Identification of a novel translocation producing an in-frame fusion of TAF15 and ETV4 in a case of extraosseous Ewing sarcoma revealed in the prenatal period.

Ewing sarcoma (ES) is a highly malignant round cell sarcoma, characterized by gene fusion involving FET (FUS, EWSR1, TAF15) and ETS family genes, respectively. The involvement of the EWSR1 gene has been reported in approximately 90% of cases of ES, with the EWSR1::FLI1 fusion being the most frequent. We report the case of a newborn with a localized soft tissue paravertebral neoplasm diagnosed prenatally. Histopathology and immunophenotype were consistent with a CD99 + , NKX2.2 + undifferentiated round cell sarcoma (URSC); whole-exome RNA-sequencing demonstrated an undescribed in-frame TAF15::E

Characterization of a novel sarcoma cell line with an EWSR1::POU2AF3 fusion.

Sarcomas with an EWSR1::POU2AF3(COLCA2) fusion are a very recently described entity of preferentially sinonasal origin and with undifferentiated round/spindle cell morphology. We established a novel cell line (PF1095) carrying a EWSR1::POU2AF3 fusion from the malignant pleural effusion of a 25-year-old sarcoma patient. The patient was first diagnosed with poorly differentiated neuroendocrine carcinoma based on tumor cell morphology and positivity to markers such as EMA, synaptophysin, and CD56. Later, the EWSR1 translocation was identified in the tumor cells with unknown partners and the patie

Extra-skeletal Ewing's sarcoma with EWSR1::ERG translocation of the breast: a case report.

BACKGROUND: Ewing's sarcoma (ES), first described by Ewing in 1921, is a highly malignant small blue round cell tumor that arises in bone or extraskeletal soft tissues. It predominantly affects patients aged 10 to 30 years. Approximately 12% of cases involve solid organs, often presenting as rapidly growing masses in deep soft tissues. The most common genetic alteration in ES is the t(11;22)(q24;q12) translocation, resulting in the EWSR1::FLI1 fusion gene, which accounts for 90% of tumors. Extraskeletal Ewing's sarcoma (EES) presented as a breast mass is extremely rare, with only a handful of

Germline Variation and Somatic Alterations in Ewing Sarcoma.

Ewing sarcoma (EwS) is a rare bone or soft tissue tumor that occurs early in life, suggesting genetic variation is likely a major contributor to EwS risk. To date, genetic investigations have identified key somatic mutations and germline variants of importance for EwS risk. While substantial progress has been made in uncovering the genetic etiology of EwS, considerable gaps in knowledge remain. Herein, we provide a summary of methodological approaches for future genomic investigations of EwS. We anticipate that this recommended analytical framework for germline and somatic investigations, alon

Homoharringtonine suppresses acute myeloid leukemia progression by orchestrating EWSR1 phase separation in an m6A-YTHDF2-dependent mechanism.

Homoharringtonine (HHT) is widely used in combination regimens for acute myeloid leukemia (AML), yet its direct cellular targets remain undefined, limiting precision application. Here, we identified EWS RNA-binding protein 1 (EWSR1) as the primary target of HHT through chemical proteomics and biophysical validation. HHT bound the RNA recognition motif of EWSR1 with micromolar affinity, inducing an allosteric conformational switch that promoted oligomerization and liquid-liquid phase separation (LLPS). EWSR1 condensates selectively recruited the N6-methyladenosine (m6A) reader YTHDF2, forming c

EWSR1::BEND2 fusion sarcoma in bone: a report of two rare cases.

The two cases, we had presented here, were extremely rare instances of the EWSR1::BEND2 fusion sarcomas in bone. The first case was a 12-year-old female with a 50 × 25-mm lytic tumor in metatarsal bone. Histopathologic examination revealed a widespread cellular, fascicular forming, spindled cell component along with a focal round cell component. CD99 was diffuse membranous positive while cytokeratin and EMA were focal positive. The second case was a 62-year-old female with a pathological fracture in femur. On MRI examination, a heterogeneous tumor measuring 96 × 54 mm was detected in the femur

DNA Methylation Profiling Distinguishes Adamantinoma-Like Ewing Sarcoma From Conventional Ewing Sarcoma.

