Sex chromosome complement affects multiple aspects of reversal-learning task performance in mice.

Determining the mechanisms by which the sex-chromosome complement (SCC) affects learning, attention, and impulsivity has implications for observed sex differences in prevalence, severity, and prognosis of psychiatric/neurodevelopmental disorders and syndromes associated with sex-chromosome aneuploidy. Here, Four Core Genotypes (FCG) mice were evaluated in order to assess the separable and/or interacting effects of gonads (testes vs. ovaries) and their secretions and/or SCC (XX vs. XY) acting via non-gonadal mechanisms on behavior. We tested FCG mice on a reversal-learning task that enables the

Reversal Learning Performance in the XY∗ Mouse Model of Klinefelter and Turner Syndromes.

Klinefelter syndrome (KS; 47, XXY) and Turner syndrome (TS; 45, XO) are caused by two relatively common sex chromosome aneuploidies. These conditions are associated with an increased odds of neuropsychiatric disorders, including attention deficit/hyperactivity disorder (ADHD), as well as impairments in cognition that include learning delays, attentional dysfunction and impulsivity. We studied cognitive functions in the XY∗ mouse model, which allows comparison of XXY to XY males (KS model), and XO to XX females (TS model). We evaluated adult mice with and without gonads, using a version of an o

Neural correlates of reversal learning in severe mood dysregulation and pediatric bipolar disorder.

OBJECTIVE: Outcome and family history data differentiate children with severe mood dysregulation (SMD), a syndrome characterized by chronic irritability, from children with "classic" episodic bipolar disorder (BD). Nevertheless, the presence of cognitive inflexibility in SMD and BD highlights the need to delineate neurophysiologic similarities and differences between the two patient groups. Functional magnetic resonance imaging was used to examine neural correlates of cognitive flexibility deficits in patients with SMD and BD versus healthy volunteers (HV). METHOD: During functional magnetic

Selective lesions of the dorsomedial striatum impair serial spatial reversal learning in rats.

Impairments in reversal learning have been attributed to orbitofrontal cortex (OFC) dysfunction in many species. However, the role of subcortical areas interconnected with the OFC such as the striatum remains poorly understood. This study directly evaluated the contribution of core and shell sub-regions of the nucleus accumbens (NAc), dorsomedial (DMS) and dorsolateral (DLS) striatum to reversal learning of an instrumental two-lever spatial discrimination task in rats. Selective NAc core, DMS and DLS lesions were achieved with microinjections of quinolinic acid and NAc shell lesions with ibote

Pharmacological or genetic inactivation of the serotonin transporter improves reversal learning in mice.

Growing evidence supports a major contribution of cortical serotonin (5-hydroxytryptamine, 5-HT) to the modulation of cognitive flexibility and the cognitive inflexibility evident in neuropsychiatric disorders. The precise role of 5-HT and the influence of 5-HT gene variation in mediating this process is not fully understood. Using a touch screen-based operant system, we assessed reversal of a pairwise visual discrimination as an assay for cognitive flexibility. Effects of constitutive genetic or pharmacological inactivation of the 5-HT transporter (5-HTT) on reversal were examined by testing

[Discrimination reversal learning in rats under the treatment of chlordiazepoxide: effect of overtraining].

A total of 48 rats were run in a black-white discrimination (original) learning task to the learning criterion of 18/20 correct responses. Correct responses were rewarded by food and a non-correction method was used. One group was intraperitoneally injected with 15 mg/kg of chlordiazepoxide (CDP) and the other with physiological saline (SAL), 30 min prior to the beginning of the daily trials. For half of the rats (NOT) of each group, reversal learning began on the next day after reaching the criterion, and for the other half (OT), 100 additional trials(overtraining) were given before the rever

Neural alterations in Pavlovian reversal learning link childhood trauma and transdiagnostic anxiety in youth.

BACKGROUND: Pavlovian reversal learning (RL) is a potential pathway linking childhood trauma (CT) with psychopathology. RL describes the ability to flexibly update associations of stimuli with threat and safety based on changing contextual significance. This study aims to characterize how CT may alter the neural underpinnings of RL in youth in ways relevant to mental health. METHODS: One hundred participants aged 9-19, half with CT (i.e., interpersonal violence) exposure, completed a RL task during an fMRI scan. One previously-neutral stimulus was paired with an aversive stimulus and predicte

Overexpression of miR-99a in hippocampus leads to impairment of reversal learning in mice.

As one of the most common human genetic disorders, Down syndrome (DS) is characterized by a mild-to-moderate cognitive disability, which mainly results from genes overexpression on chromosome 21. The expression of miR-99a, a gene harboring on chromosome 21, is increased by 50 folds in DS brain samples. This study aims to investigate the effect of miR-99a overexpression in the hippocampus on mouse behaviors and explore the underlying mechanisms. Lentivirus vectors were delivered into the hippocampus for focal miR-99a overexpression in mice. Then behaviors were observed by an open field, elevate

Evidence of prefrontal hyperactivation to food-cue reversal learning in adolescents with anorexia nervosa.

Food avoidance in anorexia nervosa (AN) has been hypothesized to occur as a result of deficits in reversal learning and altered neuronal processing of food-cue relationships. Extant findings suggest that those with AN may rely on heightened recruitment of prefrontal regions during initial formation of food-cue learning and difficulty extinguishing these relationships may result from elevated insula activity. We tested this hypothesis by comparing behavioral and neuronal responses to food-cue acquisition and reversal between adolescents with AN and healthy controls. Compared to controls, acquis

Effects of acute restraint stress on set-shifting and reversal learning in male rats.

