Oxytocin receptors in the Magel2 mouse model of autism: Specific region, age, sex and oxytocin treatment effects.

The neurohormone oxytocin (OXT) has been implicated in the regulation of social behavior and is intensively investigated as a potential therapeutic treatment in neurodevelopmental disorders characterized by social deficits. In the Magel2-knockout (KO) mouse, a model of Schaaf-Yang Syndrome, an early postnatal administration of OXT rescued autistic-like behavior and cognition at adulthood, making this model relevant for understanding the actions of OXT in (re)programming postnatal brain development. The oxytocin receptor (OXTR), the main brain target of OXT, was dysregulated in the hippocampus

Identification, localization and functional activity of oxytocin receptors in epididymis.

Oxytocin (OT) is a neurohypophysial hormone with unclear physiological functions in the male. Several previous studies indicated that OT might have a role in the ejaculatory process, stimulating sperm release from the epididymal storage. In this study we investigated on the presence and function of OT receptor (OTR) in rabbit and human epididymis. By using RT-PCR, Western and binding studies, we found that OTR gene and protein is expressed in the human epididymis and stimulates in vitro contractility. The immunolocalization of OTR suggests that the receptor is not only present in the smooth mu

Targeting the oxytocin receptor to relax the myometrium.

Oxytocin acting via its receptor is involved in the myometrial hyperactivity of preterm labour and possibly also in that of primary dysmenorrhoea. The closely related hormone vasopressin acting on its uterine receptor of type V1a may also contribute to the myometrial hyperactivity of these conditions. Several pharmaceutical compounds inhibiting these receptors are, therefore, under development and one substance, atosiban, has now been registered in many countries for the treatment of preterm labour. This compound blocks both the oxytocin and the vasopressin V1a receptor. The efficacy is at lea

Investigation of the oxytocin receptor at the molecular level.

Before the onset of labor, the myometrium becomes extremely sensitive to oxytocin, for which it is a primary target tissue, because of a tremendous increase in the number of oxytocin receptors. To study this dynamic expression and regulation of the receptor, we cloned the human oxytocin receptor cDNA in 1992 and characterized its expression in female reproductive organs and in the rat brain using the receptor cDNA and anti-oxytocin receptor monoclonal antibodies. In the uterus, we revealed that oxytocin receptor expression is mainly regulated at the transcriptional level. For the investigation

The oxytocin receptor, luteolysis and the maintenance of pregnancy.

During luteal regression episodic pulses of oxytocin secretion become coupled to the release of prostaglandin F2 alpha (PGF2 alpha) following synthesis of endometrial oxytocin receptors, but in early pregnancy the inhibition of oxytocin receptor formation by the conceptus prevents the development of the pulsatile pattern of PGF2 alpha release needed to achieve luteolysis. Oxytocin receptors are present on the luminal epithelium in ovariectomized and anoestrous ewes, in pregnant animals throughout most of gestation (day 21 to term) and in explants of endometrial tissue cultured in vitro. These

Oxytocin and the timing of parturition. Influence of oxytocin receptor gene expression, oxytocin secretion, and oxytocin-induced prostaglandin F2 alpha and E2 release.

Oxytocin receptor (OTR) gene expression was studied in various tissues of the reproductive tract of pregnant cows and compared with ligand binding activity. Myometrium, intercaruncular endometrium, caruncular endometrium, cotyledons and fetal membranes, as well as the uterine cervix of pregnant cows expressed the bovine OTR gene. Receptor concentrations, measured by ligand binding to crude microsomal pellets, were comparable to OTR mRNA signal strength in all instances indicating that the receptor protein formation is probably regulated at the transcriptional level. During bovine pregnancy OTR

Systemic oxytocin and vasopressin excite gastrointestinal motility through oxytocin receptor in rabbits.

The aim of the present study was to investigate the effect of systemic oxytocin (OT) and vasopressin (VP) on the motility of stomach and duodenum. Two plastic balloons made of condom were inserted into stomach and duodenum to monitor the change of mean pressure. Intravenous injection of OT (0.1-0.8 microg kg(-1)) or VP (0.02-0.08 IU kg(-1)) dose-dependently increased the stomach and duodenum pressure. Pretreatment of atosiban (1 microg kg(-1)), the specific OT receptor (OTR) antagonist, attenuated the excitatory effect of OT or VP on the pressure of stomach and duodenum. Pretreatment of V1880

Differential Effects of Oxytocin Receptor Antagonists, Atosiban and Nolasiban, on Oxytocin Receptor-Mediated Signaling in Human Amnion and Myometrium.

One of the most established roles of oxytocin (OT) is in inducing uterine contractions and labor. Apart from inducing contractions, our recent studies showed that OT can also activate proinflammatory pathways in both human myometrial and amnion cells, which suggests that the proinflammatory role of OT should be taken into account when developing tocolytics targeting the OT/oxytocin receptor (OTR) system. The OTR antagonist, atosiban, is currently used therapeutically for the treatment of preterm labor. We previously showed that atosiban fails to inhibit the proinflammatory effects of OT in hum

Seasonal expressions of oxytocin and oxytocin receptor in epididymis of the male muskrat (Ondatra zibethicus).

