Resistant Starch-Encapsulated Probiotics Attenuate Colorectal Cancer Cachexia and 5-Fluorouracil-Induced Microbial Dysbiosis.

5-Fluorouracil (5-FU) is commonly used as the primary chemotherapy for colorectal cancer (CRC). However, it can lead to unwanted chemoresistance. Resistant starch (RS), which functions similarly to fermentable dietary fiber, has the potential to reduce the risk of CRC. The effects of RS on improving CRC-associated cachectic symptoms and 5-FU chemotherapy-induced microbial dysbiosis remain unknown. Female BALB/cByJNarl mice were randomly divided into four groups: one tumor group (with CT26 colonic carcinoma but no treatment) and three CT26 colonic carcinoma-bearing groups that were administered

Low-Cytotoxicity, Broad-Spectrum Corn Starch-Based Antibacterial Particles that Inhibit Multidrug-Resistant Bacteria.

Antimicrobial resistance is a serious problem in biomedical applications that seriously increases the risk of medical failure. Therefore, developing highly efficient antibacterial agents that inhibit the growth of multidrug-resistant bacteria is a long-standing research goal. In this report, a low-cytotoxicity and highly efficient alternative to antibiotics was designed and prepared using edible corn starch as the scaffold and 2-hydroxypropyl-trimethylammonium chloride chitosan (HTCC) as the antimicrobial agent. The HTCC/starch particles were found to have a positively charged surface over a w

The effect of high-amylose resistant starch on the glycogen structure of diabetic mice.

Glycogen is a complex branched glucose polymer found in many tissues and acts as a blood-glucose buffer. In the liver, smaller β glycogen particles can bind into larger composite α particles. In mouse models of diabetes, these liver glycogen particles are molecularly fragile, breaking up into smaller particles in the presence of solvents such as dimethyl sulfoxide (DMSO). If this occurs in vivo, such a rapid enzymatic degradation of these smaller particles into glucose could exacerbate the poor blood-glucose control that is characteristic of the disease. High-amylose resistant starch (RS) can

Resistant starch alleviates intestinal fibrosis involving an acetate-mediated HDAC2-H3K27ac axis in fibroblasts.

Dietary fibre-based interventions are of growing interest for the prevention and treatment of digestive diseases. In this study, we investigated the effect of resistant starch (RS) on intestinal fibrosis, a stricturing condition driven by excessive extracellular matrix (ECM) accumulation. RS was found to alleviate intestinal fibrosis in a dextran sulfate sodium (DSS)-induced chronic colitis mouse model, as evidenced by restored colon length, reduced ECM deposition (fibronectin and collagen I), and decreased levels of α-smooth muscle actin. Given that RS is fermented by the gut microbiota in th

Microbiome-metabolomics analysis insight into the effects of dietary resistant starch on intestinal integrity.

Resistant starch (RS) has caught much attention for its potential to exert a beneficial impact on intestine and certain members of its resident microbiota. In this study, we examined how dietary RS promotes intestinal barrier in meat ducks by microbiome-metabolomics analysis. Ducklings were fed corn-soybean basal diet or RS diet. Dietary RS improved intestinal morphology and enhanced barrier function in ileum, evidenced by lower permeability and upregulated tight junction proteins and Mucin-2 gene expression. Microbiome analysis showed that RS administration elevated the proportion of Firmicut

Effects of resistant starch on behaviour, satiety-related hormones and metabolites in growing pigs.

Resistant starch (RS) has been suggested to prolong satiety in adult pigs. The present study investigated RS-induced changes in behaviour, satiety-related hormones and metabolites in catheterized growing pigs to explore possible underlying mechanisms for RS-induced satiety. In a cross-over design with two 14-day periods, 10 pigs (initial BW: 58 kg) were assigned to two treatments comprising diets containing either 35% pregelatinized starch (PS) or 34% retrograded starch (RS). Diets were isoenergetic on gross energy. Pigs were fed at 2.8× maintenance. Postprandial plasma response of satiety-rel

Strategy for Fabricating Starch-Triacylglycerol Complexes with Resistance to Amylolysis and Lipid Oxidation.

