Original endomorphin-1 analogues exhibit good analgesic effects.

A new class of endomorphin-1 analogues was synthesized by combining successful chemical modifications including N-terminal guanidino modification, Phe4 was chlorinated, D-Ala-Gly Substituted L-Pro2. Their bioactivities were measured by radioligand binding assay, metabolic stability and the tail-flick test. In radioligand binding assays, analogue GAGPC (Nα-Amidino-Tyr-D-Ala-Gly-Trp-p-Cl-Phe-NH2), shown a μ-opioid receptor affinity about 1.42-fold higher and a 2.51-fold higher δ-opioid receptor affinity than EM-1. In the metabolic stability assays, GAGPC had the longest half-lives which was 284m

Methods for assessing binding of a small-molecule radioligand to soluble enzyme isocitrate dehydrogenase 1 variants.

BACKGROUND: Isocitrate dehydrogenase 1 (IDH1) is a cytosolic NADP+-dependent enzyme involved in cellular redox homeostasis and 2-oxoglutarate (2-OG) metabolism. Glioma-associated mutations in IDH1 confer a neomorphic enzymatic activity, resulting in the production and accumulation of the oncometabolite D-2-hydroxyglutarate (2-HG), which contributes to metabolic and epigenetic alterations in IDH-mutant tumors. Small-molecule inhibitors of mutant IDH1 have emerged as promising therapeutic agents. In parallel, radiolabeled derivatives of these inhibitors are being investigated as potential positr

Drosophila nicotinic receptors: evidence for imidacloprid insecticide and alpha-bungarotoxin binding to distinct sites.

The principal mammalian brain nicotinic acetylcholine receptors (nAChRs) are the alpha-bungarotoxin (alpha-BGT)-insensitive alpha 4 beta 2 and the alpha-BGT-sensitive alpha 7 subtypes assayed with radiolabeled nicotinoids and alpha-BGT, respectively. Drosophila head membranes bind the insecticide radioligand [(3)H]imidacloprid ([(3)H]IMI) and [(3)H]alpha-BGT with K(D) 5.7 and 2.7 nM and B(max) 980 and 1400 fmol/mg protein, respectively. The hypothesis that [(3)H]IMI at 2.5 or 20 nM and [(3)H]alpha-BGT at 1 or 10 nM bind to distinct sites or subtypes is tested by using these radioligands alone

Isolation and identification of lysophosphatidylcholines as endogenous modulators of thromboxane receptors.

Inhibition of thromboxane receptor radioligand binding to human platelet membranes has been employed as the basis for a radioreceptor assay designed to measure thromboxane receptor binding activity in samples of biological fluids. This method was used during phase 1 clinical evaluation of the thromboxane receptor antagonist SQ 30,741. Frequently, baseline plasma samples as well as plasma samples from placebo-treated subjects showed significant inhibition of radioligand binding in the radioreceptor assay, suggesting the presence of endogenous thromboxane receptor ligands. This receptor binding

Relationship between thyroid status and Graves' disease-specific immunoglobulins.

The prevalence of TSI in Graves' disease was investigated with a radioligand receptor assay using human thyroid membranes and highly purified labeled porcine TSH or bovine TSH in 110 patients before treatment and in 39 patients after successful treatment with antithyroid drugs. In 11 patients, the assay was performed before, during, and after therapy. The results in the radioligand receptor assay were compared to thyroid function tests (TRH test, free T4 index, serum %3 level, and thyroid suppression test). Normal IgG inhibits nonspecifically, but dose dependently, the binding of [125I]TSH to

Radioreceptor assay of anticholinergic drugs.

The radioreceptor assay (RRA) of anticholinergic drugs in plasma and other biological fluids presents methodological difficulties. The specificity is not comparable with some pure chemical methods because drug metabolites which are active in binding to muscarinic receptors participate in the assay. There are also problems with serum dilution, with the lipophilicity of the radioligands and drugs, and with the protein binding of the drugs. However, the RRA of anticholinergic drugs is rapid and sensitive. In racemic drugs, only active stereoisomers show affinity for the muscarinic receptors. Resu

Systematic Investigation of Fatty Acids with Different Carbon-Chain Length as Albumin Binder to Improve Tumor Accumulation of Radioligands.

In the development of radiopharmaceuticals, high tumor accumulation is required for their diagnostic and therapeutic effectiveness. A key strategy to enhance such tumor accumulation is the incorporation of small-molecule albumin binders (ALBs) into radioligands, extending the blood circulation time through albumin interaction for efficient delivery to tumors. In this study, we systematically investigated the potential use of fatty acids (FAs) with various carbon-chain lengths (C12, C14, C16, and C18) as ALBs (FA12, FA14, FA16, and FA18). First, we synthesized four radiolabeled compounds compos

Acetaminophen protein adducts: a review.

Significant controversy surrounds the clinical and legal implications of 3-para cysteinyl acetaminophen, the protein degradation product of acetaminophen protein adducts. Versions of this test have been used for several years in animal research to help understand acetaminophen toxicity. As human research papers have appeared, the allegation has been made that the presence of 3-para cysteinyl acetaminophen in a patient with hepatic injury proves causal association of acetaminophen with the injury. It has also been suggested that quantitative adduct assays can guide the management of acute overd

Pirenzepine distinguishes between muscarinic receptor-mediated phosphoinositide breakdown and inhibition of adenylate cyclase.

