The bacterium Pseudomonas aeruginosa is an opportunistic human pathogen that uses a quorum sensing signal cascade to activate expression of dozens of genes when sufficient population densities have been reached. Quorum sensing controls production of several key virulence factors, including secreted proteases such as elastase. Cooperating groups of bacteria growing on protein are susceptible to social cheating by quorum-sensing defective mutants. A possible way to restrict cheater emergence is by policing where cooperators produce costly goods to sanction or punish cheats. The P. aeruginosa Las

Genetically attenuated pathogenic bacteria have been extensively considered as vaccine candidates. However, insufficient attenuation has been a frequent limitation of this approach. Many pathogens use quorum sensing to escape host defense mechanism. Here, we hypothesized that quorum sensing can be manipulated to diminish pathogenesis. To test this hypothesis, we modified the quorum sensing circuitry of a live cholera vaccine strain to add a second layer of attenuation. Attenuation resulted from the expression of phage PhiX174 lysis gene E on a balanced lethal plasmid from the quorum sensing-re

One relevant aspect for understanding the bottlenecks that modulate the spread of resistance among bacterial pathogens consists in the effect that the acquisition of resistance may have on the microbial physiology . Whereas studies on the effect of acquiring resistance of bacterial growth are frequently performed, more detailed analyses aiming to understand in depth the cross talk between resistance and virulence, including bacterial communication are less frequent. The bacterial quorum sensing system, is an important intraspecific and interspecific communication system highly relevant for man

African trypanosomes, parasites that cause human sleeping sickness, undergo a density-dependent differentiation in the bloodstream of their mammalian hosts. This process is driven by a released parasite-derived factor that causes parasites to accumulate in G1 and become quiescent. This is accompanied by morphological transformation to 'stumpy' forms that are adapted to survival and further development when taken up in the blood meal of tsetse flies, the vector for trypanosomiasis. Although the soluble signal driving differentiation to stumpy forms is unidentified, a recent genome-wide RNAi scr

Challenges encountered in relapse of illness caused by resistance of microorganisms to antimicrobial agents (drugs) are due to factors of severe stress initiated by random use of antibiotics and insufficient beneficial approaches. These challenges have resulted to multiple drug resistance (MDR) and, subsequently, biofilm formation. A type of intercellular communication signal called quorum sensing (QS) has been studied to cause the spread of resistance, thereby enabling a formation of stable community for microorganisms. The QS could be inhibited using QS inhibitors (QSIs) called quorum-quench

UNLABELLED: Quorum sensing (QS) is a widespread process in bacteria used to coordinate gene expression with cell density, diffusion dynamics, and spatial distribution through the production of diffusible chemical signals. To date, most studies on QS have focused on model bacteria that are amenable to genetic manipulation and capable of high growth rates, but many environmentally important bacteria have been overlooked. For example, representatives of proteobacteria that participate in nitrification, the aerobic oxidation of ammonia to nitrate via nitrite, produce QS signals called acyl-homoser

BACKGROUND: The increasing prevalence of antimicrobial resistance (AMR) among urinary tract infections (UTIs) is making clinical management challenging. Biofilm formation is a key mechanism contributing to antibiotic resistance in uropathogens. Quorum sensing is known to regulate several bacterial processes like bioluminescence, virulence, and biofilm formation which is known to play a role in the development of drug resistance. Out of three major quorum sensing systems, luxS/AI-2 is a universal system used by both Gram-positive and Gram-negative bacteria.
METHODS: Community-acquired AMR UTI

Quorum sensing is defined as a cell to cell communication between micro-organisms, which enables micro-organisms to behave as multicellular organisms. Quorum sensing enables many collaborative benefits such as synchronisation of virulence factors and biofilm formation. Both quorum sensing, as well as biofilm formation, encourage the development of drug resistance in micro-organisms. Biofilm formation and quorum sensing are causally linked to each other, playing a role in the pathogenesis of the micro-organisms. With the increasing drug resistance against the available antibiotics and antifunga

Some bacterial pathogens utilize cell-cell communication systems, such as quorum sensing (QS), to coordinate genetic programs during host colonization and infection. The human-restricted pathosymbiont Streptococcus pyogenes (group A streptococcus [GAS]) uses the Rgg2/Rgg3 QS system to modify the bacterial surface, enabling biofilm formation and lysozyme resistance. Here, we demonstrate that innate immune cell responses to GAS are substantially altered by the QS status of the bacteria. We found that macrophage activation, stimulated by multiple agonists and assessed by cytokine production and N

