Ralstonia solanacearum Alters Root Developmental Programmes in Auxin-Dependent and -Independent Manners.

Microbial pathogens and other parasites can modify the development of their hosts, either as a target or a side effect of their virulence activities. The plant-pathogenic bacterium Ralstonia solanacearum, causal agent of the devastating bacterial wilt disease, is a soilborne microbe that invades host plants through their roots and later proliferates in xylem vessels. In this work, we studied the early stages of R. solanacearum infection in the model plant Arabidopsis thaliana, using an in vitro infection system. In addition to the previously reported inhibition of primary root length and incre

Transcriptomics and overexpression analyses reveal StERF87 confers resistance to Ralstonia solanacearum in potato.

StERF87 directly activates StPR1a via GCC-box binding and integrates SA/ET signaling to enhance bacterial wilt resistance. Potato (Solanum tuberosum) is an important food crop worldwide, yet its yield is severely constrained by bacterial wilt caused by Ralstonia solanacearum. We used RNA-Seq to study gene expression in 'Z1076-1' at 0, 1, and 2 days post-inoculation (dpi) with R. solanacearum (10⁶ CFU mL⁻1). We identified 6663 differentially expressed genes at 1 dpi and 7390 at 2 dpi. Calcium signaling and MAPK cascade genes were upregulated at 1 dpi. PR protein and ROS-related genes showed str

Ralstonia pickettii: Two Cases of Bacteremia in a Moroccan Pediatric Intensive Care Unit.

Ralstonia pickettii is an emerging opportunistic pathogen in hospital settings, particularly affecting immunocompromised patients. Its ability to persist in aqueous solutions and colonize medical devices contributes to nosocomial outbreaks, complicating infection prevention and management strategies. We report two cases of R. pickettii bloodstream infections in the pediatric intensive care unit of a Moroccan university hospital. The first case involved an 11-month-old infant admitted with febrile respiratory distress. Despite initial treatment with imipenem and amikacin, R. pickettii bacteremi

Targeting flagellar-mediated virulence: The lipopeptide PW-1 suppresses pathogenicity in Ralstonia solanacearum.

Bacterial wilt caused by Ralstonia solanacearum is a devastating soil-borne disease, calling for novel green control strategies. This study reports that the natural lipopeptide PW-1 controls this disease through an "anti-virulence" mechanism without inhibiting pathogen growth. Phenotypic analysis showed that PW-1 specifically impaired bacterial motility. Transcriptome sequencing revealed that PW-1 coordinately downregulated flagellar biosynthesis-related genes, and transmission electron microscopy confirmed the loss of flagellar filaments in PW-1-treated cells. Using GFP-tagged bacteria, we de

Oxacillinases and antimicrobial susceptibility of Ralstonia pickettii from pharmaceutical water systems in Croatia.

This study evaluated antibiotic susceptibility and presence of blaOXA22 and blaOXA60 genes in 81 isolates of Ralstonia pickettii obtained from different purified and ultra-pure water systems in two different geographical areas of Croatia. E-test and disc diffusion test were performed to determine antibiotic susceptibility. Polymerase chain reaction was applied to detect genes encoding OXA-22 and OXA-60 oxacillinases previously identified in R. pickettii. The isolates were genotyped by pulsed-field gel electrophoresis. The results revealed variable susceptibility/resistance profiles. Our isolat

Metabolic adaptation of Ralstonia solanacearum during plant infection: a methionine biosynthesis case study.

MetE and MetH are two distinct enzymes that catalyze a similar biochemical reaction during the last step of methionine biosynthesis, MetH being a cobalamin-dependent enzyme whereas MetE activity is cobalamin-independent. In this work, we show that the last step of methionine synthesis in the plant pathogen Ralstonia solanacearum is under the transcriptional control of the master pathogenicity regulator HrpG. This control is exerted essentially on metE expression through the intermediate regulator MetR. Expression of metE is strongly and specifically induced in the presence of plant cells in a

Biodegradation of dichloromethane by the polyvinyl alcohol-immobilized methylotrophic bacterium Ralstonia metallidurans PD11.

A dichloromethane (DCM)-degrading bacterium, Ralstonia metallidurans PD11 NBRC 101272, was immobilized in a polyvinyl alcohol (PVA) gel to use in a bioreactor for DCM treatment. After 4-month incubation of PVA gel beads with R. metallidurans PD11 and DCM in a mineral salt medium, the cells were tightly packed in the mesh of the gel. Forty beads of the gel in 10 ml of a batch system model showed effective activity degrading 500 and 1,000 mg l(-1) DCM within 2 and 3 h, respectively. Although reduction of pH due to accumulation of chloride ion liberated from DCM decreased the activity, it was rec

Characterization of microbial polythioesters: physical properties of novel copolymers synthesized by Ralstonia eutropha.

Various samples of polythioesters with different contents of 3-mercaptopropionic acid (3MP) or 3-mercaptobutyric acid (3MB) as one comonomer and with 3-hydroxybutyric acid (3HB) as the second constituent were produced by cultivating cells of Ralstonia eutropha strain H16 in mineral salts medium containing 3MP or 3MB plus gluconate as carbon sources. Fermentations were done also at the 30-L scale. The various samples were cast as films from chloroform and the following were recorded: melting point, solid-state NMR, X-ray diffraction. The copolyester poly(3HB-co-3MP) displayed mutiple melting pe

Involvement of many chemotaxis sensors in negative chemotaxis to ethanol in Ralstonia pseudosolanacearum Ps29.

