[Progress in studies of effects on brain-derived neurotrophic factor in degenerative fundus disease].

Degenerative fundus diseases are a group of outer retinal disease caused by dysfunction of photoreceptors, some of which have the inner layer defection. Brain-derived neurotrophic factor (BDNF) has been shown to play a key role in the neuronal survival, which accomplishes its protection via receptors, including a low affinity p75 and a high affinity tyrosine kinase receptor TrkB, retinal growth cone filopodia and cofactor of Zinc. It is proved that BDNF can induce differentiation of stem cells and retinal progenitor, in case for the effective retinal transplantation. The protection by BDNF has

The classical receptive field surround of primate parasol ganglion cells is mediated primarily by a non-GABAergic pathway.

Although the center-surround receptive field is a fundamental property of retinal ganglion cells, the circuitry that mediates surround inhibition remains controversial. We examined the contribution of horizontal cells and amacrine cells to the surround of parasol ganglion cells of macaque and baboon retina by measuring receptive field structure before and during the application of drugs that have been shown previously to affect surrounds in a range of mammalian and nonmammalian species. Carbenoxolone and cobalt, thought to attenuate feedback from horizontal cells to cones, severely reduced the

Effects of mercury intoxication on the response of horizontal cells of the retina of thraira fish (Hoplias malabaricus).

Methyl mercury (MeHg) is highly neurotoxic, affecting visual function in addition to other central nervous system functions. The effect of mercury intoxication on the amplitude of horizontal cell responses to light was studied in the retina of the fish Hoplias malabaricus. Intracellular responses were recorded from horizontal cells of fish previously intoxicated with MeHg by intraperitoneal injection (IP group) or by trophic exposure (T group). Only one retina per fish was used. The doses of MeHg chloride administered to the IP group were 0.01, 0.05, 0.1, 1.0, 2.0, and 6.0 mg/kg. The amplitude

The Retina of Asian and African Elephants: Comparison of Newborn and Adult.

Elephants are precocial mammals that are relatively mature as newborns and mobile shortly after birth. To determine whether the retina of newborn elephants is capable of supporting the mobility of elephant calves, we compared the retinal structures of 2 newborn elephants (1 African and 1 Asian) and 2 adult animals of both species by immunohistochemical and morphometric methods. For the first time, we present here a comprehensive qualitative and quantitative characterization of the cellular composition of the newborn and the adult retinas of 2 elephant species. We found that the retina of eleph

Specific ablation of Hippo signalling component Yap1 in retinal progenitors and Müller cells results in late onset retinal degeneration.

Yes-associated protein (YAP) is a major component of the Hippo pathway involved in development, growth, repair and homeostasis. Nonsense YAP1 mutations in humans result in autosomal dominant coloboma. Here, we generated a conditional knockout mouse model in which Yap1 was specifically deleted in embryonic retinal progenitor cells (RPCs) and in mature Müller cells using a Chx10-Cre driver. Our data demonstrated that the conditional ablation of Yap1 in embryonic RPCs does not prevent normal retinal development and caused no gross changes in retinal structure during embryonic and early postnatal

Early alteration of retinal neurons in Aipl1-/- animals.

PURPOSE: Mutations in the photoreceptor cell-specific gene encoding aryl hydrocarbon receptor-interacting protein-like 1 (AIPL1) lead to Leber congenital amaurosis (LCA4), retinitis pigmentosa, and cone-rod dystrophy. Gene therapy appears to be promising in the treatment for AIPL1-mediated vision loss in humans. Prior to initiating these treatments, however, it is crucial to understand how the retinal neurons remodel themselves in response to photoreceptor cell degeneration. In this study, using an animal model for AIPL1-LCA, Aipl1(-/-) mice, we investigate the changes in postreceptoral retina

The Variety of Mechanosensitive Ion Channels in Retinal Neurons.

