C7-Substituted Quinolines as Potent Inhibitors of AdeG Efflux Pumps in Acinetobacter baumannii.

Efflux, mediated by a series of multidrug efflux pumps, is a major contributor to antibiotic resistance in Gram-negative bacteria. Efflux pump inhibitors (EPIs), which can block efflux, have the potential to be used as adjuvant therapies to resensitize bacteria to existing antibiotics. In this study, 36 quinoline-based compounds were synthesized as potential EPIs targeting resistance nodulation division (RND) family pumps in the multidrug-resistant pathogen Acinetobacter baumannii. In A. baumannii strains with overexpressed AdeFGH (chloramphenicol-adapted) and AdeABC (AYE, Ab5075-UW), these co

Structure-activity relationship and target investigation of 2-aryl quinolines with nematocidal activity.

Within the context of our anthelmintic discovery program, we recently identified and evaluated a quinoline derivative, called ABX464 or obefazimod, as a nematocidal candidate; synthesised a series of analogues which were assessed for activity against the free-living nematode Caenorhabditis elegans; and predicted compound-target relationships by thermal proteome profiling (TPP) and in silico docking. Here, we logically extended this work and critically evaluated the anthelmintic activity of ABX464 analogues on Haemonchus contortus (barber's pole worm) - a highly pathogenic nematode of ruminant

Novel tricyclic pyrrolo-quinolines as pharmacological correctors of the mutant CFTR chloride channel.

F508del, the most frequent mutation in cystic fibrosis (CF), impairs the stability and folding of the CFTR chloride channel, thus resulting in intracellular retention and CFTR degradation. The F508del defect can be targeted with pharmacological correctors, such as VX-809 and VX-445, that stabilize CFTR and improve its trafficking to plasma membrane. Using a functional test to evaluate a panel of chemical compounds, we have identified tricyclic pyrrolo-quinolines as novel F508del correctors with high efficacy on primary airway epithelial cells from CF patients. The most effective compound, PP02

In Vitro and In Vivo Studies of the Trypanocidal Effect of Novel Quinolines.

Therapies for human African trypanosomiasis and Chagas disease, caused by Trypanosoma brucei and Trypanosoma cruzi, respectively, are limited, providing minimal therapeutic options for the millions of individuals living in very poor communities. Here the effects of 10 novel quinolines are evaluated in silico and by phenotypic studies using in vitro and in vivo models. Absorption, distribution, metabolism, excretion, and toxicity (ADMET) properties revealed that most molecules did not infringe on Lipinski's rules, which is a prediction of good oral absorption. These quinolines showed high proba

Carbon annulation of ortho-vinylanilines with dimethyl sulfoxide to access 4-aryl quinolines.

A palladium-catalyzed annulation of ortho-vinylanilines with dimethyl sulfoxide was developed to access 4-aryl quinolines in moderate to good yields. Activated by 1,4-diazabicyclo[2.2.2]octane bis(sulfur dioxide) adduct (DABSO), DMSO served as a "[double bond, length as m-dash]CH-" fragment in this transformation. It represents a facile pathway leading to 4-aryl quinolines.

ATP Synthase Inhibitors as Anti-tubercular Agents: QSAR Studies in Novel Substituted Quinolines.

BACKGROUND: Tuberculosis (TB) is a major infectious disease caused by Mycobacterium Tuberculosis. As per the World Health Organization (WHO) report of 2019, there were 1.5 million deaths in the year 2018, mainly because of multi- and extensively drug-resistant tuberculosis (MDR & XDR-TB). Among several antitubercular drugs in clinical trials, bedaquiline (TMC207) is a highly promising drug that was approved by the FDA in 2012 and marketed in 2016 for the treatment of multidrug resistant TB in combination with other drugs. Bedaquiline acts on mycobacterial ATP synthase and is highly effective i

Determination of aniline and quinoline compounds in textiles.

A simple method for simultaneous determination of twenty-one analytes, belonging to two classes of compounds, aromatic amines and quinolines, is presented. Several of the analytes considered in this study frequently occur in textiles goods on the open market and have been related to allergic contact dermatitis and/or are proven or suspected carcinogens. The method includes an efficient clean-up step using graphitized carbon black (GCB) that simplifies and improves the robustness of the subsequent GC-MS analysis. Briefly, after solvent extraction of the textile sample, the extract is passed thr

Quinoline-based antimalarial hybrid compounds.

