Identification of a putatively novel trichomonad species in the intestine of a common quail (Coturnix coturnix).

A common quail (Coturnix coturnix) from a private keeping died unexpectedly and showed a moderate lymphocytic infiltration of the colonic mucosa associated with numerous protozoa-like objects at the pathological examination. These organisms were further identified using chromogenic in situ hybridization (ISH) and gene sequencing. ISH was performed on paraffin embedded tissue sections and produced a positive signal using a probe specific for the 18S ribosomal RNA (rRNA) gene of the order Trichomonadida, but remained negative with probes specific for the 18S rRNA gene of the common bird parasite

Ribosomal RNA analysis of Babesia odocoilei isolates from farmed reindeer (Rangifer tarandus tarandus) and elk (Cervus elaphus canadensis) in Wisconsin.

Piroplasms isolated from a farmed reindeer and elk in Wisconsin were determined to be Babesia odocoilei, based on morphology and ribosomal RNA (rRNA) analysis. Different clinical manifestations were observed in the two host species. The reindeer was parasitemic and exhibited acute babesiosis resulting in death, while the elk showed no parasites in blood smears and no overt clinical signs of babesiosis. B. odocoilei was, however, readily cultured from elk erythrocytes. Small subunit rRNA gene sequences from the two isolates were identical to that previously reported for B. odocoilei. Internal t

Phylogenetic identification of Pneumocystis murina sp. nov., a new species in laboratory mice.

Pneumocystis is a fungal genus that contains multiple species. One member of the genus that has not been formally analysed for its phylogenetic relationships and possible species status is the Pneumocystis found in laboratory mice, Pneumocystis murina sp. nov. (type strain ATCC PRA-111(T)=CBS 114898(T)), formerly known as Pneumocystis carinii f. sp. muris. To advance research in this area, approximately 3000 bp of additional DNA sequence were obtained from the locus encoding rRNAs. This sequence and others were used to determine genetic distances between P. murina and other members of the genu

Rapid detection of pathogenic fungi using loop-mediated isothermal amplification, exemplified by Fonsecaea agents of chromoblastomycosis.

Loop-mediated isothermal amplification (LAMP) was developed for rapid detection of pathogenic or allergenic fungal in the environment. Primers applied were derived from the rDNA Internal Transcribed Spacer and the 5.8S rRNA gene. The assay enabled amplification of target fungi at the level of genus or closely related species using pure cultures after 1h reaction at 65 degrees C in a water bath. No cross-reactivity to related species was observed. The DNA detection limit was 0.2fg. The method also proved to work well with fungi on non-sterile adhesive tape. Amplification products were detected

Gyrodactylidae from Fundulus heteroclitus (Fundulidae, Cyprinodontiformes) in South Carolina, USA, and insights on the historical ecology of the gyrodactylids of Fundulidae.

Mummichog (Fundulus heteroclitus) hosts a diverse array of parasites, including several species of Gyrodactylidae. We surveyed populations of mummichogs across three estuarine systems in South Carolina, USA, for gyrodactylids and studied their taxonomy, phylogenetics, and evolutionary history. Molecular analysis using concatenated sequences of 18S, ITS, and 5.8S rDNA fragments and morphology support the synonymization of Fundulotrema with Swingleus, as the former was shown paraphyletic. Five species of Gyrodactylidae were encountered. Gyrodactylus kritskyi n. sp. is described and four other sp

Salpingitis in Pekin ducks associated with concurrent infection with Tetratrichomonas sp. and Escherichia coli.

Increased mortality (1.5% per week) and low egg production (5-10% lower than normal) were observed in a flock of domestic breeding Pekin ducks (Anas platyrhynchos). At necropsy, salpingitis and peritonitis were the most significant findings. Histologically, there was accumulation of necrotic debris in the lumen of the oviduct. Numerous bacteria and trichomonads were observed histologically in the lumen of the vagina and occasionally in the shell gland. Escherichia coli and a trichomonad were isolated from the oviduct. The trichomonads were oval (6-8 microm long, 4.5-6 microm wide) and had 4 an

Noncoding mutations cause super-enhancer retargeting resulting in protein synthesis dysregulation during B cell lymphoma progression.

