Performance on a progressive-ratio schedule of food reinforcement during concurrent access to a sucrose solution or tap water.

Seven food-deprived rats were observed on a progressive-ratio 5 schedule of food reinforcement under baseline conditions and during concurrent access to tap water, a 1 % sucrose solution, or a 5 % sucrose solution. Subjects responded for the alternative reinforcer primarily during the preratio pause. Responding for the alternative reinforcer caused a local increase in pause durations in all conditions, however, the mean pause duration for the session increased only when the 5 % sucrose solution was available. Otherwise, the increase in pause durations that accompanied responding for the altern

Fault-Tolerant Scheduling Mechanism for Dynamic Edge Computing Scenarios Based on Graph Reinforcement Learning.

With the proliferation of Internet of Things (IoT) devices and edge nodes, edge computing has taken on much of the real-time data processing and low-latency response tasks which were previously managed by cloud computing. However, edge computing often encounters challenges such as network instability and dynamic resource variations, which can lead to task interruptions or failures. To address these issues, developing a fault-tolerant scheduling mechanism is crucial to ensure that a system continues to operate efficiently even when some nodes experience failures. In this paper, we propose an in

Discrimination of pentobarbital doses and drug mixtures under fixed-ratio and fixed-interval reinforcement schedules.

Pigeons were trained to discriminate among 5 mg/kg pentobarbital, 10mg/kg pentobarbital, and saline, under either fixed-interval (FI) or fixed-ratio (FR) reinforcement schedules. When baseline responding stabilized, a higher percentage of responses occurred on the key that produced the reinforcer under the FR schedule than under the FI schedule. After low doses of pentobarbital, responding shifted from the saline key to the 5 mg/kg pentobarbital key; at higher doses of pentobarbital responding shifted to the 10mg/kg pentobarbital key under both schedules. After low doses of ethanol and chlordi

Optimal Fractionation Scheduling for Radiotherapy Treatments with Reinforcement Learning, Tumor Growth Modeling and Outcome Modeling.

Objective: Radiotherapy is a primary method for cancer treatment, wherein radiation doses are divided into multiple sessions or fractions to effectively target tumors and minimize damage to surrounding tissues. Methods: In this study, we leverage reinforcement learning (RL) to enhance treatment planning with the aim of improving the adaptability and robustness of RL agents given the inherent inaccuracies in tumor growth models. A 2D simulation model of tumor growth is employed, where tabular RL techniques are used to determine the optimal treatment strategies. We emphasize the significance of

Deep Reinforcement Learning for Charging Scheduling of Electric Vehicles Considering Distribution Network Voltage Stability.

The rapid development of electric vehicle (EV) technology and the consequent charging demand have brought challenges to the stable operation of distribution networks (DNs). The problem of the collaborative optimization of the charging scheduling of EVs and voltage control of the DN is intractable because the uncertainties of both EVs and the DN need to be considered. In this paper, we propose a deep reinforcement learning (DRL) approach to coordinate EV charging scheduling and distribution network voltage control. The DRL-based strategy contains two layers, the upper layer aims to reduce the o

Prenatal methylmercury exposure increases responding under clocked and unclocked fixed interval schedules of reinforcement.

Recent experiments have suggested that developmental methylmercury exposure produces perseverative behavior in adulthood. In the present experiment, interactions between developmental low-level methylmercury (MeHg) and nutritionally relevant dietary selenium (Se) on operant behavior and its persistence were examined in aged animals. Female rats were exposed, in utero, to 0, 0.5, or 5 ppm mercury as MeHg via drinking water, approximating mercury exposures of 0, 40, and 400 microg/kg/day. They also received both pre- and chronic post-natal exposure to a diet that was marginal (0.06 ppm) or rich

Suppression of schedule-induced drinking and food-reinforced bar pressing by tail-pinch is not reversed by naloxone.

In Experiment 1 eleven food-deprived rats were tested in a schedule-induced drinking paradigm under both tail-pinch and non-tail-pinch conditions. Tail-pinch produced a strong suppression of schedule-induced drinking, licking, licks per milliliter, bar presses, and number of reinforcers received during 30-min test sessions. Experiment 2 showed that the narcotic antagonist naloxone (2 and 4 mg/kg) did not reverse the tail-pinch suppression of schedule-induced drinking and food-reinforced bar pressing. Experiment 3 demonstrated that the suppression of schedule-induced drinking by tail-pinch coul

Acute nicotine administration reduces the efficacy of punishment in curbing remifentanil consumption in a seeking-taking chain schedule of reinforcement.

