Decoding Burkholderia Cepacia complex: recA-based species differentiation and antimicrobial susceptibility pattern.

The Burkholderia cepacia complex (BCC) comprises more than twenty-four species of Gram-negative bacteria that are known to cause infections in humans, particularly in immunocompromised patients. While BCC is most commonly associated with respiratory infections in people with cystic fibrosis (CF), it can also lead to a range of infections in individuals without CF. The goal of the current study was to molecularly identify and characterize Bcc species responsible for infections in hospitalised and non-hospitalised patients in Erbil province, using PCR to amplify and sequence the recA gene of the

Preexisting variation in DNA damage response predicts the fate of single mycobacteria under stress.

Clonal microbial populations are inherently heterogeneous, and this diversification is often considered as an adaptation strategy. In clinical infections, phenotypic diversity is found to be associated with drug tolerance, which in turn could evolve into genetic resistance. Mycobacterium tuberculosis, which ranks among the top ten causes of mortality with high incidence of drug-resistant infections, exhibits considerable phenotypic diversity. In this study, we quantitatively analyze the cellular dynamics of DNA damage responses in mycobacteria using microfluidics and live-cell fluorescence ima

Zinc blocks SOS-induced antibiotic resistance via inhibition of RecA in Escherichia coli.

Zinc inhibits the virulence of diarrheagenic E. coli by inducing the envelope stress response and inhibiting the SOS response. The SOS response is triggered by damage to bacterial DNA. In Shiga-toxigenic E. coli, the SOS response strongly induces the production of Shiga toxins (Stx) and of the bacteriophages that encode the Stx genes. In E. coli, induction of the SOS response is accompanied by a higher mutation rate, called the mutator response, caused by a shift to error-prone DNA polymerases when DNA damage is too severe to be repaired by canonical DNA polymerases. Since zinc inhibited the o

A genome-wide CRISPRi screen identifies homologous recombination pathway as potential target for broad-spectrum antibiotic adjuvants.

INTRODUCTION: The widespread misuse and overuse of antibiotics have driven the emergence of multidrug-resistant and pan drug-resistant bacteria, constituting a formidable global health threat. Antibiotic adjuvants that potentiate the efficacy of existing antibiotics represent a particularly promising avenue to address this challenge. METHODS: We performed a genome-wide CRISPR interference (CRISPRi) screening to identify potential targets for broad-spectrum antibiotic adjuvants, which highlighted the homologous recombination pathway as a promising candidate. To functionally validate this pathw

Opposing regulation of Gig2 by RecA and GlrK controls redox-linked desiccation tolerance in Cronobacter sakazakii.

Cronobacter sakazakii is a desiccation-tolerant foodborne pathogen that poses a serious threat to neonatal health through contamination of powdered infant formula. Despite its clinical relevance and exceptional persistence in dry environments, the molecular basis of its desiccation tolerance remains poorly defined. Here, we systematically investigated the genetic and physiological determinants of C. sakazakii survival under desiccation stress. A functional screen of gene knockout strains revealed intricate relationships between surface hydrophilicity and desiccation tolerance, suggesting the i

The recA operon: A novel stress response gene cluster in Bacteroides fragilis.

Bacteroides fragilis, an opportunistic pathogen of humans, is a leading cause of bacteraemias and anaerobic abscesses which are often treated with metronidazole, a drug which damages DNA. This study investigated the responses of the B. fragilis recA three gene operon to the stress experienced during metronidazole treatment and exposure to reactive oxygen species simulating those generated by the host immune system during infection. A transcriptionally regulated response was observed using quantitative RT-PCR after metronidazole and hydrogen peroxide treatment, with all three genes being upregu

RecA and RadA proteins of Brucella abortus do not perform overlapping protective DNA repair functions following oxidative burst.