Previous studies have shown that starch interacts with fatty acids, monoglycerides, or diglycerides to form starch-lipid complexes. Triacylglycerols (TAG) have been considered incapable of forming complexes with starch. Here, we describe a novel complexation strategy using octenyl succinic anhydride-modified maize starch (OSAMS) and glyceryl tridodecanoate (TLG) to form OSAMS-TAG complexes for the first time. MD simulations showed that the three hydrocarbon tails of the TLG molecule were encapsulated together by amylose helices, mainly through van der Waals forces. Structural analyses showed t

Extrusion and chlorogenic acid treatment increase the ordered structure and resistant starch levels in rice starch with amelioration of gut lipid metabolism in obese rats.

Dietary interventions are receiving increasing attention for maintaining host health and diminishing disease risk. This study endeavored to elucidate the intervention effect of chlorogenic acid coupled with extruded rice starch (CGA-ES) in mitigating lipid metabolism disorders induced by a high-fat diet (HFD) in rats. First, a significant increase in resistant starch (RS) and a decrease in the predicted glycemic index (pGI) were observed in CGA-ES owing to the formation of an ordered structure (Dm, single helix, and V-type crystalline structure) and partly released CGA. Compared to a physical

Spatial constraints drive amylosome-mediated resistant starch degradation by Ruminococcus bromii in the human colon.

Degradation of complex dietary fiber by gut microbes is essential for colonic fermentation, short-chain fatty acid production, and microbiome function. Ruminococcus bromii is the primary resistant starch (RS) degrader in humans, which relies on the amylosome, a specialized cell-bound enzymatic complex. To unravel its architecture, function, and the interplay among its components, we applied a holistic multilayered approach: Cryo-electron tomography reveals that the amylosome comprises a constitutive extracellular layer extending toward the RS substrate. Proteomics demonstrates remodeling of it

Resistant starch improves Parkinson's disease symptoms through restructuring of the gut microbiome and modulating inflammation.

Alterations in the gut microbiome and a "leaky" gut are associated with Parkinson's disease (PD), which implies the prospect of rebalancing via dietary intervention. Here, we investigate the impact of a diet rich in resistant starch on the gut microbiome through a multi-omics approach. We conducted a randomized, controlled trial with short-term and long-term phases involving 74 PD patients of three groups: conventional diet, supplementation with resistant starch, and high-fibre diet. Our findings reveal associations between dietary patterns and changes in the gut microbiome's taxonomic composi

Preparation and characterization on the eco-friendly corn starch based adhesive of with salient water resistance, mildew resistance.

Starch adhesive is a commonly used bonding glue that is sustainable, formaldehyde-free and biodegradable. However, there are obviously some problems related to its high viscosity, poor water and mildew resistance. Hence, exploring a starch-based adhesive with good properties that satisfies the requirements of wood processing presents the context of the current research. Thus, corn starch was used as raw material to form oxidized starch (OCS) via oxidation using sodium periodate, it was reacted with a synthesis polyurea compound that prepared from hexanediamine-urea (HU) obtained by deamination

Individual variability shapes ex vivo responses to resistant starch in inflammatory bowel disease derived microbiomes.

Fiber-based therapies focus on butyrate production, a process often dysregulated in inflammatory bowel disease (IBD), but seldomly examine other metabolites or functional pathways. Here, we systematically profiled ex vivo responses of 66 pediatric IBD microbiomes to nine resistant starches (RS), with extensive multi-omic characterization in a subset. Our study demonstrates that inter-individual variability dominates over RS-specific effects, yielding consistent yet highly personalized fermentation phenotypes, microbial compositional shifts, and metabolite outputs. Beyond butyrate, we identify

Reviewing nano-reduction as a tool to tailor functional and nutraceutical properties of resistant starch.

One of the important sources of dietary carbohydrates present in human diet is starch. However due to its ability to cause an increase in blood sugar levels, the consumer acceptance towards starchy foods has seen a rapid decline. The consumers are mostly driven towards starchy foods with low glycemic index that has paved a way to explore new sources that could replace starch. In this context, resistant starch (RS) is a type of starch that resists digestion in the small intestine and reaches the large intestine intact. It behaves as dietary fibre in our body and is associated with various healt

Characterization of Water-Resistant Adhesive Prepared by Cross-Linking Reaction of Oxidized Starch with Lignin.