RATIONALE: Nicotine dependence is highly comorbid with opioid use disorders (OUDs). The use of nicotine-containing products increases the propensity to misuse prescription opioids and addressing both nicotine and opioid use simultaneously is more efficacious for treatment of OUDs than treating opioid use alone. OBJECTIVES: Given this extreme comorbidity, further elucidation of the effects of nicotine as a factor in promoting vulnerability to development of OUDs is needed. Here, we sought to further explore the effects of nicotine administration on operant self-administration of remifentanil (

Acute but not long-lasting antidepressant-like effect of psilocybin in differential reinforcement of low-rate 72 schedule in rats.

BACKGROUND: In clinical studies, psychedelics including psilocybin and D-lysergic acid diethylamide (LSD) demonstrate rapid and persistent antidepressant effects. Since the effective treatment with psychedelics is usually provided with psychotherapy, it is debatable whether their prolonged efficacy can be observed in infrahuman species. Preclinical reports on psychedelics' effects most often address their acute actions, and different tests and models provide inconsistent results. The goal of this study was to examine whether the treatment with psilocybin and/or LSD would demonstrate immediate

Effects of morphine, naloxone, d,l-cyclazocine, and d-amphetamine on behaviour controlled by a schedule of interresponse time reinforcement.

The separate effects of graded doses of morphine, naloxone, d,l-cyclazocine, and d-amphetamine on responding maintained by a differential reinforcement of low rate schedule of food presentation were examined in rats. Morphine did not alter response rates at doses of 1--5.6 mg/kg; at 10 mg/kg a 57% decrease in responding was observed and behaviour was even more severely depressed by 30 mg of morphine per kilogram. Naloxone did not affect responding at doses ranging from 0.1 to 10 mg/kg. d,l-Cyclazocine at doses of 3 and 5.6 mg/kg induced substantial increases in responding not observed when the

Further evaluations of functional communication training and chained schedules of reinforcement to treat multiple functions of challenging behavior.

We evaluated functional communication training (FCT) combined with a chained schedule of reinforcement procedure for the treatment of challenging behavior exhibited by two individuals diagnosed with Asperger syndrome and autism. Following functional analyses that suggested that challenging behavior served multiple functions for both participants, we implemented FCT in which mands for a discriminative stimulus (S(D); wristband) were reinforced with access to the S(D) and all three functional reinforcers. Next, we modified the procedure by incorporating delays to increase ease of implementation

Optimizing fractionation schedules for de-escalation radiotherapy in head and neck cancers using deep reinforcement learning.

PURPOSE: Patients with locally-advanced head and neck squamous cell carcinomas (HNSCCs), particularly those related to human papillomavirus (HPV), often achieve good locoregional control (LRC), yet they suffer significant toxicities from standard chemoradiotherapy. This study aims to optimize the daily dose fractionation based on individual responses to radiotherapy (RT), minimizing toxicity while maintaining a low risk of LRC failure. METHOD: A virtual environment was developed to simulate tumor dynamics under RT for optimizing dose schedules. Patients predicted to maintain LRC were selected

Choice between food and cocaine reinforcers under fixed and variable schedules in female and male rhesus monkeys.

Illicit drugs like cocaine may be uncertain in terms of the time and effort required to obtain them. Behavior maintained by variable schedules resembles excessive drug-taking compared with fixed schedules. However, no prior research has examined fixed versus variable schedules in drug versus nondrug choice. The present study evaluated cocaine versus food choice under fixed- (FR) and variable-ratio (VR) schedules. The simpler food versus food and cocaine versus cocaine arrangements also were included. Adult female (n = 6) and male (n = 7) rhesus monkeys chose between cocaine (0.01-0.18 mg/kg/in

Functional communication training and chained schedules of reinforcement to treat challenging behavior maintained by terminations of activity interruptions.

