Comparison of drug-induced responses of rabbit trachea and bronchus.

The effect of various smooth muscle stimulants and relaxants was examined on isolated rabbit trachea and bronchus. Trachea contracted maximally in response to carbachol, slightly to KCl, and there was no response to serotonin or PGF2alpha. Relaxation of carbachol-contracted trachea was elicited by papaverine, aminophylline, isoproterenol, bradykinin, or PGE2. Histamine also relaxed the rabbit trachea. However, bronchi from the same animal contracted to this amine. As was the case with trachea, the bronchi contracted maximally to carbachol and slightly to KCl; serotonin and PGF2alpha were inact

Effects of histamine H(1) receptor antagonists on delayed spatial win-shift task in rats.

We investigated the effects of histamine H(1) receptor antagonists in 3 different stages of spatial working memory in comparison with scopolamine. The effects of drugs were measured using an eight-arm radial maze with a delayed spatial win-shift task in rats. Drugs were injected 10 min before the training phase (timing 1), immediately after the training phase (timing 2), or 10 min before the test phase (timing 3). Intrahippocampal injection of pyrilamine and diphenhydramine resulted in significant effects on the number of correct choices and across-phase errors in all 3 timings. Scopolamine im

Ameliorative effects of histamine on spatial memory deficits induced by scopolamine infusion into bilateral dorsal or ventral hippocampus as evaluated by the radial arm maze task.

1. The present study examined the role of the hippocampal histaminergic system in the regulation of spatial memory deficit in rats using the radial arm maze task after scopolamine injection into the bilateral dorsal (DH) or ventral (VH) hippocampus. 2. Bilateral injection of scopolamine (5 microg/site) into both the DH and VH impaired spatial memory in the retrieval memory process. Injection of histamine (50 or 100 ng/site) in the DH and intraperitoneal injection of histidine (100 mg/kg) markedly improved working memory and reference memory deficits induced by scopolamine injection into the DH

Role of brain histamine H1- and H2-receptors in neostigmine-induced hyperglycemia in rats.

We previously reported that when neostigmine, an inhibitor of acetylcholine esterase, was injected into the third cerebral ventricle, the concentration of hepatic venous plasma glucose was increased via central muscarinic receptors in anesthetized rats. To determine whether brain histamine receptors are involved in cholinergic system transmission with regard to central nervous system (CNS)-mediated glucoregulation, we examined the effects of the H1 receptor antagonist pyrilamine and the H2 receptor antagonist ranitidine on neostigmine-induced hyperglycemia in anesthetized rats. The injection o

Effects of the NMDA antagonist MK-801 on radial maze performance in histidine-deficient rats.

We examined the effects of a histidine-deficient diet on brain histamine contents as well as on learning and memory using the eight-arm radial maze in rats. A significant decrease in histamine content in the hippocampus was observed after long-term feeding of rats with a histidine-deficient diet. At the same time, significant enhancement of the acquisition process in radial maze performance was also observed. Pyrilamine did not show a significant effect on radial maze performance in histidine-deficient rats. On the other hand, pyrilamine caused a significant spatial memory deficit in control r

Slow wave sleep-inducing effects of first generation H1-antagonists.

The present study was performed to see if first-generation histamine H1-antagonists are useful sedative-hypnotic drugs. Increases in electroencephalogram (EEG) power spectra of the delta band (0-4 Hz) at the frontal cortex and theta band (4-8 Hz) at the hippocampus in rats were used as an indexes of sleep. The H1-antagonists used in this study resulted in a decrease in sleep latency and an increase in sleep duration (slow wave sleep). The rate of REM (rapid eye movement) sleep during slow wave sleep was decreased by H1-antagonists and brotizolam. The order of potency of H1-antagonists for the

The major role of peripheral release of histamine and 5-hydroxytryptamine in formalin-induced nociception.

Formalin injected subcutaneously into the paw is a widely used model of pain. This procedure evokes a short-lasting period of flinching (phase 1) and a long-lasting period of intense flinching (phase 2) following a very short period of quiescence. Phase 2 has been extensively used to support the involvement of central (spinal cord) sensitization in inflammatory hyperalgesia. The present study evaluated the contribution of stimulation of peripheral nociceptors by the release of endogenous mediators at the site of lesion. The participation of histamine and 5-hydroxytryptamine was demonstrated by

Increased levels of extracellular dopamine in neostriatum and nucleus accumbens after histamine H1 receptor blockade.

The dopaminergic system plays a central role in the processing of reward or reinforcement since drugs that have reinforcing properties all share the ability to elevate dopamine (DA) levels in the nucleus accumbens or neostriatum. Histamine H1 receptor antagonists are known to have reinforcing effects in humans and laboratory rats. Here, we examined the effect of systemic (i.p.) treatment with two H1 antagonists, chlorpheniramine and pyrilamine, on the extracellular levels of DA and its metabolites dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in the neostriatum and nucleus acc

Effects of H1- and H2-histamine receptor agonists and antagonists on sleep and wakefulness in the rat.

The H1-receptor agonist 2-thiazolylethylamine (2-TEA) given by i.c.v. route dose-dependently increased wakefulness (W) and decreased NREM sleep (NREMS) and REM sleep (REMS) in rats prepared for chronic sleep recordings. The H1-receptor antagonists pyrilamine and diphenhydramine given by i.p. route decreased W and increased NREMS. Pyrilamine prevented the increase of W and decrease of NREMS produced by 2-TEA. However, REMS reduction was not antagonized, what tends to suggest that two different mechanisms could be involved in the 2-TEA-induced effects on NREMS and REMS. Cimetidine which blocks H

Structural Requirements for Uptake of Diphenhydramine Analogs into hCMEC/D3 Cells Via the Proton-Coupled Organic Cation Antiporter.

