms and clinical implications.

Growing evidence from preclinical and clinical studies suggests that psychosocial factors, including stress, influence cancer incidence, progression, and survival. From a psychoneuroimmunology perspective, this integrative review synthesizes the neurobiological mechanisms underlying stress-cancer interactions, together with the epidemiological evidence supporting this relationship and its methodological limitations. Chronic stress impairs tumor immunosurveillance by CD8⁺ T cells and natural killer cells through dysregulation of the hypothalamic-pituitary-adrenal axis and sympathetic activation

The Integrative Single-Case Design as a Biosemiotic-Systemic Research Tool in Psychoneuroimmunology.

Psychoneuroimmunology (PNI) is a field of research that deals with the interactions between psyche, nervous, endocrine, and immune systems. Investigating these complex PNI relationships under as ecologically valid as possible conditions ("life as it is lived") necessitates a paradigm change in research. This shift places factors such as "time" and "subjective meaning" of personal experiences at the center of the research methodology. For this purpose, the biosemiotic-systemic research design "Integrative Single-Case Study" was developed. Initial results from healthy individuals as well as pati

[Pruritus and stress: psychoneuroimmunological mechanisms, assessment of psychosocial stress, and therapeutic implications].

BACKGROUND: Chronic pruritus (CP) represents one of the most common and therapeutically challenging symptoms in dermatological care. Its relationship with psychosocial and symptom-related stress is bidirectional and clinically highly relevant: pruritus is significantly associated with psychosocial stress via the brain-skin axis and can be simultaneously exacerbated by psychosocial stress. At the behavioral level, scratching perpetuates this cycle. METHODS: This overview summarizes the current state of knowledge on psychoneuroimmunology in the context of pruritus, presents validated assessment

Nicotine from a Different Angle: Biological Effects from a Psychoneuroimmunological Perspective.

Statistical data demonstrate a concurrent rise in smoking prevalence and mental disorders, such as depression and anxiety, which may be attributed to contemporary lifestyle factors, including social media and recent global events. This indicates a potential correlation between these trends, as individuals with mental disorders may engage in smoking as a form of self-medication to alleviate anxiety. However, smoking is harmful and increases the risk of many diseases. Therefore, smoking cessation strategies are increasingly being considered. Nicotine is a naturally produced alkaloid in plants th

Chronic fatigue syndrome from vagus nerve infection: a psychoneuroimmunological hypothesis.

Chronic fatigue syndrome (CFS) is an often-debilitating condition of unknown origin. There is a general consensus among CFS researchers that the symptoms seem to reflect an ongoing immune response, perhaps due to viral infection. Thus, most CFS research has focused upon trying to uncover that putative immune system dysfunction or specific pathogenic agent. However, no single causative agent has been found. In this speculative article, I describe a new hypothesis for the etiology of CFS: infection of the vagus nerve. When immune cells of otherwise healthy individuals detect any peripheral infec

Psychoneuroimmunological correlates of persisting sciatic pain in patients who underwent discectomy.

Patients suffering from persisting sciatic pain 8 weeks following discectomy were compared with patients displaying low complaints and healthy, pain-free volunteers regarding their interleukin-6 (IL-6) levels, morning cortisol levels and degree of psychological distress. Whereas serum concentrations of IL-6 were measured by collecting blood samples between 0945 and 2400 h in intervals of 45 min, morning cortisol levels were obtained by sampling saliva on five ensuing measurements, beginning immediately after awakening. In addition, questionnaires aimed at measuring depressive mood, somatic sym

Perinatal Psychoneuroimmunology of Prenatal Stress and Its Effects on Fetal and Postnatal Brain Development.

Prenatal stress (PS) impacts early behavioral, neuroimmune, and cognitive development. Pregnant rat models have been very valuable in examining the mechanisms of such fetal programming. A pregnant sheep model of maternal stress offers the unique advantages of chronic in utero monitoring and manipulation. This chapter presents the techniques used to model single and multigenerational stress exposures and their pleiotropic effects on the offspring.

Psychoneuroimmunological changes in adults with obesity receiving chiropractic care: a single-arm pilot trial.

INTRODUCTION: Psychoneuroimmunology (PNI) is an interdisciplinary framework that merges psychology, neurology, and immunology. Obesity, a condition of increasing global prevalence, is associated with elevated risk of PNI dysfunction. The primary aim of this single-arm pilot trial was to assess the feasibility of a PNI-focused assessment battery in combination with community-based chiropractic care in adults (18-65 years) with obesity. Additionally, we explored potential changes in PNI-related outcomes. METHODS: Eighteen participants (12 females; mean age 48.7 ± 9.0 years) were enrolled and as

Psychoneuroimmunology-informed strategies for early detection of biological threat exposure: Insights from experimental rodent models.