Wood adhesives play a critical role in the wood processing industry; however, traditional formaldehyde-based adhesives pose health risks and are reliant on non-renewable resources. This study aims to develop a bio-based wood adhesive with excellent water resistance, focusing on environmentally friendly solutions. The synthesis of an oxidized starch-lignin (OSTL) composite adhesive was accomplished by modifying starch via oxidation and subsequent cross-linking with lignin. Ammonium persulfate (APS) was employed for oxidation of starch, introducing aldehyde groups that upgrade its reactivity wit

Loss-of-function of SSIIa and SSIIIa confers high resistant starch content in rice endosperm.

Rice (Oryza sativa L.) endosperm accumulates huge amounts of starch. Rice starch is highly digestible, potentially enhancing the occurrence of blood sugar- and intestine-related diseases such as type 2 diabetes. Resistant starch (RS) is hardly digestible in small intestine but can be converted into beneficial short-chain fatty acids in large intestine, potentially reducing the incidence of these diseases. However, it is still difficult to produce a high RS rice variety. Here, we report that simultaneous deficiency of soluble starch synthases IIa and IIIa confers high RS content in rice endospe

Resistant starch: Implications of dietary inclusion on gut health and growth in pigs: a review.

Starch from cereal grains, pulse grains, and tubers is a major energy substrate in swine rations constituting up to 55% of the diet. In pigs, starch digestion is initiated by salivary and then pancreatic α-amylase, and has as final step the digestion of disaccharides by the brush-border enzymes in the small intestine that produce monosaccharides (glucose) for absorption. Resistant starch (RS) is the proportion of starch that escapes the enzymatic digestion and absorption in the small intestine. The undigested starch reaches the distal small intestine and hindgut for microbial fermentation, whi

High amylose resistant starch diet ameliorates oxidative stress, inflammation, and progression of chronic kidney disease.

Inflammation is a major mediator of CKD progression and is partly driven by altered gut microbiome and intestinal barrier disruption, events which are caused by: urea influx in the intestine resulting in dominance of urease-possessing bacteria; disruption of epithelial barrier by urea-derived ammonia leading to endotoxemia and bacterial translocation; and restriction of potassium-rich fruits and vegetables which are common sources of fermentable fiber. Restriction of these foods leads to depletion of bacteria that convert indigestible carbohydrates to short chain fatty acids which are importan

Effect of dietary resistant starch and protein on colonic fermentation and intestinal tumourigenesis in rats.

Protein as well as starch is fermented in the colon, but the interaction between protein and starch fermentation and the impact on colonic oncogenesis is unknown. High-protein diets increase delivery of protein to the colon and might promote oncogenesis through generation of toxic products. We investigated the interaction of resistant starch (RS) with digestion-resistant potato protein (PP) on colonic fermentation events and their relationship to intestinal tumourigenesis. Male Sprague-Dawley rats were fed an AIN-76A-based diet for 4 weeks and intestinal neoplasms were induced by azoxymethane.

Cassava-resistant starch type 3 modulates high-fat diet-induced adiposity and transcriptional oxidative stress imbalance.

High-fat diets (HFDs) are associated with excessive weight gain and lipid accumulation, contributing to metabolic syndrome. Resistant starch (RS), an indigestible dietary fiber that undergoes colonic fermentation, is proposed to improve metabolic outcomes. This study investigated the effects of cassava-derived resistant starch type 3 (RS3), incorporated into an isocaloric HFD, on body weight, glucose metabolism, lipid deposition, and hepatic redox-related gene expression in healthy rats. Male Wistar rats were fed for 28 days with an isocaloric high-fat diet containing either native cassava sta

Functional Rice Enriched With Resistant Starch Ameliorates Metabolic Dysregulation in High-Fat Diet-Induced Obese Mice.

Resistant starch (RS) is a non-digestible fraction of dietary starch that modulates glucose and lipid metabolism. In this study, a RS-enriched high-amylose rice (R-HAR) was developed using a mild citric acid-assisted thermal process to enhance RS content and evaluated for its potential to alleviate high-fat diet (HFD)-induced metabolic disorders in mice. The RS content of the diet increased proportionally with the inclusion of R-HAR. Long-term R-HAR supplementation significantly reduced body weight gain and adiposity compared with the HFD control. Serum biochemical analyses revealed that R-HAR