Quinoline-containing compounds, such as quinine and chloroquine, have a long-standing history as potent antimalarial agents. However, the increasing resistance of the Plasmodium parasite against these drugs and the lack of licensed malaria vaccines have forced chemists to develop synthetic strategies toward novel biologically active molecules. A strategy that has attracted considerable attention in current medicinal chemistry is based on the conjugation of two biologically active molecules into one hybrid compound. Since quinolines are considered to be privileged antimalarial building blocks,

Quinolinic acid: a pathogen in seizure disorders?

The evidence for an involvement of QUIN in human seizure disorders is clearly circumstantial. Importantly, QUIN is not a classical neurotransmitter and may thus play only a negligible or no role at all in normal brain function (Foster et al., 1984). We have yet to understand if and how such a possibly inert metabolite may turn into a pathogen. Several crucial questions remain to be addressed before a case can be made for a 'quinolinic acid hypothesis' of temporal lobe epilepsy. Among the most prominent ones figure the extracellular concentration of QUIN in the human brain under normal and path

Quinoline hybrids and their antiplasmodial and antimalarial activities.

Malaria, in particular infection with P. falciparum (the most lethal of the human malaria parasite species, responsible for nearly one million deaths every year), is one of the most devastating and common infectious disease throughout the world. Beginning with quinine, quinoline containing compounds have long been used in clinical treatment of malaria and remained the mainstays of chemotherapy against malaria. The emergence of P. falciparum strains resistant to almost all antimalarials prompted medicinal chemists and biologists to study their effective replacement with an alternative mechanism

Brain quinolinic acid in Alzheimer's dementia.

Quinolinic acid (QA) content was measured in postmortem frontal and temporal cortex, putamen and cerebellum obtained from patients with senile dementia of Alzheimer type (SDAT), Huntington's disease (HD) and controls, using a gas chromatography/mass spectrometry method. There were no significant group differences in QA content of any of the regions examined. The data do not support the hypothesis that an accumulation of QA plays a role in neuronal degeneration occurring in the frontal and temporal cortex, putamen and cerebellum of patients with SDAT.

Zwitterionic-to-cationic ruthenium carbazole complexes: structure and selective hydroboration of quinolines and pyridines.

We report the synthesis of the Ru(II) zwitterionic complexes [MeRu(η6-carbazole-N-EMe2Cl)(PPh3)2] (E = Al, Ru-1; Ga, Ru-2) via a one-pot reaction of RuCl2(PPh3)3 with carbazole in the presence of excess group 13 trialkyl reagents (AlMe3 or GaMe3). Structural characterization confirms η6-coordination of ruthenium to a single fused aryl ring of the carbazole framework. The zwitterionic character of Ru-1 and Ru-2 results from deprotonation of the carbazole N-H bond to form an anionic amido fragment coordinated to a group 13 center, while the cationic Ru(II) center is supported by two phosphines,

Electrophilic Fluorine-Mediated Povarov Cyclization: Access to Fluorinated Quinolines via in Situ Generated Fluoroketenimines.

Electrophilic fluorine-mediated synthesis of mono- and difluorinated quinolines is developed via a Povarov-type cycloaddition. The reaction involves the in situ generation of highly reactive fluoroketenimine intermediates from readily accessible phosphorus ylides under metal-free and mild conditions. The electrophilic fluorinating reagent functions as both a fluorine source and an oxidant, enabling a one-pot, step-economic cascade process. This protocol exhibits broad functional group compatibility and delivers a wide array of fluorinated quinoline derivatives in good to excellent yields. Gram

Na2S-catalyzed dehydrogenative condensation of 2-aminobenzyl alcohols and ketones: a synthesis of quinolines.

We report sodium sulfide as a simple, inexpensive, yet powerful and sustainable catalyst for synthesizing quinolines and 1,8-naphthyridines. This new method involves the dehydrogenative condensation of 2-aminobenzyl alcohols with ketones and proceeds under mild heating conditions. It also demonstrates a broad substrate scope, achieving the transformation without the need for transition metals, chiral ligands, additives, or even solvents.

Dehydrogenative Cyclization of 2-Aminobenzyl Alcohols with Ketones or Secondary Alcohols to Construct Quinolines.

Herein, we present an environmentally friendly and sustainable approach for accessing functionalized quinolines through the dehydrogenative cyclization of 2-aminobenzyl alcohols with ketones or secondary alcohols. This reaction does not require transition metal catalysts, utilizes oxygen from the air as an oxidant, and is performed at room temperature. Notably, the lignin β-O-4 model compounds can be directly converted into a variety of quinolines by using this method. Mechanistic studies reveal that the reaction proceeds through a cascade of dehydrogenation, aldol condensation, and ketoamine