In this article, the authors evaluated functional communication training (FCT) and a chained schedule of reinforcement for the treatment of challenging behavior exhibited by two individuals diagnosed with Asperger syndrome and autism, respectively. Following a functional analysis with undifferentiated results, the authors demonstrated that challenging behavior was occasioned by interruptions of ongoing activities and maintained by terminations of interruptions. Next, they demonstrated the effectiveness of a treatment consisting of FCT with a chained schedule of reinforcement. Last, they modifi

MDPV "high-responder" rats also self-administer more oxycodone than their "low-responder" counterparts under a fixed ratio schedule of reinforcement.

RATIONALE: Oxycodone is one of the most commonly prescribed and most frequently abused opioid analgesics, yet little is known regarding individual vulnerabilities to oxycodone abuse. The synthetic cathinone 3,4-methylenedioxypyrovalerone (MDPV) has been shown to produce a "high-responder" phenotype characterized by increased drug intake and responding during periods of signaled drug unavailability (e.g., during post-infusion timeouts) in ~ 40% of male Sprague-Dawley rats. This phenotype also transfers to other psychostimulants (e.g., cocaine and methamphetamine), but it is unknown whether this

Effect of 6-hydroxydopamine lesions of the medial prefrontal cortex on intravenous cocaine self-administration under a progressive ratio schedule of reinforcement.

Rats were trained to self-administer intravenous cocaine under a progressive ratio (PR) schedule of reinforcement. Under this schedule, an increasing number of lever responses had to be made to obtain each subsequent reinforcement (1.5 mg/kg per injection). Once stable responding was achieved with this schedule, bilateral 6-hydroxydopamine (6-OHDA) or vehicle-only injections were delivered into the medial prefrontal cortex (mPFC). Following recovery from surgery, the animals were given access to cocaine under the PR schedule. The effect of the lesion on self-administration behaviour was examin

Choice between food and cocaine or fentanyl reinforcers under fixed and variable schedules in female and male rhesus monkeys.

RATIONALE: Illicit drugs may be unpredictable in terms of the time and effort required to obtain them, and this can be modeled with variable- (VR) vs. fixed-ratio (FR) schedules. In a recent experiment (Zamarripa et al. 2023), the potency of cocaine to maintain choice was greatest under a VR (compared with a FR) when food was available under a FR schedule. OBJECTIVES: The goal of the current study was to extend prior choice results with VR vs. FR schedules to a more efficient procedure with cocaine or fentanyl vs. food. Furthermore, the FR schedule of food delivery was manipulated to determin

Acquisition of cocaine reinforcement using fixed-ratio and concurrent choice schedules in socially housed female and male monkeys.

RATIONALE: Previous studies in socially housed monkeys examining acquisition of cocaine self-administration under fixed-ratio (FR) schedules of reinforcement found that subordinate males and dominant females were more vulnerable than their counterparts. OBJECTIVES: The present studies extended these findings in two ways: (1) to replicate the earlier study, in which female monkeys were studied after a relatively short period of social housing (~ 3 months) using cocaine-naïve female monkeys (n = 9; 4 dominant and 5 subordinate) living in well-established social groups (~ 18 months); and (2) in

A comparison of the effects of psychotomimetics and anxiolytics on punished and unpunished responding maintained by fixed interval schedules of food reinforcement in the rat.

Characterization of anxiolytic drugs often employs conflict paradigms in which the drug effects on punished and unpunished responding can be compared. In this study, a fixed interval schedule generating a range of baseline response rates allowed comparison of the effects of anxiolytic drugs with those of psychotomimetic drugs on equivalent and differing rates of punished and unpunished responding. The first response made by the rat after a 40-s fixed interval elapsed resulted in food pellet delivery. In punished intervals, signalled by the illumination of stimulus lamps above each lever, a 0.6

The interaction of d-amphetamine and naloxone differs for rats trained on separate fixed-interval or fixed-ratio schedules of reinforcement.

The effects of d-amphetamine and naloxone were investigated using two groups of rats trained on either an FR30 or F12 schedule of reinforcement. Amphetamine (0.1-1.0 mg/kg), and naloxone (1.0 and 10 mg/kg) administered separately reduced responding on the FR procedure in a dose-dependent manner. The combined administration of naloxone with amphetamine had an additive suppressive effect on responding. The same doses of amphetamine and naloxone, when given separately, did not significantly depress responding in the FI procedures. However, naloxone/amphetamine combinations produced a marked inhib
