Foliar application of plant-derived peptides decreases the severity of leaf rust (Puccinia triticina) infection in bread wheat (Triticum aestivum L.).

BACKGROUND: Screening and developing novel antifungal agents with minimal environmental impact are needed to maintain and increase crop production, which is constantly threatened by various pathogens. Small peptides with antimicrobial and antifungal activities have been known to play an important role in plant defense both at the pathogen level by suppressing its growth and proliferation as well as at the host level through activation or priming of the plant's immune system for a faster, more robust response against fungi. Rust fungi (Pucciniales) are plant pathogens that can infect key crops

Host resistance responses against Puccinia striiformis f. sp. tritici in wheat cultivars with different resistance levels: molecular, biochemical, and ultrastructural studies.

BACKGROUND: Yellow (stripe) rust of wheat, caused by Puccinia striiformis, is a serious disease that results in great economic losses. This study aimed to investigate the variation in plant responses in three wheat cultivars with different resistance levels against yellow rust. RESULTS: The highest disease severity was recorded for cv. Gemmieza-11 (95%), followed by cv. Shandweel 1 (60%), while the lowest was recorded for cv. Misr-3 (3%). qPCR results of the yellow rust-resistance genes Yr5, Yr10, Yr15, and Yr18 showed that the infection did not affect the expression of Yr5 and Yr15 in the in

Identification of physiological races of Puccinia striiformis f. sp. tritici and molecular docking of some biological treatments as prospective fungal inhibitor candidates in wheat.

Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst), represents one of the most significant challenges to the world's wheat production. This study aimed to investigate five aggressive physiological races of Pst impacting significant wheat cultivars in Egypt's northern Delta, which are essential for breeding programs. Moreover, the study evaluated the antifungal activity and mechanisms of action of chitosan nanoparticles, Trichoderma harzianum, and Sargassum latifolium against Pst using molecular docking analysis. During the 2024 growing season, an increase in the virulence of newly

Genetic behavior of controlling area under disease progress curve for stripe rust (Puccinia striiformis f. sp. tritici) in two wheat (Triticum aestivum) crosses.

Genetic effects on controlling resistance to stripe rust (Puccinia striiformis f. sp. tritici Eriksson)were determined in two wheat crosses, Bakhtawar-92 (B-92) x Frontana and Inqilab-91 x Fakhre Sarhad using area under the disease progress curve (AUDPC) as a measure of stripe rust resistance. The resistant and susceptible parents involved in developing genetic populations were identified by initial assessment of 45 wheat accessions for stripe rust reaction. Mixed inheritance model was applied to the data analysis of six basic populations (P(1), F(1), P(2), B(1), B(2), and F(2)) in the crosses

Genetic architecture and evolution of stripe rust resistance uncovered using diverse panels of wheat lines and North American Puccinia striiformis f. sp. tritici isolates.

Screening global wheat germplasm with a diverse collection of pathogen races expands the catalog of novel Yr loci and identifies new sources of broad-spectrum resistance against evolving Pst populations. Newly emerging highly virulent races of Puccinia striiformis f. sp. tritici (Pst) often defeat deployed resistance genes (Yr), highlighting the need for novel sources of durable resistance. A global diversity panel of 377 spring wheat (Triticum aestivum L.) lines was screened for all-stage resistance (ASR) against a panel of diverse 20 Pst isolates at the seedling stage and for adult-plant-sta

Cold-tolerant strains of Puccinia striiformis f.sp. tritici (MLG1 and MLG2) persist under snow, triggering early epidemic of stripe rust in the Xinjiang region, China.

Global warming has changed snowfall patterns in recent years worldwide, which affected the timing and severity of wheat stripe rust outbreaks. In wheat-growing areas of Xinjiang, China, where cold temperatures prevail and snow typically accumulates after cultivation, stripe rust epidemics have traditionally emerged after snowmelt. However, the recently delayed snowfall in some regions has led to earlier-than-expected outbreaks. The role of spores of different Puccinia striiformis f.sp. tritici (Pst) genotypes surviving under snow remains largely unexplored. We performed Pst sampling at three s

Genetic Background-Dependent Expression of Lr34, Lr67 and the Lr46-Associated Candidate Gene Lr46-Glu2 in Wheat (Triticum aestivum L.) Highlights Contrasting Transcriptional Responses Following Puccinia triticina Infection.

