Effects of ketamine and propofol on muscarinic plateau potentials in rat neocortical pyramidal cells.

Propofol and ketamine are widely used general anaesthetics, but have different effects on consciousness: propofol gives a deeply unconscious state, with little or no dream reports, whereas vivid dreams are often reported after ketamine anaesthesia. Ketamine is an N-methyl-D-aspartate (NMDA) receptor antagonist, while propofol is a γ-aminobutyric-acid (GABAA) receptor positive allosteric modulator, but these mechanisms do not fully explain how these drugs alter consciousness. Most previous in vitro studies of cellular mechanisms of anaesthetics have used brain slices or neurons in a nearly "com

Altered dendritic morphology of MEC II pyramidal and stellate cells in Rett syndrome mice.

INTRODUCTION: Mutations in the methyl-CpG-binding protein-2 gene (MECP2), which cause Rett syndrome (RTT), disrupt neuronal activity; however, the impact of the MECP2 loss-of-function on the cytoarchitecture of medial entorhinal cortex layer II (MECII) neurons-crucial for spatial memory and learning-remains poorly understood. METHODS: In this study, we utilized Golgi staining and neuron tracing in the Mecp2+/- mouse model of RTT to investigate the pyramidal and stellate cell alterations in MECII. RESULTS AND DISCUSSION: Our findings revealed that pyramidal cells displayed a significant reduc

Dendritic development of hippocampal CA1 pyramidal cells in a neonatal hypoxia-ischemia injury model.

It is believed that neonatal hypoxia-ischemia (HI) brain injury causes neuron loss and brain functional defects. However, the effect of HI brain injury on dendritic development of the remaining pyramidal cells of the hippocampus and the reaction of contralateral hippocampal neurons require further studies. The Morris water maze and Golgi-Cox staining were used to evaluate the learning and memory and dendritic morphology of pyramidal cells. The results of Golgi-Cox staining showed CA1 pyramidal neurons of HI injury models with fewer bifurcations and shorter dendrite length than the naive contro

Kv7/M channel dysfunction produces hyperexcitability in hippocampal CA1 pyramidal cells of Fmr1 knockout mice.

Fragile X syndrome (FXS), the most frequent monogenic form of intellectual disability, is caused by transcriptional silencing of the FMR1 gene that could render neuronal hyperexcitability. Here we show that pyramidal cells (PCs) in the dorsal CA1 region of the hippocampus elicited a larger action potential (AP) number in response to suprathreshold stimulation in juvenile Fmr1 knockout (KO) than wild-type (WT) mice. Because Kv7/M channels modulate CA1 PC excitability in rats, we investigated if their dysfunction produces neuronal hyperexcitability in Fmr1 KO mice. Immunohistochemical and wester

Type I TARPs promote dendritic growth of early postnatal neocortical pyramidal cells in organotypic cultures.

The ionotropic α-amino-3-hydroxy-5-methyl-4-isoxazole propionate glutamate receptors (AMPARs) have been implicated in the establishment of dendritic architecture. The transmembrane AMPA receptor regulatory proteins (TARPs) regulate AMPAR function and trafficking into synaptic membranes. In the current study, we employ type I and type II TARPs to modulate expression levels and function of endogenous AMPARs and investigate in organotypic cultures (OTCs) of rat occipital cortex whether this influences neuronal differentiation. Our results show that in early development [5-10 days in vitro (DIV)]

Cellular correlates of anxiety in CA1 hippocampal pyramidal cells of 5-HT1A receptor knockout mice.

RATIONALE: 5-HT(1A) receptor knockout (1AKO) mice have a robust anxiety phenotype. Tissue-specific "rescue" strategies and electrophysiology have implicated a critical role for postsynaptic 5-HT(1A) receptors, particularly in the CA1 region of the hippocampus. OBJECTIVES: In this study, we evaluated differences in membrane properties and synaptic activity in CA1 hippocampal pyramidal cells between 1AKOs and wild-type (WT) controls to better understand the cellular correlates of anxiety in this mouse model. METHODS: Whole-cell patch-clamp recordings were conducted in CA1 pyramidal cells in hi

Hyperexcitability in hippocampal CA1 pyramidal cells of Fmr1 knockout mice emerges during late postnatal development.