Very little is known about the role of DNA repair networks in Brucella abortus and its role in pathogenesis. We investigated the roles of RecA protein, DNA repair, and SOS regulation in B. abortus. While recA mutants in most bacterial species are hypersensitive to UV damage, surprisingly a B. abortus recA null mutant conferred only modest sensitivity. We considered the presence of a second RecA protein to account for this modest UV sensitivity. Analyses of the Brucella spp. genomes and our molecular studies documented the presence of only one recA gene, suggesting a RecA-independent repair pro

Paradigm shifts in vaccine development: lessons learned about antigenicity, pathogenicity and virulence of Brucellae.

As part of a program to support the USDA Animal Plant Health Inspection Service Bovine Brucellosis Eradication Program, the Brucellosis Research Unit of the National Animal Disease Center (NADC) sought to develop a bovine brucellosis vaccine that would allow vaccinated animals to be distinguished from virulent field infected animals. In order to meet that goal, several avenues of research were undertaken to construct and test candidate vaccines, including Brucella abortus RB51. In early vaccine development studies, a subunit preparation obtained by extracting B. abortus with salts was studied

A Novel Isothermal Assay of Borrelia burgdorferi by Recombinase Polymerase Amplification with Lateral Flow Detection.

A novel isothermal detection for recombinase polymerase amplification with lateral flow (LF-RPA) was established for Borrelia burgdorferi (B. burgdorferi) detection in this study. This assay with high sensitivity and specificity can get a visible result without any additional equipment in 30 min. We designed a pair of primers according to recA gene of B. burgdorferi strains and a methodology evaluation was performed. The results showed that the RPA assay based on the recA gene was successfully applied in B. burgdorferi detection, and its specific amplification was only achieved from the genomi

Tuberculosis vaccine strain Mycobacterium bovis BCG Russia is a natural recA mutant.

BACKGROUND: The current tuberculosis vaccine is a live vaccine derived from Mycobacterium bovis and attenuated by serial in vitro passaging. All vaccine substrains in use stem from one source, strain Bacille Calmette-Guérin. However, they differ in regions of genomic deletions, antigen expression levels, immunogenicity, and protective efficacy. RESULTS: As a RecA phenotype increases genetic stability and may contribute restricting the ongoing evolution of the various BCG substrains while maintaining their protective efficacy, we aimed to inactivate recA by allelic replacement in BCG vaccine s

Stationary-phase Pseudomonas aeruginosa fluoroquinolone persisters mostly avoid DNA double-stranded breaks.

UNLABELLED: When susceptible bacterial cultures are treated with antibiotics, some cells can survive treatment without heritable resistance, giving rise to susceptible daughter cells in a phenomenon termed antibiotic persistence. Current models of fluoroquinolone (FQ) persistence in stationary-phase cultures posit that post-treatment resuscitation is dependent on double-stranded break (DSB) repair through RecA-mediated homology-directed repair. Previously, we reported that stationary-phase P. aeruginosa does not depend on RecA to persist. In this work, we ask whether P. aeruginosa FQ persister

Whole genome sequencing reveals recA gene recombination, virulence factors, antibiotic resistance, and emergence of clonal complexes in Vibrio parahaemolyticus associated with acute hepatopancreatic necrosis disease (AHPND) in shrimp.

The AHPN disease, caused by Vibrio parahaemolyticus, is a critical threat to global shrimp aquaculture. In this study, we sequenced and analyzed 5 AHPND-associated V. parahaemolyticus genomes, referred to as CGVP strains (CGVP3, CGVP8, CGVP10, CGVP11, and CGVP18) that were isolated during the 2016 disease outbreak in Korea. These genomes, averaging ∼ 5.3 Mbp, consist of two circular chromosomes (3.3-3.4 Mbp and 1.8-2.0 Mbp) and a 69 kbp plasmid carrying pirABvp toxin genes. Within the genomes, a unique 20.6 kbp insertion in the recA gene, characterized by a lower GC content and 15 open reading

DNA recombination and repair in Wolbachia: RecA and related proteins.

