Pseudomonas aeruginosa is an opportunistic, Gram-negative bacterium, involved in severe infections associated with cystic fibrosis, pneumonia, burn wounds, ocular diseases, and immunosuppressive illnesses, and is a major cause of intrahospital infections. This bacterium is also one of the most commercially and biotechnologically significant microorganisms, since it can produce valuable biomolecules which represent a rich source of potential drug candidates. On the other hand, metal complexes have been used in medicine for both therapeutic and diagnostic purposes since ancient times. This class

Clearance of neutrophils from inflamed sites is critical for resolution of inflammation, but pathogen-driven neutrophil apoptosis can impair host defenses. We previously showed that pyocyanin, a phenazine toxic metabolite produced by Pseudomonas aeruginosa, accelerates neutrophil apoptosis in vitro. We compared wild-type and pyocyanin-deficient strains of P. aeruginosa in a murine model of acute pneumonia. Intratracheal instillation of either strain of P. aeruginosa caused a rapid increase in bronchoalveolar lavage neutrophil counts up to 18 h after infection. In wild-type infection, neutrophi

Pseudomonas aeruginosa, a ubiquitous opportunistic pathogen, coordinates virulence gene expression primarily through three quorum-sensing (QS) systems: Las, Rhl, and Pqs. Despite the functions of QS systems in P. aeruginosa being relatively well-documented, the regulatory mechanisms modulating their activities remain largely unexplored. Here, we isolated and characterized a Las-null strain, PA_HN008, which overproduces pyocyanin (PYO) at the late growth stage due to its active RhlI-RhlR-PqsE pathway. Leveraging PYO production as a phenotypic marker, we conducted a genome-wide screening for ups

Chlamydia is an obligate intracellular bacterial pathogen responsible for sexually transmitted infections and blindness worldwide. In our search for antichlamydial agents from natural sources, we identified pyocyanin, a phenazine derivative from Pseudomonas aeruginosa. Previous studies showed that pyocyanin disables elementary body (EB) infectivity at low concentrations and inhibits intracellular growth at higher concentrations. In the present study, we show that its antichlamydial activity is primarily attributed to the intracellular developmental stage. Complete inhibition of detectable infe

Camellia nitidissima Chi, known as a medicinal and edible plant in China, exhibits multiple bioactivities, especially antibacterial activity. In this study, we investigated the inhibitory effects of the dichloromethane fraction (DF) of C. nitidissima Chi flowers on the pyocyanin production, swarming motility, and swimming motility of Pseudomonas aeruginosa PAO1, at sub-minimum inhibitory concentrations. Results showed that the DF had a remarkable inhibitory effect on pyocyanin production without influencing P. aeruginosa PAO1 growth, and concentration-dependent inhibitory effects on swarming a

The aim of this study was to examine the effects of pyocyanin exposure on mitochondrial GSH, other cellular thiols (thioredoxin-1, Trx-1), and oxidant-sensitive signaling pathways hypoxia inducible factor (HIF-1α) and heme oxygenase (HO-1) in A549 and HBE cell lines. A549 human type II alveolar epithelial cells and human bronchial epithelial (HBE) cells were treated with varying concentrations of pyocyanin extracted from Pseudomonas aeruginosa bacteria. Cytoplasmic and mitochondrial thiols and oxidant sensitive signal transduction proteins (HIF-1α and HO-1) were measured. Exposure to pyocyanin

Previously, it was reported that natural phenazines are able to support the anaerobic survival of Pseudomonas aeruginosa PA14 cells via electron shuttling, with electrodes poised as the terminal oxidants (Y. Wang, S. E. Kern, and D. K. Newman, J Bacteriol 192:365-369, 2010, https://doi.org/10.1128/JB.01188-09). The present study shows that both pyocyanin (PYO) and 1-hydroxyphenazine (1-OHPHZ) promoted the anaerobic killing of PA14 Δphz cells presumably via a single-electron transfer reaction with ferrous iron. However, phenazine-1-carboxylic acid (PCA) did not affect anaerobic survival in the

Airways of immunocompromised patients, or individuals with cystic fibrosis (CF), are common ground for Pseudomonas aeruginosa and Aspergillus fumigatus infections. Hence, in such a microenvironment both pathogens compete for resources. While under limiting iron conditions the siderophore pyoverdine is the most effective antifungal P. aeruginosa product, we now provide evidence that under nonlimiting iron conditions P. aeruginosa supernatants lack pyoverdine but still possess considerable antifungal activity. Spectrometric analyses of P. aeruginosa supernatants revealed the presence of phenazin

The human pathogen Pseudomonas aeruginosa is particularly notorious for its multiple resistance mechanisms. A new concept for anti-infectives is the "pathoblocker" approach, which targets virulence factors to disarm rather than kill pathogens and thus attenuates the development of resistance. Based on the estrogen receptor modulator raloxifene, which had previously been identified as a potential biosynthesis inhibitor of the virulence factor pyocyanin via in silico screening, analogues have been developed as pathoblockers against P. aeruginosa. These compounds reduce the production of pyocyani

