Acute tolerance to the hypnotic effect of morphine in rats.

To demonstrate the development of acute tolerance to the hypnotic effects of morphine, loss and recovery of the righting reflex with a constant-rate morphine infusion was studied in rats. In one group of animals, brain and serum concentrations of morphine were detected (radioimmunoassay) at the time of loss of the righting reflex, and in another group, at the time of the reflex recovery. The morphine infusion at a constant rate of 14 mg.kg-1.h-1 caused a loss of the righting reflex in all animals that was achieved by 2.5 h. However, this level of response could not be maintained, and at 5 h it

Vagal reflex provoked by radiofrequency catheter ablation in the right aortic sinus cusp: a Bezold-Jarisch-like phenomenon.

A 66-year-old woman with idiopathic premature ventricular contractions with a left bundle branch block QRS morphology and left inferior axis underwent electrophysiologic testing. Successful radiofrequency ablation was achieved in the right coronary cusp (RCC). However, radiofrequency ablation at sites adjacent to the successful ablation site provoked sinus bradycardia followed by atrioventricular conduction block. That phenomenon might be explained by a vagal reflex through stimulation of vagal pathways or receptors in the anterior epicardial fat pads neighboring to the RCC. A vagal reflex sho

[Right pulmonary artery arising from the proximal ascending aorta. A model of reflex pulmonary hypertension of the left lung?].

The right pulmonary artery arising from the proximal ascending aorta is a rare and severe malformation. This retrospective study of 11 children with this condition was undertaken to determine the conditions of diagnosis, to analyse the results of surgery, and, above all, to clarify the mechanism of the left pulmonary arterial hypertension which was always present. Ten of these patients were 4 to 90 days old. All had severe congestive cardiac failure with iso- or suprasystemic left pulmonary arterial hypertension. The only associated lesions were ventricular septal defect (1 case) and patent du

[30% intralipid shortens the duration of anesthesia induced by etomidate in rats].

OBJECTIVE: To examine whether 30% intralipid could shorten the duration of anesthesia induced by etomidate and its underlying mechanism. METHODS: Male Sprague-Dawley rats were anesthetized by etomidate (2.85 mg/kg) through caudal vein. After loss of righting reflex, 40 rats were randomly divided into four groups, each being administered intravenously with 2 mL 30% intralipid, 2 mL normal saline, 0.5 mL 30% intralipid, and 0.5 mL normal saline, respectively. The durations from loss of righting reflex to revival of righting reflex and to normal activities were recorded. In the study in vitro, t

Acceleration registrography of head movement during alternating inclination of the support platform.

The righting reflex, essential for maintaining equilibrium, keeps the head and trunk in a state of balance with regard to gravity. Because patients suffering from vestibular disorders almost always complain of dizziness during everyday movements such as rising and walking, the righting reflex should be evaluated within the context of these dynamic states. A new method of acceleration registrography was designed to enable the testing of this reflex as the subject stands upon a moving platform. Participating in the study were 49 normal control subjects and 80 patients with peripheral vestibular

Age-dependent effects of chronic intermittent ethanol treatment: Gross motor behavior and body weight in aged, adult and adolescent rats.

The proportion of people in the population who are elderly is rapidly increasing. In addition, dangerous alcohol consumption in this demographic is rising. Approximately 33% of all people with an alcohol use disorder are diagnosed with late onset alcoholism. However, few suitable animal models for late onset alcoholism exist, making it difficult to investigate the impact of alcoholism later in life. The current study investigated if chronic intermittent ethanol exposure via intraperitoneal injections every other day for 20days in aged, adult and adolescent male rats differentially alters body

Both ethanol toxicity and thiamine deficiency are necessary to produce long-term memory deficits in the young chick.

This series of studies revealed that a 2.5-mg dose of oxythiamine administered over 2 days yielded memory deficits from 10 min following passive avoidance learning. This result existed in association with slowing of righting reflex. Administration of thiamine reversed the memory deficit and the slowing of the righting reflex. A combination of oxythiamine and peripheral alcohol administration also resulted in memory deficits appearing from 10 min after training; however, in contrast to the effect of thiamine deficiency alone, the deficits induced by thiamine deficiency in association with acute

Revisiting MAC as a static measure of anaesthesia: evidence for dynamic responses under isoflurane in male mice.

BACKGROUND: The presence of a righting reflex is known to vary over time in an individual at the population half-maximal effective anaesthetic concentration (EC50). However, it is unclear whether similar variability occurs for other anaesthetic endpoints at comparable anaesthetic concentrations. We sought to test whether individual responses to a pedal pinch, a strong aversive stimulus, vary over time in individuals at the population EC50 for stimulus response and to explore the dynamics of individual responses. METHODS: Male mice were exposed to fixed concentrations of isoflurane, and aversi

A Spinal Mechanism Related to Left-Right Symmetry Reduces Cutaneous Reflex Modulation Independently of Speed During Split-Belt Locomotion.

Task- and phase-dependent reflex modulation during locomotion is well established, but we do not know the signals driving this modulation. To determine whether signals related to left-right symmetry of the locomotor pattern modulate cutaneous reflexes, we stimulated the superficial peroneal nerve in five intact female cats and in four spinal-transected cats (spinal cats, two males and two females) during split-belt locomotion at different left-right speeds. We compared cutaneous reflexes evoked in three ipsilateral and two contralateral hindlimb muscles during split-belt locomotion with those

Congenital rubella syndrome: function of equilibrium of 80 cases with deafness.