Alterations in intraocular and external pressure critically involve the pathogenesis of glaucoma, traumatic retinal injury (TRI), and other retinal disorders, and retinal neurons have been reported to express multiple mechanical-sensitive channels (MSCs) in recent decades. However, the role of MSCs in visual functions and pressure-related retinal conditions has been unclear. This review will focus on the variety and functional significance of the MSCs permeable to K+, Na+, and Ca2+, primarily including the big potassium channel (BK); the two-pore domain potassium channels TRAAK and TREK; Piezo

Retinal ganglion cell topography in the masked greenling Hexagrammos octogrammus (Pallas, 1814) (Pisces, Scorpaeniformes, Hexagrammidae).

The spatial arrangement of retinal neurons determines to a certain extent visual acuity and sensitivity. Despite a large number of studies dealing with retinal topography, there are gaps in our knowledge on the issue. We addressed the spatial arrangement of retinal ganglion cells in the masked greenling, a marine demersal fish. The combined proportion of displaced amacrine cells and glia was 17.63%. The spatial density of ganglion cells was lowest (1900-2500 cells/mm2) in the ventronasal periphery. It increased centripetally to form a horizontal visual streak situated dorsal to the optic disc.

Human neural progenitor cells decrease photoreceptor degeneration, normalize opsin distribution and support synapse structure in cultured porcine retina.

Retinal neurodegenerative disorders like retinitis pigmentosa, age-related macular degeneration, diabetic retinopathy and retinal detachment decrease retinal functionality leading to visual impairment. The pathological events are characterized by photoreceptor degeneration, synaptic disassembly, remodeling of postsynaptic neurons and activation of glial cells. Despite intense research, no effective treatment has been found for these disorders. The current study explores the potential of human neural progenitor cell (hNPC) derived factors to slow the degenerative processes in adult porcine reti

Neurotensin actions in the retina: mechanisms and variability.

The effects of neurotensin on mudpuppy retinal cells were studied using extracellular and intracellular electrophysiological recording techniques and bath application of the peptide. Ganglion and amacrine cells (but not bipolar or horizontal cells) were reversibly depolarized by low micromolar concentrations of neurotensin. Depolarizations also occurred with neurotensin application during cobalt block of synaptic transmission and were accompanied by decreased input resistances. This suggests neurotensin may act directly on amacrine and ganglion cells as a conventional excitatory transmitter. H

Advances in Ophthalmic Epigenetics and Implications for Epigenetic Therapies: A Review.

The epigenome represents a vast molecular apparatus that writes, reads, and erases chemical modifications to the DNA and histone code without changing the DNA base-pair sequence itself. Recent advances in molecular sequencing technology have revealed that epigenetic chromatin marks directly mediate critical events in retinal development, aging, and degeneration. Epigenetic signaling regulates retinal progenitor (RPC) cell cycle exit during retinal laminar development, giving rise to retinal ganglion cells (RGCs), amacrine cells, horizontal cells, bipolar cells, photoreceptors, and Müller glia.

Restoration of vision after transplantation of photoreceptors.

Cell transplantation is a potential strategy for treating blindness caused by the loss of photoreceptors. Although transplanted rod-precursor cells are able to migrate into the adult retina and differentiate to acquire the specialized morphological features of mature photoreceptor cells, the fundamental question remains whether transplantation of photoreceptor cells can actually improve vision. Here we provide evidence of functional rod-mediated vision after photoreceptor transplantation in adult Gnat1−/− mice, which lack rod function and are a model of congenital stationary night blindness. The mechanotransduction channel Piezo2 refines axonal projections to the accessory optic system and regulates the optokinetic reflex.

The optokinetic reflex (OKR) is an evolutionarily conserved reflexive behavior that ensures image stabilization on the retina during global motion. It consists of smooth eye tracking movements in the direction of the moving stimulus interspersed with rapid resetting saccades. This reflex is driven by retinal ON direction-selective ganglion cells (oDSGCs), which comprise distinct subtypes tuned either to vertical or horizontal motion. oDSGCs convey directional signals to the brain via precise axonal projections to specific accessory optic system (AOS) nuclei. However, the mechanisms that establ

Effects of nitric oxide on horizontal cells in the rabbit retina.