Fragile X syndrome (FXS) is a neurodevelopmental disorder and the most frequent monogenic form of intellectual disability and autism. Fmr1 knockout (KO) mice have been used to study the neuronal bases of FXS. Alterations in both synaptic plasticity and electrophysiological membrane properties have been reported in young Fmr1 KO mice, but current knowledge about when and how these functional alterations emerge during postnatal development is almost lacking. To address this issue, we compared subthreshold and suprathreshold membrane properties at 8 to 28 postnatal days (P8-28) in pyramidal cells

An increased excitation and inhibition onto CA1 pyramidal cells sets the path to Alzheimer's disease.

Synapses are critical targets of Alzheimer's disease (AD), a highly prevalent neurodegenerative disease associated with accumulation of extracellular amyloid-β peptides. Although amyloidosis and aggregation of the 42-amino acid amyloid-β (Aβ 42 ) have long been considered pathogenic triggers for AD, clinical evidence linking high levels of Aβ 42 with normal cognition challenges this hypothesis. To resolve this conundrum on the role of Aβ 42 in regulating synaptic activity, we used an adeno-associated viral vector approach that triggers extracellular accumulation of Aβ 42 and spatial memory imp

Age-related plasticity of the axon initial segment of cortical pyramidal cells in marmoset monkeys.

Structural plasticity of the axon initial segment (AIS), the site of action potential initiation, is observed as part of the normal early development of the cortex, as well as in association with injury and disease. Here, we show that structural AIS plasticity also occurs with normal aging in adult marmosets. Immunohistochemical techniques were used to reveal the extent of the AIS of layer 2/3A pyramidal cells in 8 neocortical areas. We found that the AIS length varied significantly between areas in young adult (2-3 years old) marmosets, with neurons in frontal area 14C having the longest AIS,

Altered expression of CDC42 signaling pathway components in cortical layer 3 pyramidal cells in schizophrenia.

BACKGROUND: Cognitive dysfunction in schizophrenia is associated with a lower density of dendritic spines on deep layer 3 pyramidal cells in the dorsolateral prefrontal cortex (DLPFC). These alterations appear to reflect dysregulation of the actin cytoskeleton required for spine formation and maintenance. Consistent with this idea, altered expression of genes in the cell division cycle 42 (CDC42)-CDC42 effector protein (CDC42EP) signaling pathway, a key organizer of the actin cytoskeleton, was previously reported in DLPFC gray matter from subjects with schizophrenia. We examined the integrity

Somatodendritic integration under increased network activity in layer 5 pyramidal cells of the somatosensory cortex.

Integrative properties of single neurons have been extensively studied in acute brain slices. However, these preparations are characterized by extremely low levels of synaptic and action potential activity. In comparison to in vivo, reduced intracortical input and lack of subcortical modulation increase the effective difference between mean membrane potential and spiking threshold, preventing self-sustained network activity in vitro. To elicit an increased and stable network activity (INA) in vitro comparable to that found in awake animals, we mimicked subcortical cholinergic and serotoninergi

Properties of action-potential initiation in neocortical pyramidal cells: evidence from whole cell axon recordings.

Cortical pyramidal cells are constantly bombarded by synaptic activity, much of which arises from other cortical neurons, both in normal conditions and during epileptic seizures. The action potentials generated by barrages of synaptic activity may exhibit a variable site of origin. Here we performed simultaneous whole cell recordings from the soma and axon or soma and apical dendrite of layer 5 pyramidal neurons during normal recurrent network activity (up states), the intrasomatic or intradendritic injection of artificial synaptic barrages, and during epileptiform discharges in vitro. We demo

Excitatory GABA input directly drives seizure-like rhythmic synchronization in mature hippocampal CA1 pyramidal cells.

