Review: avian models for experimental myopia.

Myopia poses a serious threat to unaided vision among the human population, affecting as much as fifty per cent or more of individuals in some subpopulations and at some age levels. Because the etiology of the condition remains obscure, attention has been directed toward a search for experimental animal models for myopia: the hope is that any environmental or experimental intervention found to increase eye size, especially axial length, or to lead to negative refractive error, may suggest some parallels in clinical experience. As well, the availability of myopic animal subjects affords the opp

[Blocking action of Nephila clavata spider toxin on ionic currents activated by glutamate and its agonists in isolated hippocampal neurons].

The blocking action of the Nephila clavata spider neurotoxin was studied using the concentration clamp method in isolated neurons of the rat hippocampus. Crude venom JSTX blocked L-glutamate-, quisqualate- and kainate-activated ionic currents mediated by activation of the non-N-methyl-D-aspartate (non-NMDA) membrane receptors. Ionic currents elicited by all agonists were depressed by crude JSTX venom to 34-35% of its initial amplitude with no recovery during prolonged washing. An active fraction of JSTX venom blocked ionic currents almost completely, but its action was partially reversible. Th

Modulation of the intracellular calcium concentration in photoreceptor terminals by a presynaptic metabotropic glutamate receptor.

Fast excitatory neurotransmission in the central nervous system is mediated through glutamate acting on ionotropic glutamate receptors. However, glutamate acting on metabotropic glutamate receptors (mGluRs) can also exert an inhibitory action. Here, we report by immunocytochemistry and physiology, to our knowledge, the first glutamate receptor to be found in terminals of photoreceptors in the mammalian retina-the group III metabotropic glutamate receptor mGluR8. Glutamate is the transmitter of photoreceptors, and thus mGluR8 functions as an autoreceptor. Activation of mGluR8 by the group III m

The mouse neocortical slice: preparation and responses to excitatory amino acids.

1. An improved method of preparing wedges of cerebral cortex and corpus callosum from 500 microM thick coronal sections using mouse brain is described. 2. The characteristics of mixing in the two compartment baths is fully reported. 3. Cortical tissue could be depolarized relative to callosal tissue by superfusion of N-methyl-D-aspartate (NMDA), quinolinate, kainate or quisqualate. 4. At higher concentrations of agonists the depolarization was followed by a small hyperpolarization. This hyperpolarization was abolished by 5 microM ouabain. 5. Cyclic adenosine 3'5'-monophosphate potentiates excitatory amino acid and synaptic responses of rat spinal dorsal horn neurons.

Intracellular recordings were made from rat dorsal horn neurons in the in vitro slice preparation to study the actions of cyclic adenosine 3',5'-monophosphate (cyclic AMP). In the presence of TTX, bath application of the membrane permeable analogue of cyclic AMP, 8-Br cyclic AMP (25-100 microM) caused a small depolarization of the resting membrane potential accompanied by a variable change in membrane input resistance. In addition, 8-Br cyclic AMP caused a long-lasting increase in the spontaneous synaptic activity and the amplitude of presumed monosynaptic excitatory postsynaptic potentials ev

[Tinnitus: causes, diagnosis, therapy].

The symptom tinnitus may be due to a number of causes. Pathological processes within the sense-organ "ear" as well as non-otologic diseases may result in tinnitus. Therefore a thorough ENT diagnosis is necessary. The evaluation of the patient includes the history, ENT-status examination, audiological and vestibular findings, imaging investigations and, if necessary, examinations by other specialists. The therapy of tinnitus is difficult. If possible, the therapy should be causally orientated. Mostly, this is only the case with tinnitus caused by diseases of the external and middle ear. The the

The inhibition of pineal arylalkylamine n-acetyltransferase by glutamic acid and its analogues.

Previous studies from this laboratory have identified in bovine pineal gland a glutamate receptor site with a dissociation equilibrium constant (K(D)) value of 0.534 ?M and a receptor density (B(max)) value of 4.84 pmol/mg protein. This pH- and temperature-dependent binding site showed stereospecificity, was activated by Ca(2+) and displayed affinity for both glutamate receptor agonists and antagonists. The role of this glutamate receptor site was investigated by studying the effects of select glutamate receptor agonists and antagonists and of ?-aminobutyric acid on the basal- and on the norep

The NMDA-receptor antagonist, MK-801, suppresses limbic kindling and kindled seizures.

Excitatory amino acid receptors in the mammalian central nervous system (CNS) are divided into 3 receptor subtypes: kainate, quisqualate, and N-methyl-D-aspartate (NMDA). MK-801 is a selective, non-competitive antagonist of excitatory amino acid transmitters at the NMDA receptor site. The role of excitatory amino acid neurotransmission in electrical kindling was examined in animals stimulated daily in the amygdala following i.p. administration of low dosages of MK-801 (0.1 and 0.5 mg/kg). A second experiment evaluated the anticonvulsant properties of MK-801 in rats kindled in the hippocampus a

Excitatory amino acid receptors in commissural nucleus of the NTS mediate carotid chemoreceptor responses.

Stimulation of carotid body chemoreceptors by saline saturated with 100% CO2 elicited an increase in mean arterial pressure, respiratory rate, tidal volume, and minute ventilation (VE). Microinjections of L-glutamate into a midline area 0.5-0.75 mm caudal and 0.3-0.5 mm deep with respect to the calamus scriptorius increased VE. Histological examination showed that the site was located in the commissural nucleus of the nucleus tractus solitarii (NTS). Excitation of supraoptic vasopressin cells by stimulation of the A1 noradrenaline cell group: failure to demonstrate role for established adrenergic or amino acid receptors.