Adult plant resistance (APR) is widely used in wheat breeding, but its behaviour across genetic backgrounds remains poorly understood. In this study, we analysed the expression of three APR loci (Lr34, Lr46, Lr67) following their introgression into elite winter wheat cultivars. BC2F1 populations derived from crosses between donor lines and commercial cultivars were evaluated under controlled infection with Puccinia triticina (Pt). Gene expression was assessed using RT-qPCR, and miRNA abundance was quantified by ddPCR at five time points (0-48 hours post-inoculation, hpi). Expression patterns d

Virulence Phenotyping and Molecular Genotyping Reveal High Diversity Within and Strong Gene Flow Between the Puccinia striiformis f. sp. tritici Populations Collected from Barberry and Wheat in Shaanxi Province of China.

Emergence of new Puccinia striiformis f. sp. tritici races that overcome resistance of wheat cultivars is a challenging issue for wheat production. Although sexual reproduction of the fungus on barberry plants under field conditions in the spring in China has been reported, the diversity of the pathogen on barberry plants and the relationship to the population in wheat fields have not been determined. In the present study, two P. striiformis f. sp. tritici populations collected in western Shaanxi Province in May 2016, one from barberry plants (103 isolates) and the other from nearby wheat crop

Two ancient nuclear lineages and divergent reproductive strategies drive global success of the wheat stripe rust pathogen.

Plant pathogen evolution is driven by complex biological and ecological processes with major consequences for food security. Understanding how genetic diversity arises and is maintained is critical for predicting disease emergence. The wheat stripe rust fungus Puccinia striiformis f. sp. tritici is among the world's most destructive crop pathogens. As a dikaryotic fungus harboring two distinct haploid nuclei, Puccinia striiformis f. sp. tritici offers an excellent model to investigate nuclear-level evolution. Here, using population genomics of 507 global isolates and haplotype-phased genomes,

The control of Asian rust by glyphosate in glyphosate-resistant soybeans.

BACKGROUND: Glyphosate is a widely used broad-spectrum herbicide. Recent studies in glyphosate-resistant (GR) crops have shown that, in addition to its herbicidal activity, glyphosate exhibits activity against fungi, thereby providing disease control benefits. In GR wheat, glyphosate has shown both preventive and curative activities against Puccinia striiformis f. sp. tritici (Erikss) CO Johnston and Puccinia triticina Erikss, which cause stripe and leaf rusts respectively. RESULTS: Laboratory studies confirmed earlier observations that glyphosate has activity against Phakopsora pachyrhizi Sy

Glyphosate inhibits rust diseases in glyphosate-resistant wheat and soybean.

Glyphosate is a broad-spectrum herbicide used for the control of weeds in glyphosate-resistant crops. Glyphosate inhibits 5-enolpyruvyl shikimate 3-phosphate synthase, a key enzyme in the synthesis of aromatic amino acids in plants, fungi, and bacteria. Studies with glyphosate-resistant wheat have shown that glyphosate provided both preventive and curative activities against Puccinia striiformis f. sp. tritici and Puccinia triticina, which cause stripe and leaf rusts, respectively, in wheat. Growth-chamber studies demonstrated wheat rust control at multiple plant growth stages with a glyphosat

Mathematical Model Analysis for the Transmission Dynamics of Wheat Yellow Rust Disease: In Case of Melga Woreda, Sidama Regional State, Ethiopia.

In this work, we present a mathematical model that uses real data to characterize the transmission dynamics of wheat yellow rust (WYR) disease. The model's well-posedness, such as the existence and uniqueness, the positivity, and boundedness of the solutions of the model, is examined. Additionally, the study looks at qualitative evaluations such as the existence and stability of equilibrium points and uses the next-generation matrix to derive the basic reproduction number. The Routh-Hurwitz criteria were used to assert and demonstrate the local stability of the model's WYR free and endemic equ

Wheat Rust Diseases: Global Impact, Integrated Disease Management, and Emerging Biotechnological Innovations for Sustainable Crop Protection.

Wheat (Triticum aestivum L.) remains one of the most important staple crops worldwide and serves as a primary source of calories and protein for billions of people. Global wheat production, however, continues to be threatened by rust diseases caused by Puccinia triticina, Puccinia striiformis f. sp. tritici and Puccinia graminis f. sp. tritici. These pathogens are highly adaptive and frequently give rise to new virulent races, causing resistance in wheat cultivars to breakdown. Under favorable environmental conditions, severe epidemics can develop and substantial yield losses may occur. In res

Novel 5-(Trifluoromethyl)-1,2,4-oxadiazole-Based Pyrimidin-4-ether Histone Deacetylase Inhibitors for Controlling Rust Disease: Design, Synthesis, Activity, and Structure-Activity Relationship.

