Development of a Comprehensive Lesion Severity Classification Model for Largemouth Bass (Micropterus salmoides) Ranavirus (LMBV) Based on Machine Vision.

This study presents the development of a quantitative evaluation method utilizing machine vision technology to characterize the extent of body surface damage in largemouth bass (Micropterus salmoides) infected with largemouth bass ranavirus (LMBV). High-resolution, multi-angle images (6000 × 4000 pixels) of the body surface from 239 infected specimens were acquired at a fixed distance of 40 cm using a SONY ILCE-7RM3 digital camera within a GODOX-LST60 softbox. Key parameters, including the number of segmented injury areas, the count of body surface lesions, and the total lesion area, were anal

Dissection of the global responses of mandarin fish pyloric cecum to an acute ranavirus (MRV) infection reveals the formation of serositis and then ascites.

Mandarin fish ranavirus (MRV), a new member of the species Ranavirus micropterus1, sharing over 98% whole-genome nucleotide identity with the well-known largemouth bass virus (LMBV), is a distinct member of the genus Ranavirus within the family Iridoviridae. Our recent work showed that acute MRV infection predominantly affects the pyloric cecum, a critical visceral organ in mandarin fish, and was hypothesized to drive the characteristic external clinical sign of severe ascites. In this study, we reveal that acute MRV infection initially targets the serosal layer of the pyloric cecum of mandari

Characterization and genomic analysis of a ranavirus associated with cultured black-spotted pond frogs (Rana nigromaculata) tadpoles mortalities in China.

Ranaviruses are globally emerging pathogens causing mass mortality in both farmed and wild populations. In this study, we confirmed that a novel ranavirus was related to mass die-offs of black-spotted pond frogs (Rana nigromaculata) in Sichuan Province, China. Histological examination revealed marked degeneration or necrosis in parenchymatous organs, and intracytoplasmic inclusions can be observed in the liver and intestine cells. The virus was isolated from diseased tadpoles (tentatively named the virus as RNRV), and the ranavirus-like particles could be observed in infected EPC cells by the

A multi-model genome-wide association study identifies genetic variants underlying resistance to Largemouth Bass Ranavirus (LMBV) in Micropterus salmoides.

Largemouth bass (Micropterus salmoides) is an economically important freshwater aquaculture species, yet recurrent outbreaks of Largemouth Bass Ranavirus (LMBV) continue to impair production and cause substantial losses. The genetic basis of host variation in LMBV resistance remains insufficiently characterized. Here, we applied a multi-model genome-wide association study (GWAS) to identify loci associated with resistance following a controlled challenge with the LMBV-23PY strain. Whole-genome resequencing was performed for 146 phenotyped fish, including 72 susceptible and 74 resistant individ

Multi-route induction of Salmonella-based vaccine delivery elicits protective immunity against FV3-like ranavirus in an ornamental fish model.

Viruses belonging to the genus Ranavirus affect the ectothermic animal community worldwide. In this study, we employed a bioengineered Salmonella mutant, JOL2500, to deliver major capsid (MCP) and outer membrane proteins of frog virus 3 (FV3) to elicit FV3 specific immunity. The optimized antigen protein sequences were cloned into a prokaryotic expression vector, pJHL65. The recombinant plasmids were transformed into JOL2500, and the constructed strains were designated as JOL3171 and JOL3172 carrying MCP and membrane protein retrospectively. The constructed vaccine strains were administrated t

Andrias davidianus ranavirus 1R encoding a delayed-early protein promotes cell proliferation by driving cell cycle progression into S phase.

Andrias davidianus ranavirus 1R (ADRV-1R), a core gene of the family Iridoviridae, is predicted to encode a viral transcription factor (vTF) since the protein contains a virus late transcription factor-3 like (VLTF3 like) domain. However, its characteristics and function are still unclear. In this study, the transcription and expression of ADRV-1R were investigated in Chinese giant salamander thymus cells (GSTCs). ADRV-1R transcription starts 6 hours post-infection (hpi), while the protein expression starts 8 hpi. Drug inhibition assay showed that the transcripts are inhibited by cycloheximide

Antioxidant anthocyanin synergistic immune enhancer nanopeptide C-I20 remarkably enhances the protective effect of largemouth bass against largemouth bass ranavirus.

Largemouth bass ranavirus (LMBV) infection results in huge economic losses in largemouth bass (Micropterus salmoides) industry. Nanopeptide C-I20 and anthocyanins have a positive effect on promoting immune responses and antioxidant mechanisms in several aquatic organisms, and are therefore used to inhibit LMBV infection. In this study, we developed an LMBV immersion challenge model using three different viral concentrations (1 × 104 copies/mL, 1 × 105 copies/mL, and 1 × 106 copies/mL) to infect largemouth bass, and LMBV-MCP mRNA expression was detected in infected fish. Following infection, th

Optimization, Physicochemical Characterization, and Immunoprotective Efficacy of an Oral Inactivated Largemouth Bass Ranavirus Vaccine Encapsulated in N,O-Carboxymethyl Chitosan-Chitosan Nanoparticles.

Viral diseases pose a significant threat to aquatic animal health, and no effective treatment is currently available. Vaccines represent the most efficient approach to preventing viral infections. Because of the aquatic habitat, injection-based vaccination is time-consuming and labor-intensive, whereas oral vaccines lack a suitable delivery system. This study aimed to prepare N,O-Carboxymethyl Chitosan (NOCMS)-Chitosan (CS) nanoparticles (NOCMS-CS NPs) via a self-assembly method for the delivery of inactivated largemouth bass ranavirus (LMBV) vaccine (LMBV-iV). Single-factor experiments, the s

Rapid, sensitive, and visual detection of mandarin fish ranavirus and infectious spleen and kidney necrosis virus using an RPA-CRISPR/Cas12a system.