Legionella pneumophila is a water-borne opportunistic pathogen causing a life-threatening pneumonia called 'Legionnaires' disease'. The Legionella quorum sensing (Lqs) system produces and responds to the α-hydroxyketone signaling molecule 3-hydroxypentadecane-4-one (Legionella autoinducer-1, LAI-1). The Lqs system controls the switch between the replicative/non-virulent and the transmissive/virulent phase of L. pneumophila, and it is a major regulator of natural competence, motility and virulence of the pathogen. Yet, beyond gene regulation, LAI-1 also directly affects pathogen-host interactio

UNLABELLED: Members of the genus Burkholderia are known to be adept at biofilm formation, which presumably assists in the survival of these organisms in the environment and the host. Biofilm formation has been linked to quorum sensing (QS) in several bacterial species. In this study, we characterized Burkholderia thailandensis biofilm development under flow conditions and sought to determine whether QS contributes to this process. B. thailandensis biofilm formation exhibited an unusual pattern: the cells formed small aggregates and then proceeded to produce mature biofilms characterized by "do

Candida albicans biofilms are surface-associated, structured communities composed of yeast, hyphal, and pseudohyphal cells surrounded by an extracellular matrix. C. albicans biofilms often lead to life-threatening systemic infections and are particularly difficult to eradicate because of their high levels of resistance to antibiotics. Farnesol, an autoregulatory molecule secreted by C. albicans, inhibits hyphal growth and the expression of a number of morphology-specific genes that are necessary for robust biofilm formation. Many stages of biofilm development are impacted by farnesol including

The biochemical and biological mechanisms metastatic cancer cells use to function as communities and thwart internal and external growth control mechanisms remain undefined. In this work, we present the hypothesis that cancer cells may use a Quorum-Sensing mechanism to regulate multicellular functions and control steps in metastatic colonization. Quorum sensing is a bacterial cell-cell communication process used to track increasing cell-population density and, in response to changes in cell number, coordinate gene expression and behavior on a community-wide scale. Important parallels between t

Cystic fibrosis (CF) is a genetic lung disease in which thick mucus builds up in the airways, making patients highly vulnerable to chronic bacterial infections. One of the most problematic bacteria in these infections is Pseudomonas aeruginosa, an adaptable microbe that can survive antibiotics and persist in the lungs for many years. This bacterium uses a communication system called "quorum sensing (QS)" to coordinate group behaviors. Through this system, bacterial cells sense their population density and collectively switch on genes that control toxin production, biofilm formation, and resist

This perspective provides a comprehensive review of salicylic acid (SA) and its boron complexes, emphasizing their biological significance. Research suggests that SA forms boron complexes with water-soluble compounds, including sugars, glycerol, and organic acids, aiding in its systemic transport in plants via xylem and phloem saps. Moreover, SA possesses antibacterial properties, while its boron complexes display quorum sensing inhibition and antibiofilm activity. Through an in silico study using the self-consistent extreme classifier (SCEC), we analyzed the effects of these compounds on quor

The human gut harbors enormous microbial diversity, which influences the gut's chemical environment. As the production of virulence factors affects bacterial fitness, Vibrio cholerae has adapted to sense the gut chemical environment to selectively activate virulence factors at permissible intestinal niches. Here, we show that diffusible signal factors (DSFs) canonically used by gut proteobacteria for quorum-sensing communication can repress expression of essential ToxT-dependent virulence factors cholera toxin (ctxAB) and the toxin co-regulated pilus (tcpA-F). Unlike canonical quorum sensing,

Aeromonas species (spp.) are well-known fish pathogens, several of which have been recognized as emerging human pathogens. The organism is capable of causing a wide spectrum of diseases in humans, ranging from gastroenteritis, wound infections, and septicemia to devastating necrotizing fasciitis. The systemic form of infection is often fatal, particularly in patients with underlying chronic diseases. Indeed, recent trends demonstrate rising numbers of hospital-acquired Aeromonas infections, especially in immuno-compromised individuals. Additionally, Aeromonas-associated antibiotic resistance i

Gram-positive bacteria can regulate gene expression at the population level via a mechanism known as quorum sensing. Oligopeptides serve as the signaling molecules; they are secreted and then are either detected at the bacterial surface by two-component systems or reinternalized via an oligopeptide transport system. In the latter case, imported peptides interact with cognate regulators (phosphatases or transcriptional regulators) that modulate the expression of target genes. These regulators help control crucial functions such as virulence, persistence, conjugation and competence and have been

Group A Streptococcus (GAS, Streptococcus pyogenes) is a human-restricted pathogen with a capacity to both colonize asymptomatically and cause illnesses ranging from pharyngitis to necrotizing fasciitis. An understanding of how and when GAS switches between genetic programs governing these different lifestyles has remained an enduring mystery and likely requires carefully tuned environmental sensors to activate and silence genetic schemes when appropriate. Herein, we describe the relationship between the Control of Virulence (CovRS, CsrRS) two-component system and the Rgg2/3 quorum-sensing pat