Rust disease, an important plant pathogen, can lead to reduced crop or fruit production. Trifluoromethyloxadiazole (TFMO) is a class of histone deacetylase inhibitors (HDACs). Herein, a series of 5-(trifluoromethyl)-1,2,4-oxadiazole (TFMO)-based pyrimidin-4-ether derivatives were designed and synthesized. Antirust bioassay results of TFMOs showed that some of them possessed excellent activities against plant rust pathogens, such as Puccinia sorghi, Phakopsora pachyrhizi, and Puccinia rubigo. The most active compound, 3-(5-(((6-(difluoromethyl)pyrimidin-4-yl)oxy)methyl)thiophen-2-yl)-5-(trifluo

Unlocking the potential of spring wheat genetic resources: uncovering resistance sources against leaf rust and yellow rust.

Integrating high-throughput phenotyping with multiple complementary GWAS models reduces method-dependent bias and enables reliable identification of novel loci and elite germplasm for durable yellow and leaf rust resistance in wheat. The causal agents of yellow and leaf rust in wheat, Puccinia striiformis f. sp. tritici and Puccinia triticina, pose a serious threat to grain yield and quality worldwide. Growing durable resistant wheat cultivars is an effective protection measure contributing to sustainable agriculture. Many of the known resistance genes, however, have been overcome due to the h

Rust Control in Glyphosate Tolerant Wheat Following Application of the Herbicide Glyphosate.

In greenhouse and field trials, transgenically modified wheat (Triticum aestivum) genotypes with tolerance to glyphosate had extremely low infection types to leaf rust caused by Puccinia triticina when treated with a labeled rate of the herbicide glyphosate prior to inoculation with leaf rust. A surfactant solution and a nonglyphosate herbicide had no effect on leaf rust development on the glyphosate tolerant wheat. Glyphosate had a systemic effect in reducing leaf rust development. The leaf rust control by glyphosate decreased with reduced application rates and longer periods of time between

Stripe Rust Virulence and Host Reaction Assessment Protocols: From Lab to Field.

Puccinia striiformis, the causal agent of stripe rust disease, poses a significant threat to cereal crops worldwide. Since 2000, the emergence of heat-adapted races/lineages of the wheat-infecting form P. striiformis f. sp. tritici (Pst) has enabled the disease to spread beyond its traditional high-elevation, cool, and moist regions. It can now occur wherever wheat is cultivated, making it a pandemic concern. This chapter provides an overview of the methodologies and practices established in our lab for working with this pathogen. These methodologies include isolate collection, recovery, purif

Marker-assisted backcross breeding to develop wheat lines with stable yellow rust resistance without yield penalties.

Wheat production faces severe yield threats globally and regionally in India from rust diseases caused by Puccinia species. Genetic resistance breeding is the most sustainable approach to mitigate these losses, but traditional phenotypic selection is often constrained by environmental fluctuations and rapid pathogen evolution. Here we utilized marker-assisted backcross breeding (MABB) to introgress broad-spectrum yellow rust resistance genes, Yr10 and Yr15, from the donor genetic stock PBW703 into the elite, high-yielding wheat cultivar DBW88 to develop two high-yielding rust-resistant genotyp

ZmTPS18-mediated β-caryophyllene biosynthesis contributes to resistance against southern corn rust in maize.

Southern corn rust (SCR), caused by the obligate biotrophic fungus Puccinia polysora Underwood, is a major foliar disease of maize associated with substantial yield losses in many production regions. Terpenoids are important components of plant defense, but the terpene synthases and upstream regulators involved in pathogen-induced terpenoid biosynthesis in maize are still poorly defined. In this study, volatile profiling showed that β-caryophyllene emission was induced in the SCR-resistant maize inbred line Yanzi F7016K after pathogen inoculation, reaching levels 3.18-fold higher than those in

Brachypodium distachyon. A new model system for functional genomics in grasses.

A new model for grass functional genomics is described based on Brachypodium distachyon, which in the evolution of the Pooideae diverged just prior to the clade of "core pooid" genera that contain the majority of important temperate cereals and forage grasses. Diploid ecotypes of B. distachyon (2n = 10) have five easily distinguishable chromosomes that display high levels of chiasma formation at meiosis. The B. distachyon nuclear genome was indistinguishable in size from that of Arabidopsis, making it the simplest genome described in grasses to date. B. distachyon is a self-fertile, inbreeding