Subtypes of muscarinic cholinergic receptors have been proposed to exist, but the biochemical responses mediated by the putative subtypes are unknown. In the present study, muscarinic receptor-mediated phosphoinositide breakdown and inhibition of adenylate cyclase activity were characterized in rat brain as well as rat parotid and heart. To study whether these responses are mediated by separate subtypes of muscarinic receptors, the potencies of agonists and antagonists were determined in both assays. Antagonist potencies were calculated by Schild analysis. In the brain, the putatively selectiv

Design, Synthesis, and Biological Evaluation of Small-Molecule-Based Radioligands with Improved Pharmacokinetic Properties for Imaging of Programmed Death Ligand 1.

Small molecules offer some advantages for developing positron emission tomography (PET) tracers and are therefore a promising approach for imaging and therapy monitoring of programmed death ligand 1 (PD-L1) positive tumors. Here, we report six biphenyl PD-L1 radioligands using the NODA-GA-chelator for efficient copper-64 complexation. These radioligands contain varying numbers of sulfonic and/or phosphonic acid groups, serving as hydrophilizing units to lower the log D7.4 value down to -4.28. The binding affinities of compounds were evaluated using saturation binding and a real-time binding as

Synthesis and Characterization of Tritium-Labeled Neuropeptide Y Y5 Receptor Ligands Derived From CGP71683A.

Among the human neuropeptide Y (NPY) receptors, the Y5 receptor (Y5R) represents a potential therapeutic target for the treatment of obesity and stress-related diseases. As the Y5R is expressed in various malignant tumors, it is also considered a potential target for cancer diagnosis and therapy. Although numerous non-peptidic Y5R antagonists with high binding affinity have been reported, only labeled peptidic agonists are used in routine Y5R competition binding assays, which are needed for the development and characterization of Y5R ligands. In the present study, three radiolabeled derivative

Bacterial assay for quantitative measurement of nanomolar concentrations of a metabolite.

A mutant of Pseudomonas fluorescens was used to develop a radioligand, competitive binding assay to quantitatively measure gamma-aminobutyric acid. The highly reliable and reproducible assay was sensitive (nM detection), rapid, and easy to perform. Nonspecific activity and scatter were insignificant. Radiolabel was irreversibly fixed by the cells in an energy-dependent reaction. The finding that a bacterium was effective in quantitatively detecting nanomolar amounts of a metabolite suggests that other bacteria or their mutants might be used in competitive binding assays to detect and quantify

(-)-Stepholidine is a potent pan-dopamine receptor antagonist of both G protein- and β-arrestin-mediated signaling.

RATIONALE: (-)-Stepholidine is a tetrahydroberberine alkaloid that is known to interact with dopamine receptors and has also been proposed as a novel antipsychotic agent. Its suggested novelty lies in the fact that it has been proposed to have D1-like receptor agonist and D2-like receptor antagonist properties. Thus, it might be effective in treating both positive and negative (cognition) symptoms of schizophrenia. However, its activity on specific dopamine receptor subtypes has not been clarified, especially with respect to its ability to activate D1-like receptors. OBJECTIVES: We wished to

Synthesis of 4,6-diphenylpyrimidin-2-ol derivatives as new benzodiazepine receptor ligands.

Benzodiazepines (BZDs) have been widely used in neurological disorders such as insomnia, anxiety, and epilepsy. The use of classical BZDs, e.g., diazepam, has been limited due to adverse effects such as interaction with alcohol, ataxia, amnesia, psychological and physical dependence, and tolerance. In the quest for new benzodiazepine agonists with more selectivity and low adverse effects, novel derivatives of 4,6-diphenylpyrimidin-2-ol were designed, synthesized, and evaluated. In this series, compound 2, 4-(2-(benzyloxy)phenyl)-6-(4-fluorophenyl)pyrimidin-2-ol, was the most potent analogue in

Bovine parathyroid hormone-(41-84), a hormone fragment with desirable properties for use as radioligand.

Radioiodinated bPTH has been widely used as the labeled ligand in the radioimmunoassay of PTH. We now report the properties of a carboxyterminal fragment of bPTH that has several favorable characteristics when used as radioligand. This peptide, the chief component of a commercial preparation of bPTH, was isolated by gel filtration, where it migrated more slowly than did authentic bPTH-(1-84). It yielded lower nonspecific binding values and more sensitive hPTH assays than were seen with the intact hormone. By immunological criteria this peptide lacked the aminoterminal region of PTH, since hPTH

Brain benzodiazepine receptor characteristics in canine narcolepsy.

A regional analysis of brain benzodiazepine (BDZ) receptors was conducted in narcoleptic and normal dogs to determine whether these sites are altered in narcolepsy. It was postulated that BDZ receptors play a role in the excessive sleepiness of narcolepsy because activation of these sites in freely behaving normal animals is hypnogenic. [3H]Flunitrazepam binding sites were assessed in 11 discrete areas of the forebrain and brainstem. No consistent or statistically significant group differences in either receptor densities (Bmax) or binding affinities (Kd) were found. These findings do not supp