Wolbachia is an obligate intracellular bacterium that has undergone extensive genomic streamlining in its arthropod and nematode hosts. Because the gene encoding the bacterial DNA recombination/repair protein RecA is not essential in Escherichia coli, abundant expression of this protein in a mosquito cell line persistently infected with Wolbachia strain wStri was unexpected. However, RecA's role in the lytic cycle of bacteriophage lambda provides an explanation for retention of recA in strains known to encode lambda-like WO prophages. To examine DNA recombination/repair capacities in Wolbachia

Suramin is a potent and selective inhibitor of Mycobacterium tuberculosis RecA protein and the SOS response: RecA as a potential target for antibacterial drug discovery.

OBJECTIVES: In eubacteria, RecA is essential for recombinational DNA repair and for stalled replication forks to resume DNA synthesis. Recent work has implicated a role for RecA in the development of antibiotic resistance in pathogenic bacteria. Consequently, our goal is to identify and characterize small-molecule inhibitors that target RecA both in vitro and in vivo. METHODS: We employed ATPase, DNA strand exchange and LexA cleavage assays to elucidate the inhibitory effects of suramin on Mycobacterium tuberculosis RecA. To gain insights into the mechanism of suramin action, we directly visu

RecA-like activity in mammalian cell extracts of different origin.

The occurrence of a RecA-like activity similar to the one detected in the fibroblast cell line GM1492 derived from a patient suffering from the autosomal recessive disease Bloom's syndrome has been investigated in cell extracts of different origin. The formation of D-loop containing joint molecules from phi X174 RFI DNA and fragments of phi X174 single-stranded DNA by partially purified extracts was measured by a filter binding assay. The RecA-like activity, dependent on ATP and Mg2+, was detected at an elevated level only in the human and rodent cell lines, GM1492 and CHO respectively. The le

A novel multiplex PCR for the accurate identification of four clinically relevant Cutibacterium species.

Cutibacterium is a genus of skin commensals involved in acne inflammation, occasionally causing healthcare-associated infections. Previously established multiplex PCR assays have been unable to detect Cutibacterium modestum. However, due to the reported occurrence of infections caused by this species, a comprehensive identification method that includes C. modestum is warranted. The objective of this study was to develop a novel multiplex PCR method that can rapidly distinguish between four Cutibacterium species, thereby facilitating the appropriate management of conditions such as acne and pro

A novel antimicrobial polymer efficiently treats multidrug-resistant MRSA-induced bloodstream infection.

The present study aimed to ascertain if polymer 2a, a novel synthesized antimicrobial polyionene, could treat methicillin-resistant Staphylococcus aureus (MRSA)-induced bloodstream infection. The minimum inhibitory concentration (MIC) of polymer 2a against MRSA was detected. A time-kill assay was employed to determine the killing kinetic of polymer 2a. Potential antimicrobial mechanisms of polymer 2a, including membrane disruption and programmed cell death (PCD), were explored. A resistance development assay was introduced to determine the propensity of polymer 2a toward resistance against MRS

Ciprofloxacin-Mediated Mutagenesis Is Suppressed by Subinhibitory Concentrations of Amikacin in Pseudomonas aeruginosa.

Resistance to antibiotics is a global health problem. Activation of the SOS response, and the subsequent elevation in mutagenesis, contributes to the appearance of resistance mutations. Among currently used drugs, quinolones are the most potent inducers of the SOS response. In the present study, we show that amikacin inhibits ciprofloxacin-mediated SOS induction and mutagenesis in Pseudomonas aeruginosa.

Non-canonical double-stranded DNA break repair in Pseudomonas aeruginosa includes LigD-independent non-homologous end joining.

Pseudomonas aeruginosa is a highly adaptive Gram-negative pathogen that is commonly managed with fluoroquinolone (FQ) antibiotics. Even without heritable resistance mutations, FQ-susceptible cells of P. aeruginosa can survive treatment in a phenomenon termed antibiotic persistence. Current models of stationary-phase FQ persistence established with other bacterial species posit that persister survival is dependent on DNA double-stranded break (DSB) repair via RecA-mediated homologous recombination (HR). In P. aeruginosa, however, the effects of RecA loss on FQ persistence vary depending on the