BACKGROUND: Tackling the high demand for alternative therapeutic options, especially for multi-drug-resistant organisms, has awakened interest in naturally originated biomolecules such as pyocyanin (PYO). Herein, PYO-producing bacterium was isolated and PYO was extracted, characterized, and its antagonistic potency was scrutinized against a vast array of prokaryotic and eukaryotic water pathogens, either in free-living or biofilm lifestyles, both in vitro and in vivo. RESULTS: Initially, Pseudomonas sp. MPF-2 that was screened from agricultural soil for PYO production, was characterized and id

The rhizobacterium Pseudomonas aeruginosa 7NSK2 produces secondary metabolites such as pyochelin (Pch), its precursor salicylic acid (SA), and the phenazine compound pyocyanin. Both 7NSK2 and mutant KMPCH (Pch-negative, SA-positive) induced resistance to Botrytis cinerea in wild-type but not in transgenic NahG tomato. SA-negative mutants of both strains lost the capacity to induce resistance. On tomato roots, KMPCH produced SA and induced phenylalanine ammonia lyase activity, while this was not the case for 7NSK2. In 7NSK2, SA is probably very efficiently converted to Pch. However, Pch alone a

Pyocyanin is the primary phenazine produced by Pseudomonas aeruginosa, which possesses redox activity. This compound contributes to its virulence by generating reactive oxygen species and inducing oxidative stress in prokaryotic and eukaryotic cells. P. aeruginosa is able to resist high concentrations of pyocyanin despite its toxicity. In this work, we employed P. aeruginosa ID4365 and PAO1 rsmA mutants, which produce 276.2 μM and 6.9 μM of pyocyanin, respectively, to conduct a proteomic analysis in order to identify proteins involved in the protective response against this phenazine. We found

The redox-active exotoxin pyocyanin (PCN) can be recovered in 100 µM concentrations in the sputa of bronchiectasis patients chronically infected with Pseudomonas aeruginosa (PA). However, the importance of PCN within bronchiectatic airways colonized by PA remains unrecognized. Recently, we have shown that PCN is required for chronic PA lung infection in mice, and that chronic instillation of PCN induces goblet cell hyperplasia (GCH), pulmonary fibrosis, emphysema and influx of immune cells in mouse airways. Many of these pathological features are strikingly similar to the mouse airways devoid

In response to the urgent requirement for rapid, precise, and cost-effective detection in intensive care units (ICUs) for ventilated patients, as well as the need to overcome the limitations of traditional detection methods, researchers have turned their attention towards advancing novel technologies. Among these, biosensors have emerged as a reliable platform for achieving accurate and early diagnoses. In this study, we explore the possibility of using Pyocyanin analysis for early detection of pathogens in ventilator-associated pneumonia (VAP) and lower respiratory tract infections in ventila

Pseudomonas aeruginosa, a leading cause of sepsis, produces pyocyanin, a blue-pigmented virulence factor. Sepsis is associated with cachexia, but mechanisms are unknown and conventional nutrition approaches are not effective treatments. Pyocyanin has affinity for the aryl hydrocarbon receptor (AhR), which is expressed on adipocytes and regulates adipocyte differentiation. The purpose of this study was to define in vitro and in vivo effects of pyocyanin on adipocyte differentiation and body weight regulation as relates to septic cachexia. In 3T3-L1 preadipocytes, pyocyanin activated AhR and its

Phenazines are compounds of medical and biotechnological interest due to their antimicrobial and antitumoral properties. Pseudomonas aeruginosa, a Gram-negative bacterium, naturally produces phenazines such as pyocyanin and phenazine-1-carboxylic acid (PCA). These phenazines can generate reactive oxygen species, which increase oxidative stress in susceptible cells and ultimately lead to their death. Pyocyanin can inhibit the growth of human bacterial pathogens, while PCA serves as a potent antifungal agent that can prevent and control crop diseases. The environmental strain P. aeruginosa ID436

A series of sulfonamide acetamide derivatives 3-14 were synthesized and evaluated for their antibacterial and anti-virulence activities. The target compounds were initially screened in vitro for their antibacterial activity against multidrug-resistant strains of Gram-positive and Gram-negative bacteria. Compounds 9, 10 and 13 showed significant antibacterial activity with bactericidal effects, compared to the standard antibiotics, ampicillin and sulfamethoxazole. These derivatives were then further tested for their antibiofilm activity against biofilm-forming S. aureus and P. aeruginosa. At su

Combating Pseudomonas aeruginosa infection is challenging. It secretes pyocyanin (PCN) pigment that contributes to its virulence. Neutralizing PCN via reaction with thiol-containing compounds may represent a potential therapeutic option. This study investigates the neutralization reaction between PCN and N-acetyl cysteine (NAC) for bacterial inhibition and explores its mechanism of action. The neutralization adduct (PCN-NAC) was synthesized by reacting the purified PCN and NAC. The adduct was analyzed and its structure was elucidated. LC-MS/MS method was developed for the determination of PCN-

The present study investigated the effects of dietary supplementation with pyocyanin powder (PYP), a microbial-derived bioactive compound, and green coffee powder (GCP) as natural feed additives on growth performance, carcass traits, blood biochemical parameters, antioxidant status, meat quality, and cecal microbial populations in broiler chickens. A total of 300 one-week-old Ross 308 broiler chicks were assigned to five dietary treatments in a completely randomized design, with five replicates of 12 birds each, and reared until 35 days of age. The dietary treatments consisted of a basal diet