Iridoviruses are large cytoplasmic icosahedral viruses that contain dsDNA. Among them, mandarin fish ranavirus (MRV) and infectious spleen and kidney necrosis virus (ISKNV) are particularly notable due to their high contagiousness and pathogenicity. These viruses pose a significant threat to fish aquaculture, resulting in substantial annual economic losses for the fish farming industry. Therefore, the development of novel, rapid virus detection technologies is essential for the prevention and control of ISKNV and MRV diseases. In this study, we developed a rapid, sensitive, and visual detectio

The Protective Effects and Immunological Responses Induced by a Carboxymethyl Cellulose Microcapsule-Coated Inactivated Vaccine Against Largemouth Bass Ranavirus (LMBRaV) in Largemouth Bass (Micropterus salmoides).

BACKGROUND: Epizootics of largemouth bass ranavirus (LMBRaV) in largemouth bass (Micropterus salmoides) populations are associated with elevated mortality and significant financial losses. Given the lack of effective and safe medication to treat this disease, oral vaccination, which directly targets the intestinal mucosal immune system, is crucial for disease resistance. METHODS: This study utilized carboxymethyl cellulose (CMC) to coat LMBRaV inactivated vaccine (LIV) (micro-CMC@LIV). The morphology and characteristics of the CMC microcapsules were determined. In vitro simulated gastric and

Establishment of quantitative PCR detection methods and epidemiological investigation of ranavirus and megalocytivirus infections in hybrid snakehead (Northern Snakehead ♀ × Blotched Snakehead Channa maculata ♂).

OBJECTIVE: In China, the incidence of iridovirus infections in economically valuable fish species is increasing, but there is a paucity of reports documenting ranavirus infections and there are no reports concerning megalocytivirus in hybrid snakehead (Northern Snakehead Channa argus ♀ × Blotched Snakehead C. maculata ♂). METHODS: In this study, the major capsid protein DNA fragments of two iridoviruses were amplified from diseased hybrid snakehead, and two specific primers and TaqMan probes were designed based on the conserved sequences. Using the constructed recombinant plasmid as the stand

Infection Causes Trade-Offs between Development and Growth in Larval Amphibians.

AbstractTrade-offs between life history traits are context dependent; they vary depending on environment and life stage. Negative associations between development and growth often characterize larval life stages. Both growth and development consume large parts of the energy budget of young animals. The metabolic rate of animals should reflect differences in growth and developmental rates. Growth and development can also have negative associations with immune function because of their costs. We investigated how intraspecific variation in growth and development affected the metabolism of larval

Amphibian immunity-stress, disease, and climate change.

Like all other vertebrate groups, amphibian responses to the environment are mediated through the brain (hypothalamic)-pituitary-adrenal/interrenal (HPA/I) axis and the sympathetic nervous system. Amphibians are facing historically unprecedented environmental stress due to climate change that will involve unpredictable temperature and rainfall regimes and possible nutritional deficits due to extremes of temperature and drought. At the same time, amphibians in all parts of the world are experiencing unprecedented declines due to the emerging diseases, chytridiomycosis (caused by Batrachochytriu

The benefits of coinfection: trematodes alter disease outcomes associated with virus infection.

Coinfections are increasingly recognized as important drivers of disease dynamics. Consequently, greater emphasis has been placed on integrating principles from community ecology with disease ecology to understand within-host interactions among parasites. Using larval amphibians and two amphibian parasites (ranaviruses and the trematode Echinoparyphium sp.), we examined the influence of coinfection on disease outcomes. Our first objective was to examine how priority effects (the timing and sequence of parasite exposure) influence infection and disease outcomes in the laboratory. We found that

Amphibian (Xenopus laevis) Tadpoles and Adult Frogs Differ in Their Antiviral Responses to Intestinal Frog Virus 3 Infections.

The global amphibian declines are compounded by ranavirus infections such as Frog Virus 3 (FV3), and amphibian tadpoles more frequently succumb to these pathogens than adult animals. Amphibian gastrointestinal tracts represent a major route of ranavirus entry, and viral pathogenesis often leads to hemorrhaging and necrosis within this tissue. Alas, the differences between tadpole and adult amphibian immune responses to intestinal ranavirus infections remain poorly defined. As interferon (IFN) cytokine responses represent a cornerstone of vertebrate antiviral immunity, it is pertinent that the

Water Contaminants Associated With Unconventional Oil and Gas Extraction Cause Immunotoxicity to Amphibian Tadpoles.

Chemicals associated with unconventional oil and gas (UOG) operations have been shown to contaminate surface and ground water with a variety of endocrine disrupting compounds (EDCs) inducing multiple developmental alteration in mice. However, little is known about the impacts of UOG-associated contaminants on amphibian health and resistance to an emerging ranavirus infectious disease caused by viruses in the genus Ranavirus, especially at the vulnerable tadpole stage. Here we used tadpoles of the amphibian Xenopus laevis and the ranavirus Frog virus 3 (FV3) as a model relevant to aquatic envir

Isolation and characterisation of a novel Bohle-like virus from two frog species in the Darwin rural area, Australia.

Twelve captive magnificent tree frogs Litoria splendida and 2 green tree frogs L. caerulea on a property in the Darwin rural area (Northern Territory, Australia) either died or were euthanased after becoming lethargic or developing skin lesions. Samples from both species of frog were submitted for histopathology and virus isolation. An irido-like virus was cultured from tissue samples taken from both species and was characterised using electron microscopy, restriction enzyme digests and nucleic acid amplification and sequencing. The isolates were determined to belong to the genus Ranavirus, we