Exposure to acute stress alters cognition; however, few studies have examined the effects of acute stress on executive functions such as behavioral flexibility. The goal of the present experiments was to determine the effects of acute periods of stress on two distinct forms of behavioral flexibility: set-shifting and reversal learning. Male Sprague-Dawley rats were trained and tested in an operant-chamber-based task. Some of the rats were exposed to acute restraint stress (30 min) immediately before either the set-shifting test day or the reversal learning test day. Acute stress had no effect

Post-learning REM sleep deprivation impairs long-term memory: reversal by acute nicotine treatment.

Rapid eye movement sleep deprivation (REM-SD) is associated with spatial learning and memory impairment. During REM-SD, an increase in nicotine consumption among habitual smokers and initiation of tobacco use by non-smokers have been reported. We have shown recently that nicotine treatment prevented learning and memory impairments associated with REM-SD. We now report the interactive effects of post-learning REM-SD and/or nicotine. The animals were first trained on the radial arm water maze (RAWM) task, then they were REM-sleep deprived using the modified multiple platform paradigm for 24h. Du

Sex, but not juvenile stress, affects reversal learning and DRL performance following cocaine administration.

INTRODUCTION: Early adversity, impulsivity and sex all contribute to the risk of developing substance use disorder. Using rats, we examined how juvenile stress interacts with sex and cocaine to affect performance on a serial reversal task and a differential reinforcement of low rates 10 s (DRL10) task. The expression of dopamine-related proteins in several brain areas was also assessed. METHODS: From postnatal days (PND) 25-29, rats were exposed to a variable stress protocol. In adulthood, rats were trained on the reversal task and the effects of cocaine (0, 10, or 20 mg/kg, IP) on performanc

Reversal learning and associative memory impairments in a BACHD rat model for Huntington disease.

Chorea and psychiatric symptoms are hallmarks of Huntington disease (HD), a neurodegenerative disorder, genetically characterized by the presence of expanded CAG repeats (>35) in the Huntingtin (HTT) gene. HD patients present psychiatric symptoms prior to the onset of motor symptoms and we recently found a similar emergence of non motor and motor deficits in BACHD rats carrying the human full length mutated HTT (97 CAG-CAA repeats). We evaluated cognitive performance in reversal learning and associative memory tests in different age cohorts of BACHD rats. Male wild type (WT) and transgenic (TG

Δ(9)Tetrahydrocannabinol impairs reversal learning but not extra-dimensional shifts in rhesus macaques.

Expansion of medical marijuana use in the US and the recently successful decriminalization of recreational marijuana in two States elevates interest in the specific cognitive effects of Δ(9)tetrahydrocannabinol (Δ(9)THC), the major psychoactive constituent of marijuana. Controlled laboratory studies in nonhuman primates provide mixed evidence for specific effects of Δ(9)THC in learning and memory tasks, with a suggestion that frontal-mediated tasks may be the most sensitive. In this study, adult male rhesus monkeys were trained on tasks which assess reversal learning, extradimensional attentio

Establishing the dopamine dependency of human striatal signals during reward and punishment reversal learning.

Drugs that alter dopamine transmission have opposite effects on reward and punishment learning. These opposite effects have been suggested to depend on dopamine in the striatum. Here, we establish for the first time the neurochemical specificity of such drug effects, during reward and punishment learning in humans, by adopting a coadministration design. Participants (N = 22) were scanned on 4 occasions using functional magnetic resonance imaging, following intake of placebo, bromocriptine (dopamine-receptor agonist), sulpiride (dopamine-receptor antagonist), or a combination of both drugs. Reverse learning and the physiological basis of eye movement desensitization.

Eye movement desensitization is a new and effective procedure for post-traumatic stress disorder that requires explanation. Reverse learning is a model developed in artificial neural networks as a theoretical explanation of rapid-eye-movement sleep. It demonstrates that an overloaded node within a network can be consolidated with a series of non-specific activations. Rapid-eye-movement sleep is suspected to have a memory consolidation function. Ponto-geniculo-occipital spikes, which occur in rapid-eye-movement sleep, are a candidate for such activations in the real brain. In cats, the phasic f

Effects of cerebellar transcranial direct current stimulation on reversal learning performance during threat of shock.

According to the cerebellar lateralization hypothesis of motivational direction, relative left-to-right dominant posterior cerebellar activity is associated with avoidance motivation and anxiety, whereas relative right-to-left dominant posterior cerebellar activity is associated with approach motivation and anger. The present study tested this hypothesis in avoidance-related behavior during rule learning combined with threat of shock. In a randomized double-blind sham-controlled between-subjects design, ninety healthy right-handed adult volunteers received either active (n = 45) or sham (n = 4

Prefrontal and ventral striatal neural correlates of reversal learning in anorexia nervosa and bulimia nervosa.

Difficulty with reversal learning (RL)-appropriately shifting behavior following outcome contingency changes-may represent a shared or distinct mechanism across eating disorder (ED) diagnoses. We tested whether RL-related neural correlates differ among adults without and with EDs (anorexia nervosa, restricting subtype [AN-R], AN binge-eating/purging subtype [AN-BP], and bulimia nervosa [BN]) and whether these correlates correspond to ED-symptom severity and frequency. We hypothesized that individuals with EDs would demonstrate differential neural activation during RL relative to individuals wi

Interactions between estradiol and haloperidol on perseveration and reversal learning in amphetamine-sensitized female rats.

There are sex differences associated with schizophrenia, as women exhibit later onset of the disorder, less severe symptomatology, and better response to antipsychotic medications. Estrogens are thought to play a role in these sex differences; estrogens facilitate the effects of antipsychotic medications to reduce the positive symptoms of schizophrenia, but it remains unclear whether estrogens protect against the cognitive symptoms of this disorder. Amphetamine sensitization is used to model some symptoms of schizophrenia in rats, including cognitive deficits like excessive perseveration and s