Oxytocin (OT) is presented in the male reproductive tract and has various physiological functions, such as stimulating contraction of the reproductive tract and aiding sperm transport. In this study we investigated seasonal expressions of OT and oxytocin receptor (OTR) in the epididymis of the muskrat during the breeding and non-breeding seasons. Morphologically, the weight and length of epididymis in the breeding season were significantly higher than those in the non-breeding season. In the breeding season, the lumen diameter and epithelial thickness of epididymis increased significantly, and

Oxytocin and oxytocin receptor gene expression in the reproductive tract of the pregnant cow: rescue of luteal oxytocin production at term.

The peptide hormone oxytocin is produced both in the hypothalamus and in certain peripheral organs. The extent of extra-hypothalamic hormone synthesis in the pregnant cow has not previously been examined. In this study we have analyzed different tissues from the pregnant bovine reproductive tract and corpus luteum for the presence of mRNA encoding the oxytocin peptide as well as the oxytocin receptor. In uterine tissues oxytocin mRNA could only be detected sporadically with the help of a reverse transcription-polymerase chain reaction method, implying only very low levels of expression. The ca

Central neurokinin 3 receptors increase systemic oxytocin release: interaction with norepinephrine.

Stimulation of central tachykinin receptors contributes to neuroendocrine functions of the hypothalamo-neurohypophyseal system. However, the specific role of each tachykinin receptor subtype has not been completely characterized. Specifically, while neurokinin 3 (NK3) receptor stimulation increases systemic vasopressin, the effects on oxytocin (OT) are not known. Therefore, the present studies investigated the effect of central NK3 receptor stimulation with senktide on release of systemic and central OT. Furthermore, since central NK3 receptors activate noradrenergic systems, which contribute

Oxytocin receptors in guinea pig myometrium near term and during labor.

Oxytocin receptors in myometrium of women, rats, and rabbits rise markedly before the onset of labor, suggesting a role in the initiation of labor. In guinea pigs, a previous study reported no such rise by one-point determination of oxytocin binding. The purpose of this study was to use a more rigorous method to determine whether the binding characteristics of myometrial oxytocin receptors change in relation to labor in guinea pigs. Competitive binding studies were carried out in microsomes from inner and outer myometrium between 42 days of gestation and labor. Binding to analogs was also test

Hormonal control of concentrations of endometrial oxytocin receptors in the ewe.

Uterine oxytocin receptors have been shown to play a major role in the regulation of uterine prostaglandin F2 alpha release during the oestrous cycle and early pregnancy in sheep. The concentration of endometrial oxytocin receptors increases sharply from around Day 13 of the oestrous cycle to reach a maximum between Days 15 and 16. The high concentration of endometrial oxytocin receptors at this time coincides with the release of endogenous uterine prostaglandin F2 alpha during luteal regression and the maximum uterine prostaglandin F2 alpha response to an oxytocin stimulus. The concentration

Behavioral effects of progesterone associated with rapid modulation of oxytocin receptors.

The ventromedial nuclei of the hypothalamus (VMN) are important for the control of feminine mating behavior, and hormone action within these nuclei has been causally related to behavior. Estradiol induces receptors for oxytocin in the VMN and in the area lateral to these nuclei over the course of 1 to 2 days, and progesterone causes, within 30 minutes of its application, a further increase in receptor binding and an expansion of the area covered by these receptors lateral to the VMN. The rapid progesterone effect appears to be a direct and specific effect of this steroid on the receptor or mem

Myometrial oxytocin receptor mRNA concentrations at preterm and term delivery - the influence of external oxytocin.

The hormonal system for induction of term and preterm labour is not fully understood. Therefore, we investigated myometrial gene expressions for neurohypophyseal hormones and their receptors, prostaglandin F(2alpha) and ovarian steroid receptors in women delivered by Caesarean section. Myometrial tissue for real time PCR was collected from 39 women delivered at term before and after the onset of labour and preterm. Women delivered electively at term had significantly higher oxytocin receptor mRNA expressions (2.52 +/- 0.37 oxytocin receptor/actin; median +/- SEM) than those delivered with ongo

Coincident increases in oxytocin receptor expression and EMG responsiveness to oxytocin in the ovine cervix at oestrus.

This study has localised oxytocin receptor (OTR) mRNA expression within the cervix of non-pregnant ewes and related this to changes in the sensitivity of the cervical musculature to administered oxytocin (OT) during the oestrous cycle by recording electromyographic (EMG) activity. Cervices were collected from 34 ewes at specified time points throughout the cycle. OTR mRNA expression was localised by in situ hybridisation and results were expressed as optical density measurements from autoradiographs in each of four different cervical regions. EMG recordings were made for up to 8 h per day from

The potentialities of oxytocin receptor inhibitors for endometriosis therapy.

OBJECTIVE: Genital endometriosis (GE) is a widespread gynecological disease which requires its further pathogenesis investigation and search for new effective treatments. The known data of oxytocin receptor presence in endometrioid heterotopy smooth muscle cells give some grounds to assume oxytocin participation in the pathogenesis of endometriosis. The present study objective was to evaluate oxytocin level in peripheral blood (PB) in patients with endometriosis associated pain syndrome and to estimate the efficacy of oxytocin receptor inhibitors (IOXTR) administration based on animal endometr