The growing frequency of emerging infectious diseases, antimicrobial resistance, and accidental or deliberate biological threat releases underscores the urgent need for early detection of biological threat exposure. Current diagnostic approaches primarily rely on identification of specific pathogens or toxins, often after clinical symptoms have emerged. An alternative approach could exploit the body's conserved psychoneuroimmunological (PNI) response to biological threat. This approach could provide a host-centred framework capable of detecting real-time deviations from baseline PNI status, ev

The integration of cardiovascular behavioral medicine and psychoneuroimmunology: new developments based on converging research fields.

The immune system plays a role in the progression of coronary artery diseases and its clinical manifestations as acute coronary syndromes. It is well established that psychological factors can act as risk factors for acute coronary syndromes. This review describes psychoneuroimmunological pathways involved in coronary disease progression and documents that the stage of coronary disease is a major determinant of pathophysiological mechanisms accounting for the association between psychological risk factors, immune system parameters, and acute coronary syndromes. Chronic psychological risk facto

A psychoneuroimmunologic examination of cumulative perinatal steroid exposures and preterm infant behavioral follow-up.

PURPOSE: This study's aim was to explore relationships between preterm infant behavioral outcomes and maternal/infant glucocorticoid (dexamethasone [DEX]) treatments using a psychoneuroimmunologic approach. Research questions were (a) do relationships exist between infant cumulative perinatal steroid (PNS) exposure and child behavioral problems? and (b) do maternal/infant characteristics (e.g., immune markers and biophysiologic stressors) influence these relationships? METHODS: The convenience sample comprised 45 mother-child dyads in which the children (mean age 8 years ± 2.3) had been born

Beyond survival: psychoneuroimmunology-driven cancer psychological rehabilitation-a paradigm shift from symptom management to biobehavioral remodeling.

Cancer treatment paradigms have evolved from solely focusing on tumor eradication to optimizing survivorship trajectories. However, current psychological rehabilitation approaches predominantly target symptom relief, such as anxiety, depression, and fatigue, without direct evidence of influencing cancer's biological processes like recurrence, metastasis, or immune evasion. This review, grounded in psychoneuroimmunology, advocates for elevating psychological rehabilitation from an auxiliary support role to a synergistic component of antitumor therapy. Central to this perspective is redefining r

Through the Looking-Glass: Psychoneuroimmunology and the Microbiome-Gut-Brain Axis in the Modern Antiretroviral Therapy Era.

OBJECTIVE: Depression, substance use disorders, and other neuropsychiatric comorbidities are common in people with HIV (PWH), but the underlying mechanisms are not sufficiently understood. HIV-induced damage to the gastrointestinal tract potentiates residual immune dysregulation in PWH receiving effective antiretroviral therapy. However, few studies among PWH have examined the relevance of microbiome-gut-brain axis: bidirectional crosstalk between the gastrointestinal tract, immune system, and central nervous system. METHODS: A narrative review was conducted to integrate findings from 159 art

Psychoneuroimmunology in multiple myeloma and autologous hematopoietic stem cell transplant: Opportunities for research among patients and caregivers.

Multiple myeloma (MM) is an incurable cancer and is the leading indication for autologous hematopoietic stem cell transplantation (HSCT). To be eligible for HSCT, a patient must have a caregiver, as caregivers play a central role in HSCT preparation and recovery. MM patients remain on treatment indefinitely, and thus patients and their caregivers face long-term challenges including the intensity of HSCT and perpetual therapy after transplant. Importantly, both patients and their caregivers show heightened depressive and anxiety symptoms, with dyadic correspondence evidenced and caregivers' dis

From traditional warming and tonifying to psychoneuroimmunology: theoretical reconstruction and clinical application of moxibustion for cancer-related fatigue.

Cancer-related fatigue (CRF) is a multidimensional and highly prevalent symptom that severely impairs the quality of life in patients with cancer. Due to its complex etiology and the limited effectiveness of current therapeutic options, CRF remains a major clinical challenge. Moxibustion, a traditional thermal stimulation therapy characterized by its "warming and tonifying" effects in East Asian medicine, has shown potential in alleviating CRF, however, its underlying biological mechanisms remain insufficiently defined. This review aims to reconstruct the mechanistic basis of moxibustion in th