GABA, which generally mediates inhibitory synaptic transmissions, occasionally acts as an excitatory transmitter through intense GABA(A) receptor activation even in adult animals. The excitatory effect results from alterations in the gradients of chloride, bicarbonate, and potassium ions, but its functional role still remains a mystery. Here we show that such GABAergic excitation participates in the expression of seizure-like rhythmic synchronization (afterdischarge) in the mature hippocampal CA1 region. Seizure-like afterdischarge was induced by high-frequency synaptic stimulation in the rat

Apamin-sensitive conductance mediates the K(+) current response during chemical ischemia in CA3 pyramidal cells.

Pyramidal cells typically respond to ischemia with initial transient hyperpolarization, which may represent a neuroprotective response. To identify the conductance underlying this hyperpolarization in CA3 pyramidal neurons of rat hippocampal organotypic slice cultures, recordings were obtained using the single-electrode voltage-clamp technique. Brief chemical ischemia (2 mM 2-deoxyglucose and 3 mM NaN(3), for 4 min) induced a response mediated by an increase in K(+) conductance. This current was blocked by intracellular application of the Ca(2+) chelator, bis-(o-aminophenoxy)-N,N,N', N'-tetraa

[Dendrite-morphometry of lamina-V-pyramidal cells in premature brain damaged humans--a Golgi study].

Specimens from the area 10 of the frontal cortex collected out of 7 patients with severe mental retardation, aged 3 to 24 years and also 8 non-neuropsychiatric age-matched controls were processed according to the Golgi-Kopsch method. The total length of the apical side-dendrites and of the basal dendrites was measured on layer V pyramidal neurons. Compared with controls the total dendritic length of the apical side-dendrites as well as of the basal dendrites was significant reduced in the affected patients (p less than 0.001). The reduced dendritic length represents perhaps a morphological cor

Variation in Pyramidal Cell Morphology Across the Human Anterior Temporal Lobe.

Pyramidal neurons are the most abundant and characteristic neuronal type in the cerebral cortex and their dendritic spines are the main postsynaptic elements of cortical excitatory synapses. Previous studies have shown that pyramidal cell structure differs across layers, cortical areas, and species. However, within the human cortex, the pyramidal dendritic morphology has been quantified in detail in relatively few cortical areas. In the present work, we performed intracellular injections of Lucifer Yellow at several distances from the temporal pole. We found regional differences in pyramidal c

Pyramidal cell specialization in the occipitotemporal cortex of the Chacma baboon (Papio ursinus).

Pyramidal cell structure varies systematically in occipitotemporal visual areas in monkeys. The dendritic trees of pyramidal cells, on average, become larger, more branched and more spinous with progression from the primary visual area (V1) to the second visual area (V2), the fourth (V4, or dorsolateral DL visual area) and inferotemporal (IT) cortex. Presently available data reveal that the extent of this increase in complexity parallels the expansion of occipitotemporal cortex. Here we extend the basis for comparison by studying pyramidal cell structure in occipitotemporal cortical areas in t

Alzheimer's disease and its progression reduce pyramidal cell gain and connectivity.

INTRODUCTION: Alzheimer's disease (AD) affects neurophysiology by loss of neurons, synapses, and neurotransmitters. A mechanistic understanding of the human disease will facilitate new treatments. METHODS: Magnetoencephalography was recorded during an auditory mismatch negativity paradigm from healthy adults (n = 14) and people with symptomatic AD (n = 45, amyloid biomarker positive) at baseline and after 16 months. Fourteen people with AD had repeat magnetoencephalography at 2 weeks to assess test-retest reliability. Dynamic causal models were fitted to the evoked responses and analyzed usin

Implications of altered pyramidal cell morphology on clinical symptoms of neurodevelopmental disorders.

The prevalence of pyramidal cells (PCs) in the mammalian cerebral cortex underscore their value as they play a crucial role in various brain functions, ranging from cognition, sensory processing, to motor output. PC morphology significantly influences brain connectivity and plays a critical role in maintaining normal brain function. Pathological alterations to PC morphology are thought to contribute to the aetiology of neurodevelopmental disorders such as autism spectrum disorder (ASD) and schizophrenia. This review explores the relationship between abnormalities in PC morphology in key cortic