Retinal horizontal cells display large receptive fields as a result of extensive electrical coupling via gap junctions. There is abundant evidence that these gap junctions are dynamically regulated by changes in the adaptational state of the retina. The neuromodulator dopamine appears to play a major role in regulating gap junctional conductances of horizontal cells. Emerging evidence indicates that nitric oxide (NO) also acts as a neuromodulator in the retina and, more specifically, regulates the coupling between horizontal cells. In the present study, we examined the effects of a nitric oxid

[Anatomy of the retina].

The neuroretina is a functional unit of the central nervous system that converts a light signal into a nerve impulse. Of neuroectodermal origin, derived from the diencephalon, the neuroretina is a layered tissue composed of six types of neuronal cells (two types of photoreceptors: cones and rods, horizontal, bipolar, amacrine and ganglion cells) and three types of glial cells (Müller glial cells, astrocytes and microglial cells). The neuroretina lays on the retinal pigmentary epithelium, that together form the retina. The existence of the internal and external blood-retinal barriers and intra-

Sip1 regulates the generation of the inner nuclear layer retinal cell lineages in mammals.

The transcription factor Sip1 (Zeb2) plays multiple roles during CNS development from early acquisition of neural fate to cortical neurogenesis and gliogenesis. In humans, SIP1 (ZEB2) haploinsufficiency leads to Mowat-Wilson syndrome, a complex congenital anomaly including intellectual disability, epilepsy and Hirschsprung disease. Here we uncover the role of Sip1 in retinogenesis. Somatic deletion of Sip1 from mouse retinal progenitors primarily affects the generation of inner nuclear layer cell types, resulting in complete loss of horizontal cells and reduced numbers of amacrine and bipolar

Light stress affects cones and horizontal cells via rhodopsin-mediated mechanisms.

Retinal degenerations are a major cause of blindness in human patients. The identification of endogenous mechanisms involved in neurodegeneration or neuroprotection helps to understand the response of the retina to stress and provides essential information not only for basic retinal physiology but also for defining molecular targets for neuroprotective strategies. Here we used excessive light exposure as a model system to study mechanisms of photoreceptor degeneration in mice. Using one wild type and four genetically modified mouse strains, we demonstrate that light exposure resulted not only

Distinct cellular localization of α- synuclein and β-synuclein in the chick retina.

α-Synuclein and β-synuclein are critical regulators of synaptic function and play significant roles in physiology and disease. While their retinal expression patterns are well-characterized in mammals, limited studies exist for the chicken retina, a model valuable for studying human retinal degenerative diseases due to its cone-dominant photoreceptor structure. Notably, research on β-synuclein in chickens remains scarce, leaving synucleinopathy models underdeveloped. Using immunofluorescence, we analyzed α- and β-synuclein expression in the chicken retina. Our results show that β-synuclein has

Activity-dependent development of synaptic circuits mediates direction selectivity in an axis-specific manner.

The optokinetic reflex (OKR), which stabilizes images on the retina as a mouse navigates its environment, originates in direction-selective ganglion cells (DSGCs). A mouse model that lacks cholinergic retinal waves, the β2-nAChR-KO mouse, does not develop horizontal direction selectivity but preserves vertical direction selectivity. Here, we demonstrate that the absence of horizontal direction selectivity in β2-nAChR-KO mice results in a loss of the OKR along the horizontal axis, consistent with previous findings on optomotor response. In addition, we observe diminished asymmetric inhibition o

Imaging and quantifying ganglion cells and other transparent neurons in the living human retina.

Ganglion cells (GCs) are fundamental to retinal neural circuitry, processing photoreceptor signals for transmission to the brain via their axons. However, much remains unknown about their role in vision and their vulnerability to disease leading to blindness. A major bottleneck has been our inability to observe GCs and their degeneration in the living human eye. Despite two decades of development of optical technologies to image cells in the living human retina, GCs remain elusive due to their high optical translucency. Failure of conventional imaging-using predominately singly scattered light