The effects of adrenergic and excitatory amino acid antagonists on supraoptic nucleus (SON) neurosecretory cell responses to stimulation of the A1 noradrenaline (NA) cell group were examined in anaesthetized male rats. As in previous studies, delivery of cathodal pulses (100 microA, 1 ms pulses, 1 Hz) to the A1 region of the caudal ventrolateral medulla excited spontaneously active, antidromically identified neurosecretory cells, the majority of which were identified as arginine vasopressin (AVP) secreting on the basis of basal discharge patterns and responses to abrupt increases in arterial b

Protective effect of excitatory amino acids on cold-restraint stress-induced gastric ulcers in mice: role of cyclic nucleotides.

Previous studies have shown that excitatory amino acids (EAAs) and their receptors may play important roles in the mammalian enteric system. In this study, we investigated whether EEAs, including L-glutamate (L-Glu) and subtypes N-methyl-D-aspartate (NMDA), kainic acid (KA), and quisqualic acid (QA), reduce cyclic AMP (cAMP) levels and play a role in protecting gastric lesions in cold-restraint stress (CRS) mice. First, we found that dose-dependent administration of four selected EAAs significantly attenuated the increase of cAMP content and exhibited a protective effect on the development of

Group I, II, and III mGluR compounds affect rhythm generation in the gastric circuit of the crustacean stomatogastric ganglion.

We have studied the effects of group I, II, and III metabotropic glutamate receptor (mGluR) agonists on rhythm generation by the gastric circuit of the stomatogastric ganglion (STG) of the Caribbean spiny lobster Panulirus argus. All mGluR agonists and some antagonists we tested in this study had clear and distinct effects on gastric rhythm generation when superfused over combined oscillating or blocked silent STG preparations. A consistent difference between group I agonists and group II and III agonists was that group I agonists acted excitatory. The group I-specific agonists L-quisqualic ac

Glutamate inhibition of the adrenergic-stimulated production of melatonin in rat pineal gland in vitro.

The effect of L-glutamate on the adrenergic-stimulated release of melatonin in the rat pineal gland was examined using an in vitro perfusion system. L-Glutamate by itself had no effect on melatonin secretion whereas L-glutamate administered prior to (-)-isoproterenol (beta-adrenergic agonist) and L-phenylephrine (alpha-adrenergic agonist) inhibited melatonin production by 42%. L-Glutamate did not inhibit melatonin secretion when glands were stimulated with (-)-isoproterenol alone. Selective destruction of nitric oxide synthase neurons with quisqualate reduces damage after hypoxia-ischemia in the neonatal rat.

The vulnerability of the developing CNS to hypoxia-ischemia (H-I) differs from that of the mature brain and is due in part to release of nitric oxide (NO) from parenchymal neurons. If NO is important in the generation of excitotoxic injury after H-I in the developing CNS, then selective destruction of the neuronal nitric oxide synthase (nNOS) cells before H-I should lessen the injury seen after the insult. Using low dose quisqualic acid (QA) injected into neonatal (postnatal d 7) parietal cortex, the nNOS neurons were eliminated while sparing other neuronal and glial populations as ascertained

Excitatory and inhibitory transmission from dorsal root afferents to neonate rat motoneurons in vitro.

Intracellular recordings were made from antidromically identified motoneurons in neonate (12-22 days) rat transverse spinal cord slices and the transmitters and receptors probably involved in initiating the excitatory (EPSP) and inhibitory (IPSP) postsynaptic potentials were investigated. Stimulation of dorsal roots elicited in motoneurons an EPSP, an IPSP, or an EPSP followed by an IPSP. EPSPs in 70% of motoneurons had a short latency (less than or equal to 1 ms) and in the remaining cells a latency longer than 1 ms. The IPSPs had a long latency (greater than or equal to 1 ms). Effects of a spider toxin (JSTX) on hippocampal CA1 neurons in vitro.

The effect of a toxin (JSTX) obtained from Nephila clavata (Joro spider) on the CA1 pyramidal neurons of the hippocampus was studied using slice preparations. JSTX blocked the excitatory postsynaptic potentials (EPSPs) in the pyramidal neuron evoked by Schaffer collateral stimulation but was without effect on the antidromic action potentials or on the resting conductance. Depolarization induced by ionophoretic application of glutamate was readily suppressed by JSTX but aspartate-induced depolarization was much less sensitive to the toxin. Among preferential agonists activating 3 receptor subty

2-Methyl-3,3-diphenyl-3-propanolamine (2-MDP) selectively antagonises N-methyl-aspartate (NMA).

Using electrophoretic application to rat central neurones in vivo, and bath application to frog spinal cord in vitro, 2-methyl-3,3-diphenyl-3-propanolamine was found to be a selective antagonist of N-methyl-DL-aspartate, but not of quisqualate or kainate. In this respect the (-) isomer proved to be about three times more potent than the (+) in both preparations.

Spider toxin blocks excitatory amino acid responses in isolated hippocampal pyramidal neurons.

Using the 'concentration-clamp' technique we have investigated the action of Joro spider toxin (JSTX), as a specific blocker of glutamate receptor, on freshly isolated rat hippocampal pyramidal neurons. The neurons showed prominent responses to L-glutamate (L-Glu), quisqualate (QA) and kainate (KA) and JSTX blocked completely the both responses. The blocking action of toxin was dose-dependent at the concentrations of toxin between 4.8 X 10(-12) and 4.8 X 10(-8) M, and was remarkably similar in cell to cell trials. The kinetics of blockade was revealed by applying the toxin to the non-desensiti