Adamantinoma-like Ewing sarcoma (ALES) has traditionally been considered a variant of Ewing sarcoma because it generally harbors EWSR1::FLI1 fusions despite showing diffuse positivity for keratins and p40. However, it has become increasingly recognized that different tumors can have identical translocations, including shared fusions between carcinomas and sarcomas, raising questions as to whether ALES might represent a separate entity. Using methylation profiling, we further explored the relationship between Ewing sarcoma and ALES. The archives of multiple institutions were searched for candid

At the crossroad between Ewing sarcoma and neuroblastoma: a report of two cases of Ewing sarcoma with post treatment neuroblastoma-like differentiation.

BACKGROUND: Ewing sarcoma (ES) is the second most frequent sarcoma of bone, often affecting young patients and pursuing an aggressive clinical course. Among therapeutic choices, radio- and chemotherapy are employed in neoadjuvant setting, and they yield variable histological changes in neoplastic tissue, which mainly include necrosis and fibrosis. Cytodifferentiation is seldom observed in pediatric tumors such as nephroblastoma and rhabdomyosarcoma following treatment. Nevertheless, it represents an extremely rare phenomenon in ES. CASE PRESENTATION: In this study we present the clinico-patho

Adamantinoma‑like Ewing Sarcoma of Head and Neck: A Clinicopathologic Study of Three Cases Including One EWSR1-Negative Case with NF1 Mutation.

Adamantinoma-like Ewing sarcoma (ALES) is a rare variant of Ewing sarcoma that predominantly occurs in the head and neck region, characterized by epithelial differentiation and overlapping morphology with other small round cell tumors. We present three cases of head and neck ALES (two in the parotid gland and one in the nasal cavity) in patients aged 23-42 years. Histologically, the tumors exhibited a spectrum of features, including small round blue cells and basaloid squamous morphologies. Immunohistochemically, the tumor cells variably expressed CKpan, p63, CD99, NKX2.2, FLI1, CD56, and Syn.

[Rare case of round cell sarcoma of bone with EWSR1-NFATC2 fusion].

Undifferentiated small round cell sarcoma of bone and soft tissue are rare, malignant neoplasms, often presenting diagnostic challenges due to its overlapping features with other conditions such as osteomyelitis and Brodie abscess. Accurate diagnosis requires a combination of imaging studies, histopathology, and genetic testing. Delayed or misdiagnosis can impact the prognosis and treatment outcomes. We present the case of a 34-year-old female who initially presented with persistent left upper thigh pain. Initial imaging raised suspicion of chronic osteomyelitis, with subsequent MRIs showing a

Intracranial mesenchymal tumours with FET::CREB fusions: Two case reports and literature review.

Intracranial mesenchymal tumours with in-frame gene fusions of the FET family of RNA-binding proteins to the cyclic AMP response element-binding protein family of transcription factors are exceptionally rare primary central nervous system neoplasms, with morphological similarities to angiomatoid fibrous histiocytoma. Despite their recent inclusion in the 2021 World Health Organisation classification of tumours of the central nervous system, only a limited number of cases have been reported. We describe two cases of intracranial mesenchymal tumours harbouring fusion of the Ewing sarcoma breakpo

Ewing sarcoma breakpoint region 1 (EWSR1) protein binds to nonstructural protein 2 and non-coding regions of chikungunya virus genome to inhibit virus replication.

Chikungunya virus (CHIKV) is an alphavirus with a single-stranded RNA genome. Conserved sequence elements within the 5' and 3' non-coding regions (NCRs) of the CHIKV genome interact with host proteins to regulate viral RNA replication. Electrophoretic mobility shift assays (EMSAs) and mass spectrometry identified several host RNA-binding proteins that associate with these conserved NCR elements, among which EWSR1 uniquely bound the 3' ends of both the plus-sense genomic RNA and the minus-sense replication intermediate RNA. EMSA and biolayer interferometry (BLI) demonstrated that EWSR1 binds st

Engineering enhanced protein disaggregases for neurodegenerative disease.