There is increasing evidence that a proton-coupled organic cation (H+/OC) antiporter facilitates uptake of various central nervous system-active drugs, such as the histamine H1 receptor antagonist diphenhydramine, into the brain. The purpose of this study was to clarify the structural requirements for H+/OC antiporter-mediated uptake into hCMEC/D3 cells, an established in vitro model of the human blood-brain barrier, by using a series of diphenhydramine analogs. For this purpose, we synthesized seven tertiary amine analogs and three amide analogs. Uptake of all the amines was facilitated by an

[Roles of histamine in the exacerbated allergic dermatitis].

We established a novel dermatitis model in mice earlobes and analyzed the roles of histamine using specific antagonists for histamine receptors. After sensitization with picryl chloride (PiCl) by painting it on the earlobes of cyclophosphamide-treated mice, 12-O-tetradecanoylphorbol 13-acetate (TPA) was painted twice at the same site, and then allergic inflammation was induced by painting with PiCl. Histamine antagonists and cyclosporin A were administered i.v. The application of TPA shifted the PiCl-induced allergic inflammation from a delayed-type response to a biphasic response and increase

Different roles of histamine receptor subtypes in ampullar & isthmic segments of human fallopian tube.

We have studied the influence of both menstrual cycle phase and subtype-selective histamine antagonists on the contractions produced by histamine isolated in human fallopian tubes. Histamine produced tonic contractions of the isolated preparations of both the ampullar and isthmic segments of the human fallopian tube were analyzed. The effects of both histamine and its antagonists were not dependent on the phase of the menstrual cycle. The isolated preparations of the ampullar segment from post menopausal patients were not responsive on histamine. When responding, the ampullar segments were sli

Intestinal mucosal lymphocytes have H1 receptors: H1 antagonists reduce their proliferation and cytotoxicity.

Histamine and H1 antagonists inhibit the proliferation and natural killer activity of peripheral blood mononuclear cells, which express large numbers of H1 receptors. This study examined the presence of H1 receptors and the effects of histamine, H1 antagonists (pyrilamine and diphenhydramine), and H2 antagonists (cimetidine and ranitidine) on human intestinal lymphocyte proliferation and cytotoxicity. Intestinal lymphocytes were obtained by chemical and enzymatic treatment of surgical specimens and purified by Percoll density gradient centrifugation. Proliferation was measured by [3H]thymidine

Effect of H1 blockers alone and in combination with morphine to produce antinociception in mice.

Antinociception was assessed in male CD-1 mice by a modification of Haffner's tail-clamp procedure. The H1 blockers, including an ethylenediamine (pyrilamine), an ethanolamine (diphenhydramine), a phenothiazine (methdilazine), a piperazine (cyclizine) and an alkylamine (chlorpheniramine), all produced antinociception when given alone to mice and also caused potentiation when combined with morphine.

Characterization of a primate model of asthma using anti-allergy/anti-asthma agents.

The following study was performed to further characterize a primate model of asthma using classes of drugs that target allergy (pyrilamine, cetirizine), are bronchodilators for the treatment of asthma (salbutamol, salmeterol) or are anti-inflammatory (dexamethasone). These drugs were examined for their ability to inhibit acute, antigen-induced bronchoconstriction, the development of airway hyperresponsiveness (AHR) and the infiltration of leukocytes into the lungs of atopic cynomolgus monkeys (Macaca facsicularis) using a 10-day, multiple antigen (Ag) challenge protocol. Quantitation of phototoxic hyperemia and permeability to protein: II. Inhibition by histamine (H1 and H2) receptor antagonists in mouse skin.

Exposure of anthracene treated skin to long-wave ultraviolet radiation (UVA) will produce hyperemia, increased vascular permeability, burning and itching. This photosensitized inflammation was produced in the skin of hairless mice by the topical application of a 0.025% (w/v) solution of anthracene followed by exposure to UVA. The resultant hyperemia was quantified by the increase in 51Cr-erythrocyte content (cpm/wet weight) of skin. After 1000 sec (1.3 x 10(4)J/M2) of UVA, the blood content of the skin had increased nine-fold. Pyrilamine (H1 antagonist), administered orally, did not inhibit th

Restraint stress stimulation of prolactin and ACTH secretion: role of brain histamine.

The possible role of brain histamine in the release of prolactin, ACTH and corticosterone following acute restraint, was pharmacologically evaluated in adult male rats. Fifteen min of restraint caused marked increases in the plasma levels of these hormones. alpha-Fluoromethyl histidine (FH), a histidine decarboxylase inhibitor which depleted hypothalamic histamine, inhibited the enhancement of plasma prolactin levels. In contrast, plasma ACTH levels were not modified. FH treatment decreased plasma corticosterone concentrations in animals submitted to stress or in rest; this suggests a direct a

No development of tolerance to analgesia by repeated administration of H1 antagonists.

The effect produced by repeated administration of the H1-antihistaminics diphenhydramine, promethazine and pyrilamine on their ability to induce antinociception was evaluated in the mouse hot-plate and abdominal constriction tests. Contrary to morphine, baclofen and oxotremorine, the three H1 antagonists investigated did not promote the development of tolerance to the analgesic effect after 14 days of repeated treatment. H1 antagonists could, therefore, represent a promising pharmacological approach to the management of chronic pain.