Whole-genome sequencing of longitudinal tumor pairs representing transformation of follicular lymphoma to high-grade B cell lymphoma with MYC and BCL2 rearrangements (double-hit lymphoma) identified coding and noncoding genomic alterations acquired during lymphoma progression. Many of these transformation-associated alterations recurrently and focally occur at topologically associating domain resident regulatory DNA elements, including H3K4me3 promoter marks located within H3K27ac super-enhancer clusters in B cell non-Hodgkin lymphoma. One region found to undergo recurrent alteration upon tran

Incidence and risk factors of hookworm infection in a rural community of central Thailand.

A cohort study to identify incidence and risk factors of hookworm infection was conducted in a rural community, central Thailand from November 2005 to February 2007. Stool specimens were examined for hookworm eggs using wet preparation, Kato thick smear, and water-ethyl acetate sedimentation technique. The incidence rate of hookworm infection was 7.5/100 person-years. The independent risk factors for acquiring hookworm infection were barefoot walking (incidence rate ratio [IRR] = 4.2, 95% confidence interval [CI] = 1.2-14.5) and raising buffaloes around the house (IRR = 4.8, 95% CI = 1.9-11.8)

A Multiplex-PCR approach to identification of the Brazilian intermediate hosts of Schistosoma mansoni.

Due to difficulties concerning morphological identification of planorbid snails of the genus Biomphalaria, and given a high variation of characters and in the organs with muscular tissue, we designed specific polymerase chain reaction (PCR) primers for Brazilian snail hosts of Schistosoma mansoni from available sequences of internal transcribed spacer 2 (ITS2) of the ribosomal RNA gene. From the previous sequencing of the ITS2 region, one primer was designed to anchor in the 5.8S conserved region and three other species-specific primers in the 28S region, flanking the ITS2 region. These four p

An optimised protocol for molecular identification of Eimeria from chickens.

Molecular approaches supporting identification of Eimeria parasites infecting chickens have been available for more than 20 years, although they have largely failed to replace traditional measures such as microscopy and pathology. Limitations of microscopy-led diagnostics, including a requirement for specialist parasitological expertise and low sample throughput, are yet to be outweighed by the difficulties associated with accessing genomic DNA from environmental Eimeria samples. A key step towards the use of Eimeria species-specific PCR as a sensitive and reproducible discriminatory tool for

DNA sequence characterisation and phylogeography of Lymnaea cousini and related species, vectors of fascioliasis in northern Andean countries, with description of L. meridensis n. sp. (Gastropoda: Lymnaeidae).

BACKGROUND: Livestock fascioliasis is a problem throughout Ecuador, Colombia and Venezuela, mainly in Andean areas where the disease also appears to affect humans. Transmission patterns and epidemiological scenarios of liver fluke infection have shown to differ according to the lymnaeid vector snail species involved. These Andean countries present the vectors Lymnaea cousini, L. bogotensis and L. ubaquensis, unknown in the rest of Latin America. An exhaustive combined haplotype study of these species is performed by means of DNA sequencing of the nuclear ribosomal 18S RNA gene, ITS-2 and ITS-1

Reflexicalyptrales ordo nov., a distinct Agaricomycetes lineage discovered from orchid mycorrhizae.

Uncovering the identity of "dark fungi" is crucial for understanding fungal taxonomy, diversity, and phylogenetic relationships. Previous exploratory analysis of the mycorrhizal fungi of the partially mycoheterotrophic orchid Nervilia nipponica revealed an association with a particular group of Agaricomycetes, tentatively designated "unidentified Agaricomycetes X" (uAX). Two culture strains were successfully isolated from the mycorrhiza, which enabled the phylogenetic position of this fungus to be determined using ultrastructural and multigene phylogenetic analyses. Transmission electron micro