Protein misfolding and aggregation underpin several fatal neurodegenerative diseases, including Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), and frontotemporal dementia (FTD). There are no treatments that directly antagonize the protein-misfolding events that cause these disorders. Agents that reverse protein misfolding and restore proteins to native form and function could simultaneously eliminate any deleterious loss-of-function or toxic gain-of-function caused by misfolded conformers. Moreover, a disruptive technology of this nature would eliminate self-templating conforme

The tip of the iceberg: RNA-binding proteins with prion-like domains in neurodegenerative disease.

Prions are self-templating protein conformers that are naturally transmitted between individuals and promote phenotypic change. In yeast, prion-encoded phenotypes can be beneficial, neutral or deleterious depending upon genetic background and environmental conditions. A distinctive and portable 'prion domain' enriched in asparagine, glutamine, tyrosine and glycine residues unifies the majority of yeast prion proteins. Deletion of this domain precludes prionogenesis and appending this domain to reporter proteins can confer prionogenicity. An algorithm designed to detect prion domains has succes

Transcriptional Programs Define Intratumoral Heterogeneity of Ewing Sarcoma at Single-Cell Resolution.

EWSR1-FLI1, the chimeric oncogene specific for Ewing sarcoma (EwS), induces a cascade of signaling events leading to cell transformation. However, it remains elusive how genetically homogeneous EwS cells can drive the heterogeneity of transcriptional programs. Here, we combine independent component analysis of single-cell RNA sequencing data from diverse cell types and model systems with time-resolved mapping of EWSR1-FLI1 binding sites and of open chromatin regions to characterize dynamic cellular processes associated with EWSR1-FLI1 activity. We thus define an exquisitely specific and direct

Novel Glomeruloid Adenocarcinoma Arising in the Mandible Harboring EWSR1::YY1.

The identification of novel, molecularly defined neoplasms continue to refine diagnostic precision and illuminate tumor pathogenesis. Assessment of the EWSR1 locus has emerged as a frequently utilized diagnostic tool in the work-up of salivary and craniofacial bone tumors, due to its established perturbations in hyalinizing clear cell carcinomas, myoepithelial carcinomas, rare mucoepidermoid carcinomas, and others. Here, we report a case of a 12-year-old female who presented with a large expansile intraosseous lesion within the left body of the mandible, initially biopsied with negative FISH r

EWSR1(22q12) Translocation Positive Pediatric Adrenal Tumor with Loss of 1p, 11q, and Unbalanced Gain of 17q: Neuroblastoma or Ewing Sarcoma?

Background Although neuroblastoma and Ewing sarcoma/Primitive neuroectodermal tumor are different clinical entities, they are both a member of small round blue cell tumors and can mimic each other's behavior in clinical and molecular aspects. Case report: A 3 year-old girl with an abdominal mass was found to have a small round blue cell tumor originating from the right adrenal gland. High level of neuron specific enolase, initial genetic test results (N-Myc amplification: negative, loss of 1p, 11q, and unbalanced gain of 17q) and characteristic radiological appearance of the tumor suggested a

Targeted and molecular therapies in Ewing sarcoma: a comprehensive review of preclinical and clinical advances.

INTRODUCTION: Ewing sarcoma (EWS) is an aggressive small round blue cell malignancy driven by EWSR1::ETS fusions, with poor survival outcomes in metastatic and relapsed disease. While multimodal chemotherapy remains the cornerstone of therapy, emerging agents hold potential to overcome resistance and improve outcomes. METHODS: A systematic search of PubMed, Scopus, and Google Scholar through February 2026 identified 1,166 records. Following duplicate removal and two-stage screening, 87 preclinical and clinical studies were included. Data were extracted and synthesized narratively, with emphas

Adenoid Cystic Carcinoma With Striking Tubular Hypereosinophilia: A Unique Pattern Associated With Nonparotid Location and Both Canonical and Novel EWSR1::MYB and FUS::MYB Fusions.

The classification of salivary gland tumors is ever-evolving with new variants of tumors being described every year. Next-generation sequencing panels have helped to prove and disprove prior assumptions about tumors' relationships to one another, and have helped refine this classification. Adenoid cystic carcinoma (AdCC) is one of the most common salivary gland malignancies and occurs at all major and minor salivary gland and seromucous gland sites. Most AdCC are predominantly myoepithelial and basaloid with variable cribriform, tubular, and solid growth. The luminal tubular elements are often