Ralstonia pseudosolanacearum Ps29 showed repellent responses to alcohols including methanol, ethanol, 1-propanol, 2-propanol, 1-butanol, 2-butanol, 1,3-propanediol and prenol. R. pseudosolanacearum Ps29 possesses 22 putative chemoreceptors known as methyl-accepting chemotaxis proteins (MCPs). To identify a MCP involved in negative chemotaxis to ethanol, we measured ethanol chemotaxis of a complete collection of single mcp gene deletion mutants of R. pseudosolanacearum Ps29. However, all the mutants showed repellent responses to ethanol comparable to that of the wild-type strain. We constructed

Biodegradation of vinyl chloride and cis-dichloroethene by a Ralstonia sp. strain TRW-1.

An aerobic bacterium, Ralstonia sp. strain TRW-1, that assimilates vinyl chloride (VC) or ethene (ETH) as the sole carbon source was isolated from a chloroethene-degrading enrichment culture. Phylogenetic analysis of 16S rDNA sequence of the isolate revealed almost 99% sequence similarity to Ralstonia pickettii. To our knowledge, this is the first report describing the isolation of a member of Ralstonia that can degrade VC as the growth substrate. The measured growth yield values for VC and ETH were 11.27 and 18.90 g protein/mole, respectively. The estimated half-velocity constant K (m) values

Naringenin restricts the colonization and growth of Ralstonia solanacearum in tobacco mutant KCB-1.

Bacterial wilt severely jeopardizes plant growth and causes enormous economic loss in the production of many crops, including tobacco (Nicotiana tabacum). Here, we first demonstrated that the roots of bacterial wilt-resistant tobacco mutant KCB-1 can limit the growth and reproduction of Ralstonia solanacearum. Secondly, we demonstrated that KCB-1 specifically induced an upregulation of naringenin content in root metabolites and root secretions. Further experiments showed that naringenin can disrupt the structure of R. solanacearum, inhibit the growth and reproduction of R. solanacearum, and ex

Cometabolic degradation of bisphenol A by pure culture of Ralstonia eutropha and metabolic pathway analysis.

Bisphenol A (BPA) is a toxic compound emitting to the environment mainly by polycarbonate production facilities. In this research, BPA with the initial concentrations in the range of 1-40 mg l-1 was degraded by Ralstonia eutropha. The bacteria were unable to use BPA as the sole carbon source. Therefore, resting and growing cells of phenol-adapted R. eutropha were used for cometabolic biodegradation of BPA with phenol at the concentration of 100 mg l-1. The optimum initial concentrations of BPA were 20 mg l-1 in both approaches of cometabolism. By using resting cells, BPA removal efficiency (RE

Monitoring of Ralstonia eutropha KT1 in groundwater in an experimental bioaugmentation field by in situ PCR.

Ralstonia eutropha KT1, which degrades trichloroethylene, was injected into the aquifer after activation with toluene, and then the number of bacteria was monitored by in situ PCR targeting the phenol hydroxylase gene and by fluorescent in situ hybridization (FISH) targeting 16S rRNA. Before injection of the bacterial suspension, the total concentration of bacteria in the groundwater was approximately 3 x 10(5) cells/ml and the amount of Ralstonia and bacteria carrying the phenol hydroxylase gene as a percentage of total bacterial cells was less than 0.1%. The concentration of bacteria carryin

A Ralstonia solanacearum effector RipAD perturbs aquaporin NbPIP oligomeric status to suppress plant immunity.

Bacterial wilt caused by Ralstonia solanacearum is a devastating disease that affects a wide range of crops, including tobacco. The pathogen promotes infection by secreting type III effector (T3E) proteins into host plants to facilitate pathogenicity; however, the molecular mechanisms for many of these effectors remain elusive. Here, we characterize RipAD, a conserved T3E that significantly contributes to R. solanacearum virulence in Nicotiana tabacum and Nicotiana benthamiana. We demonstrate that RipAD dampens early immune responses, including pathogen-associated molecular pattern-triggered r

Fabrication of pH-Sensitive Tetramycin Releasing Gel and Its Antibacterial Bioactivity against Ralstonia solanacearum.

Ralstonia solanacearum (R. solanacearum)-induced bacterial wilt of the nightshade family causes a great loss in agricultural production annually. Although there has been some efficient pesticides against R. solanacearum, inaccurate pesticide releasing according to the onset time of bacterial wilt during the use of pesticides still hinders the disease management efficiency. Herein, on the basis of the soil pH change during R. solanacearum growth, and pH sensitivity of the Schiff base structure, a pH-sensitive oxidized alginate-based double-crosslinked gel was fabricated as a pesticide carrier.

Antibacterial efficacy of iron oxide and silver nanoparticles against bacterial wilt pathogen Ralstonia solanacearum.

The security of vegetable plants worldwide is threatened by bacterial wilts, one of the most infectious soil-borne bacterial plant diseases. This is caused by R. Solanacearum. Overuse of bactericides and antibiotics to combat bacterial wilt results in pesticide resistance and toxicity to beneficial living organisms. Consequently, nanoparticles are more beneficial, safe for the environment, and have strong antibacterial properties than conventional pesticides. In the present work, iron oxide nanoparticles (IONPs) and silver nanoparticles (AgNPs) were prepared by simple chemical, eco-friendly pr