The equilibrium function of 80 deaf children with congenital rubella syndrome, aged 12, was examined by the caloric and righting reflex tests. Of these 80 children, 24 (30%) had vestibular hypofunction, 6 (7.5%) were suspect, and the remaining 50 (62.5%) were normal. Of all 160 ears tested, vestibular hypofunction was proven in 21.3%, suspect in 5.6%, and normal in the rest. There was no correlation between the incidence of vestibular hypofunction and the degree of hearing loss. Of the 30 children with vestibular hypofunction, 14 (47%) showed a normal righting reflex, suggesting the working of

A fixed moderate-dose combination of tiletamine+zolazepam outperforms midazolam in induction of short-term immobilization of ball pythons (Python regius).

Laboratory animals are commonly anesthetized to prevent pain and distress and to provide safe handling. Anesthesia procedures are well-developed for common laboratory mammals, but not as well established in reptiles. We assessed the performance of intramuscularly injected tiletamine (dissociative anesthetic) and zolazepam (benzodiazepine sedative) in fixed combination (2 mg/kg and 3 mg/kg) in comparison to 2 mg/kg of midazolam (benzodiazepine sedative) in ball pythons (Python regius). We measured heart and respiratory rates and quantified induction parameters (i.e., time to loss of righting re

An evaluation of three neuroleptanalgesic combinations in rabbits.

In this study, novel combinations of analgesics and neuroleptics were used in the rabbit in an attempt to produce a surgical level of anesthesia. A commercially available mixture of fentanyl (0.06 mg/kg) and droperidol (3.0 mg/kg; F/D) was evaluated alone and in combination with either the benzodiazepine derivative, diazepam (2 mg/kg) or the alpha-2 adrenoceptor agonist, detomidine (20 micrograms/kg). Rabbits were anesthetized on consecutive weeks with one of the three regimens. Heart rate, respiratory rate, blood pressure, and arterial blood gases (pH, PCO2, PO2) were measured throughout each

Developmental differences in the acquisition of tolerance to ethanol.

This study was designed to compare the development of tolerance to ethanol in adolescent and adult rats. Rats were pretreated with ethanol (4 g/kg) twice daily by intragastric gavage for 3 or 7 days, and then challenged with a single IP dose of ethanol (5 g/kg). Throughout the pretreatment period body temperature was measured before and after the morning dosage. During the IP challenge test we measured body temperature, duration of the loss of righting reflex, and blood ethanol level upon regaining the righting reflex. The adolescent rats that were pretreated for 7 days developed greater toler

The Auto-Tilt Test in patients with positional vertigo and Menière's disease.

As a new method of testing the righting reflex, which has been considered to be closely related to the otolith function, we used the Auto-Tilt Test (ATT) on 24 patients with positional vertigo, on 5 patients with Meniere's disease before and after cochlearendolymphatic shunt, and on 10 normal subjects. The subject is required to stand on an electrically driven tilt board with an autoregulating switch in hand, and to perform bilateral tilt swings of 5 degrees and 10 degrees wearing a helmet with a Head Inclinometer or tilt sensor. The result of this test in patients with positional vertigo reve

The effects of (+)-SKF10047 and ketamine hydrochloride on stereotyped behaviour, locomotor activity and ataxia in guinea pig.

Although behavioural studies of the effects of (+/-)-N-allylnormetazocine ((+)-SKF10047) and ketamine hydrochloride have been conducted in many species, there are no data available on their effects in guinea pig. The aim of the present study was to provide an analysis of the effects of these drugs on stereotyped behaviour, locomotor activity, ataxia and righting reflex latency in guinea pigs. (+)-SKF10047 did not produce significant stereotyped behaviour nor locomotor hyperactivity at any of the doses tested (2.5-10.0 mg/kg, i.p). While righting reflex latency was not significantly affected, s

Effects of drugs acting alone and in combination on the motor activity of intact mice.

1. When administered to intact white mice, the central depressants-diphenhydramine, promethazine, chlorpromazine, gammahydroxybutyrate, gammabutyrolactone, hyoscine, and pethidine-produced sedation in small doses, but excitement and convulsions in higher doses. When given to mice pretreated with subanaesthetic doses of phenobarbitone these drugs abolished the righting reflex both in convulsant doses (hyoscine excepted) and in non-convulsant doses. These effects are similar to the effects previously observed with local anaesthetics.2. Meprobamate, diazepam and chlorpromazine produced a loss of

The indices of potency for intravenous anaesthetics.

In rat experiments, the relative potency an safety of thiopentone, diazepam and etomidate were assessed using different indices of anaesthesia - loss of righting reflex, prevention of movement and heart rate responses to an noxious stimulus. Log-probit dose-response curves for these end-points and for lethal effect were determined. Etomidate proved to be more potent than thiopentone or diazepam; its relative potency figures varied from 5 to 18 with the use of different end-points of anaesthesia. According to the ED50 level of response for the loss of righting reflex and increased of heart rate

Effects of morphine on pentobarbital-induced responses in mu-opioid receptor knockout mice.

Effects of morphine on the potentiation of pentobarbital-induced responses were investigated using mu-opioid receptor knockout mice. The duration of loss of righting reflex, hypothermia, and loss of motor coordination induced by pentobarbital were measured after pretreatment with either morphine or saline. Morphine pretreatment failed to show potentiation of both pentobarbital-induced loss of righting reflex and hypothermia in mu-opioid receptor knockout mice, while it significantly potentiated these responses in the wild-type controls. For motor incoordination test, morphine potentiated pento
